Effects of culture supernatants of Lactobacillus reuteri MG5346 (CS-MG5346) on receptor activator of nuclear factor-kappa B ligand (RANKL)-induced osteoclastogenesis were examined. CS-MG5346 treatment up to 400 ㎍/mL significantly reduced tartrate-resistant acid-phosphatase (TRAP) activity, the phenotype biomarker of osteoclast, without affecting cell viability. CS-MG5346 inhibited the expression of osteoclast specific transcriptional factors (c-fos and nuclear factor-activated T cells c1) and their target genes (TRAP, cathepsin, and matrix metallo-proteinase-9) in a dose-dependent manner (p<0.05). The administration of L. reuteri MG5346 (2×108 CFU/day) for 8 wks significantly improved furcation involvement, but no difference was observed in alveolar bone loss in ligature-induced experimental periodontitis rats. The elevated RANKL/osteoprotegerin ratio, the biomarker of periodontitis, was significantly lowered in the gingival tissue by administration of L. reuteri MG5346 (p<0.05). L. reuteri MG5346 showed excellent stability in simulated stomach and intestinal fluids and did not have antibiotic resistance. Based on the results, L. reuteri MG5346 has the potential to be a promising probiotic strain for oral health.
본 연구는 해충에 대한 생물적 방제제로서 유용한 곤충병원성 선충의 효율적 검색과 in vitro에서의 대량 증식 가능성을 조사하였으며 다음과 같은 결과를 얻었다. 1. 토양으로부터 곤충병원성 선충의 검색을 위한 trap용 곤충으로는 꿀벌부채명나방보다 누에가 감수성이 높았다. 2. Sterinernema, Rhabditidae, Diplogastroidae과 계통의 선충은 꿀벌부채명나방 유충에서의 증식은 5만 마리 이하였으나, 누에 유충에서는 15만에서 35만 마리의 선충 증식이 일어났으며, 누에 번데기에서는 25만 마리 이상의 선충이 증식되었다. 누에 증식에 있어 누에 번데기를 사용했을 때가 가장 우수하였다. 3. Sterinernema, Rhabditidae, Diplogastroidae과 계통의 선충에 대한 증식률은 열풍건조 번데기의 흡수처리 후 배양 및 기타 천연물 인공배지에서 증식은 가능하였으나 살아있는 누에 유충보다는 증식률이 낮았다. 4. 이상의 결과로 토양으로부터 곤충병원성 선충을 분리하는데 누에를 이용한 trap이 매우 우수하였으며, 선충의 증식에 있어 누에 유충 및 번데기가 효과적인 것으로 판단된다.

Receptor activator of nuclear factor-kappa B ligand (RANKL) is a critical factor in osteoclastogenesis. It makes osteoclasts differentiate and multinucleate in bone remodeling. In the present study, RANKL was expressed as a soluble maltose binding protein (MBP)-fusion protein using the Escherichia coli maltose binding domain tag system (pMAL) expression vector system. The host cell E. coli DH5α was cultured and induced by isopropyl β-D-1-thiogalactopyranoside for rRANKL expression. Cells were disrupted by sonication to collect soluble MBP-fused rRANKL. The MBP-fusion rRANKL was purified with MBP Trap affinity chromatography and treated with Tobacco Etch Virus nuclear inclusion endopeptidase (TEV protease) to remove the MBP fusion protein. Dialysis was then carried out to remove binding maltose from the cleaved rRANKL solution. The cleaved rRANKL was purified with a second MBP Trap affinity chromatography to separate unsevered MBP-fusion rRANKL and cleaved MBP fusion protein. The purified rRANKL was shown to have biological activity by performing in vitro cell tests. In conclusion, biologically active rRANKL was successfully purified by a simple two-step chromatography purification process with one column.
Acetic acid producing bacteria were isolated from deastringent persimmon vinegar and the major bacterium was identified using morphological and biochemical tests. Acetobacter sp. AH-1 was motile, gram negative rod with catalase positive and oxidase negative. The strain can grow up to 5 % ethanol and 2% NaCl as well as 25% glucose. Optimum temperature and pH for growth were 3$0^{\circ}C$ and 5.0, respectively. Volatile constituents of persimmon vinegar were analyzed by purge and trap sampling . Acetic acid adn alcohol were the largest volatile compounds quantitiatively in persimmon vinegar. Among alcohols, 20methyl-1-propanol, isoamyl alcohol and amyl alcohol were detected. Isovaleradehyde and benzaldehyde for aldehyde, isoamyl acteate, ethyl formate, propyl aceetate, and ethyl acetate for esters were likely to contribute to persimmon vivegar flavor.
In the cities nowadays, the dwelling conditions of Cackroach are provided by use of various kinds of construction material, installation of heating and cooling facilities that maintain moderate temperature, filthy sanitation, and lack of knowledge about insect as mechanical Vector. Cakroach is infectious disease Vector, and it proves to be of much importance in the public health. Considering such importance, the author has surveyed the distribution of Cackroach in the housing areas of Seoul City from May 1969 to the end of August 1969, and has come to the following conclusion: 1. House dwelling Cackroach collected in the City of Seoul are of two species, i. e. Blatella Germanica Linneaus and Periplaneta Japonica Karny. 2. Most of cackroach collected in Seoul are Blatella Germanica Linneaus and they are distributed in whole area of Seoul while that of Periplaneta Japonica Karny is distributed only in the limited area. 3. Dwelling places of Blatella Germanica Lineaus are in grog shop, Korean food restaurants, bakery shops, and patient rooms in hospitals. Many of them are found in tile kitchens and dining rooms of restaurants, and tearooms. They live in groups at older houses with much crack, warm and humid places. 4. For method of collection, the hand collection was the most effective. In trap collection during the day time, when the trap is placed at the narrow corners off the ground after confirming the dwelling places of cackroach, the match trap was more effective than the flask trap in collecting. In case of collecting during the night time, the flask trap was much more effective method than the match trap.
본 연구는 제2형 당뇨모델 쥐에서 조각자 추출물의 항당뇨 효과를 알아보기 위해 SD계 수컷 흰쥐에 streptozotocin (STZ)을 복강주사 15분 후 nicotineamide(NA)로 복강에 재주사하여 제2형 당뇨를 유발시켰다. 그리고 당뇨대조군(DC), 당뇨 유발군+acarbose 4 mg/kg(AC), 당뇨유발군+조각자 추출물(Gleditschiae Spina ethanol extract) 50 mg/kg (GSE), 그리고 정상군(NC)으로 나누어 10주간 식이와 함께 물을 자유 급여하였다. 10주 동안 매일 식이 섭취량과 음료섭취량 및 매주 체중과 공복혈당을 측정하고, 마지막 주에 경구포도당 부하검사(OGTT)를 실시하였으며, 부검 시 혈액을 채취하여 혈장 TRAP 수준을 측정하였다. 정상군에 비해 당뇨그룹(DC, AC, GSE)의 체중 증가는 유의적으로 감소하였고, 당뇨 유발군들 간에는 유의적 차이가 없었다. 식이섭취량, 음료섭취량 및 식이이용효율은 정상군에 비해 당뇨그룹 DC, AC, GSE에서 유의적으로 높게 나타났으나, 음료섭취량은 DC에 비해 AC, GSE군에서 유의적으로 감소하였다. 정상군은 10주 동안 정상 수준 공복혈당을 나타내었고, acarbose는 8주부터 공복혈당이 유의적으로 감소하였으며, 9주까지 GSE군은 DC군보다 공복혈당은 낮았지만 유의적 차이는 보이지 않다가 마지막 10주에서는 DC군보다 공복혈당이 유의적으로 감소하였다. 경구 당부하 검사 결과에서는 포도당 투여 30분 후부터 혈당이 상승되기 시작하여 정상군보다 당뇨군들에서 유의적으로 상승하였고, 당뇨군들에서는 DC군에 비해 AC와 GSE군에서 유의적으로 혈당상승이 억제되었다. 10주간의 GSE 보충은 DC군에 비해 혈장 내총항산화(TRAP) 수준을 유의적으로 증가시키는 것으로 나타났다. 따라서 제2형 당뇨병에서 조각자 추출물을 장기간 투여한다면 혈당증가 억제에 있어 확연한 효과를 보일 것으로 생각된다. 이는 조각자 추출물의 항산화 활성과도 연관이 있을 것으로 추측되며, 향후 조각자 추출물의 혈당강하작용 효과를 가지는 물질의 분리 및 정제에 대한 연구가 이루어져야 할 것으로 사료된다.
본 연구는 천연물의 효능을 미생물을 이용하여 증가시키거나 새로운 효능을 도출하고자 하는 연구를 통해 Bacillus subtilis CJH 101 및 Bacillus safensis CJH 102 로 발효한 동과 발효물(HR1901-BS, HR1901-BSaf)의 뼈 건강 관련 효능을 평가하였다. 뼈를 형성하는 조골세포의 증식을 비교한 결과, 동과 발효물은 조골세포의 증식을 농도 유의적으로 증가시키는 것으로 나타났으며 조골세포 분화 유도 및 무기질화에 관여하는 ALP 활성을 효과적으로 촉진시켰다. 또한 조골세포 분화를 조절하는 전사 인자인 ALP, OCN, Runx2의 발현이 증가됨을 확인하였다. 뼈를 흡수하는 파골세포의 활성을 확인하기 위해 TRAP 활성을 측정한 결과 동과 발효물은 TRAP 활성을 유의적으로 억제하는 것을 확인하였다. 따라서 동과 발효물(HR1901-BS, HR1901-BSaf)은 조골세포의 활성 증가 및 파골세포의 활성 억제를 통해 골대사에 긍정적인 영향을 미치므로 뼈 대사 및 골다공증 관련 기능성 식품 소재로 활용 가능할 것으로 사료된다.
We compared the preventive capacity of high intakes of vitamin C (VC) and vitamin E (VE) on oxidative stress and liver toxicity in rats fed a low-fat ethanol diet. Thirty-two Wistar rats received the low fat (10% of total calories) Lieber-DeCarli liquid diet as follows: either ethanol alone (Alc group, 36% of total calories) or ethanol in combination with VC (Alc + VC group, 40 mg VC/100 g body weight) or VE (Alc + VE group, 0.8 mg VE/100 g body weight). Control rats were pair-fed a liquid diet with the Alc group. Ethanol administration induced a modest increase in alanine aminotransferase (ALT), aspartate aminotransferase (AST), conjugated dienes (CD), and triglycerides but decreased total radical-trapping antioxidant potential (TRAP) in plasma. VE supplementation to alcohol-fed rats restored the plasma levels of AST, CD, and TRAP to control levels. However, VC supplementation did not significantly influence plasma ALT, AST, or CD. In addition, a significant increase in plasma aminothiols such as homocysteine and cysteine was observed in the Alc group, but cysteinylglycine and glutathione (GSH) did not change by ethanol feeding. Supplementing alcohol-fed rats with VC increased plasma GSH and hepatic S-adenosylmethionine, but plasma levels of aminothiols, except GSH, were not influenced by either VC or VE supplementation in ethanol-fed rats. These results indicate that a low-fat ethanol diet induces oxidative stress and consequent liver toxicity similar to a high-fat ethanol diet and that VE supplementation has a protective effect on ethanol-induced oxidative stress and liver toxicity.
Folate deficiency and hyperhomocysteinemia are found in most patients with alcoholic liver disease. Oxidative stress is one of the most important mechanisms contributing to homocysteine (Hcy)-induced tissue injury. However it has not been examined whether exogenous administration of folic acid attenuates oxidative stress and hepatic toxicity. The aim of this study was to investigate the in vivo effect of folic acid supplementation on oxidative stress and hepatic toxicity induced by chronic ethanol consumption. Wistar rats (n = 32) were divided into four groups and fed 0%, 12%, 36% ethanol, or 36% ethanol plus folic acid (10 mg folic acid/L) diets. After 5 weeks, chronic consumption of the 36% ethanol diet significantly increased plasma alanine transaminase (ALT) (P < 0.05) and aspartate transaminase (AST) (P < 0.05), triglycerides (TG) (P < 0.05), Hcy (P < 0.001), and low density lipoprotein conjugated dienes (CD) (P < 0.05) but decreased total radical-trapping antioxidant potential (TRAP) (P < 0.001). These changes were prevented partially by folic acid supplementation. The 12% ethanol diet had no apparent effect on most parameters. Plasma Hcy concentration was well correlated with plasma ALT (r = $0.612^{**}$), AST (r = $0.652^*$), CD (r = $0.495^*$), and TRAP (r = $-0.486^*$). The results indicate that moderately elevated Hcy is associated with increased oxidative stress and liver injury in alcohol-fed rats, and suggests that folic acid supplementation appears to attenuate hepatic toxicity induced by chronic ethanol consumption possibly by decreasing oxidative stress.
