  • The development of random amplified polymorphic DNA (RAPD) and expressed sequence tag-derived simple sequence repeats (EST-SSRs) provided a useful tool for investigating Korean ginseng genetic diversity. In this study, 18 polymorphic markers (7 RAPD and 11 EST-SSR) selected to assess the genetic diversity in 31 ginseng accessions (11 Korean ginseng cultivars and 20 breeding lines). In RAPD analysis, a total of 53 unique polymorphic bands were obtained from ginseng accessions and number of amplicons ranged from 4 to 11 with a mean of 7.5 bands. Pair-wise genetic similarity coefficient (Nei) among all pairs of ginseng accessions varied from 0.01 to 0.32, with a mean of 0.11. On the basis of the resulting data, the 31 ginseng accessions were grouped into six clusters. As a result of EST-SSR analysis, 11 EST-SSR markers detected polymorphisms among the 31 ginseng accessions and revealed 49 alleles with a mean of 4.45 alleles per primer. The polymorphism information content (PIC) value ranged from 0.06 to 0.31, with an average of 0.198. The 31 ginseng accessions were classified into five groups by cluster analysis based on Nei's genetic distances. Consequently, the results of ginseng-specific RAPD and EST-SSR markers may prove useful for the evaluation of genetic diversity and discrimination of Korean ginseng cultivars and breeding lines.

  • The total area of paddy field was estimated to be 55 % of the cultivated lands in South Korea, which is approximately 1 million hectares. Organisms inhabiting paddy fields if they are sensitive to environmental changes can be environmental indicator of paddy fields. Biological indicators such as phenology and distributional range are evaluated as intuitive and quantitative method to analyze the impact of climate change. This study aims to estimate flight time change of Hydrophilidae species' based on the RCP 8.5 climate change scenario. Unmanned monitoring systems were installed in Haenam, Buan, Dangjin and Cheorwon relative to the latitudinal gradient. In the three regions excepting Cheorwon, it was able to measure the abundance of flying Hydrochara affinis and Sternolophus rufipes. Degree-day for the flight time was determined based either on field measurement values and estimates of 2020s, 2050s and 2080s from KMA climate change scenario data. As a result, it is found that date of both species of initial flight becomes 15 days earlier, that of peak flight becomes 22 days earlier and that of final flight does 27 days earlier in 2080s compared to 2020s. The climate change impact on flight time is greater in coastal area, rural area and valley than inland area, urban area and plan. H. affinis and S. rufipes can be used as climate change indicator species.

  • The Discrete Element Method (DEM) is one of the useful numerical methods to analyze the behavior of the ground formation by computing the motion and interaction using particles. The DEM has not been applied in civil engineering but also a wide range of industrial fields, such as chemical engineering, pharmacy, material science, food engineering, etc. In this study, to review a performance of the spoke-type earth pressure balance (EPB) shield TBM (Tunnel Boring Machine), the commercial software based on the DEM technology was used. An analysis of the TBM during excavation was conducted according to two pre-defined excavation conditions with the different rotation speed of a cutterhead. During the analysis, the resistant torque at the face of the cutterhead, the compressive force at the cutterhead and shield surface, the muck discharge at the screw auger were measured and compared. Upon the two kinds of excavation conditions, the applicability of the DEM analysis was reviewed as a modelling method for the TBM.

Especially, we divided the surveyed region into nine representative collection areas based on floristic components were compared and analyzed. This field study discovered significant plants in various categories. The 41 surveys revealed that the vascular plants in this region comprised 815 taxa: 116 families, 429 genera, 717 species, 4 subspecies, 78 varieties and 16 forms. In this study, 80 taxa were firstly recorded from this region. Korean endemic plants numbered 22 taxa, and the fifteen taxa rare plants designated by Korean Forest Service as were found in this region. The taxa in the category II of rare and endangered plants and the taxa with the higher rank than the third degree among the floristic regional indicator plants designated by Korean Ministry of Environment were one and 56, respectively. From a geographical perspective, limited distributions of Matteuccia struthiopteris, Spiraea trichocarpa, and Aster tataricus at a latitudes higher than that of Woraksan National Park. Moreover, this study has confirmed that the Woraksan National Park is the species diversity center for Asarum koreanum, Elsholtzia angustifolia, and Allium linearifolium in the central province of the Korean peninsula. Inaddition, the naturalized plants of 54 taxa were recorded.
