Comparison of Methanol with Formamide on Extraction of Nitrogen Heterocyclic Compounds Contained in Model Coal Tar Fraction (모델 콜타르 유분 중에 함유된 질소고리화합물의 추출에 관한 메탄올과 포름아마이드의 비교)

  • The separation of nitrogen heterocyclic compound (NHC) contained in a model coal tar fraction was compared by the methanol and formamide extraction. The model coal tar fraction comprising four kinds of NHC (NHCs : quinoline, iso-quinoline, indole, quinaldine) and three kinds of bicyclic aromatic compound (BACs : 1-methylnaphthalene, 2-methylnaphthalene, dimethylnaphthalene), biphenyl and phenyl ether was used as a raw material. The aqueous solution of methanol and formamide were used as solvents. A batch-stirred tank was used as the raw material - a solvent contact unit of this work. Independent of the solvent used, the distribution coefficient of NHCs sharply increased by decreasing the initial volume ratio of water to the solvent and increasing the equilibrium operation temperature, whereas, the selectivity of NHCs in reference to BACs decreased. Decreasing the initial volume ratio of solvent to feed resulted in deteriorating distribution coefficients, but the selectivity of NHCs in reference to BAC was almost the constant. The distribution coefficient of NHCs by the methanol extraction was 3~5 times higher than that of NHCs by the formamide extraction, inversely, the selectivity of NHCs based on BACs by the formamide extraction was 3~7 times higher than that of NHCs by the methanol extraction. Furthermore, two different solvent extraction methods by adding the extraction processing speed to the balance between solvency and selectivity of NHCs were compared.

  • We have developed a direct polymerase chain reaction (PCR) and electrophoresis method for the diagnosis of bovine theileriosis from whole blood DNA analysis without DNA extraction. The technique empolyed a FoLT (formamide, low temp.) technique and was utilized in the diagnosis of bovine theileriosis. Formamide solubilize the blood cells, and the lowered incubation temperatures reduced protein coagulation. 100-200 nL of whole blood and PCR reagents were introduced directly into a PCR tube. After the amplification, the PCR product (816-bp DNA) was introduced into the electrophoresis system. The results of this analysis were consist with those obtained using purified DNA.

Effect of Experimental Factors on Reduction of Nitrogen Compounds Contained in Crude Methylnaphthalene Oil by Formamide Extraction (포름아미드 추출에 의한 조제 메틸나프탈렌유에 함유된 질소화합물의 저감에 관한 실험인자의 영향)

  • The crude methylnaphthalene oil (CMNO) contains nitrogen compounds (NCs) such as quinoline (QU), isoquinoline (IQU), and indole (IN). These NCs in the CMNO are treated as impurities contained in the CMNO due to contamination of the atmospheric environment and unpleasant odors. In order to improve the quality of CMNO, this study examined the effect of extraction experimental factors on the reduction of NCs contained in CMNO using CMNO as a raw material and an aqueous formamide solution as a solvent, respectively. The increase in the volume ratio of solvent to feed in initial (S/F)0 in initial increased the distribution coefficient of NCs and the selectivity of NCs in reference to 2-methylnaphthalene (2MNA). Additionally, an increase in operating temperature (T) increased the distribution coefficient of NCs but conversely decreased selectivity. The compositions of QU, IQU, and IN in the raffinate oil recovered through equilibrium extraction under a constant condition (volume fraction of water to solvent in initial (yw,0) = 0.1, (S/F)0 = 9, T = 303 K, liquid-liquid contacting time = 72 h) were reduced by about 58.5 wt%, 61.9 wt%, and 73.4 wt%, respectively, compared to those of CMNO. The formamide extraction method in this study was expected to be an effective reduction method for NCs contained in CMNO.

  • The growing cells of S. aureus were fractionated along the Schmidt-Thannhauser-Schneider's technique into several fractions such as TCA(trichloroacetic acid)-soluble, lipid, nucleic acid, protein and residue fraction. They were also fractionated according to their cellular structure into Sonic-supernatant, SDS(sodium lauryl sulfate)-soluble, Formamide-soluble and Residue fraction. Fractionation was carried out by orderly treatment of the Sonic pellet with 1.0% SDS and hot$(150^{\circ}C)$ formamide, and the pellet was prepared by centrifugation of the cells sonic osillated for 20 minutes at 150 watt. Sonic-supernatant fraction contained a 91.3% of total DNA while other fractions contained less than 9.5%. SDS-soluble fraction showed a high activity of malate dehydrogenase(13.67 unit/mg protein) and which was higher 22.3 times than the activity found from unsoluble fraction. Formamide-soluble fraction prepared from SDS-undoluble pellet by using the hot formamide exhibited a clear action of reducing sugars against the Anthronesulfate, while it exhibited no clear action against the ninhydrin. However, contrastly, the residue remainnning after extraction with formamide exhibited a clear action against ninhydrin and glucosamine was detected form the hydrolysate of residue by paper chromatography. From these results it is considered that the Sonic-supernatant fraction is mainly consisted of plasmic component of the cells. Other fractions, SDS-soluble, Formamide-soluble and Residue, should be consisted of plasma membrane, lipoplysaccharide and peptidoglycan of the cell, respectively.

Separation and Recovery of Indole from Model Coal Tar Fraction by Batch Cocurrent 5 Stages Equilibrium Extraction (회분 병류 5단 평형추출에 의한 모델 콜타르 유분 중에 함유된 Indole의 분리 및 회수)

  • The separation of indole from a model mixture comprising four kinds of nitrogen heterocyclic compounds [indole (In), quinoline (Q), iso-quinoline (iQ), quinaldine (Qu)], three kinds of bicyclic aromatic compounds [1-methylnaphthalene (1MN), 2-methylnaphthalene (2MN), dimethylnaphthalene (DMN)], biphenyl (Bp) and phenyl ether (Pe) was examined by batch cocurrent 4 stages equilibrium extraction. The model mixture used as a raw material in this work was prepared according to the components and compositions contained in coal tar fraction (the temperature ranges of fraction: $240{\sim}265^{\circ}C$). An aqueous solution of formamide was used as a solvent. Indole was recovered more than 99% through 4 stages of the equilibrium extraction. The range of selectivity of indole in reference to DMN obtained through the 5 stages equilibrium extraction was found to be 63~118. The process for separation and recovery of indole contained in coal tar was studied by using the experimental results obtained from this work and the previous work.

  • As a part of improving the quality of crude methylnaphthalene (CMNO), this study was experimentally examined the reduction of nitrogen-containing compounds (NC) present in the CMNO by solvent extraction. The CMNO was composed of three kinds of NC [quinolone (QU), iso-quinoline (IQU), indole (IN)], three kinds of bicyclic aromatic compound [BAC; naphthalene (NA), 1-methylnaphthalene (1MNA), 2-methylnaphthalene (2MNA)] and biphenyl (BP) etc., in addition to an aqueous formamide solution, which were used as raw materials and a solvent, respectively. The increase in the volume fraction of water to the solvent in the initial state (yw,0) caused a sharp decrease in the distribution coefficient and the yield of NC, but conversely raised the increased selectivity of NC based on 2MNA. The compositions of QU, IQU and IN in the raffinate oil recovered through the equilibrium extraction of batch co-current 5-stage under constant conditions [yw,0 = 0.1, volume fraction of solvent to feed (CMNO) at the initial state = 1, operating temperature = 303 K, liquid-liquid contacting time = 72 h] were reduced by about 51.5%, 55.2%, and 71.8%, respectively, when compared to those of CMNO. From the excellent reduction rate of NC, the formamide extraction method suggested in this study can be expected to be a useful reduction method for NC contained in the CMNO.

  • The reduction of nitrogen-containing compounds (NC) containing the model crude methylnaphthalene oil (CMNO) of 5 compounds system was experimentally studied by solvent extraction. The model CMNO consisting of 3 kinds of NC including quinolone (QU), iso-quinoline (IQU), indole(IN) and 2 kinds of bicyclic aromatic compounds such as 1-methylnaphthalene (1MNA), 2-methylnaphthalene (2MNA) as raw materials, as well as the aqueous solution of formamide as a solvent, were used. The increase in the volume fraction of water to the solvent at the initial state (yw,0) sharply decreased, but the distribution coefficient and the yield of NC conversely increased the selectivity of NC based on 2MNA, and an increase in the volume fraction of solvent to feed (S/F) simultaneously increased the distribution coefficient, yield and selectivity of NC. The yields of QU, IQU and IN under constant conditions (yw,0 = 0.1, S/F = 1, equilibrium temperature 303 K) were 30%, 31% and 10%, respectively, and selectivity was 15, 15 and 20, respectively. From the excellent yield and selectivity of NC, the formamide extraction method of this study was expected as a method for reducing the NC contained in the model CMNO.

Comparison of Phytoplankton Chlorophyll-$\alpha$ Extracted with Different Solvents

  • Algal chlorophyll-$\alpha$ is commonly determined by spectro-photometric method using 90% acetone as solvents. However, acetone has low extraction efficiency without grinding filters, and DMF (dimethyl formamide) was tested for the compatibility with acetone. Chlorophyll-$\alpha$ concentration was determined for samples from 5 reservoirs of different trophic states and phytoplankton composition, using acetone extraction with grinding and DMF without grinding. Chlorophyll-$\alpha$ measured by DMF and Acetone did not show a significant difference when using trichromatic method of UNESCO and Lorenzen, and therefore, DMF can substitute acetone. But when acidification method was applied, they showed significant difference of 8%. It can be concluded that DMF can extract chlorophylls more effectively without grinding and it can be a better alternative for the standard extraction solvent.

High-Purity Purification of Indole Contained in Coal Tar Absorption Oil by Extraction-Distillation-Crystallization Combination (추출-증류-결정의 조합에 의한 콜타르 흡수유 중에 함유된 인돌의 고순도 정제)

  • Purification of indole contained in model coal tar absorption oil was examined by extraction-distillation-crystallization combination. The absorption oil consists of nine components such as four kinds of nitrogen heterocyclic compounds (9.2% quinoline, 2.4% iso-quinoline, 4.7% indole, 2.4% quinaldine), three kinds of bicyclic aromatic compounds (14.2% 1-methylnaphthalene, 31.8% 2-methylnaphthalene, 23.5% dimethylnaphthalene), 5.5% biphenyl and 3.3% phenyl ether. 99.5% indole was recovered by combination of formamide extraction-distillation-solute crystallization using n-hexane. Furthermore, the recovery process of indole contained in coal tar absorption oil was studied by using the experimental results obtained by each operation of this work.

Effect of Extraction Solvent on the Separation of Sulfur Components in Light Cycle Oil (접촉분해경유로부터 산화황화합물의 분리에 관한 추출용매의 영향)

  • The separation of sulfone components using light cycle oil(LCO) after oxidation was carried out by solvent extraction method using various polar solvents such as water, n-methyl-2-pyrrolidone(NMP), dimethyl sulfoxide, ethyl acetate, acetonitrile, dimethyl formamide, and methyl alcohol. It was found that phase separation between LCO layer and solvent occurred under mixed solvent adding a proper amount of water. The mixture solvent of NMP and water was a promising extraction solvent due to the selective removal and high distribution coefficient of sulfone component in LCO. 99.5% over of sulfur contents in LCO can be removed by 4 stages equilibrium extraction.
