천연자원의 라디칼 소거능과 최종당화산물의 생성저해활성 검색 (Screening of Natural Resources with Inhibitory Activity on Free Radicals and Advanced Glycation end Products (AGEs) Formation)

  • Reactive oxygen species (ROS) and formation of advanced glycation end products (AGEs) play an important role in the pathogenesis of diabetic nephropathy by hyperglycemia. To find natural agents improving diabetic nephropathy, 63 natural resources which used to the treatment of diabetes mellitus in a folk remedy were investigated with an in vitro system employing radical scavenging activity and inhibitory activity of AGEs formation. In results, the extracts of Aspalathus linearis, Rubus coreanus, Rosa rugosa, and Epimedium koreanum significantly inhibited the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical with $IC_{50}$ values less than $10{\mu}g/ml$. The extracts of Zea mays, Cucurbita moschata, Cudrania tricuspidata, and Aspalathus linearis effectively reduced the formation of AGEs compared with the positive control $N-acetyl-_L-cystenine$ (NAC) and aminoguanidine (AG). In addition, the extracts of Aspalathus linearis, Commelina communis, Cornus officinalis, and Lespodeza cuneata showed the all inhibitory activity against DPPH radical and AGEs formation. Also, these resources definitely showed the radical scavenging activity against peroxynitrite $(ONOO^-)$ and hydroxyl radical $({\cdot}OH)$ relating to high glucose-induced ROS production. Thus, these results suggest that some natural resources may regulate the initiation and progression of diabetic nephropathy through inhibition of ROS production and AGEs formation.

손바닥 선인장 열매 및 줄기 추출물의 생리활성(I)-일반약리검색 (Screening on Biological Activities of the Extracts from Fruit and Stem of Prickly Pear(Opuntia ficus-indica var. saboten))

  • Prickly pear(Opuntia ficus-indica var. saboten Makino, Cactaceae) is a tropical or subtropical plant, which is widely used as folk medicine for burned wound, edema and indigestion. Screening on the biological properties of the fruits(OFS-Fr) and stems(OFS-St) of the plant was carried out to prove the pharmacological significance. By hot plate and acetic acid-inducing writhing methods, significant analgesic effects of OFS-Fr and OFS-St were found in mice and anti-edemic effect was observed in carrageenin-induced inflammatory rats. However, the extracts showed no significant actions on central and autonomic nervous system and blood circulatory system, which imply no toxic effects to animal.

갓 지상부에서 플라보노이드와 고급 알콜 화합물의 분리 (Isolations of Flavonoids and a Higher Alcohol from the Aerial Parts of Brassica juncea)

  • The aerial parts of Brassica juncea (Cruciferae) called by brown or oriental mustard have been widely used as a spice in food and also traditional folk medicine as stimulant, diuretic and expectorant agent. And Gatkimchi made of the aerial parts of this plant are very popular in Korea. The aerial parts of this plant was refluxed with MeOH and then fractionationed with $CH_2Cl_2$, EtOAc, n-BuOH and $H_2O$, successively. One higher alcohol compound and three flavonoids were isolated from the EtOAc fraction through silica gel and Sephadex LH-20 column chromatographies. Their structures were elucidated as n-hexacosanol(1), kaempferol(2), isorhamnetin $3-O-{\beta}-D-glucopyranoside(3)$ and isorhamnetin $3-O-(6'-O-acetyl)-{\beta}-D-glucopyranoside(4)$ by comparison of spectral data with those in references. And compounds 1 and 4 were firstly isolated from this plant.

나군대 잎의 약리 효과에 관한 연구 (Pharmacological Actions of Crinum folium)

  • Crinum asiaticum var. japonicum is a wild plant growing only in Jeju-island, Korea, and in Japan. The whole part of this plant has been known to have the pharmacological actions such as analgesic, anti-inflammatory, platelet-aggregation inhibitory, antitussive, and expectorant. With these assumed actions, the leaves (Crinum folium) of this plant has been used in the folk remedies for arthritis and arthralgia. There is, however, no scientific evidences for the pharmacological actions of Crinum asiaticum var. japonicum. In the present study, the analgesic, anti-inflammatory, and platelet-aggregation inhibitory actions of Crinium folium were evaluated using writhing test, tail-flick test, carrageenin antiedema test, in vitro thromboxane $B_2$ quantitation assay and in vitro platelet aggregation test. In order to obtain the partially purified fraction whose pharmacological action is excellent, the methanol extract of Crinium folium was fractionated consecutively into four biological fractions such as ether, ethyl acetate, butanol, and water fractions and their pharmacological actions of the fractions were investigated. Putting our results together, Crinium folium, especially ethyl acetate fraction was proven to have significant analgesic, anti-inflammatory and platelet-aggregation inhibitory actions by inhibition of prostanoids biosynthesis as one of its mechanism of action.

Anti Angiogenic Effects of Isorhamnetin Isolated from Persicaria thunbergii

  • Persicaria thunbergii has been utilized for the treatment of cancer as a folk medicine. We examined the effect of isorhamnetin, a flavonoid isolated from Persicaria thunbergii, on angiogenesis in vitro and in vivo. Basic fibroblast growth factor (bFGF) is a potent angiogenic factor found in various tumors. In this study, we found that the isorhamnetin decreased bFGF-induced human umbilical vein endothelial cells (HUVECs) proliferation and migration in a concentration-dependent manner (5, 10 and $20\;{\mu}M$) whereas, it did not inhibit bFGF-induced capillary-like formation of HUVECs. The chicken chorioallantoic membrane assay revealed that addition of isorhamnetin (10, 20 and $40\;{\mu}M$) displayed an antiangiogenic effect in vivo. These results suggest that the isorhamnetin inhibits the proliferation and migration of endothelial cells induced by bFGF, which may explain its anti-angiogenic properties.

제주 성읍민속마을의 귀화식물 분포현황 및 관리방안 (A Study on the Distribution Status and Management Measures of Naturalized Plants Growing in Seongeup Folk Village, Jeju Island)

  • 제주시 성읍민속마을을 대상으로 관속식물상 및 귀화식물의 현황을 조사하고, 여타 민속마을과 제주도 전역에 분포하는 귀화식물의 분포양상 및 발생특성과 비교 고찰함으로써 이곳의 귀화식물의 관리방안을 모색할 목적으로 시도된 본 연구의 결론은 다음과 같다. 성읍민속마을의 관속식물상은 93과 260속 298종 44변종 12품종으로 총 354분류군이 확인되었으며, 이 중 귀화식물은 22과 46속 53종 2변종의 총 55분류군으로 이는 제주도 전역에서 확인된 총 254분류군의 21.7%에 해당된다. 이에 따른 성읍민속마을의 귀화율은 15.5%로서 이는 경북 안동의 하회마을, 경주의 양동마을, 성주의 한개마을, 강원 고성의 왕곡마을 그리고 충남 아산의 외암마을에 비해 월등히 높은 수치였다. 마을내 발생한 귀화식물 중 분포지가 제주도에 한정되어 있는 것은 양장구채, 국화잎아욱, 애기달맞이꽃, 긴잎달맞이꽃, 솔잎미나리, 자주풀솜나물, 선풀솜나물, 큰참새피, 등심붓꽃 등 9분류군으로, 귀화식물의 이입 관문이자 선구 발생지로서의 특성을 살린 적절한 관리전략이 필요한 것으로 판단된다. 한편 본 연구를 통해 제주도 기준목록에는 포함되지 않았던 까락빕새귀 리, 삼, 눈개불알풀 등 3분류군이 성읍민속마을에서 처음으로 확인되었다. 단위문화재 정원내 발생한 귀화식물은 총 20분류군으로 이 중 유럽점나도나물의 출현율이 62.5%로 가장 높았으며, 미국자리공 선개불알풀 망초 방가지똥 큰방가지똥(37.5%)도 비교적 높게 나타났는데 이는 제주도뿐만 아니라 대부분의 민속마을과 레크레이션공간의 공통된 특성으로 보인다. 한편 경관관리적 취급이 요망되는 식물군락은 유채를 비롯하여 귀화식물인 코스모스, 토끼풀, 잔개자리, 긴잎달맞이꽃, 애기달맞이꽃, 서양벌노랑이, 호밀풀, 양장구채, 서양금혼초, 미국질경이, 큰이삭풀, 유럽점나도 나물 등으로 분석되었다. 성읍민속마을의 단기적 귀화식물 관리방안으로는 경관농업적 차원에서 식재되고 있는 코스모스와 유채의 임내 침투 현황과 개화 후 활용 방안의 모색이 필요하다. 환경부가 지정한 생태계교란야생식물인 돼지풀과 서양금혼초는 우선적으로 제거하고, 재발에 대비한 추가 발생과 확산과 팽창을 염두에 둔 모니터링의 실시를 제안한다. 특히 서양금혼초는 서식밀도 파악 및 제근 등 물리적인 방제방안이 강구되어야 할 것이다. 또한 시각적으로 녹시율이 높은 정의읍성 외벽에 자생하는 방가지똥, 약모밀, 주홍서나물, 개망초, 자주광대나물의 제거가 시급하다. 이와 함께 폐 공가에 귀화식물의 분포와 우점도가 높음에 따라 공가에 대한 보존관리 및 민박 활용방안 등을 강구해야 할 것이다.

Changes in Microbial Diversity, Methanogenesis and Fermentation Characteristics in the Rumen in Response to Medicinal Plant Extracts

  • This study evaluated the in vitro effect of medicinal plant extracts on ruminal methanogenesis, four different groups of methanogens and ruminal fermentation characteristics. A fistulated Holstein cow was used as a donor of rumen fluid. Licorice and mugwort extracts (Glycyrrhiza uralensis and Artemisia capillaris, 0.5% and 1% of total substrate DM, respectively), previously used as folk remedies, were added to an in vitro fermentation incubated with buffered-rumen fluid. Total gas production in Glycyrrhiza uralensis extract treatment was not significantly different between treatments (p<0.05) while total gas production in the Artemisia capillaris extract treatment was lower than that of the control. Artemisia capillaris extract and Glycyrrhiza uralensis extract reduced $CH_4$ emission by 14% (p<0.05) and 8% (p<0.05), respectively. Ciliate-associated methanogens population decreased by 18% in the medicinal plant extracts treatments. Medicinal plant extracts also affected the order Methanobacteriales community. Methanobacteriales diversity decreased by 35% in the Glycyrrhiza uralensis extract treatment and 30% in the Artemisia capillaris extract treatment. The order Methanomicrobiales population decreased by 50% in the 0.5% of Glycyrrhiza uralensis extract treatment. These findings demonstrate that medicinal plant extracts have the potential to inhibit in vitro ruminal methanogenesis.

전통민가와 사찰에 나타난 꽃담의 형태와 상징성 연구 - 전라북도 문화재를 중심으로 - (Comparative Study on the Shape and Symbolism of Flowered-Wall in Tradition Private Houses and Temples - Focusing on the Designated Cultural Properties of Jeollabuk-do Province -)

  • 건물의 벽이나 담에 여러 가지 문양을 넣은 담장을 '꽃담' 또는 '화초장(花草墻), 화문장(花紋牆)'이라고 한다. 꽃담을 통해서 외적으로는 소유자의 권위, 가치관 등을 짐작할 수 있고, 내적으로는 집안의 안녕과 평화를 기원하는 상징적 의미를 추적할 수 있기도 하다. 그동안 꽃담 관련 연구의 경우 궁궐은 건축, 원예, 미술 디자인 분야 등에서 다양하게 진행된 반면 지역성과 관련한 전통민가와 사찰 관련 연구는 조경분야에서 전무한 상태이다. 이러한 관점에 착안하여 본 연구는 국가 및 도지정 문화재인 전라북도 소재 전통민가와 사찰의 꽃담 사례를 조경적 관점에서 조사, 분석하였다. 전수 조사한 결과 꽃담이 도입된 전통민가 9개소와 사찰 7개소를 중심으로 연구분석하였다. 즉, 전라북도에 산재하는 문화재 16개소를 중심으로 전통민가와 사찰에 나타나는 꽃담의 형태와 기능, 재료, 문양의 상징적 의미를 추적하여 연구결과를 도출하였다. 꽃담은 위치와 형태에 따라 공간기능과 성격이 차별화 되며, 문양의 경우 담장은 물론 합각, 고막이, 굴뚝 등에 다양하게 나타난다. 민가에서는 식물은 물론 문자문양까지 다양한 반면 사찰에서는 불교의 교리와 신앙 등의 의미를 지닌 신앙관련 문양이 주로 나타난다. 상징적으로 민가의 경우 가문과 자손 번영, 벽사, 장수, 길수(吉數) 등이 나타나며, 사찰에서는 불성(佛性)과 극락왕생(極樂往生)을 기원하고 연꽃의 속성을 강조하는 등 의미적 차별성이 표출된다. 결과적으로 본 연구를 통해 향토 조경문화의 올바른 조명, 현대 공간에 꽃담을 모티브로 한 한국적 정취의 표출 가능성, 그리고 꽃담에 대한 인식 제고와 계승 방안 모색을 위한 기초자료 제공 등과 같은 연구 성과를 도출할 수 있었다.

Preliminary pharmacognostical and phytochemical evaluation of Stachys tibetica Vatke

  • Stachys tibetica Vatke (Lamiaceae) is an important medicinal plant in the folk medicine of Ladakh, India and Tibet for the treatment of various mental disorders. Infusion and decoction of the whole plant is used as a cup of tea for a severe fever, headaches and to relieve tension. The recent study is aimed to evaluate the preliminary pharmacognostical and phytochemical nature of Stachys tibetica Vatke. The whole plant material was subjected to successive soxhlet extraction with petroleum ether (40 - $60^{\circ}C$), chloroform, ethyl acetate, methanol and finally decocted with water to get the respective extracts. The fluorescence characteristics of the powdered materials were analysed under ultraviolet light and ordinary light. Different physicochemical parameters such as ash value, extractive value, foaming index, pH values, loss on drying and determination of foreign matter were carried out as per WHO guidelines. The total fat, flavonoid, saponin and volatile contents were also determined. Macroscopical studies revealed the authentication of the plant drug. Physicochemical parameters helped to standardize the plant material while preliminary qualitative chemical tests of different extracts showed the presence of Glycosides, Carbohydrates, Phytosterols/triterpenoids, Saponins, Fixed oils, Fats and phenols/tannins. Quantification of the total flavonoids and saponins and contents were determined as $54.66{\pm}0.58mg/g$ and $75.42{\pm}0.48mg/kg$ respectively, while the volatile and fat contents were 6.5% and 0.7% respectively. Results may lay the foundation for the standardization of the drug and discovery of new molecules from S. tibetica for the treatment of various diseases.

Chemical composition of different parts of ramie (Boehmeria nivea)

  • Ramie (Boehmeria nivea) is a perennial herbaceous plant belonging to the family Urticaceae. It was used in folk remedies for diuretic or anti-pyretic purposes and as an hepatoprotective, anti-oxidant, and anti-inflammation agent. In this study, we investigated the composition of free sugars, amino acids, fatty acids, organic acids, and total polyphenol contents in parts of ramie (root, stem, and leaf) and different harvest areas (10 areas). Overall, free sugars were found as sucrose, glucose, fructose, and galactose at concentrations ranging from 24.5 to 1173.8 mg/g. Amino acids lysine, threonine, tyrosine, phenylalanine, and histidine were detected at concentrations of 33.8 to 3735.3 mg/g. Major fatty acids were linoleic acid, palmitic acid, and linolenic acid at concentrations of 7.3 to 364.4 mg/g. In organic acids, the concentration of malic acid was highest of all with 672.2 mg/g. The content of free sugars, amino acids, fatty acids, organic acids, and total polyphenol was higher in leaves than in roots and stems. In contrast, the total polyphenol content was higher in roots with 33.7 - 219.4 mg/g than in leaves or in stems. As seen in collecting region, the contents of free sugars, amino acids, fatty acids, organic acids, and total polyphenol of Bn-33, 39, 55, 65, and 90 were higher than other samples. Chemical compositions of different parts of ramie are important factors to consider in manufacturing functional foods. The results of this study provide fundamental information on the chemical compositions of ramie parts and would help develop new functional foods from ramie leaves or the whole plant.
