Separation of large DNA molecules by pulsed field gel electrophoresis (역전장 전기영동장치를 이용한 대형 DNA 분리에 관한 연구)

  • Gel electrophoresis has proven to be one of the most useful of DNA separation and purification. The new technique of pulsed field gel electrophoresis (PFGE) is high resolution separation of large size DNA moleculs. Conventional continuous gel electrophresis can not be separation of large DNA fragments(20~50 k base). Field inversion gel electrophoresis(FIGE) is very useful for large DNA molecules. We have found that a pulse ratio ; 2 : 1, time ; 24hrs., volts ; $10^{volts}/_{cm}$, start ; 0.45sec, end ; 1sec, is most effectively resolves DNA fragment in the 6~50k base.

Effects of Disulfide and Thioether Linkages on Stability and Cytotoxicity of Anti-CALLA Fab-Ricin A Immunotoxins

  • Anti-CALLA Fab-RTA immunotoxins were constructed using two crosslinking agent, SMPT and SMCC, to generate a disulfide and a thioether bridge between Fab fragment of K269-65 MoAb and RTA toxin moieties, respectively. These immunotoxins were selectively immunoreative with CALLA$\^$+/ B-lineage Daudi cells. SMPT and SMCC mediated RTA immunotoxins were prepared with 49% and 53% of the RTA conjugation yields, respectively. SDS-PAGE analysis show that immunotoxins were constructed with major Fab-1 RTA (76kda), minor Fab-2RTA (106kda) and Fab-3RTA (136kda) compositions. The breakdown rates of immunotoxins were determined in the presence of glutathione by measuring the amount of reduced immunotoxins using size-exclusion HPLC. The SMCC immunotoxins were more resistant to the glutathione than SMPT immunotoxins. But, our data showed that the SMPT mediated disulfide bonded immunotoxins were much more active than the SMCC mediated thioether bonded immunotoxins to kill the target cells in vitro.

  • This paper proposes a consistency algorithm that is able to serve streaming data efficiently in VOD system. The media data is stripping into several pieces of data by the Round Robin method in order to media data service. The barrier mechanism is changed into the minimum data factor(SH. GOP) in this paper. The shared memory is allocated at one host with one fragment size. Data is combined with RTP packet transmission data format using barrier mechanism. I experiment and program the suggested algorithm on the VOD system.

  • A genomic DNA of alkaliphilic bacterium, Micrococcus sp. Y-l, was analysed using the physical mapping method of pulsed-field gel electrophoresis (PFGE). Five restriction enzymes of Sspl, Hpal, Xbal, Ndel or EcoRI, which recognize the Adenine-Thymine-rich sequences of genomic DNA, were used for the generation of few (7 to 20) distinctly separate fragments, with average sizes in the range of 200~500 kb. However, the sites for Notl and SfiI, 8 base-recognizing enzymes, were highly frequent. The genome size of this strain was determined to be 4 mega base pairs (Mb) from restriction fragments separated by PFGE. This is the first case of restriction mapping in alkaliphilic bacterium.

First Record of Callochiton foveolatus (Polyplacophora: Callochitonidae) from Korea

  • The genus Callochiton Gray, 1847 is small to medium sized chiton (up to $55{\times}36mm$ in size) and includes approximately 30 species worldwide, most of which are found on hard substrata in subtidal zone. To date, only three species of Callochiton Gray, 1847 have been reported in Japan. In this study, we found C. foveolatus(Is. Taki, 1938) as first record of the family Callochitonidae Plate, 1901 in Korea and its morphological features were described and compared with other northwestern Pacific species. In addition, the partial fragment of mitochondrial DNA cytochrome c oxidase subunit I sequences of C. foveolatus was determined as DNA barcoding record and compared with other congeneric species.

  • The nucleic acid of Synechococcus sp. cyanophage was identified as double-stranded DNA by the result of digestion with enzymes such as exonucleases, DNase, and S1 nuclease, and by acridine orange staining. The cyanophage DNA was cleaved with several restriciton ehdonucleases such as ApaI, BamHI, Bg/II, HaeIII, Eco RI, HindIII, PstI, AND aPAI gave the clearest sets of bands on agarose gels and the fragment numbers for each were 12, 20, 29, 20, and 7, respectively. The sums of the size from Bam HI and PstI digestions were estimated approximately 227$\pm$4 kb, which are in agreement with the result of the pulsed field gel electrphoresis. This virus is thought to have the largest genosome among those of known cyanophages, which corresponds to the largest haed ot 90 nm when compared with the head sizes of cyanophages discovered since 1963.

  • Natural functional compound in the textile finishing for health and amenity using fragrant material have been applied by microencapsulation method. The microcapsules containing fragrant materials as functional compound were produced by in situ polymerization using urea-formaldehyde prepolymer. The average diameter of microcapsules is 2.75$\mu$ and particle size ranges over 0.5~10$\mu$. Fragment material is extracted approximatly proportioned from microcapsule at room temperature. The adsorption of microcapsule was improved by pretreatment of cationic agent. Fragrant materials in microcapsule was revealed to have long release time.

  • In this study, the spalling and ultrasonic pulse transmission time of concrete were investigated according to compressive strength during heating. As a result, the higher the compressive strength of the concrete, the more the explosion occurs, which affects the cross-sectional loss and the spalling fragment size. Also, ultrasonic pulse transmission time was found to be strongly influenced by the section loss of concrete.

  • Using recombinant DNA technology, the vector system containing minimal fragment of a bacterial hemoglobin gene (vgb) was constructed. When this vector was inserted into Escherichia coli, the growth of the host was significantly improved in both viable cell counts and absorbance measurement, compared to that of the wild type strain. In addition, by minimizing the size of bacterial hemoglobin in the vector, the ability of vgb in growth improvement was augmented, due to the reduction of metabolic burden from the maintenance and replication of the plasmid. By using this system, it is expected that the growth of microorganisms can be improved even in the hypoxic condition.

  • The molecular interaction of $W_2(NH_2)_4Cl_4$ fragment with chlorine ligands has been studied by means of extended Huckel calculations. We have extended the Huckel calculation to unknown edge-sharing $W_2(NH_2)_4Cl_4({\eta}-X)_2$ complexes (X = CO, H) in order to compare the stability of the complexes. The calculations showed that the size and electronic property of bridged ligand are important in determining the stability. The stabilities of the related metal-metal bonded edge-sharing complexes are discussed.
