Background: The roots of Panax ginseng contain noble tetracyclic triterpenoid saponins derived from dammarenediol-II. Dammarene-type ginsenosides are classified into the protopanaxadiol (PPD) and protopanaxatriol (PPT) groups based on their triterpene aglycone structures. Two cytochrome P450 (CYP) genes (CYP716A47 and CYP716A53v2) are critical for the production of PPD and PPT aglycones, respectively. CYP716A53v2 is a protopanaxadiol 6-hydroxylase that catalyzes PPT production from PPD in P. ginseng. Methods: We constructed transgenic P. ginseng lines overexpressing or silencing (via RNA interference) the CYP716A53v2 gene and analyzed changes in their ginsenoside profiles. Result: Overexpression of CYP716A53v2 led to increased accumulation of CYP716A53v2 mRNA in all transgenic roots compared to nontransgenic roots. Conversely, silencing of CYP716A53v2 mRNA in RNAi transgenic roots resulted in reduced CYP716A53v2 transcription. HPLC analysis revealed that transgenic roots overexpressing CYP716A53v2 contained higher levels of PPT-group ginsenosides ($Rg_1$, Re, and Rf) but lower levels of PPD-group ginsenosides (Rb1, Rc, $Rb_2$, and Rd). By contrast, RNAi transgenic roots contained lower levels of PPT-group compounds and higher levels of PPD-group compounds. Conclusion: The production of PPD- and PPT-group ginsenosides can be altered by changing the expression of CYP716A53v2 in transgenic P. ginseng. The biological activities of PPD-group ginsenosides are known to differ from those of the PPT group. Thus, increasing or decreasing the levels of PPT-group ginsenosides in transgenic P. ginseng may yield new medicinal uses for transgenic P. ginseng.

The studies regarding soil bacterial community and correlation analysis of wild-simulated ginseng cultivation area are insufficient. The purpose of this study was to investigate the correlation between soil bacterial community and growth characteristics of wild-simulated ginseng for selection of suitable cultivation area. The bacterial community was investigated by high throughput sequencing technique (Illumina platform). The correlation coefficient between soil bacterial community and growth characteristics were analyzed using Spearman's rank correlation. The soil bacterial community from soil samples of 8 different wild-simulated ginseng cultivated area exhibited two distinct clusters, cluster 1 and cluster 2. The relative abundance of Proteobacteria (35.4%) and Alphaproteobacteria(24.4%) was observed to be highest in all soil samples. The lower soil pH and higher abundance of Acidobacteria resulted in increased growth of wild-simulated ginseng. Additionally, abundance of Acidobacteriia (class) and Koribacteraceae (family) demonstrated significant positive correlation with fresh weight of wild-simulated ginseng. The results of this study clearly state the correlation between growth characteristic and soil bacterial community of wild-simulated ginseng cultivation area, thereby offering effective insight into selection of suitable cultivation area of wild-simulated ginseng.

Wood-cultivated ginseng, including roots in its dried form, is produced in forest land without using artificial facilities such as light barriers. To identify suitable sites for the propagation of wood-cultivated ginseng, factor combination technique (FCT) and linear combination technique (LCT) were used with geographic information system and the results were superimposed onto an actual wood-cultivated ginseng plantation. The LCT more extensively searched for suitable sites of cultivation than that by the FCT; further, the LCT probed wide areas considering the predominance of precipitous mountains in Korea. In addition, the LCT showed the much higher degree of overlap with the actual cultivation sites; therefore, the LCT more comprehensively reflects the cultivator's intention for site selection. On the other hand, the inclusion of additional factors for the selection of suitable cultivation sites and experts' opinions may enhance the effectiveness and accuracy of the LCT for site application.

Background: Identifying suitable site for growing mountain-cultivated ginseng is a concern for ginseng producers. This study was conducted to evaluate the soil properties of cultivation sites for mountain-cultivated ginseng in Hamyang-gun, which is one of the most well-known areas for mountain-cultivated ginseng in Korea. Methods: The sampling plots from 30 sites were randomly selected on or near the center of the ginseng growing sites in July and August 2009. Soil samples for the soil properties analysis were collected from the top 20 cm at five randomly selected points. Results: Mountain-cultivated ginseng was grown in soils that varied greatly in soil properties on coniferous, mixed, and deciduous broad-leaved stand sites of elevations between > 200mand < 1,000 m. The soil bulk density was higher in Pinus densiflora than in Larix leptolepis stand sites and higher in the < 700-m sites than in > 700-m sites. Soil pH was unaffected by the type of stand sites (pH 4.35-4.55), whereas the high-elevation sites of > 700mwere strongly acidified, with pH 4.19. The organic carbon and total nitrogen content were lower in the P. densiflora stand sites than in the deciduous broad-leaved stand sites. Available phosphorus was low in all of the stand sites. The exchangeable cationwas generally higher in the mixed and low-elevation sites than in the P. densiflora and high-elevation sites, respectively. Conclusion: These results indicate that mountain-cultivated ginseng in Korea is able to grow in very acidic, nutrient-depleted forest soils.

Background: Interspecific ginseng hybrid, Panax ginseng ${\times}$ Panax quenquifolius (Pgq) has vigorous growth and produces larger roots than its parents. However, F1 progenies are complete male sterile. Plant tissue culture technology can circumvent the issue and propagate the hybrid. Methods: Murashige and Skoog (MS) medium with different concentrations (0, 2, 4, and 6 mg/L) of 2,4-dichlorophenoxyacetic acid (2,4-D) was used for callus induction and somatic embryogenesis (SE). The embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 Schenk and Hildebrandt (SH) medium. The developed taproots with dormant buds were treated with $GA_3$ to break the bud dormancy, and transferred to soil. Hybrid Pgq plants were verified by random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) analyses and by LC-IT-TOF-MS. Results: We conducted a comparative study of somatic embryogenesis (SE) in Pgq and its parents, and attempted to establish the soil transfer of in vitro propagated Pgq tap roots. The Pgq explants showed higher rate of embryogenesis (~56% at 2 mg/L 2,4-D concentration) as well as higher number of embryos per explants (~7 at the same 2,4-D concentration) compared to its either parents. The germinated embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 SH medium to support the continued growth and kept until nutrient depletion induced senescence (NuDIS) of leaf defoliation occurred (4 months). By that time, thickened tap roots with well-developed lateral roots and dormant buds were obtained. All Pgq tap roots pretreated with 20 mg/L $GA_3$ for at least a week produced new shoots after soil transfer. We selected the discriminatory RAPD and ISSR markers to find the interspecific ginseng hybrid among its parents. The $F_1$ hybrid (Pgq) contained species specific 2 ginsenosides (ginsenoside Rf in P. ginseng and pseudoginsenosides $F_{11}$ in P. quinquefolius), and higher amount of other ginsenosides than its parents. Conclusion: Micropropagation of interspecific hybrid ginseng can give an opportunity for continuous production of plants.

Wild-simulated ginseng is requires standard establishment on location environment for long-term and eco-friendly cultivation. The this study was carried out to investigate the correlation between growth characteristics and location environment of wild-simulated ginseng for selection of suitable cultivation area. Samples were collected from a 9 site of wild-simulated cultivation area and examined growth characteristics and location environment. Correlation coefficient between growth characteristics and location environment was analyzed using Spearman's rank correlation. The growth of wild-simulated ginseng was shown significantly higher in Geochang and Hamyang cultivation area. The results of correlation analysis was shown significantly correlation with percentage of conifer, total diameter at breast height, soil pH, K, Ca and Mg. The results of this study was clearly investigated the correlation between growth characteristics and location environment of wild-simulated ginseng, which is considered to will provide understanding for selection suitable cultivation area of wild-simulated ginseng.
Light environment and growth of Panax ginseng In habitat and cultivation under natural shade were renewed. Grower's experiences on ginseng stand In relation to light environment were assessed. Change of shading method and grower's concepts on light requirement of ginseng plant in Korea, Manchuria and Japan was counted. Growth of wild ginseng was better under rich light. Optimum crown density index appeared to be 0.7 In natural habitat and 0.4 to 0.7 for the cultivation in forest. Change of light Intensity in forest was greatest in May and reached near to constant value (from 40% to 3% for broad leaf deciduous forest and loom 4% to 2% for pine forest). Insufficient light condition induced long and thin stem, poor flowering and seed bearing, and sequent dormancy. Relation between light and ginseng strand was not clear but light Interception with cool weather was effective. Topography and orientation of bed have been deeply considered for light environment. Panel or bark of won were used for shading in deep forest manly In Manchuria while reed blind and straw thatch were used in Korea. Kinds and number of shades material and seasonal or daily schedule have been simplified probably by labor pressure due to eulargement of plantation. Post height has been greater while width of roof, bed and ditch has changed lisle. Scientific survey in the past omitted important light control methods (complete light hardening etc) which has been practiced in specific areas. The concept and technique of light control in the past in Korea were delicate and intensive including the control of temperature and moisture. For the application of old concept in modem cultivation precise Investigation of the technique including the measurement of light, temperature and moisture is needed.

The best environments such as crown density, temperature, light intensity and humidity have to be identified because these factors are strongly related to the growth and several physiological characteristics. The purposes of this study are \circled1 to collect basic data fer growth, \circled2 to identify the best growth environments. to achieve these purposes, oak, pine and mixed forest stands have been selected in this study. forest ginseng seeds were sown in these forest four years ago. Several environmental and growth factors have been surveyed. In general, mean tree age, DBH and average height are 20-25 years old, 14-17 cm and 7-9 m, respectively. The growths of forest ginseng grown in oak stand are better than those of pine and mixed stands.

Background: The low survival rate of in vitro regenerated Panax ginseng plantlets after transfer to soil is the main obstacle for their successful micropropagation and molecular breeding. In most cases, young plantlets converted from somatic embryos are transferred to soil. Methods: In vitro thickened taproots, which were produced after prolonged culture of ginseng plantlets, were transferred to soil. Results: Taproot thickening of plantlets occurred near hypocotyl and primary roots. Elevated concentration of sucrose in the medium stimulated the root thickening of plantlets. Senescence of shoots occurred following the prolonged culture of plantlets. Once the leaves of plantlets senesced, the buds on taproots developed a dormant tendency. Gibberellic acid treatment was required for dormancy breaking of the buds. Analysis of endogenous abscisic acid revealed that the content of abscisic acid in taproots with senescent shoots was comparatively higher than that of taproots with green shoots. Thickened taproots were transferred to soil, followed by exposure to gibberellic acid or a cold temperature of $2^{\circ}C$ for 4 mo. Cold treatment of roots at $2^{\circ}C$ for 4 mo resulted in bud sprouting in 84% of roots. Spraying of 100 mg/L gibberellic acid also induced the bud sprouting in 81% roots. Conclusion: Soil transfer of dormant taproots of P. ginseng has advantages since they do not require an acclimatization procedure, humidity control of plants, and photoautotrophic growth, and a high soil survival rate was attained.
