Expression of fox-related genes in the skin follicles of Inner Mongolia cashmere goat

  • Objective: This study investigated the expression of genes in cashmere goats at different periods of their fetal development. Methods: Bioinformatics analysis was used to evaluate data obtained by transcriptome sequencing of fetus skin samples collected from Inner Mongolia cashmere goats on days 45, 55, and 65 of fetal age. Results: We found that FoxN1, FoxE1, and FoxI3 genes of the Fox gene family were probably involved in the growth and development of the follicle and the formation of hair, which is consistent with previous findings. Real-time quantitative polymerase chain reaction detecting system and Western blot analysis were employed to study the relative differentially expressed genes FoxN1, FoxE1, and FoxI3 in the body skin of cashmere goat fetuses and adult individuals. Conclusion: This study provided new fundamental information for further investigation of the genes related to follicle development and exploration of their roles in hair follicle initiation, growth, and development.

Transgenic Efficiency of FoxN1-targeted Pig Parthenogenetic Embryos

  • The clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR associated protein (Cas9) system can be applied to produce transgenic pigs. Therefore, we applied CRISPR/Cas9 system to generate FoxN1-targeted pig parthenogenetic embryos. Using single guided RNA targeted to pig FoxN1 genes was injected into cytoplasm of in vitro matured oocyte before electrical activation. In results, regardless of the concentrations of vector, the cleavage rate were significantly (p<0.05) decreased ($4ng/{\mu}l$, 51.24%; $8ng/{\mu}l$, 40.88%; and $16ng/{\mu}l$; 45.22%) compared to no injection group (70.44%). The blastocyst formation rates were also decreased in vector injected 3 groups ($4ng/{\mu}l$, 7.96%; $8ng/{\mu}l$, 6.4%; and $16ng/{\mu}l$; 9.04%) compared to no injection group (29.07%). In addition, the blastocyst formation rates between sham injected group (13.51%) and no injection group (29.07%) also showed significant difference (p<0.05). The mutation rates were comparable between groups ($4ng/{\mu}l$, 18.4%; $8ng/{\mu}l$, 12.5%; and $16ng/{\mu}l$; 20.0%). The sequencing analysis showed that blastocysts derived from each group were successfully mutated in FoxN1 loci regardless of the vector concentrations. However, the deletion patterns were higher than the patterns of point mutation and insertion regardless of the vector concentrations. In conclusion, we described that cytoplasmic microinjection of FoxN1-targeted CRISPR/Cas9 vector could efficiently generate transgenic pig parthenogenetic embryos in one-step.

Effect of caffeine on genes expressions of developing retinas in the chick model

  • It has been reported that overconsumption of caffeine during pregnancy leads to a deleterious effect within the nervous tissues during embryonic development. In this study, we further extrapolated the effect of caffeine in the developing retinas, which is known to be one of the most sensitive tissues in chick embryos. Morphological changes of retinal thickness and organization of neuroretinal epithelium were monitored using three gene markers, Atoh7, FoxN4, and Lim1. Upon treating with a single dose of caffeine (15 µmol at embryonic day 1 [E1]), relative thicknesses of developing retinas (particularly of E7 and E9) were significantly altered. Among the three genes studied, the expression pattern of Atoh7 was notably altered while those of FoxN4, and Lim1 mRNA showed only a slight change in these developing retinas. Quantitative polymerase chain reaction results supported the most notable changes of Atoh7 but not FoxN4, and Lim1 gene in the developing retinas, particularly at E7. The effect of caffeine towards other organs during development should be extrapolated and the awareness of its intensive consumption should be raised.

Structural Characterization and Thermal Behavior of a Novel Energetic Material: 1-Amino-1-(2,4-dinitrophenylhydrazinyl)-2,2-dinitroethylene

  • A novel energetic material, 1-amino-1-(2,4-dinitrophenylhydrazinyl)-2,2-dinitroethylene (APHDNE), was synthesized by the reaction of 1,1-diamino-2,2-dinitroethylene (FOX-7) and 2,4-dinitrophenylhydrazine in N-methyl pyrrolidone (NMP) at 110 $^{\circ}C$. The theoretical investigation on APHDNE was curried out by B3LYP/6-311+$G^*$ method. The IR frequencies analysis and NMR chemical shifts were performed and compared with the experimental results. The thermal behavior of APHDNE was studied by DSC and TG/DTG methods, and can be divided into two crystal phase transition processes and three exothermic decomposition processes. The enthalpy, apparent activation energy and pre-exponential factor of the first exothermic decomposition reaction were obtained as -525.3 kJ $mol^{-1}$, 276.85 kJ $mol^{-1}$ and $10^{26.22}s^{-1}$, respectively. The critical temperature of thermal explosion of APHDNE is 237.7 $^{\circ}C$. The specific heat capacity of APHDNE was determined with micro-DSC method and theoretical calculation method, and the molar heat capacity is 363.67 J $mol^{-1}K^{-1}$ at 298.15 K. The adiabatic time-to-explosion of APHDNE was also calculated to be a certain value between 253.2-309.4 s. APHDNE has higher thermal stability than FOX-7.

장기간의 지구성 운동 또는 저항성 운동이 중년 흰쥐의 골격근 내 단백질 동화 및 이화기전에 미치는 영향 (The effects of long term endurance or resistance exercise training on anabolic and catabolic pathway in skeletal muscle of middle-aged rats)

  • 본 연구의 목적은 장기간의 지구성 운동 또는 저항성 운동이 중년 흰쥐의 골격근 내 단백질 합성과 분해에 미치는 영향을 규명하는 것이다. 50 주령의 male Wistar rat 30 마리를 이용하여 3집단(좌업, 지구성 운동집단, 저항성 운동집단)으로 무선배정한 후 12 주간 처치를 실시하였다. 연구결과 12주간의 저항성운동은 중년 쥐의 족저근 내 Akt/mTOR 신호전달체계를 활성화시켰고, FoxO1/MuRF1 단백질 발현을 저해하였다. 지구성 운동은 mTOR 신호전달체계에는 영향을 미치지 않았으나 FoxO1/MuRF1 단백질 발현을 저해하였고, AMPK/PGC-1α 발현을 증가시켰다. 요약하면 12주간의 운동트레이닝은 중년 흰쥐의 골격근 내 단백질 동화/이화반응에 긍정적인 영향을 미쳤으며, 지구성 운동은 근단백질 이화반응의 저해를 통해서 노화근육의 단백질 균형에 영향을 미치는 것으로 나타났다. 이러한 결과를 통해서 근 손실에 따른 근 기능의 저하가 가시화되기 시작하는 중년을 대상으로 실시되는 운동처방 시 근 감소증 예방 및 치료를 위해 저항성 운동만이 아니라 중강도 이상의 지구성 운동도 유효할 것으로 생각된다.

  • Making use of the multidimensional fractional calculus operators introduced in [20], this paper gives the images under these operators of the celebrated Fox's H-function. Special cases are briefly point out, and some of the results are also studied on general spaces of functions $M_{\lambda}(R_{+}^n)$.

실외 번식시설에서 여우의 번식 행동 (Breeding Behavior of Red Fox (Vulpes vulpes) in an Outdoor Breeding Facility)

  • 발정기, 교배 기간 및 발정기를 포함하는 교미 중 붉은 여우 (Vulpes vulpes)의 행동을 실외 번식시설에서 환경 요인과 함께 조사하였다. 여우의 평균 교배 지속시간은 19.95분(n = 13, 범위 = 1.17-35.25분)이었다. 번식기는 1년 미만 여우의 경우, 3월 중순(60.0%)이었고, 1년 이상 여우는 2월 초순(56.6%)으로 나타났다. 교미 지속시간은 암 수컷 모두가 1세 이상 일 때 가장 길었고($24.4{\pm}11.08$분), 교배는 나이와는 관계없이 이루어졌다. 여우는 발정기를 시작한 후 2일 이내에 두 번 교배하고 두 마리의 수컷과 교배한 암컷의 임신율이 100%였다. 또한, 짝짓기 개체 간 연령이 1년 이상 된 경우 임신율이 87%로 가장 높았다. 여우는 야간보다는 주간 교미를 선호했고 대체로 맑은 날과 부분적으로 흐린 날의 10시부터 12시 사이에 교미를 했다. 수컷 여우는 다른 암컷과 최대 5번 교미했으며, 수컷의 교미 빈도가 높아질수록 교미 지속시간이 늘어났다. 교미에 대한 관심은 수컷은 3회, 암컷은 2회 교미 후 줄어들었다. 수컷은 첫 번째와 두 번째의 교미 사이에 최소 4시간 46분의 휴식이 필요했다. 이 연구에서 우리는 여우의 복원에 있어 개체 확보를 위한 번식 프로그램에 적용 가능한 기본적인 자료를 수집하였다.

  • In this paper we proposed a modified reliability function for improving bimodal speaker identification(BSI) performance. The convectional reliability function, used by N. Fox[1], is extended by introducing an optimization factor. We evaluated the proposed method in BSI domain. A BSI system was implemented based on GMM and it was tested using VidTIMIT database. Through speaker identification experiments we verified the usefulness of our proposed method. The experiments showed the improved performance, i.e., the reduction of error rate by 39%.
