Improvement of conception rate on Hanwoo; The key hormones and novel estrus detector

  • Two field experiments were conducted to improve the conception rate of Hanwoo cow. The first experiment aimed to investigate the physiological condition of Hanwoo cows on estrus, including metabolic profiles and body condition score (BCS). The second experiment investigated the effect of a novel estrus detector on the artificial insemination (AI) conception rate for Hanwoo cows. For the first experiment, 80 Hanwoo cows (2.5 ± 0.10 of parity), approximately one month before estrus, were housed in 16 pens and offered the experimental diets twice daily with free water access. The BCS were recorded, and blood was collected from the jugular veins just before AI. The collected blood was used to measure physiological conditions, such as metabolite and hormone levels. For the second experiment, each cow was equipped with a neck-mounted estrus detector collar, which had a sensor connected through the internet. Approximately one month before estrus, three hundred sixty Hanwoo cows (2.4 ± 0.21 of parity) were assigned into groups with or without W-Tag collar treatments. The animals were managed the same as in the first experiment. The pregnancy rate reached 55% in the first experiment. The concentration of luteinizing hormone (LH) was higher (p < 0.012; 1.56 vs. 1.08 ng/mL) in cows that were not pregnant (NPG) than in cows that were pregnant (PG) after AI. The BCS and other concentrations of metabolites and hormones in the blood were not different in both NPG and PG cows. The ranges of estrogen, LH, and follicle-stimulating hormone for PG cows were 11.9 to 39.0 pg/mL, < 0.25 to 1.98 ng/mL, and < 0.50 to 0.82 ng/mL, respectively. In the second experiment, cows with the estrus detector had lower days open (p < 0.001; 78.1 vs. 84.8 d), insemination frequency (p < 0.001; 1.26 vs. 2.52), and return of estrus (p < 0.001; 70.9 vs. 79.1 d) than those in cows without the estrus detector. In conclusion, the present study indicated that lower LH concentration just before AI potentially increased the pregnancy rate of Hanwoo cows. Furthermore, the application of estrus detectors to Hanwoo cows could improve the conception success rate for AI.

붉나무 추출물의 이화학적 특성과 아질산염 소거능에 관한 연구 (A Study on the Physicochemical Properties and the Nitrite Scavenging Ability of Japanese Sumac Extracts)

  • 야산에 자생하는 붉나무(Japanese sumac)의 천연자원으로서의 유용성을 확인하기 위하여 잎, 열매 및 수피 등 세부위를 열수로 추출하고, 이들 추출물의 몇 가지 추출특성과 아질산염 소거능 등 생리활성을 측정하였다. 추출물의 수율은 부위에 따라 6.62~13.84%의 범위였고, 유리 아미노산은 잎, 열매, 수피에서 각각 24종 37.9 mg/100g, 23종 27.0 mg/100g 및 27종 39.0 mg/100g의 함량이었으며, 7종의 필수 아미노산이 잎>수피>열매의 순으로 측정되었다. Naringin 상당량으로 구한 총플라보노이드 함량은 수피>열매>잎의 순으로 379.9~587.2 mg/100g이었으며, gallic acid 상당량으로 구한 총페놀성 화합물 함량은 잎>열매>수피의 순으로 80.4~111.2 mg/100g이었다. 붉나무 추출물의 아질산염 소거능은 pH 1.2에서 수피 61.93%>열매 57.38%>잎 55.49%의 순으로 측정되었으며, BHT (butylated hydroxytoluene) 소거능의 78.1~100%에 대응되었다. 추출물의 전자공여능은 열매 47.38%>잎 43.06%>수피 38.55%로서 각각 BHT의 65.6%, 58.8% 및 53.6%의 활성으로 비교되었다. 붉나무의 잎, 열매 및 수피 추출물의 환원력은 대조구로 사용한 BHT의 환원력에 비하여 각각 37%, 43% 및 46% 더 높게 평가되었으며, 금속이온 봉쇄능은 수피 27.3%>잎 20.6%>열매 11.2%의 순으로 상당히 낮게 측정되었다.

3년근 도라지와 7년근 도라지의 이화학 특성 및 생리활성 (Physicochemical properties and biological activity of three-year-old and seven-year-old Platycodon grandiflorum extracts)

  • 3년근 및 7년근 도라지의 이화학적 특성을 확인하고자 조사포닌, 유리 아미노산, 철분, 아연 및 칼슘을 비교 확인하였고, 산화 방지 활성 및 항균활성을 측정하기 위해 물 추출물을 제조하여 총 폴리페놀, 총 플라보노이드, DPPH 및 ABTS 라디칼 소거능 측정 및 호흡기 관련 항균활성을 측정하여 7년근 도라지의 상업적 이용가능성을 확인하고자 수행되었다. 3년근 및 7년근 도라지의 조사포닌을 측정한 결과 재배년수가 증가한 7년근 도라지가 낮은 함량을 보였고, 유리 아미노산은 3년근 및 7년근 도라지 모두 아르기닌, 글루탐산, 아스파트산, 세린, 트레오닌 순으로 높게 나타났으며, 시스테인은 없는 것으로 나타났다. 또한 철, 아연 및 칼슘을 분석한 결과에서는 칼슘만 7년근 도라지가 높게 나타났다. 3년근 및 7년근 도라지 물 추출물의 총 폴리페놀, 총 플라보노이드 함량을 측정한 결과, 재배년수가 높은 7년근 도라지가 총 폴리페놀 및 총 플라보노이드 모두 유의적으로 높게 나타났고, DPPH 및 ABTS 라디칼 소거능을 측정한 결과에서도 7년근 도라지가 높은 소거능을 보였다. 또한 호흡기 질환과 관련된 세균의 항균활성을 측정한 결과에서도 7년근 도라지가 높은 항균활성을 가지는 것으로 나타났다. 결과적으로 3년근 도라지보다 7년근 도라지가 산화방지 활성 및 호흡기 질환과 관련된 항균활성이 높은 것으로 나타나, 7년근 도라지를 기능성 식품 및 화장품 소재로써 이용할 경우 산화방지 활성 및 항균활성이 증가된 천연소재로의 이용 가능성이 높을 것으로 판단하였다.

Meat quality of pork loins from Hereford×Berkshire female and intact male pigs reared in an alternative production system

  • Objective: The objective of the present study was to investigate pork quality from Hereford${\times}$Berkshire female and intact male pigs reared outdoors in an alternative production system. Methods: Berkshire purebred sows were artificially inseminated, once in the fall and again in the spring of the following year, with semen from Hereford boars and managed free of antibiotics in an outdoor hoop structure until the last month of pregnancy, after which they were moved to a pasture-based unit of 0.8 hectares with individual lots with a farrowing hut, shade, and water ad libitum. Piglets were weaned at 4 weeks of age and housed in a deep-bedded hoop structure, grouped by sex. Animals were harvested at market weight of 125 kg, approximately 200 days of age. Hot carcass weight was collected at the time of the harvest. After 24 hours of refrigeration, carcass characteristics were measured. Longissimus dorsi samples collected from the right side loin. Loins were cut into 2.54-cm thick chops and were used to measure marbling score, color score, drip loss, and ultimate pH. Sensory panel tests were conducted as well at North Carolina State University. For pork characteristics and sensory panel data, trial and sex were included in the statistical model as fixed effects. Hot carcass weight was included in the model as a covariate for backfat thickness. Results: Neither the subjective nor the objective color scores displayed any differences between the boars and the gilts. No difference was found for pH and marbling score between trials or sexes. Gilts had a thicker backfat measurement at the last lumbar and a narrower longissimus muscle area measurement when compared to the boars. The only difference in the sensory characteristics was found between the trials for texture and moisture scores. Conclusion: Consumers were not able to detect boar taint under the condition of this study, which is that the intact males were reared outdoors. Additional trials would be necessary; however, based on the results of the present study, outdoor rearing can be suggested as a solution to the issue of boar taint.

Effects of feed intake restriction during late pregnancy on the function, anti-oxidation capability and acute phase protein synthesis of ovine liver

  • Objective: An experiment was conducted to investigate the effects of feed intake restriction during late pregnancy on the function, anti-oxidation capability and acute phase protein synthesis of ovine liver. Methods: Eighteen time-mated ewes with singleton fetuses were allocated to three groups: restricted group 1 (RG1, 0.18 MJ ME/kg $W^{0.75}$ d, n = 6), restricted group 2 (RG2, 0.33 MJ ME/kg $W^{0.75}$ d), n = 6) and a control group (CG, ad libitum, 0.67 MJ ME/kg $W^{0.75}$ d, n = 6). The feed restriction period was from 90 days to 140 days of pregnancy. Results: The ewe's body weight, liver weights, water, and protein content of liver in the restricted groups were reduced compared with the CG group (p<0.05), but the liver fat contents in the RG1 group were higher than those of the CG group (p<0.05). The increased hepatic collagen fibers and reticular fibers were observed in the restricted groups with the reduction of energy intake. The concentrations of nonesterified free fatty acids in the RG1 and RG2 groups were higher than those of the CG group with the reduction of energy intake (p<0.05), but there were decreased concentrations of lipoprotein lipase and hepatic lipase in both restricted groups compared with the CG group (p<0.05). In addition, the increased concentrations of ${\beta}$-hydroxybutyric acid, triglycerides, malondialdehyde, total antioxidant capacity and activities of superoxide dismutase activity and catalase were found in the RG1 group, and the concentrations of cholinesterase in the RG1 group were reduced compared with the CG group (p<0.05). For the concentrations of acute phase proteins, the C-reactive protein (CRP) in the RG1 group were reduced compared with the CG group, but there were no differences in haptoglobin relative to the controls (p>0.05). Conclusion: The fat accumulation, increased hepatic fibrosis, antioxidant imbalance and modified synthesis of acute phase proteins were induced in ewe's liver by maternal malnutrition during late pregnancy, which were detrimental for liver function to accommodate pregnancy.

Comparative Efficacy of Synthetic and Natural Tenderizers on Quality Characteristics of Restructured Spent Hen Meat Slices (RSHS)

  • In the present study, comparative efficacy of natural as well as synthetic tenderizers on the quality characteristics of restructured spent hen meat slices (RSHS) was studied. Four different batches of RSHS viz. Control (without any tenderizer), T1 (1.25% calcium chloride replacing salt in formulation), T2 and T3 (1.5% each of pineapple rind and fig powder, replacing binder in the formulation) were developed in pre-standardized formulation. Vacuum tumbling was performed for 2.5 h and cooked product (RSHS) was assayed for quality attributes. Samples were packaged in aerobic conditions, stored for 21 days under refrigeration ($4{\pm}1^{\circ}C$) and were evaluated for pH, oxidative and microbial quality parameters at regular interval of 7 days. Water holding capacity of T2 was recorded the highest and significantly higher (p<0.05) than all other samples. The textural attributes of T2 were comparable to T1 but significantly higher (p<0.05) than C and T3. The colour attributes ($L^*$, $a^*$, and $b^*$ value) of T2 and T3 were improved due to use of natural tenderizers. During sensory evaluation, tenderness scores for T2 samples were recorded the highest. Throughout storage period, thiobarbituric acid reactive substances (TBARS), free fatty acids (FFA) and peroxide value (PV) followed an increasing trend for control as well as treated products; however, T2 showed a significantly (p<0.05) lower value than control and other treated samples. It can be concluded that good quality RSHS with better storage stability could be prepared by utilizing 1.5% pineapple rind powder as natural tenderizer.

도라지 분획물의 항산화 및 탈모예방 효과 (Evaluation of Antioxidant Fractions and Hair Loss Prevention Effects of Platycodon grandiflorum)

  • 도라지의 탈모예방 효과를 검증하기 위해 몇가지 용매를 이용하여 도라지 분획물을 준비하였다. 산화적 스트레스는 두피혈관을 좁게 하여 모근으로의 영양공급을 방해함으로써 탈모를 유발한다. 본 연구에 사용된 분획물인 BF와 WF는 DPPH 라디칼 소거능의 $IC_{50}$값이 각각 16.2 mg/ml와 121.8 mg/ml로, 모두 농도의존적으로 소거능이 증가하는 것으로 나타났다. 또한 ABTS 라디칼 소거능 실험결과에서도 BF와 WF 처리시 $IC_{50}$값이 각각 4.9 mg/ml와 39.8 mg/ml로 높은 항산화활성을 나타내었다. 또한 인간피부세포인 HaCaT cell 증식 실험결과, 24시간 BF와 WF 처리 시 각각 최대 31%($1{\mu}g/ml$)와 18%($1{\mu}g/ml$)로 HaCaT cell 증식을 촉진시키는 것으로 나타나 추출물이 피부재생효과가 있음을 증명하였다. 탈모의 원인중의 하나인 두피의 염증에 대한 분획물의 효능을 확인하고자, RAW264.7 cell을 이용하여 염증반응 생성물인 NO와 $PGE_2$ 생성정도를 관찰하였다. 그 결과, BF와 WF 각각 최대 88.5%($0.1{\mu}g/ml$)와 88.0%($50{\mu}g/ml$)까지 NO와 $PGE_2$ 생성을 저해하는 것으로 나타났다. 모낭을 구성하는 모유두세포인 HFDPC cell 증식 실험 결과, 4, 48, 72시간 처리 시 모두 HFDPC cell 증식을 농도의존적으로 증가시키는 것으로 나타났다. 이상의 결과를 토대로 본 연구에 사용된 도라지로부터 추출한 부탄올 분획물과 물 분획물이 탈모예방에 효과적이며, 그중에서도 특히 부탄올 분획물이 탈모예방제품의 유용한 천연재료로써의 가치가 있음을 증명하였다.

Gestational Exposure to Pesticides Induces Oxidative Stress and Lipid Peroxidation in Offspring that Persist at Adult Age in an Animal Model

  • Pesticide exposure may induce biochemical alterations including oxidative stress and lipid peroxidation. However, in the context of developmental origin of health and disease, putative trans-generational effect of exposure to pesticides are insufficiently studied. We therefore aimed to evaluate the biochemical effect of gestational exposure to four pesticides on female Wistar rats and their offspring at adult age. We studied 30 female nulliparous Wistar rats divided into 5 equal groups. Group 1 served as the control group and received distilled water while group 2, 3, 4 and 5 received orally pesticide 1 (imidacloprid), pesticide 2 (chlorpyrifos), pesticide 3 (imidacloprid + lambda cyhalothrin) and pesticide 4 (oxamyl) respectively once daily throughout gestation at a dose equivalent to 1/10 lethal dose 50. The mothers were followed up until one month post gestation. The offspring were followed up from birth until adult age (12 weeks). In all animals at each time point we evaluated malondialdehyde (MDA), oxidative stress and liver function enzymes. There was similar variation of total body weight in all the groups during and after gestation. However, Female Wistar rats of the exposed groups had significant alterations in liver SOD (-30.8% to +64.1%), catalase (-38.8% to -85.7%) and GSH (-29.2% to -86.5%) and; kidney catalase (> 100%), GSH (> 100%). Moreover, MDA, alanine transaminase (ALT) and aspartate transaminase (AST) levels were significantly higher in pesticide exposed rats compared to the control group. Similar alterations in antioxidant enzymes, MDA and liver function enzymes were observed in offspring of treated rats evidenced at weaning and persisting until adult age. Exposure to pesticides causes oxidative stress and lipid peroxidation in exposed female Wistar rats and their offspring. The persistence in offspring at adult age suggests transgenerational adverse effects.

비무장지대(DMZ) 자연유산 남북 공동협력의 의의와 과제 (South and North Korean Collaboration for Natural Heritage Conservation across Demilitarized Zone : Its Significance and Challenges)

  • 비무장지대(Demilitarized Zone : DMZ)는 군사분계선(Military Demarcation Line : MDL)을 경계로 남북으로 각각 2km씩을 비무장지대로 설정하기로 협정으로 정했지만 지난 65년 동안 무장이 가득한 지대로 유지되어왔다. 남쪽의 경우 민간인 통제선(Civilian Control Zone : CCZ) 이북 지역과 그 배후의 접경지역은 사람들의 출입이 오랫동안 제한되어 오히려 비무장지대보다 자연이 상대적으로 더 잘 보전되어왔다. 자연유산(natural heritage)은 자연생태계의 보호할 가치가 있는 생물과 그 서식지, 그리고 무생물 자원으로 정의된다. 비무장지대에서는 다양한 생물들과 습지를 비롯한 서식지와 지형들이 있음이 여러 조사를 통해 확인되고 있다. 비무장지대는 완벽하고 보전이 잘 되어 있는 서식 공간은 아니었다. 하지만 한반도에서 그만한 다양성과 규모를 가진 공간이 없고, 향후 관리 방향에 따라 복원이 가능한 최적의 장소로서 의의가 있다. 그러므로 비무장지대와 잘 보호된 일부 민통선 이북 지역을 포함한 그 일원의 서식지 유형(habitat type)을 파악하고 지도를 만들어 관리한다면 한반도에서 멸종 위기에 있는 종들이나 멸종한 종들을 서식하게 하는 최적 장소가 될 것이다. 이때 한강 하구와 동해의 석호(lagoon) 등을 포함하는 것이 필요하다. 비무장지대 일원의 조사와 보호 노력을 남북 공동으로 진행해야 하므로, 특히 비무장지대 내부를 남북 과학자와 국제 전문가를 포함하여 국제 과학조사를 추진하고 장기 계획을 세워 보전과 지속 가능한 이용 방안을 모색하는 것이 필요하다.

운동과 영양 조절이 남성 연령별 체조성 및 비만에 미치는 영향 (Effects of Exercise and Nutrition on Male Body Composition and Obesity)

  • 본 연구는 운동과 영양 조절을 통한 남성 체 구성을 조사하였다. 훈련을 수행하는데 문제가 없는 건강한 40명을 임의로 선정하여 20명에게는 운동과 영양조절 그룹으로 선정하였고, 나머지 20명에게는 운동만 6개월간 수행하도록 하였다. 실험조건 동안 운동강도를 정확하게 측정하기 위해 최대산소 소모량($VO_2$ max)을 측정하였다. 운동과 영양조절일 실시한 20대 남자와 운동만 수행한 남자는 6개월 후 체중이 2.3 kg과 0.4 kg이 감소하였다. 0~3개월 간 실시한 결과에서 체중 측정값에 p값은 0.697이였으며, 3~6개월 측정값의 p값은 0.535로 나타났고, 0~6개월 측정 값의 p값은 0.617로 나타났다. 20대의 골격근은 약간 증가하였다. 30대의 체중은 6개월 후 운동과 영양조절 그룹이 4.0 kg, 운동만 수행한 그룹에서 1.3 kg이 감소하였다. 비록 체중의 감소는 골격근의 증가와 연관성이 있지만 유의성은 없었다. 체중의 변화는 지질 감소와 수분(세포 내 및 세포 간 수분) 감소와 연관이 있었다. 30대를 제외한 20대, 40대, 50대의 체중 감소는 있었지만 유의성은 없었다.
