환경요인을 이용한 제주도 토양예측의 정량적 연구 (Quantitative Approach of Soil Prediction using Environment Factors in Jeju Island)

  • 토양의 생성에는 모재, 기후, 지형, 생물요인, 시간 등의 5가지 요인이 작용하고 있다고 알려져 왔다. 이 연구는 제주도 토양을 대상으로 환경요인만으로 토양통을 예측하는 알고리즘을 이용하여, 정량적인 분석 방법을 이용해 환경요인과 토양분포의 관계를 알아보고자 하였다. 분석을 위한 환경요인으로는 온도, 강수량, 지표지질, 해발고도, 경사도, 경사향, 주변과의 고도차, 지형습윤지수, 해안으로부터의 거리, 산정상으로부터의 거리, 토지이용현황을 선택하였다. 토양통별로 각각의 환경요인의 평균과 표준편차 또는 면적비를 구하고, 이를 이용하여 토양통이 분포할 수 있는 환경요인의 범위와 확률을 구하였다. 환경요인별 확률을 결합시켜 분포 가능한 토양통의 순위를 제시하는 알고리즘을 작성하였고, 이를 이용하여 토양통을 예측한 결과 1순위 내에 33%, 2순위 내에 62%, 5순위 내에 74%를 정확하게 기존의 토양도와 일치시킬 수 있었다. 알고리즘을 이용하여 제주 전 지역의 환경요인을 이용하여 예측되는 토양도를 토양목, 대군 등으로 분류하 여 나타낸 결과 북부 해안가에 Entisols이 분포하고 해발고도가 높아지면서 Alfisols, Andisols이 나타난 반면, 서부지역에서는 해안가의 Alfisols에서 해발고도가 높아지면서 Ultisols, Andisols의 순으로 나타나고 있어, 토양의 생성과 관련되어 모형을 이용한 추가적인 연구가 필요하였다.

정책형성단계에서 생태계서비스에 관한 이론적 고찰 (A Study on the Contexts of Ecosystem Services in the Policymaking Process)

  • Industrialization and modernization of the $20^{th}$ century have brought convenience to human life. However, such development produced unintentional global environmental issues such as destruction of natural ecosystem through excessive use of the natural environment beyond its capacity. The United National (UN) recognized such global environment issues and coordinated with worldwide experts to conduct a comprehensive assessment of the global ecosystem, "Millennium Ecosystem Assessment (MA)", to resolve the issues. The UN conducted the MA primarily to comprehend the correlation between the ecosystem and human welfare. The results demonstrated that the ecosystem provides various direct and indirect benefits to human. This new concept was introduced as the "ecosystem services". Introduction of the ecosystem services highlighted the significance of the ecosystem and led to increased research and active discussions among experts from various discipline over the recent 10 years. It has been considered that the ecosystem service concept could be incorporated in policymaking process, as primary criteria to assess and weigh the benefits of using natural resources against the effects on value of the ecosystem. The current policymaking process is affected by the contradicting logics of modernization and preservation of ecosystem. The ecosystem services concept could be the answer to forming a rational policymaking process, allowing for nature preservation while wisely using the available natural resources. The general consensus has been formed on the need of incorporating the ecosystem services concept in the policies and globally various researches on the ecosystem services have been actively conducted. However, introduction of the ecosystem services concept is still at a very early stage and research on real application of the ecosystem services is yet almost nonexistent. Also there are no clear and agreed definition, classification scheme and value appraisal methodology regarding the ecosystem services. Therefore, this research explains the basic concept of the ecosystem services and highlights specific issues for further researches, as further researches on real application of the ecosystem services would find ways for the ecosystem services approach to be valuable in the policymaking process.

피페리딘계 이온성 액체와 포스페이트 염으로 구성된 수상이성분계를 이용한 숙신산의 추출 (Liquid Extraction of Succinic Acid by Aqueous Two-Phase Systems Composed of Piperidinium Ionic Liquids and Phosphate Salt)

  • 본 연구에서는 숙신산 추출을 위한 효과적인 분리공정으로서 피페리딘계 이온성 액체/$K_2HPO_4$에 의한 수상이성분계를 적용하여 수상이성분계 형성 특성 및 추출특성을 고찰하였다. 실험 결과 $K_2HPO_4$ 수용액에 적정량의 피페리딘계 이온성 액체를 첨가함에 따라 안정한 수상이성분계가 형성됨을 확인할 수 있었으며 이성분계 형성능력은 이온성 액체 내 양이온의 알킬 사슬길이에 따라 증가하였다. $K_2HPO_4$ 수용액을 기준으로 피페리딘계 이온성 액체의 수상이성분계 형성 능력은 이미다졸계나 피롤리딘계에 비해 우수하였다. 피페리딘계 이온성 액체를 이용한 수상이성분계 추출에 있어 숙신산 추출 효율은 75~95%의 범위를 가지며 이 값들은 이미다졸계나 피롤리딘계와 큰 차이가 없었다. 그러므로 피페리딘계 이온성 액체를 이용한 수상이성분계를 숙신산 추출에 적용할 경우 이미다졸계 및 피롤리딘계에 비해 적은 양의 이온성 액체를 사용하여 높은 추출효율을 얻을 수 있음을 확인할 수 있었다.

축산물 브랜드에 대한 고객기반브랜드자산이 브랜드 신뢰, 고객만족도 및 재구매의도에 미치는 영향 - 횡성한우를 중심으로 - (The Effects of Customer-Based Brand Equity(CBBE) for Stock Farm Products Brand on Brand Trust, Customer Satisfaction and Repurchase Intentions - Focus on Hoengsung Hanwoo Brand -)

  • 본 연구에서는 횡성한우를 중심으로 축산물 브랜드의 고객기반브랜드자산이 브랜드 신뢰와 고객만족도 및 재구매의도에 미치는 인과관계를 고찰하였다. 실증연구를 위해 확보된 301개의 표본을 바탕으로 연구모형의 신뢰성, 적합성 등을 검토하였고, 구조방정식 모형을 사용하여 총 5개의 가설을 검증하였으며, 모형의 적합도는 ${\chi}^2=635.175$(p<0.001), df=233, CMIN/DF=2.726, GFI=0.863, NFI=0.859, CFI=0.903, TLI=0.885, RMSEA=0.068로 조사되었다. 연구결과, 고객기반브랜드자산의 하위 차원인 지각된 품질(${\beta}=.562$), 브랜드 이미지(${\beta}=.377$)는 브랜드 신뢰에 유의한 영향을 주는 것으로 조사되었으며, 이러한 브랜드 신뢰는 고객만족도(${\beta}=.525$)와 재구매의도(${\beta}=.669$)에 유의한 영향을 주는 것으로 나타났다. 따라서 본 결과를 통해 확고한 브랜드관리는 브랜드 신뢰를 형성하게 되어 결과적으로 고객만족도와 재구매 의도에 긍정적인 영향을 미치는 것을 확인할 수 있었다. 아울러 지역브랜드 한우를 지속적으로 구매하기 위한 효과적인 마케팅 관리의 전략 수립과 함께 지역경제의 활성화를 위한 시사점을 제시하였다.

Palladium 촉매를 이용한 5-Norbornene-2-carboxylic Acid Esters의 부가 중합: 단량체의 합성, 단량체의 Stereochemistry(Endo-, Exo-이성질체)가 고분자의 중합 거동에 미치는 영향 (Addition Polymerization of 5-Norbornene-2-carboxylic Acid Esters Using Palladium Catalyst System: Synthesis of Monomers, Effect of Their Stereochemistry on Polymerization Behavior)

  • Palladium(II) 촉매를 이용한 norobornene carboxylic acid estsers의 중합 시 단량체의 알킬 작용기의 종류, endo/exo 비율이 촉매의 활성도 및 중합 특성에 미치는 영향을 조사하였다. Norbornene esters 단량체는 5-norborene-2-carboxylic acid와 다양한 알코올을 반응시켜 합성하였다. 중합 촉매로는 di-$\mu$-chloro-bis(-methoxybicyclo[2,2,1]-hept-2-ene)palladium(II)(DCBMP)를 합성하여 사용하였고, 짝음이온으로 실버 헥사플루오로안티모네이트($AgSbF_6$)를 이용하였다. 고분자의 분석을 위해 젤 투과 크로마토그래피(gel permeation chromatography, GPC), 열 중량 분석법(thermogravimetric analysis, TGA), 시차주사 열량측정법(differential scanning calorimetry, DSC), 화학구조 분석을 위해 $^1H$ NMR spectroscopy를 이용하였다. 분자량 분석 결과 모든 작용기의 경우 endo-이성체의 비율이 exo-이성체의 비율보다 높을 경우 촉매의 구조적 방해로 인하여 반응성이 감소됨을 보였다. 또한 단량체와 촉매의 비율이 중합 거동에 미치는 영향을 조사하기 위해 단량체와 촉매의 몰비율을 100:1, 200:1, 300:1로 변화시켜 실험을 진행하였으며, 이 때 exo-norbornene carboxylic acid octyl ester의 경우 300:1 촉매비에서 필름형성이 가능한 높은 분자량($M_n=27500g/mol$)의 고분자를 합성할 수 있었다.

EH36-TM강의 대입열 EGW 용접부 저온 인성에 미치는 미세 조직의 영향 (Effects of Microstructures on the Toughness of High Heat Input EG Welded Joint of EH36-TM Steel)

  • The characteristics of high heat input (342kJ/cm) EG (Electro Gas Arc) welded joint of EH36-TM steel has been investigated. The weld metal microstructure consisted of fine acicular ferrite (AF), a little volume of polygonal ferrite (PF) and grain boundary ferrite (GBF). Charpy impact test results of the weld metal and heat affected zone (HAZ) met the requirement of classification rule (Min. 34J at $-20^{\circ}C$). In order to evaluate the relationship between the impact toughness property and the grain size of HAZ, the austenite grain size of HAZ was measured. The prior austenite grain size in Fusion line (F.L+0.1 mm) was about $350{\mu}m$. The grain size in F.L+1.5 mm was measured to be less than $30{\mu}m$ and this region was identified as being included in FGHAZ(Fine Grain HAZ). It is seen that as the austenite grain size decreases, the size of GBF, FSP (Ferrite Side Plate) become smaller and the impact toughness of HAZ increases. Therefore, the CGHAZ was considered to be area up to 1.3mm away from the fusion line. Results of TEM replica analysis for a welded joint implied that very small size ($0.8\sim1.2{\mu}m$) oxygen inclusions played a role of forming fine acicular ferrite in the weld metal. A large amount of (Ti, Mn, Al)xOy oxygen inclusions dispersed, and oxides density was measured to be 4,600-5,300 (ea/mm2). During the welding thermal cycle, the area near a fusion line was reheated to temperature exceeding $1400^{\circ}C$. However, the nitrides and carbides were not completely dissolved near the fusion line because of rapid heating and cooling rate. Instead, they might grow during the cooling process. TiC precipitates of about 50 ~ 100nm size dispersed near the fusion line.

Fermentation Quality of Italian Ryegrass (Lolium multiflorum Lam.) Silages Treated with Encapsulated-glucose, Glucose, Sorbic Acid and Pre-fermented Juices

  • This experiment was carried out to evaluate the effects of adding encapsulated-glucose, glucose, sorbic acid or prefermented juice of epiphytic lactic acid bacteria (FJLB) on the fermentation quality and residual mono- and disaccharide composition of Italian ryegrass (Lolium multiflorum Lam) silages. The additive treatments were as follows: (1) control (no addition), (2) encapsulated-glucose addition at 0.5% for glucose, (3) glucose addition at 1%, (4) sorbic acid addition at 0.1%, (5) FJLB addition at a theoretical application rate of $2.67{\times}10^5$ CFU (colony forming unit) $g^{-1}$, on a fresh weight basis of Italian ryegrass. Although control and encapsulated-glucose treatments had higher contents of butyric acid (33.45, 21.50 g $kg^{-1}$ DM) and ammonia-N/Total nitrogen (114.91, 87.01 g $kg^{-1}$) as compared with the other treated silages, the fermentation in all silages was clearly dominated by lactic acid. This was well indicated by the low pH (4.38-3.59), and high lactic acid/acetic acid (4.39-22.97) and lactic acid content (46.85-121.76 g $kg^{-1}$ DM). Encapsulated-0.5% glucose and glucose addition increased lactic acid/acetic acid, and significantly (p<0.05) decreased ammonia-N/total nitrogen, and the contents of butyric acid and total volatile fatty acids (VFAs) as compared with the control. However, there were higher butyric acid and lower residual mono-and di-saccharides on the two treatments as compared with sorbic acid and FJLB addition, and their utilization efficiency of water soluble carbohydrates (WSC) was lower than that of both sorbic acid and FJLB additions. Sorbic acid addition showed the lowest content of ethanol and ammonia-N/total nitrogen, and the highest content of residual fructose and total mono-and disaccharides as well as the higher lactic acid/acetic acid value. Sorbic acid addition decreased the loss of mono-and disaccharides, and inhibited the activity of clostridial and other undesirable bacteria, and greatly increased the utilization efficiency of fermentable substrates by epiphytic LAB. FJLB addition had the lowest pH value and the highest lactic acid content among all additive treatments, with the most intensive lactic acid fermentation occurring in FJLB treated silage. This resulted in the faster accumulation of lactic acid and faster pH reduction. Sorbic acid and FJLB addition depressed clostridia or other undesirable bacterial fermentation which decreased the WSC loss and saved the fermentable substrate for lactic acid fermentation.

백작약 조다당분획에 의한 B 세포 증식의 특성 (Characteristics of B cell proliferation by polysaccharide fraction of Paeonia japonica miyabe)

  • Background : Paeonia japonica Miyabe is a medicinal plant which has been widely used as a component of blood-building decoctions (Chinese medicinal concept : Bu-Xie). The immunopharmacological characteristics of the extract of Paeonia japonica (PJ) were investigated. Methods : The effects of fractions of PJ extract on lymphocyte proliferation were measured by $H^3$-thymidine incorporation assay. The proliferated lymphocyte subsets were analyzed in flow cytometry. The subset cell populations of spleen cells were separated by magnetic cell separation system, and their proliferation by the extract were investigated. The effect of the extract on antibody production was determined in mice challenged with sheep red blood cells (SRBC) using hemolytic plaque forming cell assay. Results : Spleen cells were proliferated by water extract of PJ. Polysaccharide fraction (PJ-P) of the extract was most active in the proliferation. It was found in flow cytometry that the lymphocyte subset proliferated by PJ-P was B cell population. Among the separated subset cell populations, T cell-depleted cell population and macrophage-depleted cell population were most proliferated by PJ-P. However, positively selected populations of B cells and T cells were not proliferated by PJ-P. These results indicate that B cell proliferation by PJ-P may require the assistance of macrophages or T cells. These results suggest that firstly PJ-P may stimulate macrophages or T cells, and then B cells are activated. The number of antibody-secreting cells was increased by administration of PJ-P in mice immunized with SRBC as a T-dependent antigen. Conclusion : These results suggest that macrophages and accessory cells are directly activated by PJ-P and then helper T cells and B cells are indirectly activated. As the results, immune responses might be coordinately improved. In conclusion, PJ-P, a polysaccharide of P. japonica, may be a characteristic immunostimulator, which is analogous to polysaccharides such as lentinan, PSK and ginsan.

Overlapping Region of p53/Wrap53 Transcripts: Mutational Analysis and Sequence Similarity with microRNA-4732-5p

  • Background: Although the majority of investigations concerned with TP53 and its protein have focused on coding regions, recently a set of studies highlighted significant roles of regulatory elements located in p53 mRNA, especially 5'UTR. The wrap53${\alpha}$ transcript is one of those that acts as a natural antisense agent, forming RNA-RNA hybrids with p53 mRNA and protecting it from degradation. Materials and Methods: In this study, we focused on the mutation status of exon $1{\alpha}$ of the WRAP53 gene (according to exon 1 of p53) in 160 breast tumor tissue samples and conducted a bioinformatics search for probable miRNA binding site in the p53/wrap53 overlapping region. Mutations were detected, using single stranded conformation polymorphism (SSCP) and sequencing. We applied the miRBase database for prediction of miRNAs which target overlapping region of p53/wrap53 transcripts. Results: Our results showed all samples to have wild type alleles in exon 1 of TP53 gene. We could detect a novel and unreported intronic mutation (IVS1+56, G>C) outside overlapping regions of p53/wrap53 genes in breast cancer tissues and also predict the presence of a binding site for miR-4732-5p in the 5'UTR of Wrap53 mRNA. Conclusions: From our findings we propose designing further studies focused on overexpression of miRNA-4732-5p and introducing different mutations in the overlapping region of wrap53 and p53 genes in order to study their effects on p53 and its ${\Delta}N$ isoform (${\Delta}$40p53) expression. The results may provide new pieces in the p53 targeting puzzle for cancer therapy.

중간크기 탄소사슬의 지방산으로 이루어진 인지질에 의한 tolaasin의 용혈활성 촉진 (Facilitation of tolaasin-induced hemolysis by phospholipids composed of medium-chain fatty acids)

  • Tolaasin은 Pseudomonas tolaasii에 의해 생성되어 pore를 형성하는 펩티드 독소이며, 인공재배 버섯의 막 구조를 파괴하여 갈반병을 일으킨다. Tolaasin이 막에서 pore를 형성하는 기작이나 특성은 자세히 알려지지 않았으나, 인공 지질이중막에서 tolaasin에 의한 pore 형성은 제시되었다. Tolaasin에 의한 지질막에서의 pore 형성은 드물게 나타났고, 형성된 pore는 불안정하기에 tolaasin pore의 길이와 지질막의 두께가 서로 일치하지 않을 수 있음이 제안되었다. 그러므로, 탄소수가 다른 지방산으로 이루어진 인지질들을 첨가하여 tolaasin에 의한 용혈활성 변화를 측정하였다. 두 개의 decanoic acids (C10:0, 1,2-didecanoyl-sn-glycero-3-phosphoethanolamine; DDPE)와 myristic acids (C14:0, 1,2-dimyristoyl-sn-glycero-3-phosphoethanolamine), stearic acids(C18:0, 1,2-distearoyl-sn-glycero-3-phosphoethanolamine)로 이루어진 phosphatidylethanolamine들을 적혈구와 tolaasin 펩티드가 포함된 반응용액에 가했을 때, DDPE만이 tolaasin에 의한 용혈활성을 촉진하였으며, 나머지 두 인지질은 효과를 보이지 않았다. DDPE를 다양한 농도로 처리하였을 때, tolaasin에 의한 용혈활성은 농도의존적으로 증가하였다. 중간길이의 지방산으로 구성된 인지질은 tolaasin pore 구조와 막지질 사이에 결합하여 pore 주변의 막을 얇게 함으로써 tolaasin pore를 안정화시킬 것으로 여겨진다. 본 연구의 결과는 중간 크기의 지방산으로 구성된 인지질이 tolaasin pore를 막 구조에서 안정화시킴으로써 활성을 증가시키며, 이것은 적혈구 막에서 tolaasin pore의 길이가 막의 두께보다 조금 짧을 것이라는 사실을 제안한다.
