Optimization of ${\gamma}-Polyglutamic$ Acid Production by Bacillus subtilis ZJU-7 Using a Surface-response Methodology

  • The components of the media used to elicit the biosynthesis of $poly-{\gamma}-glutamic$ acid $({\gamma}-PGA)$ by Bacillus subtilis ZJU-7 were investigated, particularly the carbon and nitrogen sources Of the 7 carbon sources investigated, sucrose induced the highest rate of ${\gamma}-PGA$ productivity; among the nitrogen sources, tryptone had the best effect for ${\gamma}-PGA$ production. A $2^{6-2}$ fractional factorial design was used to screen factors that influence ${\gamma}-PGA$ production significantly, and a central composite design was finally adopted to formulate the optimal medium. ${\gamma}-PGA$ productivity improved approximately 2-fold when the optimal medium was used compared with the original nonoptimized medium, and volumetric productivity reached a maximum of 58.2 g/L after a 24-h cultivation period.

  • Performances of multiple surrogate models are evaluated in a turbomachinery blade shape optimization. The basic models, i.e., Response Surface Approximation, Kriging and Radial Basis Neural Network models as well as weighted average models are tested for shape optimization. Global data based errors for each surrogates are used to calculate the weights. These weights are multiplied with the respective surrogates to get the final weighted average models. The design points are selected using three level fractional factorial D-optimal designs. The present approach can help address the multi-objective design on a rational basis with quantifiable cost-benefit analysis.

  • This paper presents operating characteristics of internal heat exchanger(IHX) for $CO_2$ geothermal heat pump in the heating mode. Mass flow rate of $CO_2$, inlet temperatures of $CO_2$ at high and low pressure side were selected as main effect factors by using fractional factorial DOE(Design of Experiments). And RSM(Response Surface Method) was used in optimization phase. The results show that heat transfer rate of IHX increases when either inlet temperature of low pressure side decreases or inlet temperature of high pressure side increases. Effectiveness of IHX increases with increasing of inlet temperature of either high pressure side or low pressure side. Finally, performance contour map was provided over the operation ranges of the main design factors.

혼합물 실험계획법을 이용한 이차전지의 최적설계 (An Optimum Design of Secondary Battery using Design of Experiments with Mixture)

  • Secondary batteries with high performance are essential in widespread use of modern portable devices such as cellular phones and laptop computers. High energy density, long cycle life, and safety are some of important requirements for secondary battery. To achieve such characteristics, a mixing proportion of electrolyte solution ingredients in the battery should be carefully chosen. In this paper, using statistical design of mixture experiments (DOME), we attempt to find an optimum condition of designing the secondary battery. DOME has a distinct feature in that the experimental region is represented by simplex, rather than hypercube, because the sum of blend proportions should be unity. Several designs based upon this point have been proposed for mixture experiments. Among them, an extreme vertices design is employed in this paper because there are a couple of blend constraints to be considered. In order to investigate how the mixing proportion interacts with other manufacturing factors, a fractional factorial design is also included across the extreme vertices design. As a result, we find that the blend proportion of solution ingredients has a significant effect on battery performances. By simultaneously optimizing two battery capacities, this paper proposes an optimum blend proportion according to process factor settings.

대두 단백질 및 배아를 이용한 Bifidobacterium lactis BL740의 균체성장 및 이소플라본 비배당체 생산를 위한 통계적 배지 최적화 (Statistical optimization of culture media contained soy proteins and hypocotyl for the growth of Bifidobacterium lactis BL 740 and production of soy isoflavone aglycones)

  • 대두가공공정 중 발생하는 부산물인 대두배아 및 대두 단백질을 이용하여 Bifidobacterium lactis BL740의 생균수 및 대두 이소플라본 비배당체 생산 최대화를 위한 배지최적화를 수행하였고 이를 위하여 통계적 방법인 fractional factorial design (FFD) 및 central composite design(CCD)를 이용, 대두 유래 성분들이 포함된 배지 (S-medium)의 최적 조성물을 확언하였다. FFD의 경우 glucose, cellobiose, fructooligosaccharidde, soy peptone, soy protein, 대두배아를 이용 총 6가지 요소를 2수준에서 적용하였으며 이 중 soy protein, soy peptone 그리고 대두 배아가 B. lactis BL740의 균성장 혹은 대두 이소플라본 비배당체 전환에 중요 인자로 확인이 되었다. FFD에서 확인된 3가지 인자를 CCD에 적용하였으며 이를 통해 균성장의 경우 최적의 배지조성함량이 soy peptone 29.55 g/L, soy protein 12.73 g/L, 대두 배아 130.67 g/L로 확인되었으며 비배당체 전환의 경우 soy peptone 1.25 g/L, soy protein 2.06 g/L, 대두 배아 60.02 g/L로 최적화 되었다.

Optimization of Medium Composition for Growth of Leuconostoc mesenteroides

  • The MRS medium is widely used as an optimized medium for the growth of Lactobacillus spp. and also used for the growth of Leuconostoc spp. Leuconostoc mesenteroides shows quite different physicochemical properties compared to Lactobacilli spp. and it is one of the major strain of kimchi fermenting microorganisms with its usefulness in our traditional foods and availability in biotechnology in the future, specifically tailor-made medium is necessary for the growth of Leuconostoc mesenteroides. Sequential experimental designs (Plackett-Burman, fractional factorial, steepest ascent, central composite design and response surface methodology) were introduced to optimize and improve the Leuconostoc medium. Fifteen medium ingredients were investigated and fructose, sodium acetate and ammonium citrate were determined to give a critical and positive effect for cell-growth. The yield of biomass using the optimal medium was improved more than that of the MRS medium and the result of fed-batch culture showed the capability of the improvement in cell mass similar to the E.coli system.

컨조인트 분석을 통한 새만금 관광개발 전략에 관한 연구 (A Study on the Semangum Tourism Development Strategy Using Conjoint Analysis)

  • This study aims to develop Semangum tourism package that can be most suitable for the tourist. The data were obtained by a survey carried out between August, 17 and August, 24, 2009 with a sample of latent tourist of Semangum. Conjoint analysis has been used to estimate the relative importance of attributes. Four main attributes and two or three attributes levels for each determinant were chosen for conjoint analysis. Eleven model package were made using fractional factorial design, and presented to the latent tourist, who were asked to reply in order of their like. Results show that the most important attributes for tourist were in order of cost, connected tourism, accommodation and report activity.

Satistical Analysis of SiO2 Contact Hole Etching in a Magnetically Enhanced Reactive Ion Etching Reactor

  • Plasma etching of $SiO_2$ contact holes was statistically analyzed by a fractional factorial experimental design. The analysis revealed the dependence of the etch rate and DC self-bias voltage on the input factors of the magnetically enhanced reactive ion etching reactor, including gas pressure, magnetic field, and the gas flow rates of $CHF_3$, $CF_4$, and Ar. Empirical models of the DC self-bias voltage and etch rate were obtained. The DC self-bias voltage was found to be determined mainly by the operating pressure and the magnetic field, and the etch rate was related mainly to the pressure and the flow rates of Ar and $CHF_3$.

  • Mahalanobis Taguchi System (MTS) is a pattern information technology, which has been used in different diagnostic applications to make quantitative decisions by constructing a multivariate measurement scale using data analytic methods without any assumption regarding statistical distribution. The MTS performs Taguchi's fractional factorial design based on the Mahahlanobis Distance (MS) as a performance metric. In this work, MTS is used for analyzing Wisconsin Breast Cancer data which has ten attributes. Ten different tests are conducted for the data to determine if the patient has cancer or not. Also, MTS is used for reducing the number of test to define the relationship between each attribute and diagnosis result. The accuracy of diagnosis is compare with two different previous research.
