  • Fungal endophytes were isolated from 1-, 2-, 3-, and 4-year-old ginseng roots (Panax ginseng Meyer) cultivated in Korea. The isolated fungal endophytes were identified based on sequence analysis of the internal transcribed spacer and morphological characterization by microscopic observations. A total of 81 fungal endophytes were isolated from 24 ginseng roots. Fungal endophytes were classified into 9 different fungal species and 2 unknown species. Ginseng roots that were 1-, 2-, 3-, and 4-years old were colonized by 2, 6, 8, and 5 species of fungal endophytes, respectively. While Phoma radicina was the most frequent fungal endophyte in 2-, 3-, and 4-year-old ginseng roots, Fusarium solani was the dominant endophyte in 1-year-old ginseng roots. The colonization frequencies (CF) varied with the host age. The CF were 12%, 40%, 31%, and 40% for 1-, 2-, 3-, and 4-year-old ginseng roots, respectively. We found a variety of fungal endophytes that were distributed depending on the age of ginseng plants.

  • A climatic feature, growth condition and incidence phase of disease and pest in Korean ginseng (Panax ginseng C. A. Meyer) were surveyed, both the conventional cultivation and culture in the forest located in Jinan. The results were as follows. The forest field was lower $1^{\circ}C$ in the air temperature, lower $1.9^{\circ}C$ in the soil temperature and 11.8% lower in the soil temperature compared with the conventional field. The wind velocity of the forest field was three times slower, but the dew condensation time was 3.3 hours longer than the conventional field. Particularly, the amounts of solar radiation in the forest was remarkably lower, while the rainfall was little lower with no significance. The incidence of disease and pest in the forest field was severe compared with that of the conventional field. Among them, Colletotrichum gloeosporioides, Cylindrocarpon destructans, and Alternaria panax was severe in the forest field while, Botrytis cinerea was severe in the conventional field. The growth condition of forest field in Panax ginseng C. A. Meyer was 50% worse in general and the root weight was 87.2% less than the 3-year-old ginseng in the conventional field.

  • Panax ginseng is one of the most important medicinal plants in the world. The international trade of ginseng is increasing yearly. The disguise of Chinese and American ginseng into Korean ginseng became a problem in recent years in abroad and Korea. An effective method to authenticate the Korean Panax ginseng from others at a DNA level is necessary for the healthy development of the ginseng market. Amplified fragment length polymorphism (AFLP) analysis was applied to develop a method for the identification of Korean ginseng between Chinese ginseng and American ginseng. It is very difficult to detect the different polymorphic bands among Korean field cultivated ginseng, and between field and wild-cultivated ginseng. The genetic distance coefficient by AFLP analysis between field- and wild cultivated Korean ginseng was very low, 0.056. Whereas, polymorphic bands between Korean and Chinese wild-cultivated ginseng was significantly different. The genetic distance coefficient between wild-cultivated Korean and Chinese ginseng was 0.149. The genetic distance coefficients between the P. ginseng and P. quinquefolius were ranging from 0.626 to 0.666. These results support that the AFLP analysis could be applied to authenticate Korean P. ginseng from others Chinese P. ginseng and American ginseng (P. quinquefolius).

  • Background: Panax ginseng C. A. Meyer is wood-cultivated ginseng (WCG) in Korea which depends on an artificial forest growth method. To produce this type of ginseng, various P. ginseng cultivars can be used. To obtain a WCG similar to wild ginseng (WG), this method is usually performed in a mountain using seeds or seedlings of cultivated ginseng (CG) and WG. Recently, the WCG industry is suffering a problem in that Panax notoginseng (Burk.) F. H. Chen or Panax quinquefolium L. are being sold as WCG Korean market; These morphological similarities have created confusion among customers. Methods and Results: WCG samples were collected from five areas in Korea. After polymerase chain reaction (PCR) amplification using the primer pair labeled with fluorescence dye (FAM, NED, PET, or VIC), fragment analysis were performed. PCR products were separated by capillary electrophoresis with an ABI 3730 DNA analyzer. From the results, WCG cultivated in Korea showed very diverse genetic background. Conclusions: In this study, we tried to develop a method to discriminate between WCG, P. notoginseng or P. quinquefolium using simple sequence repeat (SSR) markers. Furthermore, we analyzed the genetic diversity of WCG collected from five cultivation areas in Korea.

  • Objectives: Wild ginseng pharmacopuncture is widely used in oriental medicine. However, there is no standard method for efficiently extracting the active ingredient. In this study, in order to determine an efficient extraction method, wild ginseng was extracted by the distillation and 70% ethanol reflux methods, respectively. In comparing each extract, the index compounds were analyzed, and antioxidant activity was measured. Methods: The index compounds, ginsenoside Rg1 and ginsenoside Rb1, were detected using high performance liquid chromatography (HPLC). Antioxidative activities of total phenolic compounds, DPPH (${\alpha}$, ${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl), ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)) and FRAP (ferric reducing antioxidant power) were measured to compare their bioactivities. Since saponin is known to be hemolytic, the hemolytic activity of each extract was compared. Results: The index compounds were analyzed. Nothing was detected in the wild ginseng distilled extracts (WGDE). In the wild ginseng 70% ethanol reflux extracts (WGEE), ginsenoside Rg1 was 3.66 mg/g, and ginsenoside Rb1 was 16.70 mg/g. WGEE showed higher levels than WGDE in all antioxidative activities. In the hemolytic test, the extracts showed almost no toxicity, but WGEE showed lower toxicity than WGDE. Conclusions: In this study, it was concluded that WGEE is more advantageous than WGDE in the detection of index compounds and bioactivity. However, additional studies of additional extraction methods and other bioactivity tests are needed.

  • Background: The production method of ginseng seedlings for ginseng cultivation is very important to ensure healthy rooting system as well as high quality, and yield of the resultant plants. This study was carried out to compare the growth characteristics of 2-year-old ginseng plants that were produced from seedlings grown in self soil nursery (SSN), nursery soil (NS) or hydroponic culture (HC). Methods and Results: The shading prop used was composed of four-layered 4 polyethylene (blue 3 + black 1) shade screen. The management of main field was done by inserting oil cake (1,200 kg/10 a) and then allowing Sudan grass to grow for a year. Seedling transplantation was carried out on April 6. Root growth was measured on October 25. Root weight was observed to be excellent at 6.0 g, following SSN transplantation. Root length was 21.2 cm for HC seedlings, but these plants had a physiological disorder (i.e., rusty root), in 83.5% plants of this treatment. The ratio of PD/PT (protopanaxadiol saponins / protopanaxatriol saponins) was higher in NS seedlings. Plant analysis revealed that Fe content was lower in HC seedlings with high rustiness. The growth of 2-years-old ginseng was different following these varying seedling cultivation methods, but seedlings from NS were not different from those grown in SSN. Conclusions: For the propagation of 2-year-old ginseng plants, NS seedlings may be a good substitute for SSN seedlings.

  • In order to develop a novel system for the discrimination of five ginseng cultivars (Panax ginseng Meyer), single nucleotide polymorphism (SNP) genotyping assays with real-time polymerase chain reaction were conducted. Nucleotide substitution in gDNA library clones of P. ginseng cv. Yunpoong was targeted for the SNP genotyping assay. From these SNP sites, a set of modified SNP specific fluorescence probes (PGP74, PGP110, and PGP130) and novel primer sets have been developed to distinguish among five ginseng cultivars. The combination of the SNP type of the five cultivars, Chungpoong, Yunpoong, Gopoong, Kumpoong, and Sunpoong, was identified as 'ATA', 'GCC', 'GTA', 'GCA', and 'ACC', respectively. This study represents the first report of the identification of ginseng cultivars by fluorescence probes. An SNP genotyping assay using fluorescence probes could prove useful for the identification of ginseng cultivars and ginseng seed management systems and guarantee the purity of ginseng seed.

  • Background: Ginsenosides are the unique and bioactive components in ginseng. Ginsenosides are affected by the growing environment and conditions. In New Zealand (NZ), Panax ginseng Meyer (P. ginseng) is grown as a secondary crop under a pine tree canopy with an open-field forest environment. There is no thorough analysis reported about NZ-grown ginseng. Methods: Ginsenosides from NZ-grown P. ginseng in different parts (main root, fine root, rhizome, stem, and leaf) with different ages (6, 12, 13, and 14 years) were extracted by ultrasonic extraction and characterized by Liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry. Twenty-one ginsenosides in these samples were accurately quantified and relatively quantified with 13 ginsenoside standards. Results: All compounds were separated in 40 min, and a total of 102 ginsenosides were identified by matching MS spectra data with 23 standard references or published known ginsenosides from P. ginseng. The quantitative results showed that the total content of ginsenosides in various parts of P. ginseng varied, which was not obviously dependent on age. In the underground parts, the 13-year-old ginseng root contained more abundant ginsenosides among tested ginseng samples, whereas in the aboveground parts, the greatest amount of ginsenosides was from the 14-year-old sample. In addition, the amount of ginsenosides is higher in the leaf and fine root and much lower in the stem than in the other parts of P. ginseng. Conclusion: This study provides the first-ever comprehensive report on NZ-grown wild simulated P. ginseng.
