Endothelium-Dependent Vasorelaxation Effects of Rubus Coreanus extract on Rabbit Carotid Artery (복분자(覆盆子)의 내피세포의존성 혈관이완효과)

  • This study was investigated to evaluate the vasorelaxant effect of Rubus coreanus(RC) extract on contracted rabbit carotid artery and its mechanism. To study the effect of RC extract on contracted rabbit carotid arterial strips, arterial strips with intact or damaged endothelium were used for experiment using organ bath. The pre-contracted arterial strips with norepinephrine(NE) or potassium chloride(KCl) was treated with various concentrations of an extract of RC(0.01, 0.03, 0.1, 0.3 and 1.0 $mg/m{\ell}$). To determine the mechanisms of RC-induced vasorelaxant, RC extract was infused into contracted arterial rings which had been pretreated by indomethacin(IM), tetraethylammonium chloride(TEA), $N{\omega}$-nitro-L-arginine (L-NNA), methylene blue(MB). And calcium chloride(Ca) 1 mM was infused into precontracted arterial ring induced by NE or KCl after treatment of RC extract in $Ca^{2+}$-free krebs solution. Cytotoxic activity of RC extract on human umbilical vein endothelial cell(HUVEC) was measured by MTT assay, and nitric oxide(NO) prodution was measured by Griess reagent. RC extract revealed significant relaxation on NE-induced arterial contraction, but didn't relax on KCl-induced arterial contraction. RC extract also had an effective relaxation to the intact endothelium arterial ring, but not the damaged endothelium arterial ring. Treatment of IM, TEA, L-NNA, MB reduced the relaxation of RC extract. Pretreatment of RC extract inhibited the contraction by influx of extracellular $Ca^{2+}$ in contracted arterial ring induced by NE, but it didn't work the contraction by influx of extracellular $Ca^{2+}$ in contracted arterial ring induced by KCl in $Ca^{2+}$-free krebs solution. RC extract increased nitric oxide production on HUVEC. This study indicated that the relaxation effect of RC extract on contracted rabbit carotid artery is related with NO-cGMP pathway, EDHF, prostacyclin.

Resistance of Cementitious Binders against a Fall in the pH at Corrosion Initiation

  • At the onset of corrosion of steel in concrete, hydrogen ions usually evolve in the process of electrochemical reaction, thereby decreasing the pH of the pore solution, which can be buffered by cement hydration products, as being representatively illustrated by calcium hydroxide. Hence, a fall in the pH is dependent on properties of cement hydration (i.e. hydration products and degree of hydration). The present study tested acid neutralization capacity (ANC) of cementitious binders of OPC(Ordinary Portland Cement), 30% PFA(Pulverized Fuel Ash), 60% GGBS(Ground Granulated Blast Furnace Slag), 10% SF(Silica Fume) to quantify the resistance of cement matrix to a pH fall. Cement pastes were cast at 0.4 of a free W/C ratio with 1.5% chlorides by weight of binder in cast. Powder samples obtained crushed and ground specimen after 200 days of curing were diluted in still water combined with different levels of 1M nitric acid solution, ranging from 0.5 to 20 mol/kg. Then, the pH of diluted solution was monitored until any further change in the pH did not take place. It was seen that the pH of the diluted solution gradually decreased as the molar amount of nitric acid increased. At some particular values of the pH, however, a decrease in the pH was marginal, which can be expressed in the peak resistances to a pH fall in the ANC curve. The peaks occurred at the variations in the pH, depending on binder type, but commonly at about 12.5 in the pH, indicate a resistance of precipitated calcium hydroxide. The measurement of water soluble chloride at the end of test showed that the amount of free chloride was significantly increased at the pH corresponding to the peaks in the ANC curve, which may reflect the adsorption of hydration products to chlorides.

A Selectivity Character for the Phase Transfer Reactions of Alcohols by Tetra-n-Butyl Ammonium Chloride (Tetra-n-Butyl Ammonium Chloride에 의한 알코올류의 상전이 반응에 대한 선택 특이성)

  • Only trace amounts of hydroxide ion can be extracted from aqueous phase into organic phase by Tetra-n-Butyl Ammonium Chloride(TBAC). Addition of small amounts of primary alcohols, particularly certain dials, dramatically changes the behavior of Phase Transfer Catalysis systems, and surprising amounts of base can be found in the organic phase. Quantitative measurements were carried out for the extraction amounts of primary alkoxides, secondary alkoxides, and diol anions into organic phase. On the other hand, the selectivity constants for extraction of primary alcohols and benzylalcohol could be separated to the equilibrium constants of the ion pairs such as $Q^+RO^-$ and $Q^+Cl^-$ in the aqueous and organic phases and this distribution coefficients between phases on anions respectively. In a word, the colligated property for the selectivity of $Q^+RO^-$ in which $Q^+$ is quaternary cation and $RO^-$ alkoxide ion could be discussed in more detail by using of the corresponding free energies to the various constants mentioned.

Proximate, Free Sugar, Fatty acids Composition and Anthocyanins of Saekso 2 Corn Kernels (옥수수 색소 2호 품종의 일반성분, 유리당, 지방산 및 안토시아닌 색소의 함량)

  • This study was performed to provide basic data of Saekso 2 corn kernels. Proximate composition, free sugars, fatty acids and anthocyanin content of Saekso 2 corn kernels were analyzed. Proximate composition of dried Saekso 2 corn kernels were represented 8.84% moisture, 1.44% crude ash, 5.46% crude lipid, 10.31% crude protein. Free sugars composition by HPLC/ELSD showed that sucrose (1.00%), glucose (0.63%), maltose (0.52%), fructose (0.44%) were present. The composition of fatty acids in Saekso 2 corn kernels was analyzed by GC/FID. 11 species of fatty acids were analyzed in Saekso 2 corn kernels. The ratio of saturated fatty acids and unsaturated fatty acids 16.09 : 83.91. Content of linoleic acid was the highest in fatty acids. The total anthocyanin content in Saekso 2 corn kernels was 0.24% by UV/Vis. Anthocyanin components separated and quantified using HPLC/MS/MS. Cyanidin 3-O-glucoside chloride (C-3-G), pelargonidin 3-O-glucoside chloride (Pg-3-G) and peonidin 3-O-glucoside chloride (Pn-3-G) of anthocyanin were analyzed in Saekso 2 corn kernels.

Determination of Free 4-hydroxyproline with Dansylchloride by HPLC in Human Urine (소변 중 4-hydroxyproline 분석에 관한 연구)

  • Objectives : The level of 4-hydroxyproline (4-Hyp) in human urine was measured using high performance liquid chromatography (HPLC) with a fluorescence detector. This method is useful for medical examinations and investigating the radicals induced by physical, chemical, mental stresses. This method is superior to many published several methods in terms of its low cost and ability to analyze many samples. Methods : The urine from workers in a tire manufacturing company (22 male pre- and post-shift workers) and 18 office-workers as controls were analyzed. Data concerning age, the cumulative drinking amount and the cumulative smoking amount was collected with a questionnaire. The optimum applied amount of dansyl-Cl, the optimum reaction temperature and time, the recoveries and the optimum pH of the eluent and buffer were determined.4-Hyp from human urine was derivatized with dansyl-Cl (dimethylamino-naphthalene-1-sulfonyl chloride) after removing the a-amino acid by a treatment with phthalic dicarboxaldehyde (OPA) and cleaned with Bond Elut C18 column. The 4-Hyp derivatives were separated on a reversed phase column by gradient elution with a phosphate buffer (5 mmol, pH 8.0) and acetonitrile, and detected by fluorescence measurements at 340 nm (excitation) and 538 nm (emission). Results : The detection limit for the urinary free 4-Hyp was $0.364{\mu}mol/l$. The recovery rate of 4-Hyp was 99.7%, and the effective pH of the phosphate buffer and borate buffer were 3.0 and 8.0, respectively. From statistical analysis, age, drinking and smoking did not affect the urinary free 4-Hyp in both the controls and workers. The range of urinary 4-Hyp in the controls, pre-shift, and post-shift workers were 0.33-16.44, N.D-49.06, and $0.32-56.27{\mu}mol/l$. From the pared-sample t-test, the urinary 4-Hyp levels in post-shift workers ($11.82{\pm}6.73\;nmmol/mg\;Cre$) were 2-fold higher than in pre-shift workers ($5.36{\pm}5.53\;nmol;/mg\;Cre$) and controls ($4.91{\pm}4.89\;nmol;/mg\;Cre$). Conclusions : This method was developed with high sensitivity, accuracy, and precision. The present method was effectively applied to analyze the urinary free 4-Hyp in both controls and workers.

Growth Responses of seven Intestinal Bacteria Against Phellodendron amurense Root-Derived Materials

  • The growth responses of Phellodendron amurense root-derived materials against seven intestinal bacteria were examined, using an impregnated paper disk agar diffusion method and spectrometric method under $O_2$-free condition. The biologically active constituent of the P. amurense root extract was characterized as berberine chloride ($C_{20}H_{18}NO_{41}Cl$) using various spectroscopic analyses. The growth responses varied depending on the bacterial strain, chemicals, and dose tested. At 1 mg/disk, berberine chloride strongly inhibited the growth of Clostridium perfringens, and moderately inhibited the growth of Escherichia coli and Streptococcus mutans without any adverse effects on the growth of three lactic acid-bacteria (Bifidobacterium bifidum, B. longum, and Lactobacillus acidophilus). The structure-activity relationship revealed that berberine chloride exhibited more growth-inhibiting activity against C. perfringens, E. coli, and S. mutans than berberine iodide and berberine sulfate. These results, therefore, indicate that the growth-inhibiting activity of the three berberines was much more pronounced as chloridated analogue than iodided and sulphated analogues. As for the morphological effect caused by 1 mg/disk of berberine chloride, most strains of C. perfringens were damaged and killed, indicating that berberine chloride showed a strong inhibition against C. perfringens. As naturally occurring growth-inhibiting agents, the P. amurense root-derived materials described could be useful as a preventive agent against diseases caused by harmful intestinal bacteria such as clostridia.

A Study on Corrosion Resistance of CA2-Mixed Paste (CA2 혼입 페이스트의 부식저항성에 관한 연구)

  • Deterioration in durability of structures due to the steel corrosion is difficult to determine whether or not corrosion is initiated and how much propagated, and moreover, repair and maintenance are not easy to deal with. Therefore, preventive treatments can be the best option to avoid the deterioration. Various methods for preventing corrosion of steel, such as electrochemical treatments, anti-corrosion agents and steel surface coatings, are being developed, but economic and environmental aspects make it difficult to apply them to in-situ field. Thus, the purpose of this study was to improve corrosion resistance by using CA-based clinker that are relatively simple and expected to be economically profitable Existing CA-based clinkers had problems such as flash setting and low strength development during the initial hydration process, but in order to solve this problem, CA clinker with low initial reactivity were used as binder in this study. The cement paste used in the experiments was replaced with CA2 clinker for 0%, 10%, 20%, and 30% in OPC. And the mixture used in the chloride binding test for the extraction of water-soluble chloride was intermixed with Cl- 0.5%, 1%, 2%, and 3% by weight of binder content. To evaluate characteristic of hydration heat evolution, calorimetry analysis was performed and simultaneously chloride binding capacity and acid neutralization capacity were carried out. The identification of hydration products with curing ages was verified by X-ray diffraction analysis. The free chloride extraction test showed that the chlorine ion holding ability improved in order OC 10 > OC 30 > OC 20 > OC 0 and the pH drop resistance test showed that the resistance capability in pH 12 was OC 0 > OA 10 > OA 20 > OA 30. The XRD analyses showed that AFm phase, which can affect the ability to hold chlorine ions, tended to increase when CA2 was mixed, and that in pH12 the content of calcium hydroxide (Ca(OH)2), which indicates pH-low resistance, decreased as CA2 was mixed

In Vitro Free Radical Scavenging and Hepatoprotective Activities of Taraxacum mongolicum (포공영의 자유라디칼 소거 및 간세포 보호활성)

  • The methanol (MeOH) extract and ist fractions of Taraxacum mongolicum (Compositae) were examined for their scavenging effects on 1,1-diphenyl-2- phenylhydrazyl (DPPH) and superoxide radicals, and hepatoprotective effects on tacrine-induced cytotoxicity in human hepatoma cell line, Hep G2 cells. Both methylene chloride $(CH_2Cl_2)$ and butanol (n-BuOH) soluble fractions of the MeOH extract showed the free radicals scavenging and hepatoprotective effects. From these results, it is suggested that hepatoprotective effect of these fractions partly relies on their free radical scavenging activity.

Investigation on Chain Transfer Reaction of Benzene Sulfonyl Chloride in Styrene Radical Polymerization

  • The free radical polymerization of styrene was initiated with azobis(isobutyronitrile) in the presence of benzene sulfonyl chloride. Analysis of the terminal structures of the obtained polystyrene with $^1H$ NMR spectroscopy revealed the presence of a phenyl sulfonyl group at the ${\alpha}$-end and a chlorine atom at the ${\omega}$-end of each polystyrene chain. The terminal chlorine atom in the polystyrene chains was further confirmed through atom transfer radical polymerization (ATRP) of styrene and methyl acrylate using the obtained polystyrenes as macroinitiators and CuCl/2,2'-bipyridine as the catalyst system. GPC traces of the products obtained in ATRP at different reaction times were clearly shifted to higher molecular weight direction, indicating that nearly all the macroinitiator chains initiated ATRP of the second monomers. In addition, the number-average molecular weights of the polystyrenes increased directly proportional to the monomer conversions, and agreed well with the theoretical ones.

Evidence for Singlet Oxygen Involvement in Cell-free Myeloperoxidase/$H_2O_2$/ Chloride Sytem: Exclusion of Hydroxyl Radical Involvement (Cell-free Myeloperoxidase/$H_2O_3$/Chloride System에서 Singlet Oxygen이 관여한다는 실험적 증거)

  • The present study was performed to determine oxygen metabolites involved in cell-free MPO/$H_2O_2/Cl^-$ system by observing the effects of their scavengers on NADH oxidation and ethylene production from methional by the action of MPO prepared from human leukocytes. It was clearly demonstrated that NADH was oxidized by the cell-free MPO/$H_2O_2/Cl^-$ system as evidenced by complete inhibition of the oxidation of the substrate in the presence of eiher azide or catalase, or by omitting $Cl^-$. The MPO-mediated oxidation of NADH was completely abolished by a $^1O_2$ quencher, DABCO but not by $OH{\cdot}$ scavengers, mannitol, benzoate, formate and methanol. In ethylene assay, no ethylene was detected from methional in the MPO/$H_2O_2/Cl^-$ system with evident production of the gas by xanthine-oxidase and $Cu^{++}-H_2O_2$ systems which are suggested to generate $OH{\cdot}$. From the results obtained, it is concluded that $^1O_2$ is a major mediator with exclusion of $OH{\cdot}$ involvement in the cell-free MPO-mediated oxidation.
