  • A thoredoxin (CTRX) gene was cloned and characterized from a full length cDNA library prepared from taproot of three-year old Codonopsis lanceolata. A CTRX was 666 nucleotides long and has an open reading frame of 372 bp with 124 amino acid residues (pI = 4.92). The deduced amino acid sequence of the CTRX matched to the previously reported plant thioredoxin h genes. The deduced amino acid sequence of CTRX exhibited the similarity of 33-67% among previously registered thioredoxin genes. The expression of CTRX in leaves of Codonopsis lanceolata was increased by wounding and 1 mM $H_2O_2$, but decreased by 0.1 mM cadmium.

  • A class I type 2 metallothionein (CMet2) cDNA from taproot of Codonopsis lanceolata was isolated and characterized. A CMet2 cDNA was 572 nucleotides long and had an open reading frame of 234 bp with a deduced amino acid sequence of 78 residues (pI = 4.99). The deduced amino acid sequence of CMet2 matched to the previously reported type 2 metallothionein-like protein genes and showed 74% identity with that of G. max (BAD18377) and C. arietinum (CAA65009). Expression of CMet2 by the RT-PCR was increased at 1 hr after cadmium and hydrogen peroxide treatment, respectively.

  • The stochastic finite element method(SFEM) based structural optimal design is presented. Random system response including uncertainties for the design variable is calculated with first order perturbation method. A method for calculating the sensitivity coefficients is developed using the equilibrium equation and first-order perturbed equation. Numerical results are presented for a truss, frame and plate structures with displacement and stress constraints. The sensitivity calculation proposed here is compared with finite difference method. A nonlinear programming technique is used to solve the problem. The procedure is easily incorporated with existing deterministic structural optimization.

  • An integrated framework of modeling the human operator cognitive behavior during nuclear power plant accident scenarios is presented. It incorporates both plant and operator models. The basic structure of the operator model is similar to that of existing cognitive models, however, this model differs from those existing ones largely in too aspects. First, using frame and membership function, the pattern matching behavior, which is identified as the dominant cognitive process of operators responding to an accident sequence, is explicitly implemented in this model. Second, the non-task-related human cognitive activities like effect of stress and cognitive biases such as confirmation bias and availability bias, are also considered. A computer code, OPEC is assembled to simulate this framework and is actually applied to an SGTR sequence, and the resultant simulated behaviors of operator are obtained.

  • In this paper, LS-SynRM (Line Start-Synchronous Reluctance Motor) has been attracting attention in replace of induction motor which hardly provides high efficiency. Compared to induction motor, LS-SynRM has better efficiency per unit area. This study demonstrated the electromagnetic design methods of LS-SynRM while maintaining the frame of existing IE3 induction motor for blower. We documented the design procedures for generating high saliency which is the most essential and mechanical stress analysis is also treated. In conclusion, we proved the validity of our design by manufacturing and testing our LS-SynRM models.

  • A base isolation system is proposed for earthquake protection of structures. The system incorporates spherical supports for the base, a specially designed spring-cam system to keep the base rigidly supported under normal condition and to allow it to move for the duration of the earthquake under the constraint of a spring with optimized non-linear characteristics. A single-story model is constructed to investigate the feasibility of the concept. Numerical simulations of the system as well as experimental results show that 95% reduction of the transmitted force to the structure can be achieved. To demonstrate the effectiveness of this isolation mechanism, the maximum dynamic bending stress developed at predetermined critical points within the frame of the structure is measured. Significant reduction of the dynamic stresses is obtained.

  • Given a body undergoing a stress-strain status as consequence of external excitations, sensors can be deployed on the accessible lateral surface of the body. The sensor readings are regarded as input of a numerical model of reduced order (i.e., the number of sensors is lower than the number of the state variables the full model would require). The goal is to locate the sensors in such a way to minimize the deviations from the response of the true (full) model. One adopts either accelerometers as sensors or devices reading relative displacements. Two applications are studied: a plane frame is first investigated; the focus is eventually on a 3D body.

  • The seismic performance of six types of weak-axis steel moment connections was investigated through cyclic testing of six full-scale specimens. These weak-axis moment connections were the column-tree type, WUF-B type, FF-W type, WFP type, BFP-B type and DST type weak-axis connections. The testing results showed that each of these weak-axis connection types achieved excellent seismic performance, except the WFP and the WUF-B types. The WFP and WUF-B connections displayed poor seismic performance because a fracture appeared prematurely at the weld joint due to stress concentrations. The column-tree type connection showed the best seismic behavior such that the story drift ratio could reach 5%.

  • The critical load of a continuous compression member was determined by the beam-analogy method. The proposed method utilizes the stress-analysis results of the analogous continuous beam, where imaginary concentrated lateral load changing its direction is applied at each midspan. The proposed method gives a lower bound error of critical load and can predict the span that buckles first. The effective length factors for braced frame columns can be easily determined by the present method, but result in the upper bound errors in all cases, which can lead to a conservative structural design.

  • The objectives of this study is intended to analyze stresses using the boundary element method and probability analysis for agricultural structure. Loads and material properties are an important factor when analyzing the structure. Until now, designing structure, loads and material properties are applied deterministic value. However, load and material properties involve uncertainties due to those change probabilistic and deterministic methods could not consider uncertainties. To solve these problems, the reliability analysis based on probability properties scheme was developed. Reliability analysis is easy to approach to analysis frame structure, however it has limitation when solving plane stress strain problems a kind of agricultural structures. The BEM (Boundary Element Method) is able to analysis plane strain problems by boundary conditions. Thus, this study applied boundary element method to analysis plane strain problem, load and material properties as a probabilistic value to calculate the analytical model using Monte Carlo simulations were developed.
