젖소 말초혈액 림프구로부터 소백혈병 바이러스 배양 및 전자현미경적 관찰 (Cultivation and Electron Microscopy of Bovine Leukemia Virus from Peripheral Blood Lymphocytes of Holstein-Friesian Dairy Cattle)

  • 국내 젖소의 54.2%가 BLV에 감염되어 있지만 현재까지 국내에서는 소백혈병 바이러스 (BLV) 입자를 확인한 연구 보고가 없기 때문에 BLV 항체 양성 소의 말초혈액 림프구를 배양, BLV를 발현시켜 전자현미경으로 바이러스 입자를 검출하였고 배양조건에 따른 바이러스 발현율 및 발현 시간을 비교하였다. 검사 결과 전형적인 C-형 바이러스를 확인할 수 있었으며 BLV 단크론 항체를 이용한 면역염색결과 BLV 항원 양성으로 확인되었다. BLV는 대부분 세포 외부에 분포하고 있었으며 세포질 막에서 생성, 발아되어 나오는 것도 관찰되었다. 전체 바이러스의 크기는 $90{\sim}100$ nm였으며 nucleocapsid는 $40{\sim}60$nm였다. 소태아혈청 (FBS)과 T- 및 B-림프구 분열촉진물질(mitogen)을 각각 첨가하여 배양한 결과 두 군 모두에서 BLV 발현이 확인되었다. Lipopolysaccharide 첨가군은 배양 12시간, Conconavalin A 첨가군은 배양 24시간에 각각 림프구의 10%에서 바이러스가 관찰되었다. 또한 FBS만 첨가한 군과 FBS와 mitogen을 모두 첨가하지 않은 군에서도 관찰되었으나 바이러스의 수는 적었다. 본 연구에서 확립된 BLV 배양 기법을 활용하면 BLV에 감염된 소 중 바이러스를 발현하는 소, 즉 전파능이 있는 개체를 찾아내어 우선적으로 도태할 수 있기 때문에 BLV 감염으로 인한 피해를 막는데 효과적으로 이용될 수 있을 것으로 사료된다.

Usefulness of Milk Urea Concentration to Monitor the Herd Reproductive Performances in Crossbred Karan-fries Cows

  • The study was undertaken to investigate the validity of milk urea concentration as an index of the reproductive performances in crossbred Karan-Fries (Holstein Friesian${\times}$Tharparkar) cows under farm condition. Milk urea was analysed in noon milk samples (1200 to 1300 h) to interrelate with the interval from parturition to first service, number of insemination per conception, first service conception rate and service period. Milk progesterone (P4) was analysed in noon milk samples on the day 1, 10, 20 and 30 post insemination to study the effect of milk urea concentration on early embryonic mortality. The interval from parturition to first service was found significantly (p<0.01) higher ($77.2{\pm}5.5$ days) when milk urea concentration was ${\geq}63.4mg/dl$. The average milk urea concentrations (mg/dl) were found $42.1{\pm}2.5$, $47.9{\pm}1.5$ and $50.3{\pm}3.1$, respectively in cows that conceived at $1^{st}$, $2^{nd}$ and $3^{rd}$ insemination. However, the variation was not statistically significant. The first insemination conception rate was found significantly (p<0.05) higher (68.8%) when milk urea level was ${\leq}32.4mg/dl$. The service period was found significantly (p<0.05) higher ($125.4{\pm}8.8$ days) when milk urea concentration was ${\geq}45.1mg/dl$. The milk P4 level indicated that the cows, those were detected as non-pregnant on day 60 post insemination were initially pregnant but the pregnancy was terminated sometime during the day 30 to 60 post insemination. The study indicates that the milk urea values may be used as an index of reproductive performances in dairy herd when individual animals are not being monitored for nutritional status. The altered milk urea values may be utilised by the farmers as ready reference to rectify the protein and energy nutrition in cows to achieve the better reproductive performances in herd.

Effects of Linseed Oil or Whole Linseed Supplementation on Performance and Milk Fatty Acid Composition of Lactating Dairy Cows

  • The objective of this study was to determine the effects of linseed oil or whole linseed supplementation on performance and milk fatty acid composition of lactating dairy cows. Thirty six Holstein Friesian crossbred lactating dairy cows were blocked by milking days first and then stratified random balanced for milk yields and body weight into three groups of 12 cows each. The treatments consisted of basal ration (53:47; forage:concentrate ratio, on a dry matter [DM] basis, respectively) supplemented with 300 g/d of palm oil as a positive control diet (PO), or supplemented with 300 g/d of linseed oil (LSO), or supplemented with 688 g/d of top-dressed whole linseed (WLS). All cows were received ad libitum grass silage and individually fed according to the treatments. The experiment lasted for 10 weeks including the first 2 weeks as the adjustment period, followed by 8 weeks of measurement period. The results showed that LSO and WLS supplementation had no effects on total dry matter intake, milk yield, milk composition, and live weight change; however, the animals fed WLS had higher crude protein (CP) intake than those fed PO and LSO (p<0.05). To compare with the control diet, dairy cow's diets supplemented with LSO and WLS significantly increased milk concentrations of cis-9,trans-11-conjugated linoleic acid (CLA) (p<0.05) and n-3 fatty acids (FA) (p<0.01), particularly, cis-9,12,15-C18:3, C20:5n-3 and C22:6n-3. Supplementing LSO and WLS induced a reduction of medium chain FA, especially, C12:0-C16:0 FA (p<0.05) while increasing the concentration of milk unsaturated fatty acids (UFA) (p<0.05). Milk FA proportions of n-3 FA remarkably increased whereas the ratio of n-6 to n-3 decreased in the cows supplemented with WLS as compared with those fed the control diet and LSO (p<0.01). In conclusion, supplementing dairy cows' diet based on grass silage with WLS had no effect on milk yield and milk composition; however, trans-9-C18:1, cis-9,trans-11-CLA, n-3 FA and UFA were increased while saturated FA were decreased by WLS supplementation. Therefore, it is recommended that the addition 300 g/d of oil from whole linseed should be used to lactating dairy cows' diets.

Effect of Elemental Sulfur Supplementation on Rumen Environment Parameters and Utilization Efficiency of Fresh Cassava Foliage and Cassava Hay in Dairy Cattle

  • Effect of sulfur (S) on utilization efficiency of fresh cassava foliage and cassava hay in dairy cows was evaluated using thirty-two $1^{st}-2^{nd}$ lactation Holstein-Friesian crossbred dairy cows. The experimental treatment was a 2${\times}$2 factorial arrangement in a randomized complete block design (RCBD) using two roughages (rice straw+fresh cassava foliage (FCF) and rice straw+cassava hay (CH)) and two elemental sulfur (S) levels (0.15 and 0.4% S of dry matter (DM)), respectively. Four dietary treatments (FCF+0.15, FCF+0.4, CH+0.15 and CH+0.4) were offered ad libitum in the form of a total mixed ration (TMR) with concentrate to roughage (chopped rice straw+chopped cassava foliage) ratio at 60:40. Fresh cassava foliage or cassava hay resulted in similar dry mater intake, rumen ecology parameters, total tract digestibility, blood chemistry, milk production and composition. However, HCN intake, blood and milk thiocyanate concentration were significantly higher (p<0.01) in cows fed fresh cassava foliage with no sign of potential toxicity. Dry matter intake, body weight changes, molar percentage of propionate in rumen, neutral detergent fiber (NDF) digestibility and nitrogen (N) retention of cows tended to be increased while DM digestibility (65.6, 72.7, 68.6 and 72.1% of total DM intake for the respective treatments), rumen bacteria population (1.4, 1.7, 1.6 and $1.7{\times}10^{11}$ cell/ml for respective treatments), fungal zoospore population (0.4, 0.6, 0.4 and $0.5{\times}10^{6}$ cell/ml for respective treatments), urinary allantoin (25.3, 28.0, 26.3 and 27.6 g/d for respective treatments), microbial N yield (136.0, 154.6, 142.8 and 151.3 g N/d for respective treatments) and milk protein content (3.4, 3.5, 3.2 and 3.5% for respective treatments) were significantly (p<0.05) higher in cows fed on supplemented sulfur at 0.4% of DM in comparison with 0.15% S-supplemented diets. Based on these results, it is concluded that cassava foliage could be used as a portion of roughage for dairy cows and supplementation of S would be nutritionally beneficial.

Effects of Fermented Total Mixed Ration and Cracked Cottonseed on Milk Yield and Milk Composition in Dairy Cows

  • Four lactating Holstein Friesian crossbred cows, with an average initial weight of 450 kg, 48${\pm}$12 days in milk and initial milk yield of 18 kg/h/d, were randomly arranged according to a 2${\times}$2 factorial arrangement in a 4${\times}$4 in Latin square design with 21-d period to investigate the effects of type of total mixed ration (TMR) and type of whole cottonseed (WCS) on intake, digestibility and milk production. The dietary treatments were i) TMR and WCS supplementation at 0.5 kg/h/d, ii) TMR and cracked WCS (cWCS) supplementation at 0.5 kg/h/d, iii) fermented TMR (FTMR) and WCS supplementation at 0.5 kg/h/d, and iv) FTMR and cWCS supplementation at 0.5 kg/h/d. Voluntary feed intake was 15.9, 15.2, 15.4 and 15.6 kg DM/d in dietary treatment 1, 2, 3 and 4, respectively. Digestibility of DM, OM, CP, EE, NDF and ADF were not significantly different among dietary treatments. Ruminal pH, $NH_{3}-N$ and volatile fatty acids in the rumen were also not significantly different among type of TMR or type of WCS. Blood urea-N concentration was not significantly different among dietary treatments. Ruminal bacteria population tended to increase but ruminal protozoa population tended to decrease with supplementation of cWCS, but they were not affected by FTMR. Milk yield and 3.5% FCM were not statistically different among treatments (16.6, 16.2, 17.0, 16.3 kg/d and 18.0, 18.6, 19.9 and 19.0 kg/d, respectively). Milk composition was not significantly different among dietary treatments. However, unsaturated fatty acids in milk fat in cows fed FTMR were lower (p<0.05) than in cows fed TMR. In conclusion, fermentation is a conceivable method to improve the quality of TMR for long-time storage and the cracking method is suitable to release the fat from cottonseed for enhancing fatty acid deposition in milk. Thus, the combination of FTMR and cWCS supplementation would be an alternative strategy to improve performance of lactating cows.

Effects of Molasses at Different Levels in Concentrate Supplement on Milk Yield of Dairy Cows Grazing Setaria Grass (Setaria Sphacelata) Pasture in Fiji

  • Effects of different levels of molasses in a concentrate supplement on milk yield of cows grazing Setaria grass pastures were investigated. Thirty Friesian cows, 6-7 years old with mean pre-experimental body weight of $428{\pm}6.5$ kg, in early stage of lactation were randomly allotted to five dietary treatments in a completely randomized design experiment that lasted for 126 days. Experimental dietary treatments were forage alone and forage/concentrate mixtures with molasses included at 0, 5, 10 or 15% levels designated as $T_1$, $T_2$, $T_3$, $T_4$ and $T_5$, respectively. The parameters studied were voluntary dry matter (DM) intake, average daily live weight change (LWC), milk yield, body condition score (BCS) and apparent nutrient digestibility coefficients. The DM and energy contents of Setaria grass were low compared to the concentrate diets. Fibre fractions-NDF, ADF, ADL, hemicellulose and cellulose; and gross energy were higher in concentrate mixtures than in the forage. Total DM intake (forage+concentrate mixtures) was significantly higher (p<0.001) in cows on the concentrate mixtures. LWC was not significantly different (p>0.05) between the cows. Average milk yields were significantly different (p<0.05) between cows. Fat corrected milk (FCM) was similar among cows in the treatments. BCS was better (p<0.001) in cows on concentrate mixtures. Digestibilities of DM, CP, NDF, ADF, ADL, OM, and energy were significantly higher (p<0.001) in cows on $T_2$, $T_3$, $T_4$ and $T_5$ than in those on $T_1$. There were no significant differences in the digestibility of DM, CP, NDF, ADF and ADL (p>0.001) in cows on concentrate mixtures. This study therefore demonstrated that lactating dairy cows in Fiji need a level of readily fermented energy source such as molasses in their diets; however, a level above 10% is not nutritionally suitable for lactating dairy cows. Based on data on production parameters-milk yield, fat corrected milk, body condition score and apparent nutrient digestibility coefficients-molasses levels that range between 5-10% are recommended, however, 10% is the best and therefore recommended for inclusion in the concentrate mixture of lactating dairy cows on a basal diet of Setaria sphacelata in Fiji.

Urinary Estrone Sulfate for Monitoring Pregnancy of Dairy Cows

  • The purpose of this study firstly was conducted to establish a radioimmunoassay (RIA) of estrone sulfate ($E_1S$), secondly to monitor the reproductive status of dairy cows using their urine samples. Urine and blood samples were collected in series within a day from four pregnant Holstein-friesian cows to evaluate the relationship between $E_1S$ levels in blood and urine with or without urinary creatinine basis. The urine was then collected biweekly from three cows in estrous and those artificially inseminated; collection from pregnant cows was made on a monthly basis. Results indicated that sensitivity for the $E_1S$ RIA was 5 pg/tube and the recovery rate was 100%. The daily urinary creatinine concentrations fluctuated within a day, but changes were slighter in midday, whereas the changes of concentrations of $E_1S$ in urine were relatively smaller. The concentrations of serum $E_1S$ during the estrous cycle were undetectable due to the limitation of assay, but the urinary $E_1S$ level could be measured with no obvious changes during the cycle. The urinary $E_1S$ levels increased remarkably around 7.7 to 8.3 ng/ml, 80 to 100 days after pregnancy but the serum $E_1S$ levels did not elevate until 120 to 150 days. The level of $E_1S$ increased gradually during pregnancy and eventually reached its peak before parturition at around 40 ng/ml and finally decreased to its basal level 2 days postparturition. During pregnancy, $E_1S$ concentrations of urine increased earlier than those in blood. The correlation coefficients between urinary and serum $E_1S$ concentration during pregnancy and postparturm were higher than those adjusted with creatinine (creatinine ratio). The concentrations of $E_1S$ in urine could be maintained unchanged for 8 days storing the samples in room temperature, which was extended to 8 days when the samples were pretreated by boiling for 30 minutes or treated with autoclave. In conclusion urinary $E_1S$ concentrations can be used directly for monitoring the pregnant status and fetal viability of dairy cows and can assist accurate confirmation of pregnancy in cows at least 80 to 100 days after insemination much earlier than by serum $E_1S$.

Variance Components and Genetic Parameters for Milk Production and Lactation Pattern in an Ethiopian Multibreed Dairy Cattle Population

  • The objective of this study was to estimate variance components and genetic parameters for lactation milk yield (LY), lactation length (LL), average milk yield per day (YD), initial milk yield (IY), peak milk yield (PY), days to peak (DP) and parameters (ln(a) and c) of the modified incomplete gamma function (MIG) in an Ethiopian multibreed dairy cattle population. The dataset was composed of 5,507 lactation records collected from 1,639 cows in three locations (Bako, Debre Zeit and Holetta) in Ethiopia from 1977 to 2010. Parameters for MIG were obtained from regression analysis of monthly test-day milk data on days in milk. The cows were purebred (Bos indicus) Boran (B) and Horro (H) and their crosses with different fractions of Friesian (F), Jersey (J) and Simmental (S). There were 23 breed groups (B, H, and their crossbreds with F, J, and S) in the population. Fixed and mixed models were used to analyse the data. The fixed model considered herd-year-season, parity and breed group as fixed effects, and residual as random. The single and two-traits mixed animal repeatability models, considered the fixed effects of herd-year-season and parity subclasses, breed as a function of cow H, F, J, and S breed fractions and general heterosis as a function of heterozygosity, and the random additive animal, permanent environment, and residual effects. For the analysis of LY, LL was added as a fixed covariate to all models. Variance components and genetic parameters were estimated using average information restricted maximum likelihood procedures. The results indicated that all traits were affected (p<0.001) by the considered fixed effects. High grade $B{\times}F$ cows (3/16B 13/16F) had the highest least squares means (LSM) for LY ($2,490{\pm}178.9kg$), IY ($10.5{\pm}0.8kg$), PY ($12.7{\pm}0.9kg$), YD ($7.6{\pm}0.55kg$) and LL ($361.4{\pm}31.2d$), while B cows had the lowest LSM values for these traits. The LSM of LY, IY, YD, and PY tended to increase from the first to the fifth parity. Single-trait analyses yielded low heritability ($0.03{\pm}0.03$ and $0.08{\pm}0.02$) and repeatability ($0.14{\pm}0.01$ to $0.24{\pm}0.02$) estimates for LL, DP and parameter c. Medium heritability ($0.21{\pm}0.03$ to $0.33{\pm}0.04$) and repeatability ($0.27{\pm}0.02$ to $0.53{\pm}0.01$) estimates were obtained for LY, IY, PY, YD and ln(a). Genetic correlations between LY, IY, PY, YD, ln(a), and LL ranged from 0.59 to 0.99. Spearman's rank correlations between sire estimated breeding values for LY, LL, IY, PY, YD, ln(a) and c were positive (0.67 to 0.99, p<0.001). These results suggested that selection for IY, PY, YD, or LY would genetically improve lactation milk yield in this Ethiopian dairy cattle population.

지방산 조성 비율이 다른 유기농 사료 급여가 홀스타인 원유의 CLA 및 지방산 함량에 미치는 영양 (Effects of Feeding Organic Diets with Different Fatty Acid Composition Ratio on CLA and Fatty Acid Contents in Raw Milk of Holstein-Friesian Dairy Cows)

  • 본 연구는 지방산 함량 및 배합사료와 조사료 비율을 기초로 한 유기농 사료 급여가 홀스타인 착유우에서 생산된 원유의 CLA 및 지방산 함량에 미치는 연구를 조사하기 위한 목적으로 실시하였다. 총 290두의 홀스타인 착유우를 산차 및 유량에 따라 3개 group으로 나누었다. 대조구는 C16:00, C18:2 그리고 SFA를 높게 설계하였고, 처리구 1은 C18:1, C18:2 그리고 UFA 함량을 높게 설계하였으며 처리구 2는 MUFA, C18:3 그리고 PUFA 함량을 높게 설계하였다. 결과를 요약하면 다음과 같다. 유기농 원유 내 C16:0 함량은 처리구 2에서 가장 높은 것으로 나타났다(p<0.05). 그 이유는 반추위 내 미생물의 de novo 생합성 때문인 것으로 판단된다. 처리구 2의 C18:0 함량은 7.92%로 대조구(11.39%)와 처리구 1(10.88%)보다 높았다(p<0.05). CLA 함량도 처리구 2가 처리구 1이나 대조구에 비하여 높은 것으로 나타났다(p<0.05). 원유 내에서 검출된 대부분의 CLA는 착유우 유선조직내의 ${\Delta}^9$-desaturase에 의하여 합성된 것으로 판단된다. n-3/n-6 비율도 처리구 2에서 가장 높은 것으로 나타났다(p<0.05). 본 연구 결과를 종합해 보면, 착유우에게 혼합 목건초 등을 급여하면 CLA, n-3 농도는 증가하며 C18:0 농도는 낮아지는 것으로 조사되었다. 본 연구는 유기농 인증된 조사료 및 농후사료를 이용하여 결과를 도출하였다. 그러나 유기농 사료를 사용할 경우에만 원유 내 CLA 및 n-3 농도는 증가한다고 볼 수 없다. 원유 내 고농도의 CLA 및 n-3 지방산 생산을 위해서는 반추위 미생물 및 유선세포의 지방 대사를 통한 CLA 생산 메커니즘에 대한 충분한 이해와 급여 사료 내 지방산 구성 등이 중요한 것으로 판단된다.

제독 유황의 반추위 발효성상 및 메탄 저감 효과 연구 (Effects of Detoxified Sulfur as a Feed Supplement on in Vitro Rumen Fermentation and Methane Mitigation)

  • 제독유황은 황산염의 환원제로서 황화수소로 환원되면서 메탄생성과정에서 에너지 이용 경쟁을 통해 저감 할 수 있다. 본 연구는 유독성분이 제거된 유황을 in vitro를 통해 반추위 발효 성상 및 메탄 저감 효과를 평가하였다. 반추 위액은 케뉼라가 부착되어 있는 홀스타인 젖소에서 채취하여 이용하였으며, 기질 사료는 농후사료 1 g (DM)을 사용하였다. 처리구는 0% (대조구), 0.2% (처리구 1), 0.4% (처리구 2), 0.6% (처리구 3), 0.8% (처리구 4) 및 1.0% (처리구 5)로 실시하였다. pH, 총가스발생량 (TG), 메탄, 건물 소화율, 유기물 소화율 및 휘발성지방산을 분석하였다. 메탄생성량은 0.2% 처리구에서(13.78 ml) 가장 낮았으며, 대조구에서(20.16 ml)로 가장 높았다(p<0.05). 휘발성 지방산은 0.2% 처리구에서 64.28 mM 높았다. A/P 비율은 0.2% 처리구에서 1.90으로 가장 낮았으며, 0.8%에서 2.44로 유의성이 있었지만, pH, 총가스발생량, DM 소화율, OM 소화율, acetate, propionate 및 butyrate에서는 차이가 없었다. 총 세균의 DNA는 대조구보다 모든 처리구에서 낮았다. 이 연구를 통해 0.2% 처리구에서 반추위 발효 성상 및 메탄 저감에 효과가 있었다.
