Effects of Persimmon (Diospros kaki L.) Vinegar as a Dietary Supplement on Feed Intake, Digestibility, and Ruminal Fermentation Indices in Sheep

  • This study estimated the effect of fermented persimmon (Diospros kaki L.) extract (FPE) supplement on feed intake, digestibility, nitrogen (N) balance, and rumen fermentation characteristics in sheep. Five male sheep (Corriedale${\times}$Polwarth) with average body weight of $48.6{\pm}1.3\;kg$ were housed in metabolism crates and assigned to a 5${\times}$5 Latin square design with five consecutive 20-d periods which consisted of 14-d adaptation and 6-d data collection. The sheep were fed ad libitum a diet containing concentrate and rice straw (3:7). The five treatments were FPE supplemented at 0 (Control), 5, 10, 20, and 30 g/kg of concentrate. Intakes of dry matter (DM, p<0.01), organic matter (OM, p<0.01), neutral detergent fiber (NDF, p<0.05), acid detergent fiber (ADF, p<0.05), and nitrogen-free extract (NFE, p<0.01) increased quadratically with increasing intake of FPE supplement and maximized (p<0.05) at 10 g/kg FPE. The digestibilities of DM (p<0.05), OM (p<0.05), crude protein (p<0.01), and NFE (p<0.01) increased quadratically with increasing amount of FPE supplement, and sheep fed 5 and 10 g/kg diets had greater (p<0.05) DM, OM, and NFE digestibilites than the Control treatment. By increasing FPE supplement concentration, N intake (p<0.01) and fecal N (p<0.05) increased linearly, whereas retained N (p<0.05) and retained N ratio (p<0.05) increased quadratically. The retained N was maximized (p<0.05) in sheep fed 5 and 10 g/kg diets. The mean rumen pH was not affected by FPE supplement, but there was a quadratic increase (p<0.05) of mean rumen ammonia N concentration and a linear increase (p<0.01) in mean rumen total volatile fatty acid (VFA) and acetate concentrations. The mean concentration of rumen propionate in sheep fed all FPE supplemented diets was greater (p<0.05) than the Control, but the mean ratios of rumen acetate to propionate in sheep fed 5 and 10 g/kg diets were lower (p<0.05) than that of Control sheep. In conclusion, FPE supplemented at 5-10 g/kg of concentrate improved feed intake, the digestibilites of OM and NFE, N metabolism, and rumen fermentation indices of sheep.

The Effect of a Natural Zeolite (Clinoptilolite) on the Performance of Broiler Chickens and the Quality of Their Litter

  • The objective of this study was to assess the effect of clinoptilolite (a natural zeolite) on growth and performance of broilers as well as on the quality of their litter. A total of 5,200 one-day old broiler chickens (Cobb 500) were used in two consecutive rounds lasting 42 days each. The broilers were given free and continuous access to a nutritionally non-limiting diet (in meal form) that was either a basal diet (B) or a 'zeolite diet' (Z; the basal diet supplemented with clinoptilolite at a level of 2%). A $2{\times}2$ factorial design consisted of two feeding treatments (B and Z) and two bedding types, sawdust (S) and sawdust with zeolite (Sz, which was the result of adding $2kg\;zeolite/m^2$), was used. In each round the broilers were randomly assigned to one of four (n = 650), treatment groups: two fed on the basal diet (B) and had bedding of either sawdust (group BS) or sawdust and zeolite (group BSz) and two fed on the Z diet and had as bedding either sawdust (ZS) or sawdust and zeolite (ZSz). Average growth rates were significantly (p<0.05) different between broilers of different groups; broilers that were fed on the 'zeolite diet' (Z) and were placed either in a compartment with sawdust bedding or sawdust bedding and zeolite (ZS and ZSz) as well those that were fed on the basal diet in a compartment with sawdust bedding and zeolite (BSz) grew at a faster rate (p<0.05) compared with those of the control group (BS). The incorporation of NZ in broilers diets and in their bedding material decreased the organic content in litter samples throughout the experimental period. The lowest organic content was recorded in group ZSz where NZ was added in both feed and litter. Mean ammonia concentration (ppm) was significantly higher in group ZS in comparison to groups BSz and ZSz (27.00 vs. 20.55 and 21.71 respectively). The results of this study showed that the incorporation of the clinoptilolite both in feed and into the litter had a positive effect on broiler growth and also on the quality of their litter.

Growth Performance and Blood Chemistry of Starry Flounder Platichthys stellatus Fed Extruded and Moist Pellet (공급 사료에 따른 강도다리 (Platichthys stellatus)의 성장과 혈액성분의 변화)

  • This study was evaluated individual growth and hematological changes of young starry flounder, using hyperpigmentation marks on the blind side of body after feeding extruded pellet (EP) and moisture pellet (MP) for 45 days. As a result, the fish grew up to $220.1{\pm}47.8g$ in the EP treatment and $224.7{\pm}42.4g$ in the MP treatment, so the weight gain was significantly higher in the MP treatment ($74.6{\pm}17.3%$) than in the EP treatment ($63.7{\pm}16.9%$) (P<0.05). Osmolalities of flounder after 45 days culture were similar between the treatments (P>0.05), showing values of $359.8{\pm}4.2mOsm\;kg^{-1}$ in the EP treatment and $358.5{\pm}6.2mOsm\;kg^{-1}$ in the MP treatment. Similarly, other blood chemistry indicators such as sodium, chloride, total protein, albumin, glucose, total cholesterol, BUN, creatinine, ammonia, triglyceride, cortisol, GOT, GPT and hematocrit showed no differences between the treatments. However, the free fatty acid concentration was significantly higher in the MP treatment than in the EP treatment (P<0.05).

Desalination of Traditional Soy Sauce Using Electrodialysis (전기투석에 의한 재래식 간장의 저염화)

  • Korean traditional soy sauce was desalted using electrodialyzer, and their physicochemical properties were analyzed. The salt content of soy sauce significantly decreased from initial 18.9 (w/v) to 0.47% (w/v) and the volume also decreased to 330 mL when 1,000 mL of soy sauce was desalted for 450 min at a current of 2 A. During the desalting process, the numbers of viable cells of total bacteria, yeasts, and molds increased due to the concentration effect of the electrodialysis. Turbidity increased from 3.1 to 8.5, and total nitrogen, total sugar, and reducing sugar contents also increased. The pH, and amino nitrogen and volatile base nitrogen contents slightly decreased, with ammonia content significantly decreasing from 19.4 to $1.3\;{\mu}mol/mL$. Free amino acids content increased twofold after electrodialysis. Gas chromatograms of soy sauce showed that profiles of the flavoring compounds did not change when electrodialyzed for 360 min, but most were removed after 540 min.

Physicochemical Composition of Baked Garlic (구운 마늘의 이화학적 성분)

  • This study was conducted to investigate the major chemical components of dried baked garlic powder. The proximate compositions of baked garlic powder as a dry-matter basis were 5.30% moisture, 36.89% crude protein, 12.60% crude fat, 4.36% crude ash, 2.88% dietary tiber, and 37.97% carbohydrate, respectively. The major free sugars were identified as fructose and galactose. In the analysis of the total amino acids, 18 kinds of components were isolated from baked garlic powder. The essential amino acid contained in baked garlic powder accounted for 36.60% of total amino acids, while the non-essential amino acids accounted for 63.40%. The major unsaturated fatty acids of total lipids were linoleic acid. The ratio of polyunsaturated fatty acids to saturated fatty acids (P/S ratio) was 5.77. Oxalic acid was the major organic acids. The contents of vitamin A, C, and E were 0.064, 1.452, and 0.037 mg%, respectively. The mineral contents of baked garlic powder were greater, in order of K>Mg>Ca>Na>Zn>Fe. The total polyphenol, flavonoids and thiosulfates contents of baked-garlic ethanol extract were $2.85{\pm}0.05$ mg/100 g, $0.97{\pm}0.04$ mg/100 g and $0.61{\pm}0.02$ OD/g, respectively.

A Study on High Strength Nitrogen Wastewater Treatment and Sludge Granulation Characteristics in a Pilot-scale Air-lift Sequencing Batch Reactor (파일럿 규모의 공기 유동 연속회분반응기에서 고농도 질소제거 및 슬러지 그래뉼화 특성 연구)

  • Selective nitrification and granulation have been carried out in a pilot scale air-lift sequencing batch reactor (SBR) for stable and economical nitrogen removal from wastewater. The SBR showed about 100% nitrification efficiency up to 1.0 kg ${NH_4}^+-N/m^3{\cdot}d$, about 90% efficiency at 1.0-2.0 kg ${NH_4}^+-N/m^3{\cdot}d$, and it was less than 90% when the load was higher than 2.0 kg ${NH_4}^+-N/m^3{\cdot}d$. Nitrite accumulation was induced by selective inhibition of nitrite oxidizing bacteria by free ammonia inhibition and dissolved oxygen limitation. For the purpose, high nitrite ratio (> 0.95) was obtained by keeping the pH higher than 8.0 and dissolved oxygen lower than 1.5 mg/L. In addition, sludge granulation was achieved by keeping reactor settling time to 5 minutes to wash out poor settling sludge and to promote the growth of granulation sludge. The operation accelerated sludge granulation and the sludge volume index (SVI) decreased and stably maintained to less than 75 in 60 days.

Desorption of Water, Ammonia, and Methylamines on $K^+$ Ion Exchanged Zeolite L (칼륨 이온 치환 제올라이트-L 에서 물, 암모니아 및 메틸아민류의 탈착)

  • The potential energy of adsorbate molecules in the main channel of $K^+$ ion exchanged zeolite L(K-L) was calculated. In K-L which adsorbs three molecules per unit cell, the interaction energies of $H_2O,\;NH_3,\;CH_3NH_2,\;(CH_3)_2NH,\;and\;(CH_3)_3N$ molecules with zeolite lattice are 61.11, 62.31, 65.68, 74.65, and 79.88kJ/mol, respectively. These values are less by 3.7∼12.6kJ/mol than $K^+$ ion affinities with adsorbing molecules. These results may be due to the facts that the electrostatic energies are reduced by the negative charge of the lattice oxygens. The distribution of adsorption sites of $NH_3$ and $CH_3NH_2$ in K-L was investigated by a technique of temperature programmed desorption. The experimental value of desorption energies of $NH_3$ and $CH_3NH_2$ on K-L are in good agreement with the theoritical values. It is concluded that the desorption of $NH_3$ and $CH_3NH_2$ on K-L is the first-order desorption with free readsorption.

Effect of Dietary Supplementation of Two Tonic Formula on the Forced-Swimming Capacity of Rats (자양강장용 기능성 식품이 흰쥐의 강제수영 능력에 미치는 영향)

  • Effects of two types of formula designed for commercial product on forced-swimming capacity and related biochemical parameters of Sprague-Dawley rats were evaluated. Treatment groups were fed diet supplemented with 4.56% formula A (EFA: exercise-trained and formula A-supplemented group) or B (EFB: exercise-trained and formula B-supplemented group) for 4 weeks. All groups were regularly trained twice a week to be accustomed to swimming. After 4 weeks, each group was divided into two subgroups: 90 min swimming (90-min subgroups) and swimming until exhaustion (all-out subgroups). Serum was collected, and skeletal muscles were dissected out after swimming. Forced-swimming time until exhaustion was significantly longer (p<0.05) in EFA and EFB groups than that of exercised control group. Among 90-min subgroups, serum ammonia level of EFB group was significantly lower than that of exercised control group (p<0.05). Among all-out subgroups, serum lactate levels were not significantly different among exercised control and treatment groups, whereas serum-free fatty acid levels of EFA and EFB groups were significantly lower than that of exercised control group (p<0.05). In conclusion, EFA and EFB groups enhanced forced swimming capacity of rats by increasing fat utilization and delaying accumulation of serum fatigue factor.

Effect of Animal-Welfare Environment on the Metabolomic Properties of Breast and Thigh Meat from Two Broiler Strains (동물복지 사육환경이 두 육계 품종의 가슴육 및 다리육의 대사체학적 특성에 미치는 효과)

  • This study investigates the metabolomic changes in breast and thigh meat from Cobb and Ross 308 chickens regarding the rearing environment. One-day-old Cobb and Ross broilers were raised for 35 days in conventional and animal welfare farms with, amongst others, different floor sizes, stock densities, and ammonia concentrations. One-dimensional 1H nuclear magnetic resonance, orthogonal partial least squares-discriminant analysis (OPLS-DA), and pathway analyses were performed to analyze the metabolomic properties of broiler meat. For breast meat, only those from the Ross strain could be separated according to the environment in the OPLS-DA plot. Ross breast meat from animal welfare farms showed significantly higher acetate, anserine, creatine, and inosine monophosphate content than those from conventional farms (P<0.05). In contrast, for thigh meat, the Cobb strain was differentiated using OPLS-DA. The contents of five metabolites, such as glucose and lactate, were higher in thigh meat from animal welfare farms; however, nine metabolites, including seven free amino acids, were lower compared to those from conventional farms (P<0.05). Pathway analysis was performed to interpret the biological changes in chicken meat based on environmental factors. The results indicated that the animal welfare environment led to significant changes in four metabolic pathways in Ross breast meat and in 20 metabolic pathways in Cobb thigh meat (P<0.05). In conclusion, the animal welfare environment could influence the metabolomic properties of Ross breast meat and Cobb thigh meat, which may affect the sensory quality of meat.

A Study on The Content of Liver Protein, Nucleic Acids, and Guanine Deaminase Activity of Mouse During Acute Starvation (급성(急性) 기아(饑餓)마우스의 간단백질(肝蛋白質), 핵산(核酸) 및 Guanine Deaminase 활성(活性)에 관(關)한 연구(硏究))

  • Number of aspects, not only nutritional but social as well as political involved in human starvation pose nowadays global problems. In order to help establish the minimum nutritional requirements in the daily life of a man and to free people as well from either undernourishment, malnutrition or even starvation many workers have devoted themselves so far on the research programs to know what and how number of metabolic events take place in animals in vivo. It is the purpose of the present paper to examine in effect to what extent both of the protein and nucleic acids (DNA & RNA) together with an enzyme, guanine deaminase, which converts guanine into xanthine and in turn ends up to uric acid as an end product, undergo changes, quantitatively during acute starvation, using the mouse as an experimental animal. The mouse was strictly inhibited from taking foods except drinking water ad libitum and was sacriflced 24, 48, and 72 hours following starvation thus acutely induced. The animals consisted of two experimental groups, one control and another starvation groups, each being consisted of 6-24 mice of whose body weights ranged in the vicinity of 10 g. The animals were sacriflced by a blow on the head, followed by immediate excision of their livers into ice-cold distilled water, washing adherent blood and other contaminant tissues. The liver was minced foramin, by an all-glass homogenizer immersing it in an ice-bath, followed by subsequent fractionatin of the homogenate (10% W/V in 0.25M sucrose solution made up with 0.05M phosphate buffer of pH 7.4). For the liver protein and guanine deaminase assay, the 10% homogenate was centrifuged at 600 x g for 10 minutes to eliminate the nuclear fraction; and for the estimation of DNA and RNA, the homogenate was prepared by the addition of 10% trichloroacetic acid in order to free the homogenate from the acid-soluble fraction, the remaining residue being delipidate by the addition of alcohol and dried in vacuo for later KOH (IN) hydrolysis. The changes in body and liver wegihts during acute starvation were checked gravimetrically. Protein contents in the liver were monitored by the method of Lowry et al; and guanine deaminase activities were followed by the assay of liberated ammonia from the substrate utilizing the Caraway's colorimetry. The extraction of both DNA and RNA was performed by the Schmidt-Thannhauser's method, which was followed by Marmur's method of purification for DNA and by Chargaff's method of purification for RNA. The determinations of both DNA and RNA were carried out by the diphenylamine reaction for the former and by the orcinol reaction for the latter. The following resume was the results of the present work. 1. It was observed that the body as well as liver weights fall abruptly during starvation, and that the loss of body weight showed no statistical correlation with the decreases in the content of liver protein. 2. The content of liver protein and activity of liver guanine deaminase activity as well decline dramatically, and the specific activities of the enzyme (activity/protein), however, decreased gradually as starvation proceeded. 3. Both of the nucleic acids, DNA and RNA, showed decrements in the liver of mouse during acute starvation; the latter, however, being more striking in the decline as compared to the former. 4. The decreases in the liver protein content as resulted from the acute starvation had no statistically significant correlation with the decrements of DNA in the same tissue, but had regressed with a significant statistical correlation with the fall of RNA in the tissue. 5. The decrease in the activity of guanine deaminase in the liver of mouse during acute starvation was functionally more proportional to the decrease in RNA than DNA, and moreover correlated with the changes in the content of the liver protein. 6. The possible mechanisms involved during in this acute starvation as bring the decreases in the contents of DNA, protein, and guanine deaminase were discussed briefly.
