한우 난포낭종에서 증가되는 섬유소원 유전자 발현 (Fibrinogen mRNA Expression Up-Regulated in Follicular Cyst of Korean Cattle)

  • 난포낭종은 소 번식 장애의 주요 원인 중의 하나이며, 다양한 유전자의 변화는 여러 세포와 조직 기능에 영향을 준다. 이러한 유전자 변화는 낭종성 난소에서도 나타날 수 있다. 이온 및 수송체와 관련된 유전자 변화가 한우의 난포낭종을 유발할 수 있을 것이라는 가설 하에 난포낭종성 난포에서 발현 변화를 보이는 유전자를 찾기 위하여 마이크로어레이 분석을 수행하였다. 마이크로어레이 분석 결과, 난포낭종성 난포에서 FGG와 LRP8이 증가하고, SLC44A4, SLC27A5, ANXA8 및 aquaporin 4는 감소하였다. 반정량적 역전사중합효소 연쇄 반응으로 마이크로어레이 분석 결과를 재확인하였다. 6개의 DEG 중 3개의 DEG(FGG, SLC44A4 및 aquaporin 4)는 마이크로어레이 분석 결과와 동일하게 증가와 감소를 보였다. 마이크로어레이와 역전사중합효소 반응에서 동일한 결과를 보이는 3개의 유전자 중 가장 크게 변화를 보인 섬유소원에 중점을 두고 연구를 수행하였다. 마이크로어레이와 역전사중합효소 연쇄 반응은 난포낭종성 난포에서 섬유소원 유전자 발현을 각각 8.4배와 1.7배 증가시켰다. 그러나 난포 및 과립층세포에서 섬유소원의 단백질 양은 웨스턴 블랏 분석으로 분석한 결과, 정상에 비하여 낭종에서 유의한 차이를 보이지 않았다. 본 연구에서 섬유소원은 유전자와 단백질 발현에 있어 상관관계는 보이지 않았지만 섬유소원 유전자는 정상 조직으로부터 난포낭종을 구별하는데 있어서 중요한 생물표지자가 될 수 있는 가능성을 제시한다.

난소낭종 및 자궁내막염 한우에서 염증유래 유전자 발굴 (Identification of Inflammation-related Genes Altered in the Cystic Ovary and Endometritis of Korean Cattle)

  • This study was carried out to investigate inflammation-related gene expression altered in ovary and endometrium of Korean cattle with reproductive disorders using microarray. In the present study, nine inflammation-related differential1y expressed genes (DEGs) were identified in the cystic ovary and endometrium with endometritis. In the follicular cyst, eotaxin and alpha-2-HS-glycoprotein (AHSG) were up-regulated, whereas complement component 3 (C3) and oxidised low density lipoprotein (lectin-like) receptor 1 (OLR1) were down-regulated. Complement component 4A (C4A) was up-regulated in luteal cyst. In the endometritis, chemokine 1igand l and 2 (CXCL1 and CXCL2), protein C (inactivator of coagulation factors Va and VIIIa), and complement component C5 were up-regulated, whereas kininogen was down-regulated. Of these genes, we focused on eotaxin and kininogen, which were highly regulated in the follicular cyst and endometritis, respectively and on C3 commonly regulated in both reproductive disorders. The microarray data of eotaxin, kininogen, and C3 were validated by semi-quantitative PCR. Consistent with microarray data, eotaxin was up-regulated by 4-fold in the follicular cyst, while kininogen was down-regulated by 5-fold in the endometritis. C3 was down-regulated in the both follicular cyst and endometritis. Our results suggest that these inflammation-related genes could be useful markers for diagnosis of cystic ovary and endometritis of Korean cattle.

  • Eighteen multiparous and 8 nulliparous cows with cystic ovarian disease (19 having follicular cysts, 7 with luteal cysts) were administered either 3000 I. U. HCG or $300{\mu}g$ LHRH. Ten animals responded satisfactorily leading to normal estrus within a period of 15 to 30 days and subsequent conception. In 8 animals cysts recurred and no effect of treatment was observed in 3 patients. Plasma progesterone concentration was significantly lower in animals having follicular cysts compared to those with luteal cysts.

초음파검사에 의한 소의 번식장애 감별진단 및 치료법 개발 II. 무발정우의 감별진단 (Development of Differential Diagnosis and Treatment Method of Reproductive Disorders Using Ultrasonography in Cows II. Differential Diagnosis of Subestrous Dairy Cows)

  • Accuracy of rectal palpation and ultrasonography for differential diagnosis of subestrous dairy trows were investigatedl using the result of pIRsma progesterone assay. The ovaries were examined 2 times of 10 days interval in 520 posearom and postinsemination subestroHs dairy cows, using rectal palpation and B-mode transrectal ultrasonography. The results of rectal palpation, ultrasonographic examination and measurement of plasma progesterone profiles in 520 subestrous dairy cows were silent brat or error of estrus detection 303 (58.3%), persistent corpus luteum 59 (11.3%), follicular cyst 37 (7.1%), luteal cyst 16 (3.1%), inactive ovary 9 (1.7%), granulosa tumor 1 (0.2%), hydmsalphinx 1 (0.2%), endomehris 81 (15.6%), pyometra 12 (2.3%) and mummified fetus 1 (0.2%), respectively. Accuncy of rectal palpation and ultrasonography for diagiosing ovarian disordeir based on plasma progesterone profiles were silent heat or error of estrus detection 80.5% and 96.7%$\boxUl$ persistent corpus luteum 57.6% and 94.9%, follicular cyst 62.5% and 91.9%1 luteal cyst 62.5% and 87.5%, maclive ovary 55.6% and 88.9% and granulosa cell tumor 100% and 100%, respectively. Acnuucy of rectal palpation for diagnosing uterine disorders based on ultrasonography was pyometra 75.0%1 endometritis 51.9% and mummified fetus 100%, respectively. Cbaracteristic ultrasonographic appearances of ovaries in subestrous dairy cows were as follows; Silent heat or error of estrus detection: anechoic follicle or hypoechoic corpus luteum than ovarian stroma was alternately present on Day 0 (first examination) and Day 10. Follicular cyst: uniformly nonechogenic ovarian structure $\geq $ 25 mm in diameter with a wall < 3 mm was present in ipsilateral on Day 0 and Day 10. Luteal cyst: luteal cyst was similar to follicular cysts but thickness of cystic wall was $\geq $ 3 mm. Inactive ovary : structures within ovaries was not present on Day 0 Bnd Day 10. Characteristic uthssonograpsc appearances of uterus in subestrous dairy cows were as follows; Endometritis: characterized by uterine lumen containing fluid in which 'snowy'echogenic particles art suspended. Pyometra: ultrasonographic appearance of pyometra was diffuse echogenic particles distributed in fluid within the distended uterus, and a thickened uterine wall. These results indicated that ultrasonography was practical far diagnosing reproductive disorders. To diagnosing ovarian disorders, ultrasonography should be carried out 2 times of 10 days interval and rndometritis should be differentiated with uterus of luteal phase in normal cycling cows.

하악골에 발생한 낭종에 관한 임상적 연구 (A Clinical Study of cysts on Mandible)

  • Cystic lesions of the mandible are fairly common and usually a presumptive diagnosis is made readily, more often on the basis of roentgenographic appearance than by any other means. Occasionally, however, it is easy to fall into errors of diagnosis which may affect one's choice of treatment. The diagnosis in these situations are usually resolved by the histopathologic examination of material obtained by surgical exploration. The present discussion to the lesions which can be most easily confused in the roentgenographic interpretation, that is, the traumatic bone cyst, the dentigerous cyst, the radicular cyst. These lesions often grow to considerable size before they cause any subjective or objective symptoms. Less frequently, perhaps, the patient presents himself with the complaint of enlargement of the affected part, a discharge, or pain. On rare occasions the first sign is fracture through the cyst cavity. In any case, an adequate roentgenographic survey by means of extraoral films is essential. This is the primary means of diagnosis. The three lesions in mandible, reported here, resembled each other roentgenographically in that they were osteolytic lesions. The follicular and radicular cysts usually have a smooth periphery and may be surrounded by a white line. The follicular cyst, slow-growing lesions, is usually associated with an unerupted tooth. The radicular cyst, the most common type of the odontogenic cyst, is always associated with a nonvital tooth, or it may persist as a "residual" radicular cyst after the causative tooth has been extracted. The traumatic bone cyst can often be differentiated from the first two in that the periphery is less definite and is irregular. It is thought that because it does not occur in older people, the lesion is self-limiting and heals spontaneously if left alone.

도축 한우에서 난소낭종의 기능성 평가 (Evaluation of Functional Status of Ovarian Cysts in Slaughtered Korean Native Cows)

  • 도축장에서 도축된 한우 중 난소낭종에 이환된 소 86두를 대상으로 수침법으로 초음파 검사를 실시하여 낭종의 직경, 낭종내강의 직경, 낭종벽의 두께를 측정하였으며 낭종액 중 $P_4$농도와 $E_2$농도를 측정하여 다음과 같은 결과를 얻었다. 난소낭종에 이환된 소 86두는 해부학적 및 초음파 검사에 의하여 단포성 난포낭종 51두(59.3%), 다포성 난포낭종 19두(22.1%), 단포성 황체낭종 13두(15.1%), 다포성 황체낭종 3두(3.5%)의 발생률을 나타내어 황체낭종보다 난포낭종의 발생률이 더 높았다. 해부학적 및 초음파 검사에 의해 분류된 난소낭종은 낭종액 중 호르몬 농도 측정에 의하여 기능성 난포낭종 54두, 기능성 황체낭종 16두, 비기능성 낭종 16두로 나타났으며 황체낭종에서는 모두 황체낭종으로 확인되었지만 난포낭종에서는 70두 중 16두가 기능을 하지 않은 낭종으로 확인되었다. 기능을 하지 않은 낭종을 가지고 있는 16두에서 단포성 낭종에서는 13두가, 다포성 낭종에서는 3두가 비기능성 낭종을 가지고 있었다. 난소낭종으로 진단된 예에서 낭종벽의 두께와 혈중 $P_4$ 농도 사이에는 유의성 있는 양의 상관 관계를 나타내었다(p<0.05). 또한 낭종벽의 두께와 낭종액 중 $P_4$ 농도와의 상관 관계는 난포낭종이 0.59(p<0.001), 황체낭종은 0.65(p<0.001), 비기능성 낭종은 0.52(p<0.06) 이었으며 난포낭종과 황체낭종 전체를 대상으로 하였을 때는 0.69로 유의성 있는 양의 상관관계를 나타내었다(p<0.001). 이상의 결과로 볼 때 난소낭종에는 그 변성 과정이 다양한 단계로 나누어지며 황체낭종은 초음파 검사에 의해 낭종벽의 두께를 측정함으로써 감별진단이 가능하지만 난포낭종은 낭종의 크기와 낭종벽의 두께만으로 감별 진단하는 데는 한계가 있음을 확인하였다. 따라서 난소낭종 특히 난포낭종의 진단에 있어서 단순히 형태학적인 소견에 의해 낭종을 진단하기보다는 임상증상과 더불어 종합적인 진단이 필요하다고 생각된다.

난소낭종 시 변화되는 세포사멸 관련 유전자 (Apoptosis-related Genes Altered in Bovine Cystic Ovary)

  • 본 연구는 한우의 난포낭종 및 황체낭종에서 세포자멸사 관련 유전자의 발현 변화를 조사함으로써 난소낭종과 세포자 멸사간의 관계를 정리하고자 한다. 마이크로어레이 분석 결과, 난포낭종에서 PIK3R2와 AKT1가 유의하게 증가하였고, 황체낭종에서는 증가되는 세포자멸사 관련 DEGs, TNF-RAF2, PRLR, FOXL2, STK4 및 COL4A3와 감소하는 DEGs, INHA, CIDEB, BCL10 및 FASLG가 포함되었다. 정량적 역전사 중합 효소 반응을 이용하여 mRNA 발현 변화를 검증한 결과 INHA, CIDEB, BCL10 및 FASLG만 마이크로어레이와 동일한 결과를 보였다. INHA의 경우 마이크로어레이 결과에서는 1.43배 감소하고 정량적 역전사 중합효소 반응 결과에서는 12.3배 감소하였다. 세포사멸 확인을 위하여 TUNEL 형광염색과 ERK, JNK, p38, PI3K 및 Akt/PKB의 웨스턴 블랏 분석을 실시한 결과 정상난소와의 유의한 차이를 확인할 수 없었다. Caspase의 활성 및 Bax, Bcl-2, Bcl-xL의 단백질 분석 역시 정상난소와 유의한 차이를 보이지 않았다. 오히려 PI3K와 p-Akt의 발현이 정상에 비하여 감소하는 경향을 보였다. 이러한 결과들로 미루어 보아 난소낭종 시 볼 수 있는 세포자멸사의 낮은 발현율은 세포 사멸인자의 발현 감소로 나타날 수 있을 것이라 생각된다.

번식장애 한우의 렙틴 및 인슐린 농도 변화 (Alteration in Concentrations of Leptin and Insulin in Korean Cattle with Reproductive Disorders)

  • The objective of this study is to investigate the changes in concentrations of leptin and insulin in serum of Korean cattle (Hanwoo) with reproductive disorders and to examine the relationship among leptin, insulin, and body condition score (BCS). The concentration of leptin in serum of pregnant Hanwoo showed insignificant difference from that in serum of Hanwoo with reproductive disorder, such as repeat breeding, follicular cyst, corpus luteum cyst, ovarian atrophy, and feeble estrus (p>0.05). However, the concentrations of leptin and insulin in serum were changed with different BCS value. In emaciated Hanwoo (BCS $2.0\sim2.9$), they were significantly decreased compared to BCS $3.0\sim3.4$ (p<0.05). The leptin showed different genotypes with different BCS value. In BCS $2.0\sim2.9$, C/T genotype was expressed (83.3%) more than C/C (16.7%) or T/T (0%) genotype, whereas C/C genotype was expressed (62.5%) more than C/T (25.0%) or T/T (12.5%) genotype in BCS $3.5\sim4.0$. The insulin concentration in follicular fluid obtained from ovary with follicular cyst which has follicles having diameter of $25\sim40 mm$ was significantly higher (p<0.05) than those in normal follicle fluid which has follicles having diameter of $3\sim10 mm$. These results showed that concentration of leptin and insulin in serum were related to BCS value and follicular size and suggest that the changes in concentration of leptin and/or insulin in serum could be a potent biomarker for diagnosis of bovine reproductive disorder.

번식 장애 한우의 혈액요소태질소 및 성 스테로이드 호르몬 농도 변화 (Alteration in Concentrations of Blood Urea Nitrogen and Sex Steroid Hormone in Korean Cattle with Reproductive Disorders)

  • The objective of this study is to investigate the changes in concentrations of blood urea nitrogen (BUN) and sex steroid hormones, such as estrogen, progesterone, and testosterone in Korean cattle (Hanwoo) with reproductive disorders and to examine the relationship between BUN and body condition score (BCS) in Hanwoo. The concentration of BUN was 16.2 mg/dl, 17.8 mg/dl, 15.1 mg/dl, 17.9 mg/dl, and 28.3mg/dl in pregnancy, repeat breeding, follicular cyst, luteal cyst, and ovarian atrophy, respectively. In Hanwoo with BCS $2.0{\sim}2.9$, $3.0{\sim}3.5$ and $3.6{\sim}4.0$, the concentration of BUN was 15.8 mg/dl, 17.0 mg/dl, and 17.6 mg/dl, respectively. Fluoroimmunoassay showed that serum estrogen and progesterone levels were decreased in reproductive disorders Hanwoo, such as ovarian atrophy, endometritis, and weak estrus. The testosterone level was significantly decreased in Hanwoo with reproductive disorders compared to that in pregnant Hanwoo ($0.02{\sim}0.03\;ng/ml$ vs 0.13 ng/ml, p<0.05). The progesterone and estrogen concentrations in follicular fluid obtained from ovary with follicular cyst were significantly higher (p<0.05) than those in normal follicle fluid. These results show that there is no relationship between BUN and BCS in Hanwoo, and the concentration of sex steroid hormone in serum and follicular fluid are changed in reproductive disorders Hanwoo.

한우의 정상 난포와 난포낭종 난포에서 Aquaporin7 발현 양상 (Patterns of Aquaporin 7 Expression in Normal Follicles and Follicular Cyst Follicles of Hanwoo)

  • Alteration in ion channel or transporter expression levels affects cell volume which is produced by movement of water and ion across the plasma membrane. In particular, aquaporin (AQP) channels among ion channels play a crucial role in movement of water across the cell membrane. This study was performed to identify whether AQP expression is changed in bovine follicular cystic follicles using microarray, RT-PCR and Western blotting analyses. In microarray data, AQP4 expression was decreased, whereas AQP7 was increased in cystic follicles. Additional experiments were focused on the AQP7 expression increased in cystic follicles. The microarray data was confirmed by semi-quantitative polymerase chain reaction (PCR) and Western blot analysis. AQP7 mRNA and protein expressions were significantly increased in the cystic follicles (p<0.05). Application of estrogen ($10{\mu}g/ml$) to bovine ovarian cells showed a trend of increase in AQP7 expression. From these results, we suggest that the increase in AQP7 expression in cystic follicles may play an important role in movement of water in bovine ovary. In addition, AQP7, a aquaglyceroporin permeating water and glycerol, could be a good target in development of methods for the cryopreservation of bovine ovary.
