Occurrence and antibiotic resistance of Enterococcus spp. from retail fresh-cut products in Korea (국내 신선 편이식품으로부터 분리한 Enterococcus의 항생제 저항성)

  • Enterococcus spp. have been considered major indicator organisms for antibiotic resistance due to their ability to easily acquire and to harbor antibiotic resistance. In this study, Enterococcus spp. were isolated from 174 retail fresh-cut products (fresh vegetable salads, microgreens, and sprouts) in Korea. Among the 20 Enterococcus isolates obtained, 18 (90.0%) were Enterococcus faecalis and 2 (10.0%) were Enterococcus faecium. The patterns of antibiotic resistance against nine antimicrobials were analyzed. Most of the isolates (85.0%) were resistant to quinupristin/dalfopristin, 40.0% and 50.0% of the isolates showed intermediate resistance to two critically important antimicrobials for human medicine, ciprofloxacin and erythromycin, respectively. Vancomycin-resistant enterococci were not detected in this study. Given the importance of antimicrobial resistance of enterococci in food safety as well as in public health, our results regarding the occurrence (level of contamination) and antimicrobial resistance of Enterococcus spp. could provide useful information that aids the risk analysis of antibiotic resistance.

  • The purpose of this study was to evaluate the preference trend of frequently served meals in the elementary school lunch program. Therefore, this evaluation was surveyed on frequently served meals in the elementary school lunch program in Inchon. Also, this study was surveyed on food preference using questionnaire including frequently served food items. Statistical analysis of data was completed using SAS program. The results of this survey was as follows: 1. Freqrenctly served foods were 56 food items including 5 boiled rice, 3 one course dishes, 10 soup & pot stewes, 10 fresh & boiled salads, 8 stirred fries, 7 stewes, 6 grill & fries, 3 kimchies and 4 desserts. 2. Students liked barley bab more than the other kinds of cooked rice. One course dishes were preferred most by the subjects. Among soup & pot stewes, fermented soybean paste stew showed lower preference than the other kind of stewes. Among fresh & boiled salads, fruits salad was the most favore, whereas, root of bellflower salad was the most hated. Among side-dishes by different preparation method, stir fries, stewes, grill & fries, kimchies, dessert were prefered most by the subjects. There were significant difference between male and female students for most given foods except soup & pot stewes. Female students showed higher preference for most given foods except fresh & boiled salads, kimchies, dessert than male students.

Quality Characteristics of Fresh Gastrodia elata according to Different Steaming Time (증자시간에 따른 생천마의 품질특성 변화)

  • Gastrodia elata has been used in traditional Chinese medicine for treating headaches, dizziness, and convulsive illness for centuries. G. elata has traditionally been processed by steaming or blanching to increase the content and quality of its main ingredients. This study aimed to identify changes in physicochemical properties and active ingredients of G. elata depending on the steaming time. Data of this study could be used to develop traditional medicine and health foods. No steaming was used as a control. Steaming time was 5, 10, 20, 30, 60, or 120 min. The drying yield according to the steaming time ranged from 20.2% to 22.9%, with the lowest drying yield at 120 min. As the steaming time increased, gastrodin content increased more than that in fresh G. elatadue to inhibition of β-glucosidase enzyme activity, 4-hydroxybenzyl alcohol condensation, and parishin decomposition. Steamed G. elatadid not show higher total polyphenols, total flavonoids, or ABTS radical scavenging activities than fresh G. elata even with an increase of steaming time. The steaming time to improve the quality of G. elata may varied depending on the size of G. elata. Thus, it is important to set the steaming time taking these characteristics into consideration.

Quality Characteristics of Sulgidduk Added with Fresh Sweet Potato (생고구마를 첨가한 설기떡의 품질특성)

  • In this study, we evaluated the quality characteristics of sweet potato Sulgidduk produced with varying amounts of fresh sweet potato, after three days of storage. The more fresh sweet potato was added, the higher were the levels of crude protein and crude lipid; however, crude ash contents were lowest in the control sample, and no significant differences in this value were detected among the samples to which fresh sweet potatoes were added. Moisture contents evidenced a tendency to decrease with increases in the amount of added fresh sweet potato and increased storage time, but pH rose with increases in the amount of added sweet potato. Total cell counts showed a tendency toward decrease with increases in the amount of added fresh sweet potato. L values tended to be low with increases in the amount of added fresh sweet potato and a values were lowest immediately after its production, although no consistent tendencies were noted in correlation with the amount of added fresh sweet potato. b values tended to increase directly with the amount of added sweet potato . With increasing storage time, the L and a values decreased, whereas the b value tended to increase. Upon textural assessment, it was observed that hardness, adhesiveness, springiness, gumminess, and chewiness(all textural components except for cohesiveness) increased with increasing quantities of added fresh sweet potato, and these factors also tended to increase with the progression of storage time. After observation via scanning electron microscopy(SEM), it was noted that the cohesiveness also increased with increasing amounts of added sweet potato. With regard to the sensory evaluation, the samples to which 15% fresh sweet potato had been added evidenced the highest acceptability in terms of color, flavor, and s0weetness, and softness and moistness in these samples decreased with increasing percentages of added sweet potato. It has been previously demonstrated that the addition of 15% fresh sweet potato resulted in optimal overall acceptability. In accordance with the aforementioned results, it has been verified that the use of fresh sweet potato in Sulgidduk is possible and probably desirable, and an addition of 15% sweet potato appears to be the optimal approach in terms of overall quality and functionality.

Biological Activity of Fresh Juice of Wild-Garlic, Allium victorialis L. (산마늘 생즙의 생리활성)

  • Wild-garlic (Allium victorialis L.) is a perennial plant found in worldwide and has been considered as a favorite vegetable due to its particular smell and taste. However, the study of biological activity of wild-garlic and the development of processed food are in rudimentary. In this study, we evaluated several biological activities, including antioxidant, antimicrobial, and inhibitory activities against human thrombin, $\alpha$-amylase and $\alpha$-glucosidase, of Ulrung wild-garlic. Analysis of the composition showed that Ulrung wild-garlic is nutritive although it is perishable. The color of fresh juice was stably maintained during 10 days-storage at $4^{\circ}C$, but rapidly discolored by heat treatment at $70^{\circ}C$ for 1 h. During heat treatment, the contents of total sugar and total polyphenol were decreased to 75% and 50%, respectively, and acidity was increased from 0.069% to 0.111%. In a while, the brix, reducing sugar, and total flavonoids showed minor changes. The fresh juice showed strong DPPH scavenging activity, reducing power and antibacterial and antifungal activity, but the heat-treated juice lost the antioxidant and antimicrobial activities. The inhibitory activities against human thrombin and $\alpha$-amylase and $\alpha$-glucosidase was negligible in both fresh juice and heat-treated juice. These results suggested that the antioxidant and antimicrobial components in wild-garlic are heat-liable and volatile. Based on our results, we propose non-heat treatment products for processed wild-garlic, for example, fresh juice-added beverage or fermented liquors using wild-garlic.

Studies on the chages in Free Amono Acids of Yellow Corvenia(Pseudosciaena manchurice) during Gulbi processing (굴비제조중 유리아미노산의 변화에 관한 연구)

  • Gulbi were processed by salting Yellow corvenia (Pseudosciaena manchurica) with in three ways: the dry salting method with bar salt, the dry salting method with purified salt or with the abdominal brine injection method with purified salt. The half of the sample was dried by the control system of temperature and humidity; the other part was dried by the natural condition. The moisture content of fresh Yellow corvenia muscle and eggs were 76.8%, and 68.2% while those of dried samples decreased to 57.7% and 45.3%, respectively. The total nitrogen content of fresh muscle and eggs were 11.0g% and 7.6g%, respectively (dry weight basis), which decreased slightly during salting and showed no significant changes during drying prosess. The protein nitrogen content of fresh muscle and eggs were 10.2g% and 7.5g%, which decreased during Gulbi processing. On the other hand, the nonprotein nitrogen content of both muscle and eggs increased. The content of free amino acids of fresh muscle and eggs were 508.8mg/100g and 1,110.6mg/100g, which increased to between 5.3 and 2.7 times, respectively after 25 days of drying. The composition patterns of free amino acids in muscle and eggs were similar to each other. The four amino acids - Ala, Glu, Lys and Leu - were most abundant in both fresh and dried samples. These amino acids are known as taste and flavour constituents.

Effects of Combined Treatment with High $CO_2$ Concentration and Ascorbic acid on Browning of Fresh-cut 'Fuji' Apples (고농도 $CO_2$와 Ascorbic acid 처리가 신선절단 사과의 갈변에 미치는 영향)

  • This papers were studies the effects of high $CO_2$ concentration pretreatment and ascorbic acid on the browning of fresh-cut apples. The prepared samples were dipped in 1% (w/v) ascorbic acid solution (AA). Fresh-cut apples (3 pieces) were packed in polypropylene bags (0.03 mm, $20{\times}15$ cm), and stored at $10^{\circ}C$ for 16 days. AA-treated samples showed higher L value than those of non-treated and control (dipped in water without $CO_2$ pretreatment), while non-treated samples showed higher hardness than AA-treated sample. Total soluble solids, pH, titratable acidity were not affected by high $CO_2$ pretreatment and AA treatment. These result shows high $CO_2$ concentration pretreatment with dipping in 1% ascorbic acid solution (AA) has an effects on controlling enzymatic browning during the storage of fresh-cut 'Fuji' apples.

Effect of Ionizing Radiation of Physiological Characteristics of Fresh Mushrooms (Agaricus bisporus L.) (양송이 버섯의 생리적 특성에 대한 감마선 조사 영향)

  • Fresh mushroom (Agaricus bisporus L.) was irradiated (0, 1, 2, 3kGy) and kept for 20 days at 9{\pm}1^{\circ}C\;and\;80{\pm}7%$ RH. Parameters of qualities were investigated on the physical and physiological characteristics. The pileus and stipe on nonirradiated mushroom were expanded and elongated from the 3rd day of storage, there by losing the acceptability as edible samples. After 5 days of storage, 2 to 3kGy of gamma irradiation were especially effective for controllong natural maturation and senescence of fresh mushrooms and so irradiated mushrooms were acceptable more than 20days storage. The texture of irradiated samples was superior to that of nonirradiated samples, even though softening of the tissue occurred during storage. Weight loss was greatest in the nonirradiated sample due to evaporation from an increased surface area resulting from expansion and ripending, which were retarded in the 2 to 3kGy irradiated samples after 5days of storage. These results suggest that the irradiation dose of 2 to 3kGy is apparently effective to extend the shelf life of fresh mushrooms stored at the above-mentioned condition.

Antioxidative and $Lp-PLA_2$ Inhibitory Activities in 29 Fruits and Vegetables (시중 유통 과일 및 채소류 29종의 항산화 효과 및 $Lp-PLA_2$ 활성 저해 연구)

  • Total phenolic content and the antioxidative activity (DPPH radical-scavenging capacity) of 29 fruits and vegetables were determined. The total phenolic contents of plum (1.06 mg GAE/g fresh weight) and cherry (0.95 mg GAE/g fresh weight) were high, whereas those of banana, carrot, and cucumber (each at 0.07 mg GAE/g fresh weight) were low. In the DPPH radical-scavenging assay, apple mango extract showed the highest value(92.8%) among the 29 ethanol extracts tested, followed by paprika (75.4%), beet (75.3%), and cherry (75.1%). In the $Lp-PLA_2$ inhibition assay, cherry (34.95% inhibition) and grape (31.03%) showed the highest $Lp-PLA_2$ activities among the 29 fruits and vegetables examined.

Effects of UV-C and Ethanol Treatment on Quality of Fresh-cut Seedless Watermelon (UV-C, 에탄올 처리가 신선편이(Fresh-cut) 씨 없는 수박의 품질에 미치는 영향)

  • To apply UV-C as a non-heating sterilization method to increase the microbiological safety of fresh seedless watermelon products, reductions in E. coli and quality changes by treatment dose (0, 2, 4, 8, 14, 20 kJ/m2) were investigated. The pH, sugar content, and hardness of watermelon inoculated with E. coli were not significantly different according to the UV-C treatment dose, but the polyphenol content was significantly decreased compared to the controls (425.4 GAE ㎍/g F.W.). When treated with 2 and 4 kJ/m2, the lycopene content was 31.6 and 30.9 ㎍/g F.W., respectively, which was increased compared to the controls (28.5 ㎍/g F.W.). The arginine and citrulline content was also significantly increased compared to the controls. The number of E. coli was significantly decreased compared to the controls following UV-C treatment. Considering the degree of E. coli reduction, lycopene content, arginine content, citrulline content, and UV-C irradiation time, subsequent experiments were conducted by selecting a UV-C treatment dose of 2 kJ/m2. The results of confirming the degree of reduction in the number of E. coli colonies by a single treatment and combined treatment with UV-C 2 kJ/m2 and 70% ethanol showed that the combined treatment was most effective as colonies were decreased by 2.3 log CFU/g compared to the controls. Therefore, it is judged that UV-C 2 kJ/m2 radiation and combined treatment with 70% ethanol could be applied as a non-heating sterilization method for fresh watermelon slices.
