Background: The treatment of rheumatoid arthritis still depend on conserve therapy in major. Recent studies report that n-3 polyunsaturated fatty acids(PUFA) could modulate the incidence and progress of arthritis. The purpose of this study was to investigate the effects of n-3 PUFA on the development of collagen-induced arthritis in rats. Materials and Methods: Female Louvain rats were used for this experiment. Rats were randomly assigned into either normal (n=8) or collagen-immunized groups, and collagen immunized groups were divided into control(n=8, normal diet) and n-3 PUFA(n=8, 5% n-3 PUFA in diet) groups. One week after feeding n-3 PUFA to rats, they were immunized with type II collagen emulsified in incomplete Freund's adjuvant into tail and back. Development of arthritis was confirmed by x-ray and microscopic examination. Results: Incidence of arthritis at the 5th week after immunization was 38% in control and 0% in n-3 PUFA. Rats with arthritis showed edema in hind paws and inflammation in synovial membrane of the knee joint. Plasma glucose and insulin were not changed by both of immunization and diet. Plasma triglycerides and cholesterol concentrations were decreased by n-3 PUFA. Conclusion: n-3 PUFA may prevent or treat collagen-induced arthritis m rats. Further studies are needed for action mechanism of it.

The effect of a chronic inflammation (cell-mediated immune response) on energy metabolism and growth performance was assessed in weanling piglets. Twenty four barrows of 4 wk of age were assigned to one of two immunization treatments : Control group [CON: immunized with Incomplete Freund's Adjuvant (lFA)] or Immunization group [IMMU: immunized with Complete Freund's Adjuvant (CFA)]. On d0, piglets were weaned and subcutaneously immunized at the medial side of the femur with 2 ml of IFA or CFA, respectively. Energy and nitrogen balances were measured per group during 13-d balance period, and total $(HP_{tot})$, activity-related ($(HP_{act})$) and non-activity-related $(HP_{cor})$ heat production were determined every 9-min by indirect calorimetry. Ig total titers to Mycobacterium butyricum, which is present in CFA, were higher (p<0.01) in IMMU than in CON on d13 (2.5 vs 1.8) and d20 (2.9 vs 1.8) after immunization. There were no differences (p>0.10) between treatments in rectal temperature, performance, feed intake, and availability and partitioning of energy during the balance period. Average daily feed intake was numerically higher in IMMU than in CON (0.34 vs 0.32 kg/d), but there was no difference (p>0.10) in metabolizability of the dietary energy between treatments. $HP_{act}/HP_{tot}$ was 16.24 and 16.89%, and retained energy was 251 and 268 $268\;kJ{\cdot}kg^{0.75}{\cdot}d^{-1}$ for CON and IMMU, respectively. Numerically, maintenance requirement of IMMU was even lower than that of CON $(419\;vs\;427\;kJ{\cdot}kg^{0.75}{\cdot}d^{-1})$. The present study suggests that a chronic inflammation has no effect on energy metabolism and growth performance, in spite of the difference in systemic antibody responses. The reason was considered to be due to locally induced immune response, resulting from the possible encapsulation at the site of injection, and/or to a low systemic immune stress which is within a functionally acceptable physiological range for the piglets.

The effect of beta-glucan ($Polycan^{TM}$) derived from Aureobasidium pullulans SM-2001 were observed on, collagen-induced rheumatoid arthritis (RA) in DBA mice. Six week-old male DBA/1J mice were immunized by the intradermal injection of $200\;{\mu}g$ of bovine type II collagen with the equal volume of complete Freund's adjuvant at the tail base on day 1. On day 21, the mice were boosted by the intradermal injection of $200\;{\mu}g$ of bovine type II collagen with incomplete Freund's adjuvant. From the first immunization, mice had been administered $Polycan^{TM}$ (21.25, 42.5 and 85 mg/kg), diclofenac and vehicle once a day for 4 weeks, respectively. Collagen-induced hyperimmunities and arthritis signs were markedly and dose-dependently inhibited by treatment of $Polycan^{TM}$ compared with RA control except for tibial cartilages of $Polycan^{TM}$ 21.25 group. $Polycan^{TM}$ effectively inhibited the histopathological changes of collagen-induced arthritis and hyper-immunities.
Der f 2 is the group 2 major allergen of a house dust mite (Dermatophagoides farinae) and its function has been recently suggested. To determine the optimal condition of sensitization to recombinant Der f 2 (rDer f 2) in murine model of asthma, we compared the effectiveness with different adjuvants in BALB/c and C57BL/6 mice. Mice from both strains sensitized with rDer f 2 by intraperitoneal injection or subcutaneous injection on days 1 and 14. The dosage was $20{\mu}g$. Freund's adjuvants with pertussis toxin (FP) or alum alone were used as adjuvants. On days 28, 29, and 30, mice were challenged intranasally with 0.1% rDer f 2. We evaluated airway hyperresponsivenss, eosinophil proportion in lung lavage, airway inflammation, and serum allergen specific antibody responses. Naive mice were used as controls. Airway hyperresponsiveness was increased in C57BL/6 with FP, and BALB/c with alum (PC200: $13.5{\pm}6.3$, $13.2{\pm}6.7$ vs. >50 mg/ml, p<0.05). The eosinophil proportion was increased in all groups; C57BL/6 with FP, BALB/c with FP, C57BL/6 with alum, BALB/c with alum ($24.8{\pm}3.6$, $20.3{\pm}10.3$, $11.0{\pm}6.9$, $5.7{\pm}2.8$, vs. $0.0{\pm}0.0$%, p<0.05). The serum allergen specific IgE levels were increased in C57BL/6 with FP or alum (OD: $0.8{\pm}1.4$, $1.1{\pm}0.8$, vs. $0.0{\pm}0.0$). C57BL/6 mice were better responders to rDer f 2 and as for adjuvants, Freund's adjuvant with pertussis toxin was better.

Objectives : In order to study the effects of Harpagophyti Radix Aqua -acupuncture in rats with arthritis induced by Freund's complete adjuvant, performed several experimental items : that is edema rate, number of WBC, hemazocrit, platelet, total protein and albumin. Methods : In this study rats were divided into five groups : Normal group (non treated group), Control group (non-injected group in rats with arthritis induced by FCA), ST group (0.2cc normal Saline-injected group at right Chok-Samni(Zusanli,$ST_{36}$) once in every two days during the two weeks in rats with arthritis induced by FCA), HRS group (0.2cc Harpagophyti Radix-subcutaneous injected group once in every two days during the two weeks in rats with arthritis induced by FCA), HRST group (0.2cc Harpagophyti Radix-injected group at Chok-Samni($ST_{36}$) once in every two days during the two weeks in rats with arthritis induced by FCA). Results : 1. In the right plantar edema, HRS group showed a decrease with statistical significance on the 2 weeks and HRST group showed a decrease with statistical significance on the 1 weeks as compared with control group. 2. In the WBC, HRST group showed a meaningful decrease with statistical significance and also HRS group showed a decrease with statistical significanae as compared with control group. 3. In the platelet, HRST group showed a decrease with statistical significance as compared with control group. 4. In the hematocrit, HRS group showed a increase with statistical significance as compared with control group. 5. In the serum total protein and albumin, HRS group showed a increase with statistical significance as compared with control group. Conclusions : According to the above result, it is concluded that Harpagophyti Radix Aqua-acupuncture showed the therapeutic effect of antiinflammation in adjuvant arthritis and it is suggested that further studies and researchs for clinical use be needed.
Objectives : This study was to investigate the effect of high frequency electroacupuncture at $ST_{36}$ acupuncture point on the Complete Freund's Adjuvant (CFA) induced rat arthritis pain model. Methods : Arthritis was induced by intradermal injection of CFA into base of tail. Experimental groups were divided into 4 groups; Normal, Control and Jok-Samri ($ST_{36}$) and Non-Acupuncture point (NA). Normal group, non-arthritic group, was injected with normal saline,and the others groups were injected CFA. $ST_{36}$ group was treated by 120 Hz electroacupuncture at $ST_{36}$ acupuncture point, and NA group was treated by 120 Hz electroacupuncture at non-acupuncture point. Each groups were evaluated by the change of c-fos positive neurons in periaqueductal gray (PAG) and hippocampus by using an image analyzer and a microscope. Results: - In the PAG region, the number of fos-positive cells in the $ST_{36}$ group ($42.37{\pm}5.08$) were significantly (p<0.05) decreased compared with the control group ($64.56{\pm}6.35$). - In the PAG region, the number of fos-positive cells in the NA group were meaninglessly decreased compared with the control group - In the Cornu Ammonis(CA)l region of hippocampus, the number of fos-positive cells in the $ST_{36}$ group ($7.00{\pm}1.08$) and NA group ($5.56{\pm}2.01$) were significantly (p<0.05) decreased compared with the control group ($13.81{\pm}1.24$). - In the dentate gyrus region of hippocampus, the number of fos- positive cells in the $ST_{36}$ group ($10.75{\pm}0.98$) and NA group ($6.56{\pm}0.78$) were significantly (p$26.45{\pm}1.82$). Conclusions : It is expected that high frequency electroacupuncture can be used a treatment of arthritic pain.
This study was performed to assess that clinco-therapeutic effect of natural Italian honeybee (Apis mellifera) venom in adjuvant-induced arthritic rat. Ninety Sprague- Dawley rats of male were injected with complete Freund's adjuvant (CFA). Adjuvant arthritis was produced by a single subcutaneous injection of 1 mg Mycobacterium butyricum suspended in 0.1 ml paraffin oil into the right hindpaw. Righting reflex was uniformly lost and considered to be the point of arthritis development on day 14 after CFA injection. Experimental groups were divided into three groups. When arthritis was developed in the rat hind-paw, tested groups were administrated with prednisolone (10 mg/kg, p.o) and honeybee venom (one bee, s.c) at an interval of two days. Control group was subcutaneously injected with 0.1 ml of physiological saline solution in the rat at an interval of two days. Clinical findings, hematological values and histopathological findings were observed during or after the drugs administration. In tested groups, the development of inflammatory edema and polyarthritis on day 14 after treatment was suppressed. No significant differences of hindpaw edema volume and lameness score between prednisolone and honeybee venom groups were observed during or after therapeutic drugs treatment. WBC counts of prednisolone and honeybee venom treatment groups as compared with the control group were getting remarkably decreased during or after the therapeutic drugs administration(p<0.01). Erosions of articular cartilage and inflammatory cell infiltrations during or after the therapeutic drugs treatment was effectively suppressed in natural honey venom.
In order to evaluate the improvement effects of head of Panax ginseng on chronic arthritis, we have investigated the activity of butanol fraction (BuOH fraction) in vitro and in vivo system. BuOH fraction showed significant inhibition on the elastase activity. Anti-arthritic activity of BuOH fraction was also examined on type II collagen-induced arthritis in DBA/1J mice. Mice were immunized with injection of type II collagen emulsified in Freund's complete adjuvant, followed by a booster injection 21 days later. BuOH fraction(BHPG) was administered at an oral dose of 500mg/kg for 2 weeks from the 1st day boost. The hind paw edema was significantly decreased in the group of treatment with BuOH fraction compared to control. In collagen-induced DBA/1J mice, BuOH fraction did not affected the collagen antibody titer but significantly inhibited the tumor necrosis factoralpha(TNF-${\alpha}$) activity. These results were confirmed with histological evaluation of joint tissues. This study may raise the possibility that the usage of BuOH fraction of head of Panax ginseng as alternative medicine for the relief and prevention of rheumatoid arthritis symptoms.
Present work was undertaken to investigate the origin of milled wood lignin(MWL) in the wood cell wall using immunocytochemical techniques, which can provide the information on the localization of specific antigens(MWL in the present study) to be examined. Spruce MWL dissolved in DMSO and emulsified with Freund adjuvant was injected directly into the mouse spleen. The animals were boostered at two-week intervals after the initial immunization. Blood samples were purified in standard procedures. The characteristics of antibodies against MWL were tested by indirect ELISA. Visualization of MWL was carried out using conventional indirect immunogold-labelling methods on the ultrathin sections of spruce wood. Immuno-TEM observations showed that the immunogold probes were selectively attached to secondary cell walls of spruce wood. The most intense labelling was frequently observed in the S2 layer. In contrast, gold labelling in the lignin-rich regions, such as middle lamella and cell corner was not found. The immuno-TEM provides an indication that spruce MWL originates from the S2 layer.
A polymorphic fungus, Candida albicans, causes various forms of infections such as disseminated candidiasis and vaginitis. Recent reports indicate that the fungus is a main etiological agent for the arthritis. In search of new sources for treatment of the fungal arthritis, we examined $18{\beta}$-glycyrrhetinic acid ($18{\beta}$-GA) against C. albicans-caused septic arthritis. The compound is isolated from Glycyrrhizae Radix that is known to have various immunomodulating activities and is one of the most popular herbal medicines. For induction of animal model of a septic arthritis, mice were given an emulsion form of C. albicans cell wall mixed with Complete Freund's Adjuvant (CFA) via footpad-injection. To determine prophylactic and therapeutic effects, the component was given to the animals before or after the induction of the arthritis, respectively. Data showed that intraperitoneal administration of $18{\beta}$-GA resulted in reduction of the inflammation, indicating the component had both prophylactic and therapeutic activities. For investigation of mechanism of the $18{\beta}$-GA, inhibitory effects on NO (nitiric oxide) and on T-lymphocyte proliferation were determined. Results demonstrated that $18{\beta}$-GA suppressed NO production from LPS (lipopolysaccharide)-treated macrophages and also inhibited proliferation of Con A (concanavalin A)activated T-cells. Taken together, $18{\beta}$-GA, a pentacyclic triterpene, has anti-arthritic activity against C. albicans-caused septic arthritis, possibly by blocking NO production and T-cell suppression.
