Determination of Branched-Chain α-Keto Acid Dehydrogenase Activity in Rat Tissues

  • The branched-chain ${\alpha}$-keto acid dehydrogenase (BCKAD) complex is a rate limiting enzyme which catalyzes the oxidative decarboxylation of branched-chain ${\alpha}$-keto acids. Numerous studies have suggested that BCKAD is subject to covalent modification in vitro via phosphorylation and dephosphorylation, which are catalyzed by a specific kinase and phosphatase, respectively. The biggest difficulty in the assay of BCKAD activity is to arrest the interconversion between the active and inactive forms. BCKAD activity was determined from fresh rat heart and liver tissues using homogenizing and assay buffers containing inhibitors of phosphatase and kinase. The results suggest that a radiochemical assay using ${\alpha}$-keto[1-$^{14}C$]-isovalerate as a substrate for the enzyme can be applied as a reliable method to determine in vitro enzyme activity with arrested interconversion between the active and inactive forms of the BCKAD complex.

Effects of Dietary β-Glucan on Short Chain Fatty Acids Composition and Intestinal Environment in Rats (식이 베타-글루칸이 흰쥐의 장내 단쇄지방산 조성 및 장내환경 개선에 미치는 영향)

  • The effects of dietary ${\beta}$-glucan, obtained from bacterial fermentation, on the intestinal mass, short chain fatty acids, lactate production and pH in Sprague-Dawley (SD) rats were evaluated. SD rats fed with 0% (control group), 1% or 5% ${\beta}$-glucan supplemented diets (w/w) for 3 weeks. The presence of ${\beta}$-glucan in the diets resulted in a significant increase in colonic contents in a dose dependent manner. The amount of short chain fatty acids increased in rats fed ${\beta}$-glucan diets. Rats fed the 5% ${\beta}$-glucan diets had higher levels of acetate, propionate and butyrate by 1.8, 1.7 and 3.0 fold of the control group in the cecum, and 2.2, 2.9 and 3.1 fold of the control group in the colon, respectively. The ${\beta}$-glucan diets also significantly increased the levels of cecal and colonic lactate by 1.4~3.4 fold, when compared to the control diet, indicating that dietary ${\beta}$-glucan stimulated the growth of lactic acid bacteria within the intestine. These results suggest that dietary ${\beta}$-glucan, by providing short chain fatty acids and reducing the cecal and colonic pH, may be beneficial in improving gut health, and provide evidence for the use of ${\beta}$-glucan as a dietary supplement for human consumption.

  • The global halal market is forecast to grow at an annual average of 5.2 percent from 2017 to $3.07 trillion in 2023 due to the high growth rate of the world's Muslim population, the spread of halal-certified food consumption and the economic growth of the Muslim world. Through this study, the difficulty of obtaining halal certification can be overcome through accurate understanding of the general supply chain and other halal supply chain. Also, by examining the trends and requirements of halal logistics standards in countries with advanced halal logistics systems, halal logistics certification agencies, and halal port logistics, we can help establish our own halal logistics system by finding areas that can be benchmarked in Korea and differentiated from those that can be found. For the safe supply chain management of halal products between logistics Supply Chain, an integrated logistics system shall be developed to manage customs and customs as one-stop, while maintaining a complete halal condition on a series of logistics processes such as storage, transportation, customs clearance, etc. Korea, entry into the halal logistics market through halal integrity guarantee solution or platform development can also be considered, taking advantage of the strength of IT and packaging.

  • This study proposed a real and strategical business model from a supply chain perspective to heighten the competitiveness of agri-food export to promote export agriculture of Korea. The proposed export logistics business model can efficiently integrate and manage supply chain members in the agri-food export logistics center. The export logistics center is an integrated model to increase agri-food export which consists of product mixture that simultaneously distributes large companies' processed food and small companies' agri-food, a material and process system of export logistics, opening up foreign markets, and access to target markets on the basis of efficient agri-food export such as the construction of an export logistics system, finding overseas markets, and the launch and development of strategic goods for export.

  • Purpose: Research on supply chain sustainability is important for exporters When the factor of sustainable development is considered by the businesses as well as governments of all countries. Research on supply chain sustainability is important for exporters. Sustainable supply chain management and supply chain dynamics will help enterprises adapt to changes in the business environment. This study analyzes the impact of sustainable supply chain management, and supply chain dynamic capabilities on the sustainable development of exporting enterprises in Vietnam. Research design, data, and methodology: The research model and survey are designed based on previous studies after surveying export enterprises. With 185 samples collected from export enterprises. The Structural Equation Modeling (SEM) analysis technique is used. Data analysis is performed on SPSS and AMOS software (Reliability test, Confirmatory Factor Analysis, SEM). Results: Sustainable supply chain management and supply chain dynamic capabilities all have positive effects on the sustainable development of businesses (sustainable development is measured by distribution: measuring economic efficiency, social efficiency, and environmental performance). Conclusions: From the results of this study, the authors also made several recommendations to help export enterprises develop sustainability based on sustainable supply chain management and supply chain dynamic capabilities.

Modulation of Branched-Chain Amino Acid Metaolism by Exercise in Rats

  • A variety of important roles for branched-chain amino acids in metabolic regulation has been suggested. Branched-chain $\alpha$-keto acid dehydrogenase(BCKAD) complex is a rate limiting enzyme in branched-chain amino acid metabolism. The purpose of this study was to examine the effects of exercise on the activity and activity state of branched-chain $\alpha$-keto acid dehydrogenase in rat hert and liver thssues. Forty-eight Sprague-Dawley rats were assigned into three experimental groups : sedentary control, exercised, or exercised-rested. Submaximal exercise(running) for two hours significantly increased basal activity without a change in total activity in both tissues, with a concomitiant increase in activity state of the enzyme complex. At 10 min post-exercise, heart enzyme activity significantly decreased, though not to the control level, while liver enzyme activity remained unchanged. These data suggested that the exercise-induced increase in branched-chain $\alpha$-keto acid decarboxylation in rat tissues may not be the result of enzyme synthesis, but rather is due to increased activity of the BCKAD.

Thermic Effect of Food, Macronutrient Oxidation Rate and Satiety of Medium-chain Triglyceride (중쇄중성지방(MCT)의 식이성 발열효과, 영양소 산화율 및 포만도)

  • Objectives: The objective of this study was to evaluate the thermic effects, the macronutrient oxidation rates and the satiety of medium-chain triglycerides (MCT). Methods: The thermic effects of two meals containing MCT or long-chain triglycerides (LCT) were compared in ten healthy men (mean age $24.4{\pm}2.9years$). Energy content of the meal was 30% of resting metabolic rate of each subject. Metabolic rate and macronutrient oxidation rate were measured before the meals and for 6 hours after the meals by indirect calorimetry. Satiety was estimated by using visual analogue scales (VAS) at 8 times (before the meal and for 6 hours after meal). Results: Total thermic effect of MCT meal (42.8 kcal, 8.0% of energy intake) was significantly higher than that (26.8 kcal, 5.1% of energy intake) of the LCT meal. Mean postprandial oxygen consumption was also significantly different between the two types of meals (MCT meal: $0.29{\pm}0.35L/min$, LCT meal: $0.28{\pm}0.27L/min$). There were no significant differences in total postprandial carbohydrate and fat oxidation rates between the two meals. However, from 30 to 120 minutes after consumption of meals, the fat oxidation rate of MCT meal was significantly higher than that of the LCT meal. Comparison of satiety values (hunger, fullness and appetite) between the two meals showed that MCT meal maintained satiety for a longer time than the LCT meal. Conclusions: This study showed the possibility that long-term substitution of MCT for LCT would produce weight loss if energy intake remained constant.

Development of Multiplex Polymerase Chain Reaction Assay for Identification of Angelica Species (Multiplex Polymerase Chain Reaction을 이용한 당귀 종 판별)

  • Background: Angelica gigas, A. sinensis, and A. acutiloba are commercially important in the herbal medicine market, and among them, A. gigas has the highest economic value and price. However, their similar morphological traits are often used for fraud. Despite their importance in herbal medicine, recognition of the differences between Angelica species is currently inadequate. Methods and Results: A multiplex polymerase chain reaction (PCR) method was developed for direct detection and identification of A. gigas, A. sinensis, and A. acutiloba. The gene for the distinction of species was targeted at ITS in the nucleus and trnC-petN gene in chloroplasts. The optimized multiplex PCR in the present study utilized each Angelica species-specific primer pairs. Each primer pair yielded products of 229 base pairs (bp) for A. gigas, 53 bp for A. sinensis, 170 bp for A. acutiloba. Additionally non-specific PCR products were not detected in similar species by species-specific primers. Conclusions: In the present study, a multiplex-PCR assay, successfully assessed the authenticity of Angelica species (A. gigas, A. sinensis, and A. acutiloba). and whole genome amplification (WGA) was performed after DNA extraction to identify, the species in the product. The detection method of raw materials developed in the present study could be applied to herbal medicine and health functional food management.

Relationship Between Management Practices of Hamburger Chain Restaurants and Job Satisfaction of Restaurant Employees (햄버거 체인점의 관리실태와 종업원 직무만족과의 관계)

  • The purpose of this study was to evaluate the management practices and to measure the degree of job satisfaction of employees of hamburger chain restaurants in Seoul. Management practices were evaluated in terms of service, quality, cleanliness and shift management in 30 hamburger restaurants of 3 domestic and 3 foreign chain brands. Employee job satisfaction was measured by using questionnaires of Job Descriptive Index. 200 employees from the same 30 restaurants were participated in the survey. Results were analyzed by using SAS program in terms of t-test, one-way ANOVA, and Pearson's correlation. The results are as follows: 1. The factor of quality got the highest score and cleanliness the second, service the third, and shift management got the lowest score in the evaluation of management practiecs of hamburger chain restaurant. The foreign brands showed significantly higher scores than the domestic ones in every measured factors. 2. The factor of coworkers got the highest score and then supervision, the work-itself, promotion and pay got the next highest scores in descending order in measured Job satisfaction. The foreign brands showed significantly higher scores than domestic ones in JDI. 3. The foodservice employees rated work-itself was considered by employees the most important job satisfaction factor, and the next important factor was pay and then coworkers, supervision, and promotion. 4. The significant positive relationship between management practice factor of shift management and all the factors of job satisfaction except the factor of pay were found.

Research Trends of Food Chain Transfer of Nanomaterials in Freshwater and Marine Ecosystems (담수 및 해양생태계에서 나노물질의 먹이사슬전이 연구추세)

  • Nanomaterials are used in a range of fields, including industry, medicine, aerospace, and manufacturing, due to their unique and useful properties. In recent years, nanotechnology has developed rapidly, and the amount of nanomaterials used in various fields has increased consistently. As a result, nanomaterials are released into the aquatic and soil ecosystem, posing potential risks to organisms and environment. These materials can enter the cells and may cause serious damage to organisms. Furthermore, they can be transferred through trophic levels and food web, thereby leading to bioconcentration and biomagnification. In this study, we analyzed the trends in research on food chain transfer of nanomaterials and investigated the techniques used in the research. Although many studies have been underway, there is a need for further advanced studies on higher trophic levels and complex microcosm and mesocosm. Furthermore, study topics should be expanded to include various types of nanomaterials and varied species and trophic levels.
