Fluorescent Magnetic Silica Nanotubes with High Photostability Prepared by the Conventional Reverse Micro-Emulsion Method

  • Magnetic fluorescent silica nanotubes were fabricated using reverse micro-emulsions coupled with conventional sol-gel methods. Anodic aluminum oxide templates were used to separate spatially the magnetic and the fluorescent moieties on individual nanotubes and so prevent quenching of the fluorescence. C18 and fluorescent layers were deposited sequentially on silica. Magnetism was then obtained by the introduction of pre-made magnetic nanoparticles inside the nanotubes. The photo- and chemical stabilities of nanotubes were demonstrated through dye release and photobleaching tests. The produced nanotubes did not show fluorescence quenching upon the addition of the nanoparticles, an advantage over conventional spherical fluorescent magnetic nanoparticles. High photostability of nanotubes, magnetism and biocompatiblily make them potentially useful in bioanalysis.

An off-on Fluorescent Sensor for Detecting a Wide Range of Water Content in Organic Solvents

  • This paper describes the synthesis and water sensing properties of a fluorescent photoinduced electron transfer (PET) sensor (5) with an extended operating sensing range. The 1,8-naphthalimide derivative (5) attached with a piperazine group and a carboxylic group was synthesized and applied as a fluorescent water sensor in water-miscible organic solvents. The fluorescence intensity of the dye 5 increased with increasing water content up to 80% (v/v) and the fluorescence intensities were enhanced 45-, 67- and 122-fold in aqueous EtOH, DMF and DMSO solutions, respectively. In aqueous acetone solution, the enhancement of the fluorescence intensities was somewhat lower (30-fold) but the response range was wider (0-90%, v/v).

  • Extensive tumor resection accompanied by radiotherapy and chemotherapy is the standard of care for malignant gliomas. However, there is a significant obstacle to the complete resection of the tumor due to the difficulty of distinguishing tumor and normal brain tissue with a conventional surgical microscope. Recently, multiple studies have shown the possibility of fluorescence-guided surgery in malignant gliomas. The most used fluorescence dyes for brain tumor surgery are 5-aminolevulinic acid (5-ALA) and indocyanine green (ICG). In this paper, a new fluorescence guided operation system, which can detect both 5-ALA and ICG fluorescent images simultaneously, is presented. This operation system consists of light emitting diodes (LEDs) which emits 410 nm and 740 nm wavelengths. We have performed experiments on rats in order to verify the operation of the newly developed operation system. Oral administration and imaging were performed to observe the fluorescence of 5-ALA and ICG fluorescence in rats. When LEDs at wavelengths of 410 nm and 740 nm were irradiated on rats, 628 nm wavelength with a violet fluorescence color and 825 nm wavelength with a red fluorescence color were expressed in 5-ALA and ICG fluorescent material, respectively, thus we were able to distinguish the tumor tissues easily. Previously, due to the poor resolution of the conventional surgical microscope and the fact that the color of the vein is similar to that of the tumor, the tumor resection margin was not easy to observe, thus increasing the likelihood for cancer recurrence. However, when the tumor is observed through the fluorescence guided operation system, it is possible to easily distinguish the color with the naked eye and it can be completely removed. Therefore, it is expected that surgical removal of cancerous tumors will be possible and surgical applications and surgical microscopes for cancer tumor removal surgery will be promising in the future.

백서의 대망을 혈관경으로 이식하여 생성시킨 이차적 도서형 피판 (The Secondary Island Flap Using Omental Vascular Carrier in Rats)

  • The omental pedicle based on right gastroepiploic vessels is designed new experimental model for prefabrication(revasculirization) of skin flaps in rats. A $2.5{\times}4cm$ pack of omentum with right gastroepiploic vessels was transferred under a bipediceld panniculocutaneous flap which is $2.5{\times}8cm$ size. At day 7, all four margin was divided and the flap was rasied as an secondary island flap connected only by its vascular pedicle, then the composite flap sutured back in place. The flap perfusion was examined by dermofluorometry and flap survival area was measured at day 12. The Secondary island flap demonstrated a dye fluorescence index(DFI%) of $31.38{\pm}12.33$ and survival rate $80.47{\pm}9.61$ The survival rate was increased when DFI% and contact surface between vascular carrier and skin flap was increased. An india ink injection and histologic examination provided visual evidence of revasculization. The omental pedicle is a promising and safe model for revasculirization of other tissues.

Near-infrared (NIR) 영상기법을 이용한 생체 내 수지상세포의 이동 (In vivo Dendritic Cell Migration Tracking Using Near-infrared (NIR) Imaging)

  • Matured dendritic cells (DCs) begin migration with their release from the bone marrow (BM) into the blood and subsequent traffic into peripheral lymphoid and non-lymphoid tissues. Throughout this long movement, migrating DCs must apply specialized skills to reach their target destination. Non-invasive in vivo cell-tracking techniques are necessary to advance immune cell-based therapies. In this study, we used a DiD cell-tracking solution for in vivo dendritic cell tracking in naive mice. We tracked DiD (non-invasive fluorescence dye)-labeled mature dendritic cells using the Near Infrared (NIR) imaging system in normal mice. We examined the immunophenotype of DiD-labeled cells compared with non-labelled mature DCs, and obtained time-serial images of NIR-DC trafficking after mouse footpad injection. In conclusion, we confirmed that DiD-labeled DCs migrated into the popliteal lymph node 24 h after the footpad injection. Here, these data suggested that the cell tracking system with the stable fluorescence dye DiD was useful as a cell tracking tool to advance dendritic cell-based immunotherapy.

유비퀴틴 단백질의 부분적으로 폴딩된 구조에 대한 분광학적 분석 (Spectroscopic Analysis of Partially Folded State of Ubiquitin)

  • Hydrophobic core가 변이된 유비퀴틴 단백질이 pH 2 용액에서 보이는 구조적인 특성을 여러 분광학적 방법으로 측정하였다. 낮은 pH값을 갖는 용액에서 이 변이 유비퀴틴의 intrinsic tryptophan fluorescence emission spectrum은 unfolded 상태보다 약간 blue shift되어 있고 또한 그 intensity도 상당히 낮게 나타났다. 이는 이 용액 조건에서 이 변이 유비퀴틴의 삼차구조가 약간 남아 있는 것을 의미한다. 같은 용액에서 이 변이 유비쥐틴의 far-UV circular dichroic spectrum은 native 상태나 unfolded 상태의 spectrum과 현저히 달랐으며 220 nm 에서의 molar ellipticity 값을 통하여 볼 때 pH 2인 용액에서 상당량의 이차구조를 지니고 있었다. 또한 같은 용액에서 이 변이 유비퀴틴은 hydrophobic dye인 8-anilinonaphthalene-1-sulfonic acid(ANS)외 fluorescence emission intensity를 증가시키고 fluorescence emission maximum이 짧은 파장에서 나타나게 하였다(blue shift). 이러한 현상은 pH 2 용액에서 이 변이 유비퀴틴의 hydrophobic core가 느슨하여져서 hydrophobic dye인 ANS가 결합할 수 있는 구조를 띠고 있음을 나타낸다. 이러한 분광학적인 관찰은 이 변이 유비귀틴이 pH 2인 용액에서 상당량의 이차구조를 지니고 있지만 hydrophobic core는 느슨하게 형성된 molten globule과 같은 형태를 지니고 있음을 나타낸다. 이 변이 유비퀴틴의 molten 히obule 형태는 단백질 폴딩 반응의 경로를 연구할 수 있는 좋은 모델이 될 수 있을 것으로 생각된다.

Rhodamine 560을 이용한 rhodamine 6G 색소 레이저의 출력 증가 (Output Enhancement of Rhodamine 6G Dye Laser by Rhodamine 560 Energy Transfer Dye)

  • 형광 스펙트럼이 Rhodamine 6G의 흡수 스펙트럼과 일치하는 Rhodamine 560을 첨가 색소로 하여 Rhodamine 6G 색소 레이저의 에너지 전달에 의한 출력 증가를 조사하였다. 펌핑 광원으로 펄스형의 경우 아르곤 기체를 주입한 동축 섬광관을 실험실에서 제작하여 사용하였으며 연속형은 아르곤 레이저를 이용하였다. 펄스형의 경우 주색소인 Rhodamine 6G의 종도가 $1\times10^{-4}mol/l$일 때 첨가 색소인 Rhodamine 560의 농도가 Rhodamine 6G농도의 1% 정도에서 에너지 전달효율이 가장 높았다. 연속형의 경우는 Rhodamine 6G의 종도가 $2\times10^{-3}mol/l$ 일 때 Rhodamine 560의 농도가 Rhodamine 6G 농도의 2.5%에서 에너지 전달효율이 가장 높았다. 아르곤 레이저의 multiline발진에 의한 출력이 3.6W일 때 18% 정도 출력이 증가하였으며 488nm와 514.5nm의 단색광으로 펌핑할 경우 아르곤 레이저의 최대 출력이 1.6W일 때 각각 72%와 88%의 출력 증가율을 보였다.

Optimization of SNP Genotyping Assay with Fluorescence Polarization Detection

  • Single nucleotide polymorphisms (SNPs) are valuable DNA markers due to their abundance and potential for use in automated high-throughput genotyping. Numerous SNP genotyping assays have been developed. In this report, one of effective and high throughput SNP genotyping assays, which was named the template-directed dye-terminator incorporation with fluorescence polarization detection (FP-TDI) was described. Although the most of this assay succeed, the objective of this work was to deter­mine the reasons for the failures, find ways to improve the assay and reduce the running cost. Ninety $F_2$-derived soybean, Glycine max (L.) Merr., RILs from a cross between 'Pureunkong' and 'Jinpumkong 2' were genotyped at four SNPs. FP measurement was done on $Victot^3$ microplate reader (perkinelmer Inc., Boston, MA, USA). Increasing the number of thermal cycles in the single-base extension step increased the separation of the FP values between the products corresponding to different genotypes. But in some assays, excess of heterozygous genotypes was observed with increase of PCR cycles. We discovered that the excess heterozygous was due to misincorporation of one of the dye­terminators during the primer extension reaction. After pyrophosphatase incubation and thermal cycle control, misincoporation can be effectively prevented. Using long amplicons instead of short amplicons for SNP genotyping and decreasing the amount of dye terminator and Acyclopol Taq polymerase to 1/2 or 1/3 decreased the cost of the assay. With these minor adjustments, the FP-TDI assay can be used more accurately and cost-effectively.

Sentinel lymph node mapping using tri-modal human serum albumin conjugated with visible dye, near infrared fluorescent dye and radioisotope

  • We developed an evans blue-indocyanine green-$^{99m}Tc$-human serum albumin conjugate for sentinel lymph node mapping and we describe its unique potential usage for clinical implications. This conjugate has combined the strengths of visible blue dye, near-infrared fluorescence and radioisotope into one single conjugate without any additional weakness/disadvantage. All the components of evans blue-indocyanine green-$^{99m}Tc$-human serum albumin are safe and of low cost, and they have already been clinically used. This conjugate was stable in the serum, it showed a long retention time in the lymphatic system and the lymph nodes showed a much higher signal-to-noise ratio after the conjugate was injected intradermally into the paw of mice. Both the single-photon emission computed tomography and near-infrared fluorescent images of the mice were successfully obtained at the same time as the excised sentinel lymph nodes showed blue color. The visual color, near-infrared fluorescence and gamma ray from this agent could be complementary for each other in all the steps of sentinel lymph node sampling: exploring and planning sentinel lymph node before excision with visualization of the exact sentinel lymph node location during an operation. Therefore, the triple modal agent will possibly be very ideal for sentinel lymph node mapping because of the high signal-to-noise ratio for non-invasive imaging and its complementary multimodal nature, easy preparation and safety. It is promising for clinical applications and it may have great advantages over the traditional single modal methods.

7 Diethylaminocoumarin (DEAC) 레이져 염료의 광물리적 파라미터, 광분해, 형광 소광 및 Convolutive Voltammetry (Photophysical Parameters, Photodecomposition, Fluorescence Quenching and Convolutive Voltammetry of 7-Diethylaminocoumarin (DEAC) Laser Dye)

  • 7 diethylaminocoumarin (DEAC) 레이저 염료의 광물리적 특성들을 다양한 용매에서 측정하였다. DEAC의 방출 스펙트럼은 양이온성(CTAC) 미셀과 음이온성(SDS) 미셀에서 측정하였고, 레이저 파라미터는 아세톤, 다이옥산, 에탄올 및 dimethylforamide (DMF)과 같은 여러 가지 용매에 대하여 계산하였다. DEAC의 광반응성은 366 nm 빛을 사용하여 $CCl_4$ 용매에서 연구하였고, 광화학 수득률 (${\Phi}_c$)과 속도 상수(k)도 결정하였다. picric acid (PA), tetracyanoethylene (TCNE), 7,7,8,8-tetracynoquinonedimethane (TCNQ)등의 유기 수용체와 DEAC의 상호 작용에 대한 연구는 acetonitrile ($CH_3CN$) 용액에서 형광을 측정하므로 써 수행하였다. DEAC의 전기화학적 연구는 백금 전극을 사용하여 0.1 mol/L tetrabutyl ammonium perchlorate (TBAP)/$CH_3CN$ 용액에서 순환 전류-전압법, convolutive voltammetry 및 디지털 시뮬레이션을 조합하여 수행하였고, 전기화학 파라미터도 결정하였다.
