Characterization of Fatty Acids Extracted from Brachionus rotundiformis Using Lipase-catalyzed Hydrolysis

  • Lipids were extracted from marine rotifer, Brachionus rotundiformis in order to examine the functionality of lipid enzymatic modification. The fatty acids, palmitic, linoleic, oleic and stearic acids were the dominant forms accounting for approximately 35.8%, 21.5%, 15.9% and 7.7% of the total lipid content, respectively. Lipid fractions were categorized as neutral lipids (38.5%), glycolipids (45.9%) and phospholipids (17.6%), and after extraction from the rotifer were isolated by thin-layer chromatography (TLC) as free fatty acids (FFA), monoacylglycerol (MAG), diacylglycerol (DAG) and triacylglycerol (TAG). The production of polyunsaturated fatty acid (PUFA) concentrate from rotifer lipids was studied using lipase-catalyzed hydrolysis. In addition, rotifer lipids were modified by hydrolysis using lipases such as porcine pancreas, Candida rugosa and Rhizomucor miehei. The lipase from Rhizomucor miehei was effective in extracting linoleic acid (C 18:2), while the lipase from Candida rugosa was effective in palmitic acid (C16:0) extraction.

Sea tangle (Laminaria japonica) supplementation on meat quality of Korean native black goat

  • The supplementation effects of sea tangle powders were determined for the meat quality in Korean native black goats. A total 90 castrated male black goats at 3-month age were divided into 3 dietary treatment groups: control (basal diet + mineral block), T1 (0.3% sea tangle feeding with the basal diet), T2 (0.9% sea tangle feeding with the basal diet). At 9-months feeding, 10 goats per treatment group were slaughtered, and the longissimus dorsi muscle samples were vacuum-packed, and subsequently analyzed for physicochemical evaluations. Analysis revealed decrease in the shear force and TBARS values of meat in the sea tangle dietary groups (p < 0.05). The T2 group exhibited increased levels of unsaturated fatty acids such as C16:1, C18:1, C18:2, and C20:4 (p < 0.05). The content of free amino acids with desirable taste such as aspartic acid, glutamic acid, glycine, histidine, and serine were higher in T2, whereas alanine content was higher in both sea tangle dietary groups, as compared to control (p < 0.05). These data indicate that feeding dietary sea tangle as an alternative mineral source results in an improvement in the physicochemical profiles of goat meat.

전통식 녹차된장과 일부 시판된장의 이화학적 특성 (Physicochemical Quality Comparison of Commercial Doenjang and Traditional Green Tea Doenjang)

  • 본 연구는 일반 시판된장과 녹차된장의 일반성분, 유리아미노산, 지방산, 유기산의 함량과 항산화 활성도를 비교하였으며, 그 결과는 다음과 같다. 수분함량은 일반된장에 비해 녹차된장이 높게 나타났으며, 조 단백 함량은 일반된장에 비해 녹차된장이 낮게 나타났다. 조 지방 함량의 경우 일반된장과 녹차된장이 비슷하게 나타났으며, 회분함량은 녹차된장이 일반된장의 평균값을 나타냈고, 당질 함량은 녹차된장이 가장 낮게 나타났다. 섬유소함량은 일반 시판 된장 중 H제품을 제외하고는 비슷하게 나타났으며, 열량 역시 녹차된장이 가장 낮았다. 녹차된장의 색도는 일반된장에 비해 비교적 낮게 나타났다. 녹차된장중의 총 카테킨 함량은 2.3%로 나타났다. 유리 아미노산의 경우 시판된장 중 C사 제품은 sarcosine이 337.8 mg%로 가장 많은 함량을 나타냈고, 다음으로 proline(268.4 mg%), glutamic acid(268.0 mg%)순으로 나타났으며, H사 제품은 glutamic acid가 831.6 mg%로 가장 높았고, leucine(491.0 mg%), phenylalanine(364.3 mg%)순이었고, S사 제품은 역시 glutamic acid(855.2 mg%)가 시료중에서 가장 높게 나타났으며, 다음으로 alanine(567.9 mg%), phenylalanine(413.9 mg%), valine(400.3 mg%) 순이었다. 녹차된장 역시 glutamic acid(326.24 mg%)가 가장 많이 함유되었고, 다음으로 proline(320.9 mg%), leucine(221.49 mg%)순으로 나타났다. 포화지방산은 녹차된장이 일반된장에 비해 약간 높았으며, 불포화지방산중 단일 불포화지방산은 일반된장에 비해 녹차된장이 약간 낮았고, 다 불포화지방산은 녹차된장이 일반된장의 가장 낮은 값과 비슷하게 나타났다. 총 지방산중에 linoleic acid(51%)가 가장 높게 나타났으며, 다음으로 oleic acid(21∼24%), palmitic acid(10∼11%), stearic acid(3∼4%) 순으로 나타났다 된장의 유기산은 주된 유기산이 oxalic acid였으며, lactic, succinic acid를 제외하고는 대체로 일반 시판된장이 녹차된장에 비해 1.6∼2.7배 높게 나타났다. 녹차된장의 항산화 활성도는 타 일반된장에 비하여 높게 나타났다. 본 연구결과 기능성을 가진 녹차된장이 일반시판된장에 비해 열량이 낮으며, 항산화 활성도가 높으므로 장기간 섭취시 비만예방 및 노화방지에 효과가 있을 것으로 사료된다.

  • 개화후 15일된 대두미숙종자를 배양하였을 때 callus의 형성에 적당한 온도는 $24{\sim}27^{\circ}C$였으며, NAA를 생장조절제로 사용하였을 때 embryogenic callus(EC)가 많이 형성되었다. 또한 EC를 계대배양하였을 때 BA 2mg/1 사용한 배지에서는 shoot가, IAA 2mg/1 사용한 배지에서는 root가 많이 형성되었으나, BA 2mg/1 IAA 2mg/1를 동시에 사용한 배지에서는 callus의 형성만 왕성할 뿐, 기관분화의 기미는 보이지 않았다. 배양체에 함유되어 있는 total lipid는 온도가 낮은 배양조건일수록 많았으며, callus의 생육이 왕성한 $24{\sim}27^{\circ}C$에서 glycolipid, phospholipid, free sterol의 함량이 다소 많아졌으며, embryogenic structure가 많이 생겨날 때에는 free sterol의 함량이 많았다. 배양체의 지방산 조성은 EC에서 불포화 지방산의 함량이 많았으며, 불포화도는 0.837이었다. 또한 sterol조성에서도 EC에서 cholesterol의 함량이 다른 배양체에서보다 현저히 높았다.

죽순의 영양성분 및 죽순의 식이섬유가 장내미생물에 미치는 영향 (The Nutritional Composition of Bamboo Shoots and the Effects of its Fiber on Intestinal Microorganisms)

  • This study evaluated the composition two popular species of edible bamboo shoots in Korea (Phyllostachyspubescens and Sinoarundinarianigra) and the effect of their abundant dietary fiber on intestinal microorganisms in healthy young women. The ranges of total moisture, crude protein, crude lipid, crude ash, and dietary fiber content were 87.190.8, 2.943.5, 0.150.39, 0.411.05, and 4.206.15% (wet weight basis), respectively. Moisture and crude ash content increased after heat treatment; however, crude protein, crude lipid, and dietary fiber content were reduced after heating. The major minerals found in bamboo shoots were potassium, phosphorous, sulfur, magnesium, and calcium. In addition, glucose and fructose were abundant free sugars, while asparagine and tyrosine were the most abundant free amino acids. Approximately 70% of the total free fatty acids found in bamboo shoots were linoleic acid and linolenic acid. The ascorbic acid content was 6.60~17.56 mg/100 g (wet weight basis), and one phenolic compound, p-hydroxy benzoic acid, was 0.10.2% (wet weight basis) and detected by HPLC analysis. The intake of bamboo shoots for seven days significantly increased viable cell counts of Lactobacillus spp. and reduced viable cell counts of Bacteriodes spp. in feces (p<0.05). In our data, bamboo shoots may be useful in the food industry as high dietary fiber ingredients.

밤의 저장중 지방질 조성의 변화 (Changes of Lipids in Chestnut during Storage)

  • 밤을 $20^{\circ}C$에서 9주동안, $1^{\circ}C$에서 15주동안 저장하면서 저장중 지방질의 변화를 측정하여 다음과 같은 결과를 얻었다. 밤의 총지질중에는 중성지질이 43.4%, 당지질이 26.5%, 인지질이 30.1% 함유되었으며 밤의 저장 동안에 이들의 함량이 증가되었다. 중성지질의 구성지질성분으로는 triglyceride (TG), 1,2-diglyceride (1,2-DG), 1,3diglyceride (1,3-DG), sterolester (SE), sterol (S) 및 free fatty acid(FFA) 등 6종이었으며, 당지질의 구성지질성분으로는 digalactosyldiglyceride (DGDG), monogalactosyl diglyceride (MGDG), sterylglycoside (SG) 및 acylsterylglycoside (ASG) 등 4종이었고 인지질의 주요 구성지질로는 phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylinositol (PI) 등이었다. 저장중의 지질의 변화를 보면 중성지질의 함량은 저장 5주후에 감소되었고, 당지질은 7주까지는 증가되다가 그 이후 감소되었으며 인지질은 $20^{\circ}C$$1^{\circ}C$ 양온도구에서 저장기간동안 가장 많이 증가되었다. 그리고 TG, SE, S, DGDG, MGDG의 함량은 $20^{\circ}C$$1^{\circ}C$ 양온도구에서 저장기간에 따라 증가되었다. 총지질, 중성지질, 당지질 및 인지질, 그리고 TG, DG, ASG, PC 및 PE의 주요 지방산으로는 linoleic acid, oleic acid, palmitic acid 및 linolenic acid 등이었으며 저장기간에 따라서는 $20^{\circ}C$$1^{\circ}C$ 양온도에서 linoleic acid는 증가하는 반면에, oleic acid 는 감소하였다.

Variation of Chemical Components and Their Interaction with Isoflavones in Maturing Soybean Seeds

  • This study was focuses on the variation of isoflavone contents during seed development and their interaction with major chemical components such as protein, amino acids, saccaharides, lipid and fatty acids. During maturing, lipid, protein, and amino acid contents in soybean seeds showed the highest values at R7 stages, but isoflavone contents were increased until R8 stage. It was noted that malonyl glucosides $(64.2\%)$ are predominant forms among conjugated isoflavones followed by glucosides $(30.7\%)$, acetyl glucosides $(4.1\%)$ and aglycones $(0.9\%)$. Sucrose and stachyose were presented as a major saccharide in soybean seeds. As maturing days progressed, they were constantly increased and the highest contents were observed at R8 stage. While small quantities of raffinose, fructose, glucose, maltose, DP3 (DP: degree of polymerization), DP6, and DP7 were detected. These results showed that saccharide composition at the beginning of seed development is primarily monosaccharides with little sucrose and oligosaccharides, but as maturing days proceeds, sucrose and starch increase with concomitant decrease in monosaccharides. Sucrose and stachyose were positively correlated with isoflavone (r=0.780, 0.764 at p<0.01, respectively), while fructose, glucose, maltose, and DP7 were negatively correlated (r=-0.651, -0.653, -0.602, and -0.586 at p<0.05, respectively). Soybeans at R8 stage were high in protein and amino acid, but low in free amino acid contents. Protein and amino acid contents showed positively significant correlations with isoflavone (r=0.571 and 0.599 at p<0.05, respectively), but free amino acid content were negatively correlation with isoflavone (r=-0.673, p<0.01). The lipid content reaches its final content relatively early stage of seed development and remains constant as compared with other chemical components. Among the fatty acids, although varietal difference was presented, stearic acid and linolenic acid were gradually decreased, while oleic and linoleic acid were increased as seed maturing progressed. Lipid was significantly correlated (r=0.754, p<0.01) with isoflavones. However, neither saturated fatty acid nor unsaturated fatty acids significantly affected the isoflavone contents of maturing soybean seeds.

Increased Hepatic Lipogenesis Elevates Liver Cholesterol Content

  • Cardiovascular diseases (CVDs) are the most common cause of death in patients with nonalcoholic fatty liver disease (NAFLD) and dyslipidemia is considered at least partially responsible for the increased CVD risk in NAFLD patients. The aim of the present study is to understand how hepatic de novo lipogenesis influences hepatic cholesterol content as well as its effects on the plasma lipid levels. Hepatic lipogenesis was induced in mice by feeding a fat-free/high-sucrose (FF/HS) diet and the metabolic pathways associated with cholesterol were then analyzed. Both liver triglyceride and cholesterol contents were significantly increased in mice fed an FF/HS diet. Activation of fatty acid synthesis driven by the activation of sterol regulatory element binding protein (SREBP)-1c resulted in the increased liver triglycerides. The augmented cholesterol content in the liver could not be explained by an increased cholesterol synthesis, which was decreased by the FF/HS diet. HMG-CoA reductase protein level was decreased in mice fed an FF/HS diet. We found that the liver retained more cholesterol through a reduced excretion of bile acids, a reduced fecal cholesterol excretion, and an increased cholesterol uptake from plasma lipoproteins. Very low-density lipoproteintriglyceride and -cholesterol secretion were increased in mice fed an FF/HS diet, which led to hypertriglyceridemia and hypercholesterolemia in Ldlr-/- mice, a model that exhibits a more human like lipoprotein profile. These findings suggest that dietary cholesterol intake and cholesterol synthesis rates cannot only explain the hypercholesterolemia associated with NAFLD, and that the control of fatty acid synthesis should be considered for the management of dyslipidemia.

옥수수 색소 2호 품종의 일반성분, 유리당, 지방산 및 안토시아닌 색소의 함량 (Proximate, Free Sugar, Fatty acids Composition and Anthocyanins of Saekso 2 Corn Kernels)

  • 본 연구는 흑자색 알곡 품종으로 등록된 색소 2호 옥수수 알곡의 일반성분 및 색소 함량을 분석하여 기초자료로 제공하고자 수행되었다. 색소 2호 알곡의 일반성분 분석결과, 수분, 조회분, 조지방, 조단백질은 각각 8.84%, 1.44%, 5.46%, 10.31%이었다. 색소 2호 알곡의 유리당 성분으로 fructose, glucose, maltose 및 sucrose가 검출되었으며, 함량은 sucrose (1.00%) > glucose (0.63%) > maltose (0.52%) > fructose (0.44%) 순이었다. 색소 2호 알곡의 지방산 분석결과 총 11종의 지방산이 검출되었으며 포화 지방산과 불포화 지방산의 비율은 16.09 : 83.91로 linoleic acid가 50.86%로 가장 높게 나타났다. 색소 2호 알곡 추출물의 총 안토시아닌 함량은 0.24%였고, 알곡 추출물을 HPLC-MS/MS로 분석한 결과 색소 2호 알곡에 함유된 안토시아닌 색소는 C-3-G, Pg-3-G, Pn-3-G인 것으로 확인되었다.

Fatty acid composition and antioxidant capacity of some medicinal mushrooms in Turkey

  • Turkey has a very rich fungal flora due to its phytogeographical position. The screening of chemical content and active substances of mushrooms becomes an important subject not only for Turkey but also for all over the world. In the last decade, Analyses on phytochemical and biological activity of fungi have gradually increased as a result of improvement in the number and quality of facilities. In the scope of the present research, four medicinal mushrooms; Morchella elata, Lactarius volemus, Cantharellus cibarius and Tricholoma terreum were analyzed for their fatty acid compositions and antioxidant capacities. The fungal species have been found with unsaturated fatty acid/saturated fatty acid ratio of 6.73 for Morchella elata, 4.12 for Lactarius volemus, 5.21 for Cantharellus cibarius, 3.73 for Tricholama terrum. In addition, the concentration of malondialdehyde which was an indicator of lipid peroxidation was also determined in these species. According to the results, free radical scavenging activity of Morchella elata and Lactarius volemus were found higher than the other species. Any of the mushroom species investigated were found having very high metal chelating activity. The results showed that the extract of Morchella elata and Lactarius volemus exhibited significant antioxidant activities. Hence, the mushrooms have a potential to be a natural antioxidant in food industries as antioxidant agent.
