Production of Biological Activator by Microorganisms - II. Extraction of Cellular Materials from Rhodotorula rubra - (미생물을 이용한 생리 활성 물질의 생산 - 제 2 보 : Rhodotorula rubra로 부터 균체 성분의 추출 -)

  • The extraction mechanism and extraction conditions of cellular lipid of freeze-dried cells of Rhodotorula rubra YUFE 1526, which was reported as coenzyme Q producing microorganism, were studied. 1. Methyl alcohol was the most appropriate solvent for extraction of cellular lipids and the resulting total lipid was 19.17 weight %. 2. When 1 % (w/v) of freeze-dried cell was extracted by methyl alcohol, the extraction yield was 90.49 % at $50^{\circ}C$ for 2 hr. 3. The diffusivity varied with extraction temperature and the empirical equation was derived as follows : $2.2e^{0-02907T}{\times}10^{-9}\;mm^2/min.$ 4. The diffusivity of cellular lipid of Rhodotorula rubra YUFE 1526 was $9.4{\times}10^{-9}\;mm^{2}/min.$ 5. The extraction model was well fitted on the extraction of cellular lipid.

Isolation of a Fermenting Microorganism Involved in Formation of ortho-Dihydroxyisoflavones in Doenjang (Korean Fermented Soybean Paste)

  • A fermenting microorganism involved in formation of ortho-dihydroxyisoflavones (ODIs) during aging of doenjang (Korean fermented soybean paste) has been investigated. Microorganisms in ODI-containing doenjang were isolated by cultivating on yeast mold (YM) agar medium containing 0-7% NaCl. ODI formation of the isolated strains was examined by gas chromatography/mass spectrometry (GC/MS) analysis after cultivation in modified YM broth or soybean extract medium. An ODI-producing microbe was identified as Bacillus subtilis HS-1 based on 16S rRNA gene sequence analysis. The strain has produced 8-hydroxydaidzein as a major product during growth in the modified YM broth or soybean extract medium. Therefore, it was concluded that one of the microorganisms involved in the formation of ODIs in doenjang was B. subtilis HS-1.

Progressive Screening of Thermostable Yeasts for Phytase Production

  • Biotechnological phytase preparations are commercially available and are currently used in animal feeding. However, thermostability constraints, low yields, and the high cost of the enzyme have limited its use. This study represents a new perspective for the food enzyme market. The research screened thermostable yeast strains for their ability to produce phytase. The screening was carried out with a gradual increase in temperature ($30-48^{\circ}C$). Sixteen strains (1 strain identified as Saccharomyces cerevisiae) maintained the ability to produce phytase at $48^{\circ}C$ and their phytase activity was confirmed using 2 phytase assay methodologies. The yeast strains tested in this study seem to be potential efficient producers of phytase, indicating a possible new source of thermostable phytase of commercial interest, particularly that from S. cerevisiae.

  • The need to determine the permeability of organic vapors to polymeric films such as aromas, flavors, etc. has significantly increased in the food industry because of preservation and safety issues along with migration problems. However, due to the complex nature of the permeation process, there have been few investigations compared to nonorganic compounds. In this paper, we review the history of permeability studies and typical methods of permeability measurement such as the isostatic method and quasi-isostatic method for organic vapors. New instrumental developments and significant findings are also introduced and discussed.

Formulation and Shelf Life of a Nutritional Supplement for Undernourished Elderly People

  • A nutritional supplement was developed aiming at correcting the most common nutrient and caloric deficiencies encountered in elderly people (${\geq}60$ years old). The protein source was a mixture of whey protein isolates (WPI) and bovine collagen hydrolysate (BCH) with high nutritional and functional qualities making up 12% of the formulation. The carbohydrate fraction was composed of sucrose, inulin (soluble fiber), and fructo-oligosaccharide (prebiotic). The most commonly deficient essential minerals and vitamins were also included. Acceptance of the product was good according to both an elderly panel and a laboratory panel composing of both sexes and various ages. The stability of the formulations was evaluated and the estimated shelf life at room temperature (ca. $27^{\circ}C$) was approximately 4 months.

Detection and Identification of Bacteriocins Produced by Propionibacteria Isolated from Commercial Swiss Cheese Products

  • Wild propionibacteria isolated from different commercial swiss cheese samples were tested for antimicrobial activities. In initial screening, six of these Propionibacterium isolates showed antagonistic activity against 10 selected indicator organisms by the deferred method. In next, only two Propionibacterium strains JW6 and JW14 showed antibacterial activity in the cell-free supernatants by the modified well diffusion method. Propionibacterium strains JW6 and JW14 were finally identified as bacteriocin producers which exhibited a bactericidal effect against closely related species. The antimicrobial substances were proteins, since their activities were completely destroyed following several degradative enzyme treatments. The bacteriocins showed a narrow inhibitory spectrum of activity against two propionibacteria and two bacilli of strains tested in this study.

Measurement of Spray Deposit Amount Using Spectrophotometer and Food Dye as Tracer

  • Measurement of spray deposit is necessary for evaluation of a chemical application technology. However it is not easy and time consuming. A simple method for measuring the deposition amount of spray using a tracer and a spectrophotometer was developed. Various materials were tested to determine an adequate tracer. Food dye was selected as a tracer, because it was cheep and easily treatable. Using NIRS(Near Infrared Reflection Spectrophotometer), a regression curves between maximum absorbance of a solution and concentration of the tracer were obtained. Yellow food dye solution showed a peak of spectrum at 452 nm, and absorbance of peak showed a tendency to increase as concentration increased. Green or pink food dye were tested and judged to be good tracers. However, tracer concentration should not exceed certain limits in order to measure maximum absorption. Using spraying liquid with known tracer concentration and known amount of washing liquid, spray deposit amount on real targets on leaves could be estimated at less than 13% error level.

Antibacterial Activity of Bioconverted Linoleic Acid Produced by Pseudomonas aeruginosa PR3

  • Crude extract of bioconverted linoleic acid using Pseudomonas aeruginosa PR3 was evaluated for its antibacterial activity against food-borne pathogenic bacteria. Crude extract showed antibacterial activity against four Gram-positive bacteria, Staphylococcus aureus (ATCC 6538), S. aureus (KCTC 1916), Listeria monocytogenes (ATCC 19166), and Bacillus subtilis (ATCC 6633), and one Gramnegative bacterium, Pseudomonas aeruginosa (KCTC 2004), with minimum inhibitory concentrations ranging from 750 to $1,500\;{\mu}g{\cdot}ml^{-1}$. S. aureus and B. subtilis were selected for growth inhibition assays with bioconverted linoleic acid. Major antibacterial effects occurred at lag phase.

Studies on Linerboard Compositions of Corrugated Fiberboard Boxes for Oriental Melons (참외 포장용 골판지상자의 재질구성에 관한 연구)

  • The study aimed to review linerboard compositions of corrugated fiberboard boxes for Oriental melons(Cucumis melon L. var makuwa), analyzing relationships among material compositions, price, weight of boxes, theoretical compression and bursting strength of various liner boards. 19 and 22 different liner boards were currently used for the bleached and the unbleached corrugated boards, respectively. The corrugated board containers with the bleached liners were mostly used in the market although the average price was 10 percents higher than the unbleached. The average compression strength of the bleached. We strongly recommend to redesign the boxes because the average compression and bursting strength of the current boxes were much higher than national packaging standards as much as 70 and 82, respectively.
