Development and Quality Characteristics of Seasoned Dried Pen Shell Atrina pectinata Adductor (키조개(Atrina pectinata) 패주를 이용한 조미 중간수분제품의 제조 및 품질특성)

  • To develop the high-value added seafood products from a regional speciality seafood, the seasoned dried pen shell Atrina pectinata adductor (SDPA) and seasoned smoke-dried pen shell adductor (SSPA) samples were prepared, and their optimal processing conditions, quality metrics, and shelf-life characteristics were examined. SDPA and SSPA samples were produced by thawing of frozen pen shell adductor, and cutting it into 6-7 mm slices, hot-air drying ($60^{\circ}C$, 20 min) or smoking ($110^{\circ}C$, 20 min), seasoning ($4^{\circ}C$, 12 h) with seasoning powder (60% sorbitol, 15% sucrose, 16% salt and 9.0% monosodium glutamate), hot-air drying ($60^{\circ}C$, 3 h), torching, vacuum-packaging in a laminated plastic film bag, heat treating with hot-water ($85^{\circ}C$, 15 min), and cooling. The moisture content of SDPA and SSPA samples was 44.5 and 43.0%, respectively, and the water activity was 0.845 and 0.842. The total amino acids in SDPA and SSPA samples were 20,986.8 and 21,312.4 mg/100 g, respectively, and the major amino acids in both products were aspartic acid, serine, glutamic acid, proline, glycine, alanine, valine, leucine, phenylalanine, lysine and arginine. The primary minerals were Na, S, K and P. Incubating tests indicated that the quality of SDPA and SSPA samples was maintained for 30 days of storage.

  • One of yellowed rice toxins, luteoskyrin, was investigated with respect to its identification, quantitation and producibility by Penicillium islandicum isolated from deteriorated rice. 1) Luteoskyrin was best resolved by thin-layer chromatography with silica gel G plate impregnated with 0. 5 N oxalic acid and acetone : n-hexane : water (6 : 3 : 1.5, upper layer) solvent system. The isolated yellow spot showed maximum absorption bands at 426 and 448 nm and changed to purple color upon exposure to sunlight for $2{\sim}3$ hours. 2) Detection limit for luteoskyrin was 4 ppm in elution-colorimetry and 0.1 ppm in densitometry after TLC. Assuming that the tolerance for luteoskyrin in rice is set below 3.68 ppm, densitometry is usable for its screening in grain samples 3) Producibility of luteoskyrin by Pen. islandicum was shown to be 11 mg/g mycelial mat in liquid culture and 40 mg/g autoclaved rice.

Volatile Components of Cornsilk(Zea mays L.) (옥수수 수염의 휘발성 성분)

  • Volatile components of Cornsilk(Zea mays L.) were isolated by purge and trap headspace method and were analyzed by GC and GC/MSD. A total of 44 components were identified in the cornsilk volatile coponents including 9 alcohols 7 aldehydes and ketones 14 terpenes and terpene alcohols 3 pyrazines 5 hydrocarbons and 6 miscellaneous components. The major components were 2-propanol(8.08%) pen-tanol(1.82%) hexanol(2.86%) hexanal(3.68%) heptanal(7.40%) nonanal(7.93%) decanal (2.04%) $\alpha$-copaene(2.20%) limonene(1.68%) $\alpha$-selinene(1.03%) $\beta$-selinene(1.03%)

  • Seasonal variation in fatty acid profiles was examined in the visceral mass and the posterior adductor muscle of the female pen shell, Atrina pectinata. Total percentages of saturated fatty acids were similar between the two organs, and there was not a prominent seasonal change in both tissues. While the percentages of highly unsaturated fatty acids (HUFAs) in the visceral mass were higher than those in the posterior adductor muscle, monounsaturated fatty acids (MUFAs) were high in the posterior adductor muscle. HUFA contents, especially in $20: 5\omega3$, $22: 5\omega3$ and $22: 6\omega3$, markedly decreased in September in the visceral mass, and this decrease was associated with a corresponding total MUFAs in the same organ. A similar pattern of change in September was noted in the posterior adductor muscle MUFAs and HUFAs. These results indicate that $20:\omega3$ and $22:\omega3$ HUFA changes in the visceral mass and posterior adductor muscle reflect the reproductive stages in pen shell.

  • An attempt was made to analyze the factors affecting mating performance of rams in pen mating systems. Due to many types of social interactions, mating performance of rams may decrease or increase. Six intact Muzaffarnagari rams were used and divided into three dominant subordinate pairs by food and ewe competition test. For dominant rams subordinate ram became competitor and vice versa. In the first experiment, ram was exposed to ewe but the competitor ram was kept outside the pen with the facility of visibility only. In the second experiment each ram was exposed to ewe along with the competitor ram and in third experiment ram was exposed to ewes (two) along with competitor ram in observation pen. Recordings of different mating and agonistic behaviour were done in all the experiments. It was found that subordinate ram's mating behaviour was inhibited by mere presence of a dominant ram out side without physical contact. However, when ram exposed to oestrus ewe(s) along with competitor ram, both dominant and subordinate rams spent much of their time in guarding activities instead of mating. In addition dominant ram tried to curtail the subordinate ram mating by agonistic interaction like fight and butting. Different guarding activities were observed either as active or passive type depending on level of dominanace. It was concluded that in a limited space the interaction of two or more rams might affect the number of services adversely.

  • Effects of combined treatments of radiation-heat and radiation-chemicals on the survival of mycotoxin-producing molds Aspergillus flavus and Penicillum islandicum conidia were investigated. Combined treatments of gamma-radiation and heat (10 minutes at $50^{\circ}C\;or\;55^{\circ}C$) showed a synergistic effect, causing remarkable decreases in $D_{10}$ values and induction doses in the two molds. Combined treatments of gamma-radiation and chemicals (sorbic acid) had no synergistic effect. Asp. flavus was more resistant to heat and sorbic acid than was Pen. islandicum.

A Study of Exploiting Raw Material of Seasoning by Using Fish and Shells -1. On Composition of Seasoning Material in Cooking By-product- (어패류를 이용한 조미료 소재 개발에 관한 연구 -1. 패류 자숙액즙의 성분조성-)

  • The possibility of shellfish utilization of juice as seasoning materials was examined through the analyzing proximate compositions and N-containing materials. The amounts of the sucking liquid wastes were estimated as 58,819 tons for oyster, 764 tons for pen-shell, 604 tons for cockle and 3,896 tons for blue-mussel from 1985 to 1990 in Korea. Water contents ranged from 95.2 to $97.4\%$ and crude protein level was $1.0{\sim}1.4\%$. Taurine was the most abundant nitrogenous compound($40{\sim}56\%$) in extracts, and glycine($10.4{\sim}36.1\%$), alanine($4.8{\sim}8.6\%$) and glutamic acid($3.1{\sim}5.3\%$) followed. More than $40\%$ of ADP and AMP as mucleotide were contained in all shellfish samples but pen-shell and cockle had $20\%$ of inosine, also. Glycinebetaine was present in corcentrations of 139.8mg/g in oyster and $45.5{\sim}61.9mg/100g$ in pen-shell, cockle and blue-mussel. Total creatinine and trimethylamine oxide levels were $30.4{\sim}59.1mg/100g$ and $0.82{\sim}47.2mg/100g$, respectively.

Cytoprotective Effect of a Neutrase Enzymatic Hydrolysate Derived from Korea Pen Shell Atrina pectinata Against Hydrogen Peroxide -Induced Oxidative Damages in Hepatocytes (산화적 손상에 대한 키조개(Atrina pectinata) 효소 가수분해물의 간세포 보호 효과)

  • In this study, we investigated the protective effects of a Neutrase enzymatic hydrolysate derived from Korea pen shell Atrina pectinata (APN) against hydrogen peroxide (H2O2)-induced oxidative damage in hepatocytes. First, we confirmed that APN has antioxidant activities by scavenging 2,2-azino-bis (3-ethylbenzthiazoline)-6-sulfonic acid radical (ABTS+) and H2O2 and increasing oxygen radical absorbance capacity (ORAC) value. Also, the treatment of APN increased the cell viability by reducing the intracellular reactive oxygen species (ROS) production in H2O2-stimulated hepatocytes. In addition, APN decreased the sub-G1 DNA contents and the apoptotic body formation increased by H2O2 stimulation. Moreover, APN modulated the protein expression of apoptosis related molecules (Bcl-2, Bax and p53) by suppressing the activation of nuclear factor NFkB and ERK/p38 signaling in H2O2-stimulated hepatocytes. Furthermore, APN led to the activation of Nrf2/HO-1signaling known as antioxidant systems. These results suggest APN protects hepatocytes against oxidative damages caused by H2O2 stimulation.

Studies on Preparation of a Cheese-like product from Soybean Milk (콩을 이용한 치-즈제조에 관한 연구)

  • 1) Among five lactic acid bacteria examined, Str. thermophilus and Str. diacetilactis produced remarkably greater amount of acids in soybean milk than Str. lactis, Str. cremoris and L. bulgaricus. 2) Soybean milk and skimmed dry milk were combined in the ratio of 7 : 3 and were carried out in lactic acid fermentation for 24 hours at optimum temperature. The result indicated that the yield of precipitation and protein content of it were the most, the moisture content was the least and curd structure formed was considered too hard. 3) Based on these and other results, following procedure was used for manufacturing: soybean milk and skimmed dry milk were combined in the ratio of 7 : 3, heated at $121^{\circ}C$ for 20 min., cooled, added Str. thermophilus as lactic acid starter and incubated for 24 hours and $37{\pm}1^{\circ}C$. The curd was cooked, hooped, and pressed for 24 hours, to the surface of which, Penicillium caseicolum and sodium chloride were spread. During ripening of the curd at $15^{\circ}C$ and $85{\sim}90%$ RH for 21 days, Pen. caseicolum was highly developed after 7 days, pH was increased and proteolytie activity has reached to the peak point after 14 days. After 7 days of ripening total water soluble nitrogen, water soluble protein nitrogen and amino acids nitrogen were begun to increase. After 21 days of ripening total water soluble nitrogen, water soluble protein nitrogen and amino-N reached to 52%, 32% and 14% of total nitogen. In the soybean cheese, after 21 days of ripening, 17 or more kinds of amino acids were detected by two-dimentional paper chromatography. The product contained 63.2% of moisture, 17.5% of crude protein, 13.2% of crude fat, 2.8% of crude ash and 2.5% of sodium chloride.

Dietary Requirement of True Digestible Phosphorus and Total Calcium for Growing Pigs

  • Sixty healthy growing pigs ($Duroc{\times}Landrace{\times}Yorkshire$ with an average BW of 21.4 kg) were used to determine the true digestible phosphorus (TDP) requirement of growing pigs on the basis of growth performance and serum biochemical indices. Pigs were assigned randomly to one of five dietary treatments (12 pigs/diet), representing five levels of TDP (0.16%, 0.20%, 0.23%, 0.26% and 0.39%). There were three replications per treatment, with four pigs (2 barrows and 2 gilts) in each replication (2 pigs/pen) A randomized-block design was used, with pen as the experimental unit. Experimental diets were formulated to provide the 5 TDP levels with a total calcium (Ca) to TDP ratio of 2:1, and offered to pigs at 5% BW for 28 d. The total Ca contents of the five diets were 0.33, 0.38, 0.45, 0.51 and 0.79%, respectively. During the 28-d experimental period, the ADG of pigs was affected by dietary TDP levels as described by Equation 1: y = $-809,532x^4+788,079x^3-276,250x^2+42,114x-1$,759; ($R^2$ = 0.99; p<0.01; y = ADG, g/d; x = dietary TDP, %). The feed:gain ratio for pigs was affected by dietary TDP levels as described by Equation 2: y = $3,651.1x^4-3,480.4x^3+1,183.8x^2-172.5x+10.9$ ($R^2$ = 0.99; p<0.01; y = feed:gain ratio; x = dietary TDP, %). Total P concentrations in serum were affected by dietary TDP levels as described by Equation 3: y = $-3,311.7x^4+3,342.7x^3-1,224.6x^2+195.6x-8.7$ ($R^2$ = 0.99; p<0.01; y = total serum P concentration and x = dietary TDP, %). The highest ADG (782 g/d), the lowest feed:gain ratio (1.07), and the highest total serum P concentration (3.1 mmol/L) were obtained when dietary TDP level was 0.34%. Collectively, these results indicate that the optimal TDP requirement of growing pigs is 0.34% of the diet (e.g., 5.1 g/day for a 30-kg pig that consumed 1.5 kg feed daily) at a total Ca to TDP ratio of 2:1.
