Inhibition of HBV ]Replication by the Extract of Phyllanthus ussuriensis (여우주머니(Phyllanthus ussuriensis) 추출물의 B헝 간염바이러스 증식 억제)

  • Abstract -Phyllanthus ussuriensis which belongs to Euphorbiaceae has been used as a component of Korean traditional remedy against hepatitis and jaundice. Aqueous methanolic extract of P. ussuriensis was tested for antiviral activity of hepatitis B virus (HBV) in HepG2 2.2.15 cells which were derived through transfection of cloned HBV DNA into HepG2 human hrpatoblastoma cells. P. ussuriensis extract decreased the levels of extracellular HBV virion DNA and inhibited HBV replication at concentrations ranging from 50 to 500$\mu$g/ml. Partitioning of water suspension of the extract revealed that the activity mainly resides in the EtOAc fraction. Consecutive purification of the EtOAc fraction by silica-gel column chromatography resulted in the two active anti-HBV fractions. Southern blot analysis shows that the action mechanisms or active site of the two fractions seems to be different in terms of their inhibition of HBV replication steps.

Identification of Anticoagulant Components in Korean Red Ginseng

  • In this study, the anticoagulant compounds in Korean red ginseng (KRG) were investigated. KRG powder was extracted using hot methanol, and the methanol extract was fractionated into n-hexane, ethylacetate, n-butanol, and aqueous fractions by solvent partitioning. The remains from the methanol extraction were further extracted with water and then dialyzed to obtain low and high molecular weight fractions. The anticoagulant activities of the seven fractions were evaluated in terms of thrombin time, prothrombin time, and activated partial thromboplastin time. Among these fractions, the ethylacetate fraction showed the most potent anticoagulant activity. The active components in the ethylacetate fraction were identified as the phenolic compounds vanillic, caffeic, ferulic, and p-coumaric acid via TLC and HPLC. These findings suggest that the anticoagulant activities of phenolic compounds contribute to the cardiovascular effects of KRG.

Attenuation of Brain Injury by Water Extract of Goat's-beard (Aruncus dioicus) and Its Ethyl Acetate Fraction in a Rat Model of Ischemia-Reperfusion

  • Ischemic stroke constitutes about 80% of all stroke incidences. It is characterized by brain cell death in a region where cerebral arteries supplying blood are occluded. Under these ischemic conditions, apoptosis is responsible for the cell death, at least in part. Goat's-beard (Aruncus dioicus var. kamtschaticus) is a perennial plant that grows naturally in the alpine regions of Korea. In the present study, we first determined whether water extract of goat's-beard (HY1646) and some of its fractions prepared by partitioning with organic solvents could improve the viability of human hepatocellular carcinoma cells (HepG2) cultured under hypoxic condition by blocking apoptotic pathways. Based on the in vitro findings, we subsequently investigated whether HY1646 and the ethyl acetate fraction (EA) selected from cell culture-based screening could attenuate brain injury in a rat middle cerebral artery occlusion (MCAO) model of ischemia (2 hr), followed by 22 hours of reperfusion. The cell number was sustained close to that initially plated in the presence of HY1646 even after 24 hr of cell culture under hypoxic condition (3% $O_2$), at which time the cell number reached almost zero in the absence of HY1646. This improvement in cell viability was attributed to the delay in apoptosis, identified by the formation of DNA ladder in gel electrophoresis. Of fractions soluble in hexane, ethyl acetate (EA) and butanol, EA was chosen for the animal experiments because EA demonstrated the best cell viability at the lowest concentration (10 ${\mu}g$/mL). HY1646 (200 mg/kg) and EA (10 and 20 mg/kg) significantly reduced infarct size, an index of brain injury, by 16.6, 40.0 and 61.0%, respectively, as assessed by 2,3,5-triphenyl tetrazolium chloride staining. The findings suggest that prophylactic intake of goat's beard might be beneficial for preventing ischemic stroke.

Influence of Annealing Temperature on Microstructure and Pitting Corrosion Behavior of the 27Cr-7Ni Hyper Duplex Stainless Steel

  • Influence of annealing temperature on the microstructure and resistance to pitting corrosion of the hyper duplex stainless steel was investigated in acid and neutral chloride environments. The pitting corrosion resistance is strongly dependent on the microstructure, especially the presence of chromium nitrides ($Cr_2N$), elemental partitioning behavior and volume fraction of ferrite phase and austenite phase. Precipitation of deleterious chromium nitrides reduces the resistance to pitting corrosion due to the formation of Cr-depleted zone. The difference of PREN (Pitting Resistance Equivalent Number) values between the ferrite and austenite phases was the smallest when solution heat-treated at $1060^{\circ}C$. Based on the results of electrochemical tests and critical crevice temperature tests, the optimal annealing temperature is determined as $1060^{\circ}C$.

  • Sediments were collected from eight stations in JJJJinhae Bay and heavy metals were fractionated into the adsoibed reducible, oxidizable and residual fractions. Cd, Cu, Pb and Zn in each fraction were determined by atomic absorption spectriphotmetry. Cd was shown to be mainly in the adsorbed form, Cu in the oxidizable and resedual fractions, and Pb and Zn mainly in the seducible fractions. The total concentrations of Cu, Pb and Zn in sediments decreased gradually with increasing distance from the heak of the bay, and the relationship of linear segression was obtained.

Plant Molecular Farming Using Oleosin Partitioning Technology in Oilseeds

  • Plant seed oil-bodies or oleosomes ate the repository of the neutral lipid stored in seeds. These organelles in many oilseeds may comprise half of the total cellular volume. Oleosomes are surrounded by a half-unit membrane of phospholipid into which are embedded proteins called oleosins. Oleosins are present at high density on the oil-body surface and after storage proteins comprise the most abundant proteins in oilseeds. Oleosins are specifically targeted and anchored to oil-bodies after co-translation on the ER. It has been shown that the amino-acid sequences responsible for this unique targeting reside primarily in the central hydrophobic tore of the oleosin polypeptide. In addition, a signal-like sequence is found near the junction of the hydrophobic domain and ann N-terminal hydrophilic / amphipathic domain. This "signal" which is uncleaved is also essential for correct targeting. Oil-bodies and their associated oleosins may be recovered by floatation centrifugation of aqueous seed extracts. This simple partitioning step results in a dramatic enrichment for oleosins in the oil-body fraction. In the light of these properties, we reasoned that it would be feasible to create fusion proteins on oil-bodies comprising oleosins and an additional valuable protein of pharmaceutical or industrial interest. It was further postulated that if these proteins were displayed on the outer surface of oil-bodies, it would be possible to release them from the purified oil-bodies using chemical or proteolytic cleavage. This could result in a simple means of recovering high-value protein from seeds at a significant (i.e. commercial) scale. This procedure has been successfully reduced to practice for a wide variety of proteins of therapeutic, industrial and food no. The utillity of the method will be discussed using a blood anticoagulant, hirudin, and industrial enzymes as key examples.

Photofragment Translational Spectroscopy of CH₂I₂ at 304 nm: Polarization Dependence and Energy Partitioning

  • The photodissociation dynamics of CH2I2 has been studied at 304 nm by state-selective photofragment translational spectroscopy. Velocity distributions, anisotropy parameters, and relative quantum yields are obtained for the ground I(2P3/2) and spin-orbit excited state I*(2P1/2) iodine atoms, which are produced from photodissociation of CH2I2 at this wavelength. These processes are found to occur via B1 ← A1 type electronic transitions. The quantum yield of I*(2P1/2) is determined to be 0.25, indicating that the formation of ground state iodine is clearly the favored dissociation channel in the 304 nm wavelength region. From the angular distribution of dissociation products, the anisotropy parameters are determined to be β(I)=0.4 for the I(2P3/2) and β(I*)=0.55 for the I*(2P1/2) which substantially differ from the limiting value of 1.13. The positive values of anisotropy parameter, however, show that the primary processes for I and I* formation channels proceed dominantly via a transition which is parallel to I-I axis. The above results are interpreted in terms of dual path formation of iodine atoms from two different excited states, i.e., a direct and an indirect dissociation via curve crossing between these states. The translational energy distributions of recoil fragments reveal that a large fraction of the available energy goes into the internal excitation of the CH2I photofragment; < Eint > /Eavl=0.80 and 0.82 for the I and I* formation channels, respectively. The quantitative analysis for the energy partitioning of available energy into the photofragments is used to compare the experimental results with the prediction of direct impulsive model for photodissociation dynamics.

Study on Adsorption Characteristics of Perfluorinated Compounds(PFCs) with Structural Properties (과불화화합물 구조적 속성에 따른 흡착 특성 연구)

  • Perfluorinated compounds(PFCs), an emerging environmental pollutant, are environmentally persistent and bioaccumulative organic compounds that possess a toxic impact on human health and ecosystems. PFCs are distributed widely in environment media including groundwater, surface water, soil and sediment. PFCs in contaminated solid can potentially leach into groundwater. Therefore, understanding PFCs partitioning between the aqueous phase and solid phase is important for the determination of their fate and transport in the environment. In this study, the sorption equilibrium batch and kinetic experiment of PFCs were carried out to estimated the sorption coefficient(Kd) and the fraction between aqueous-solid phase partition, respectively. Sorption branches of the PFDA(Perfluoro-n-decanoic acid), PFNA(Perfluoro-n-nonanoic acid), PFOA(Perfluoro-n-octanoic acid), PFOS(Perfluoro-1-octane sulfonic acid) and PFHxS(Perfluoro-1-hexane sulfonic acid) isotherms were nearly linear, and the estimated Kd was as follow: PFDA(1.50) > PFOS(1.49) > PFNA(0.81) > PFHxS(0.45) > PFOA(0.39). The sorption kinetics of PFDA, PFNA, PFOA, PFOS and PFHxS onto soil were described by a biexponential adsorption model, suggesting that a fast transport into the surface layer of soil, followed by two-step diffusion transport into the internal water and/or organic matter of soil. Shorter times(<20hr) were required to achieve equilibrium and fraction for adsorption on solid(F1, F2) increased with perfluorinated carbon chain length and sulfonate compounds in this study. Overall, our results suggested that not only the perfluorocarbon chain length, but also the terminal functional groups are important contributors to electrostatic and hydrophobic interactions between PFCs and soils, and organic matter in soils significantly affects adsorption maximum capacity than kinetic rate.

Spatial Partitioning for Query Result Size Estimation in Spatial Databases (공간 데이터베이스에서 질의 결과 크기 추정을 위한 공간 분할)

  • The query optimizer's important task while a query is invoked is to estimate the fraction of records in the databases that satisfy the given query condition. The query result size estimation in spatial databases, like relational databases, proceeds to partition the whole input into a small number of subsets called “buckets” and then estimate the fraction of the input in the buckets. The accuracy of estimation is determined by the difference between the real data counts and approximations in the buckets, and is dependent on how to partition the buckets. Existing techniques for spatial databases are equi-area and equi-count techniques, which are respectively analogous in relation databases to equi-height histogram that divides the input value range into buckets of equal size and equi-depth histogram that is equal to the number of records within each bucket. In this paper we propose a new partitioning technique that determines buckets according to the maximal difference of area which is defined as the product of data ranges End frequencies of input. In this new technique we consider both data values and frequencies of input data simultaneously, and thus achieve substantial improvements in accuracy over existing approaches. We present a detailed experimental study of the accuracy of query result size estimation comparing the proposed technique and the existing techniques using synthetic as well as real-life datasets. Experiments confirm that our proposed techniques offer better accuracy in query result size estimation than the existing techniques for space query size, bucket number, data number and data size.

Exploring fraction knowledge of the stage 3 students in proportion problem solving (단위 조정 3단계 학생의 비례 문제 해결에서 나타나는 분수 지식)

  • The purpose of this study is to explore how students' fractional knowledge is related to their solving of proportion problems. To this end, 28 clinical interviews with four middle-grade students, each lasting about 30~50 minutes, were carried out from May 2021 to August 2021. The present study focuses on two 7th grade students who exhibited their ability to coordinate three levels of units prior to solving whole number problems. Although the students showed interiorization of three levels of units in solving whole number problems, how they coordinated three levels of units were different in solving proportion problems depending on whether the problems required reasoning with whole numbers or fractions. The students could coordinate three levels of units prior to solving the problems involving whole numbers, they coordinated three levels of units in activity for the problems involving fractions. In particular, the ways the two students employed partitioning operations and how they coordinated quantitative unit structures were different in solving proportion problems involving improper fractions. The study contributes to the field by adding empirical data corroborating the hypotheses that students' ability to transform one three levels of units structure into another one may not only be related to their interiorization of recursive partitioning operations, but it is an important foundation for their construction of splitting operations for composite units.
