Malaria Parasite Separation from White Blood Cells Using Conductive Liquid-Based Standing Surface Acoustic Wave (CL-SSAW) (전도성 액체기반 정상표면탄성파(CL-SSAW)를 이용한 백혈구로부터의 말라리아 기생충 분리)

  • An acoustofluidic device using conductive liquid-based electrodes was developed for malaria parasite separation from white blood cells. In this device, the electrode channels filled with a conductive liquid were used to generate standing surface acoustic waves (SSAWs) in a fluidic channel, which can overcome the limitation of conventional patterned metal electrodes. Separation performance of the device was evaluated using fluorescent polystyrene particles with two different sizes (2 and $10{\mu}m$ diameters), which were successfully separated. In addition, a mixture of malaria parasites and white blood cells were also efficiently separated with high purity of ~98% in the CL-SSAW device at the flow rate of $12{\mu}l/min$.

  • We performed a dynamic characterization of passive flow-rate regulators, which compensate for inlet pressure variation and maintain a constant flow rate for precise liquid control in microfluidic systems. To measure the flow rate for a short time, much less than the period of the dynamic inlet pressure, we use the particle image velocimetry (PIV) method. DI water containing fluorescent beads with a $0.7-{\mu}m$ diameter was supplied to the flow-rate regulators, and two successive images of the particles were taken by a pulse laser and a fluorescent microscope to measure the flow velocity. For a dynamic inlet pressure of frequency 60 Hz, the flow velocity was constant with an average of 0.194 ${\pm}$ 0.014 m/s as the inlet pressure varied between 20 kPa to 50 kPa. The flow-rate regulators provided a constant flow rate of $5.82{\pm}0.29\;{\mu}l/s$ in the frequency range of the inlet pressure from 1 Hz to 60 Hz.

  • Nano sized ZnO particle as 20-30nm applies for material, pigments, rubber additives, gas sensors, varistors, fluorescent substance as well as new material such as photo-catalyst, sensitizer, fluorescent material. ZnO with a particle size in the range 20-30nm has provided to be an excellent UV blocking material in the cosmetics industry, which can be used in sunscreen product to enhance the sun protection factor and natural makeup effect. But pure ZnO particles application limits for getting worse wearing feeling. We make high-functional inorganic-composite that coated with nano-ZnO on the plate-type particle such as sericite, boron nitride and bismuthoxychloride. In this experiment, we synthesized composite powder using hydrothermal precipitation method. The starting material was ZnCl$_2$ Precipitation materials were used hexamethylenetetramine(HMT) and urea. We make an experiment with changing as synthesis factors that are concentrations of starting material, precipitation materials, nuclear formation material, reaction time, and reaction temperature. We analyzed composite powder's shape, crystallization and UV-blocking ability with FE-SEM, XRD, FT-IR, TGA-DTA, In vitro SPF test. The user test was conducted by product's formulator. In the results of this study, nanometer sized ZnD was coated regardless of the type of plate-powder at fixed condition range. When the coated plate-powders were applied in pressed powder product, the glaze of powder itself decreased, but natural make-up effect, spreadability, and adhesionability were increased.

Deterministic Lateral Displacement as a Function of Particle Size Using a Piecewise Curved Planar Interdigitated Electrode Array (다중예각 평면 교차전극을 이용한 입자 크기에 따른 측면방향 변위)

  • This paper presents the lateral displacement of a particle passing over a planar interdigitated electrode array at an angle as a function of particle size. The lateral displacement was also measured as a function of the angle between the electrode and the direction of flow. A simplified line charge model was used for numerical estimation of the lateral displacement of fluorescent polystyrene (PS) beads with three different diameters. Using the lateral displacement as a function of particle size, we developed a lateral dielectrophoretic (DEP) microseparator, which enables the continuous discrimination of particles by size. The experiment using an admixture of 3-, 5-, and $10-{\mu}m$ PS beads showed that the lateral DEP microseparator could continuously separate out 99.86% of the $3-{\mu}m$ beads, 98.82% of the $5-{\mu}m$ beads, and 99.69% of the $10-{\mu}m$ beads. The lateral DEP microseparator is thus a practical device for the simultaneous separation of particles according to size from a heterogeneous admixture.

Study on Persistent Infection of Japanese Encephalitis Virus Beijing-l Strain in Serum-free Sf9 Cell Cultures

  • Sf9 cells have obvious advantages for the conventional production technology of vaccine. They are useful tools for high concentration and large-scale cultures. Sf9 cells were grown to maximal concentration, 8${\times}$l0$\^$6/ cells/$m\ell$ in a 500$m\ell$ spinner flask, with a doubling time at the exponentially growing phase of 24.5 hours, using serum-free media. To explore the ability of Sf9 cells to be infected by the Japanese encephalitis (JE) virus Beijing-l strain, Sf9 cells were infected with the virus. By 4-5 days post-infection, 10-15 % of the Sf9 cells showed cytopathic effect (CPE), from granularity to the formation of syncytia and multinucleated giant cells continuously observed over a period of 35 days. Positive fluorescent reactions were detected in 30-40% of cells infected with the JE virus Beijing-l strain, and the uninfected Sf9 cells were completely negative. Virus particles, propagated in Sf9 and Vero cells, were concentrated by sedimentation on 40% trehalose cushions by ultracentrifugation, and showed identical patterns of viral morphogenesis. Complete virus particles, 40 to 50 nm in diameter, were observed, and JE virus envelope (E) proteins, at 53 kDa, were found in the western blot analysis to the anti-JE virus E protein monoclonal antibody and reacted as a magenta band in the same position to the glycoprotein staining. To evaluate whether the infectious virus was produced in Sf9 cells inoculated with the JE virus Beijing-l stain, Sf9 cells were inoculated with the virus, and sample harvested every 5 days. The titers of the JE virus Beijing-l strain rose from 1.0${\times}$l0$\^$5/ to 1.5${\times}$l0$\^$6/ pfu/$m\ell$. The infected Sf9 cells could be subcultured in serum-free medium, with no change in the plaque sizes formed by the JE virus Beijing-l strain in the plaque assay. It is suggested that the ability of the JE virus Beijing-l strain to infect Sf9 cells in serum-free media will provide a useful insect cell system, where the JE virus replication, cytopathogenicity and vaccine immunogen can be studied.

  • The efficacy of berberine sulphate, a fluorochrome having binding properties with both DNA and RNA, was investigated for the detection of mycoplasma-like organisms(MLOs) in jujube(Zizyphus jujuba), paulownia(Paulownia tomentosa), mulberry(Morus alba) trees and periwinkle (Catharanthus roseus) plant. When examined under fluorescence microscope, berberine sulphate-stained sections of diseased samples showed distinct MLO-specific fluorescent particles in the phloem area, while such fluorescence was absent in the healthy ones. This staining technique was proved to be a very accurate method for the diagnosis of MLO infections in woody and herbaceous plants. Furthermore, the cheap and easy procedure could be used to test a great number of samples on MLO infections with reliability and rapidity.

  • Compared with bulk material, quantum dots have received increasing attention due to their fascinating physical properties, including optical and electronic properties, which are due to the quantum confinement effect. Especially, Luminescent CdSe quantum dots have been highly investigated due to their tunable size-dependent photoluminescence across the visible spectrum. They are of great interest for technical applications such as light-emitting devices, lasers, and fluorescent labels. In particular, quantum dot-based light-emitting diodes emit high luminance. Quantum dots have very high luminescence properties because of their absorption coefficient and quantum efficiency, which are higher than those of typical dyes. CdSe quantum dots were synthesized as a function of the synthesis time and synthesis temperature. The photoluminescence properties were found strongly to depend on the reaction time and the temperature due to the core size changing. It was also observed that the photoluminescence intensity is decreased with the synthesis time due to the temperature dependence of the band gap. The wavelength of the synthesized quantum dots was about 550-700 nm and the intensity of the photoluminescence increased about 22~70%. After the CdSe quantum dots were synthesized, the particles were found to have grown until reaching a saturated concentration as time increased. Red shift occurred because of the particle growth. The microstructure and phase developments were measured by transmission electron microscopy (TEM) and X-ray diffractometry (XRD), respectively.

The Effect of Demineralized Freeze - Dried Bone Allograft in Guided Bone Regeneration on Supra - Alveolar Peri - Implant Defects in Dogs (성견의 치조 연상 임플란트주위 결손부에서의 탈회냉동건조골과 e-PTEE막의 효과)

  • The purpose of this study was to evaluate the adjunctive combined effect of demineralized freeze-dried bone allograft(DFDB) in guided bone regeneration on supra-alveo-lar peri-implant defect. Supra-alveolar perio-implant defects, 3mm in height, each including 4 IMZ titanium plasma-sprayed implants were surgically created in two mongrel dogs. Subsequently, the defects were treated with 1 of the following 3 modalities: Control) no membrane or graft application, Group1) DFDB application, Group2) guided bone regeneration using an expanded polytetra-fluoroethylene membrane, Group3) guided bone regeneration using membrane and DFDB. After a healing period of 12-week, the animals were sacrificed, tissue blocks were harvested and prepared for histological analysis. Histologic examination were as follows; 1. New bon formation was minimal in control and Group 1, but considerable new bone formation was observed in Group 2 and Group 3. 2. There was no osteointegration at the implant-bone interface in the high-polished area of group2 and Group 3. 3. In fluorescent microscopic examination, remodeling of new bone was most active during week 4 and week 8. There was no significant difference in remodeling rate between group 2 and group 3. 4. DFDB particles were observed, invested in a connective tissue matrix. Osteoblast activity in the area was minimal. The results suggest that guided bone regeneration shows promising results in supra-alveolar peri-implant defects during the 12 week healing period although it has a limited potential in promoting alveolar bone regeneration in the high-polished area. There seems to be no significant adjunctive effect when DFDB is combined with GBR.

Dust-scattered FUV halo around Spica

  • The far ultraviolet (FUV) wavelength (900-1750A) range includes a wealth of important astrophysical information related to the cooling of hot gas, fluorescent emission from H2 molecules, and starlight scattered off dust particles. Among these, we would like to focus on the scattered emission of the central star by dust with the example of the FUV halo surrounding ${\alpha}$ Vir (Spica). While scattering properties of dust have been studied with the GALEX data, the improved dataset of STSAT-1 revealed many detailed structures of this interesting region. For example, the FUV continuum map obtained from the STSAT-1 observations shows enhanced emission in the southern part of the Spica halo region, where the dust level is also high. In fact, the FUV continuum intensity is seen to have a good correlation with the IRAS 100${\mu}m$ emission data. It is also seen that the scattered spectrum is softer than the original one emitted by the central star, which is attributed to the increase in the dust-scattering albedo with wavelength. We have developed a Monte Carlo code that simulates dust scattering of light including multiple encounters. The code is applied to the present Spica halo region to obtain the scattering properties such as the albedo and the phase function asymmetry factor.

Therapeutic efficacy of the photoactivated sickle cells as novel drug delivery vehicle (약물전달 시스템 개발을 위한 여기된 광감응제의 응용)

  • Sickle cells possess a unique combination of traits that may enable their use as models for novel synthetic tumor targeting controlled release drug carriers with the ability to treat disseminated tumors in advanced metastatic disease. In this study, we assess the ability of light-activated release sickle cells to enhance tumor delivery of the fluorescent dye calcein by delayed photolysis controlled release compared to free systemic administration of calcein. Sickle cells from mouse models of the disease were shown to preferentially accumulate in tumors compared to adjacent tissue, in 4T1 tumors in mice on a time scale about 12 hours. Sickle cells photosensitized with protoporphyrin IX achieved delayed release of 50% of contents 8-16 hours after photoactivation, which was deemed useful for in vivo delivery of cargo to tumors given the tumor accumulation time of the sickle cells. Sickle cells may be useful as a model for new synthetic drug carrier particles with delayed photolysis controlled release properties.
