DEAE-Trisacryl 크로마토그래피법에 의한 IgG1 Type 쥐 단일클론 항체의 분리정제 (Purification of IgG1 Type Mouse Monoclonal Antibodies with DEAE-Trisacryl Chromatography)

  • 하이브리도마를 쥐의 복강이나 in-vitro에서 배양한 뒤 생산된 IgG1 type의 쥐 단일클론 항체를 정제하기 위하여 음이온 교환 크로마토그래피를 이용하였다. 배양이 끝난 배지를 원심분리하여 세포를 제거하고 50-60% ammonium sulfate로 침전물을 만든 다음 0.025M Tris-HCI(pH8.2)용액으로 투석하여 salt가 제거된 sample을 DEAE-Trisacryl M에 부하하였다. Column에 결합된 항체는 30-40mM NaCl 을 포함하는 0.025M Tris-HCI(pH8.2)용액으로 용출하였다. 혈청농도가 높은 배지 (10% FBS)에서는 50% ammonium sulfate 처리로 90% 이상의 항체가 회수되었으나 저혈청 배지 (2% FBS)에서는 60% ammonium sulfate 처리에도 회수율이 84%에 그쳤다. 후자의 경우 한외여과법 (ultrafiltration)을 이용하여 항체 회수율을 91%까지 증가시킬 수 있으나 농축된 항체를 크로마토그래피로 정제하였을 때 그순도가 ammonium sulfate 침전법에 비해 낮아졌다. 하이브리도마 Alps 25-3, HCGK, A4W, KW를 여러가지 배양조건에서 배양한 뒤 생산된 항체를 DEAE-Trisacryl M chromatography를 이용하여 정제해 본 결과 대체로 순도 70-80%의 항체를 얻을 수 있었고 이때 항체 회수율은 65% 선이었다. 항체의 순도를 높이기 위해서는 affinity chromatography 혹은 gel filtration과 같은 2차적인 방법이 필요 할 것으로 보이며 한 예로 affinity chromatography를 이용하여 순도 95%의 항체를 얻었다.

초임계 이산화탄소와 보조용매를 이용한 미역으로부터 색소 추출 (Extraction of Pigment from Sea Mustard ( Undaiia pinnatinda) using Supercritical Carbon Dioxide and Entrainer)

  • 동결 건조된 미역 가공 부산물로부터 색소 및 지방산의 추출 및 조건은 시료 30g, 압력 $10.3\~17.2MPa$, 유량 15mL/min와 온도 $30\~45^{\circ}C$에서 추출 실험을 실시하였다. 지방산의 경우 초임계 처리하지 않은 것에 비해 불포화 지방산의 함량이 높게 나타났고, 온도 $40^{\circ}C$의 조건에서 보조용매 (entrainer)로 에탄올을 첨가하여 압력의 변화에 따른 회수율을 실험한 결과 12.4MPa에서 최대 $92\%$의 회수율을 보였다. 그리고 압력이 증가하면서 초임계 이산화탄소에 대한 용질의 용해도가 감소하는 retrograde 현상이 나타났다 시료입자의 크기를 $500\~1,000{\mu}\;m$와 12.4MPa에서 온도의 변화를 주어 실험한 결과 입자의 크기 $500{\mu}m$$1000{\mu}m$에 비해 색소의 추출량이 1.8배정도 많았다 최적 조건 (압력 12.4MPa,온도 $35^{\circ}C$, 입자크기 $500{\mu}m$)에서 추출시간에 따른 chlorophyll a 함량을 비교한 결과 에탄올을 각각 3mL/min, 5mL/min 첨가하였을 때 모두 20분까지 함량이 증가한 후 감소하였으며, 최적 추출시간은 30분까지로 판단되었다. 유기용매 추출과 초임계 추출을 비교한 결과 유기용매로 48시간 추출시 chlorophyll a 함량이 150mg/L였지만 초임계로 1시간 추출시 최대 172mg/L로 약 $20\%$ 증가하였다.

전개판에 대한 수직해법 - 3 . 슬롯에 따른 단순만곡형전개판의 성능분석 - (Computational Fluid Analysis for the Otter Boards - 3 . Efficiency Analysis of the Single Cambered Otter Boards for the Various Slot Position -)

  • 현재 사용되고 있는 전개판의 성능을 보다 향상시키기 위한 방안으로 비행기의 익이론에 기초를 두고, 전개판의 경계층을 흡입하거나 제거하는 방법 중에서 역류역을 흡입하는 슬롯(slot)을 전개판에 적용하였다. 성능분석을 위해서 단순만곡형전개판에 슬롯이 없는 기준형과 슬롯의 위치에 따른 5종의 모형전개판에 대하여 성능실험과 가시화실험을 통하여 그 성능과 유체역학적 특성을 비교해 보았다. 실험결과를 요약하면 다음과 같다. \circled1 최대전개력계수는 0.6C, 0.4C, 기준형, 0.8C, 0.2C의 순으로, 슬롯의 위치 0.6C인 전개판이 영각 27$^{\circ}$에서 1.59로 가장 우수하였다. \circled2 항력계수는 슬롯의 위치가 후연으로 갈수록 감소하며, 슬롯을 준 경우가 기준형보다 작게 나타났다. \circled3 양항비는 0.6C, 0.4C, 0.8C, 0.2C, 기준형의 순으로 모두 기준형보다 높게 나타났다. \circled4 0.2C, 0.4C 및 0.6C의 전개판이 기준형과 0.8C의 전개판보다 박리가 후연에서 발생하였다. \circled5 전개판의 전.후면에서 유속차는 영각 15$^{\circ}$~25$^{\circ}$까지는 점차 증가하고, 30$^{\circ}$에서는 감소하는 것으로 나타났다. \circled6 박리역의 크기는 0.6C의 전개판에서 가장 적게 나타났다.

소결조제로 나노크기 γ-알루미나 입자의 도입에 따른 α-알루미나 중공사 분리막의 기계적 강도 향상 (Improvement in Mechanical Strength of α-Alumina Hollow Fiber Membrane by Introducing Nanosize γ-Alumina Particle as Sintering Agent)

  • 수처리 및 의약바이오 분야에서 유효물질 분리에 활용되고 있는 알루미나 중공사 분리막은 얇은 두께로 인해 취급 및 적용시 쉽게 파괴되는 단점이 있기 때문에 분리막의 강도를 100 MPa 이상으로 향상시키기 위한 연구가 필요하다. 본 연구에서는 나노입자의 함량을 0, 1, 3, 5 wt%로 증가시켰을 때 제조된 중공사 분리막의 특성을 평가하였다. 그 결과, 나노입자의 함량이 증가함에 따라 중공사 분리막의 강도는 79 MPa에서 115 MPa로 증가하였으며, 밀도는 1.76 g/m3에서 1.88 g/m3으로 증가하였고 기공률과 평균기공크기는 각각 51%에서 48%로, 416 nm에서 352 nm로 감소한 것을 확인하였다. 스폰지구조가 발달하고 스폰지구조의 기공크기가 향상된 알루미나 중공사 분리막은 100 MPa 이상으로 기계적 강도가 향상되었으며, 약 100000 GPU의 높은 질소 투과도 및 약 3000 L/m2h의 높은 물 투과도를 나타내었다. 따라서, γ-알루미나 나노입자를 소결조제로 첨가하는 것은 α-알루미나 중공사 분리막의 기계적 강도를 효과적으로 증진시키고 높은 투과성능을 유지할 수 있는 매우 유효한 방법임을 확인하였다.

Hydrocyclone Filter 장치를 이용한 강우유출수내 미세입자 제거특성 분석 (Performance Evaluation of Hydrocyclone Filter for Treatment of Micro Particles in Storm Runoff)

  • Hydrocyclone은 높은 수면적부하율 운전이 가능하고, 구동부분이 없고, 운전 및 유지관리비가 적게 소요되어 미세입자물질제거에 효과적인 장치로 다양한 산업 분야에서 활용되고 있다. Hydrocyclone에 나선형 유입부를 통해 직각으로 유입되어 중력이 아닌 원심력에 의해 입자물질이 하부배출구로 분리되고 처리수는 vortex finder를 통해 배출된다. 입자분리에 hydrocyclone은 많은 장점을 가지고 있음에도 불구하고 도시지역 강우유출수 처리사례는 드문 실정이다. 본 연구에서는 변형된 hydrocyclone과 perlite 여재를 조합한 hydrocyclone filter (HCF)을 이용하여 강우유출수내 미세입자의 제거능을 분석하였다. 입자물질들은 인공입자들을 이용하여 강우유출수내 입자농도를 모의 실험하였다. 인공입자들을 물에 분산시켜 강우유출수를 재현하였는데 사용한 입자들은 이온교환 수지, 도로측구 퇴적물질, 상업지역 맨홀퇴적물질, 그리고 실리카겔 등이다. 하부배출부와 vortex finder 구조를 달리하여 HCFopen system과 HCF-closed system으로 구분하여 처리능을 분석하였다. HCF장치는 아크릴 수지를 이용하였는데 hydrocyclone의 직경은 120 mm이고 여과조의 직경은 250 mm이고 전체적인 높이는 800 mm로 제작하였다. 운전조건별 유입수 농도와 입경을 다양하게 적용하였고 SS와 COD농도를 분석하여 처리효율을 산정하였다. HCF-open system의 경우 운전 가능한 최대 수면적부하율은 700 $m^3/m^2$/day이었고 HCF-closed system의 경우 수면적부하율 1,200 $m^3/m^2$/day까지이다. HCF-open system 운전결과 HCFclosed system에 비교하여 평균 수면적부하율은 2배 이상 높게 운전이 가능하며 처리효율도 8~20% 이상 향상되는 것으로 분석되었다. 또한 유입수 SS농도가 높을수록 처리효율이 증가하며, 입경이 클수록 수면적부하율의 변화에 대한 처리효율의 영향이 적은 것으로 분석되었다. HCF-closed system의 실험을 통한 수면적부하율 변화에 대한 실리카겔입자의 제거 효율과 CFD입자추적기법을 이용한 예측 처리효율을 비교한 결과 CFD에 예측효율이 실제실험결과와 비교하여 다소 높게 나타나지만 처리효율의 경향은 매우 유사하게 나타나 CFD추적기법을 이용한 HDS유형 처리장치 설계 시 유용한 도구로 활용될 수 있을 것으로 판단된다.

초임계 이산화탄소를 이용한 미세전자기계시스템의 식각, 세정, 건조 연속 공정 (Continuous Process for the Etching, Rinsing and Drying of MEMS Using Supercritical Carbon Dioxide)

  • 기존의 초임계 이산화탄소를 이용하여 식각 및 건조하는 공정은 고압 건조기 외부에서 용매를 이용하여 웨이퍼를 식각한 후 고압 건조기로 이동시켜 초임계 이산화탄소를 이용하여 세정 및 건조 하는 2단계 공정으로 구성되어 있다. 이 공정을 이용하여 본 연구에서 실험을 수행한 결과 점착 없이 식각, 세정 및 건조가 가능함은 확인되었지만, 반복 실험 결과 재현성이 떨어지는 것을 확인하였다. 이것은 외부에서 식각한 후 건조기로 이동할 때 식각용 용매가 기화하여 구조물이 점착되는 문제가 발생하기 때문이었다. 본 연구에서는 이 문제를 개선하기 위하여 웨이퍼를 이동시키지 않고, 고압 건조기 내에서 초임계 이산화탄소를 이용하여 미세전자기계시스템 웨이퍼의 식각, 세정 및 건조공정을 연속적으로 수행하고자 하였다. 또한, 연속공정 수행 시 식각 공정에서 사용하는 이산화탄소의 상태(기체, 액체, 초임계상태)에 따른 영향을 알아보고자 하였다. 기체 이산화탄소를 이용하여 식각하는 경우(3 MPa, $25^{\circ}C$)에는 점착 없는 식각, 세정 및 건조를 할 수 있었고 반복 실험을 통하여 공정의 최적화 및 재현성을 확인하였다. 또한 기존의 2단계로 이루어진 공정에 비해 세정용 용매의 양을 절감 할 수 있었다. 액체 이산화탄소를 이용하여 식각하는 경우(3 MPa, $5^{\circ}C$) 액체 이산화탄소와 식각용 공 용매(아세톤)간의 층 분리가 일어나 완전한 식각이 이루어지지 않았다. 초임계 이산화탄소를 이용하여 식각 하는 경우(7.5 MPa, $40^{\circ}C$) 점착 없는 식각, 세정 및 건조를 할 수 있었고 기존 2단계 공정에 비해 세정용 용매의 절감 뿐 아니라 기체 이산화탄소를 이용한 연속공정에 비하여 공정시간도 단축시킬 수 있었다.

지역화된 동북아시아지역의 지구자기장 영년변화 모델: 1997-2011 (A Localized Secular Variation Model of the Geomagnetic Field Over Northeast Asia Region between 1997 to 2011)

  • 동북아시아에 위치한 4개의 지자기 관측소의 3성분 지구자기장 관측 자료를 이용하여 1997년부터 2011년 동안의 지구자기장의 영년변화를 표현하는 지역화된 영년변화 모델을 제작하였다. 외핵의 움직임으로 발생되는 지구자기장은 장소와 시간에 따라 변화하며 지자기 관측소에서는 이를 시계열로 측정하게 된다. 따라서 공간적인 변화를 함께 파악하기 위해서는 이를 시공간모델로 제작하여 시간에 따른 지구자기장의 변화를 공간적으로도 표현할 수 있도록 해야 한다. 전지구 범위인 경우 관측소의 분포가 제한되어 있고 이를 공간적으로 보완하기 위해 위성 자료를 활용한다. 하지만 방대하고 복잡한 위성 자료의 처리와 세계의 모든 지자기 관측소의 자료를 활용하여 전 지구적 지구자기장의 모델을 제작하는 일은 상당한 작업과 노력을 요구한다. 또한 계속해서 들어오는 자료들을 이용하여 모델을 업데이트 하는 일 역시 같은 양의 시간과 노력을 필요로 하게 된다. 더불어, 각 관측소 자료의 오차범위와 위성 자료 처리 오차 (processing error) 역시 지역에 따라 다르게 나타남으로 이러한 오차값들이 전지구 모델의 정확도에 영향을 미칠 수 있다. 본 논문에서는 이러한 문제점을 감안하여 동북아시아 지역을 중심으로 하는 지역화된(localized) 모델링 기법을 소개하고 이를 통해 지구자기장의 영년변화 모델을 제작하는 데 적용하였다. 전 세계의 지구자기장 관측망인 INTERMAGNET에 가입된 3개의 일본 관측소와 1곳의 중국 관측소 자료를 활용하여 1997년부터 2011년까지 6개월 간격으로 지구자기장의 변화를 파악하고 이를 전지구 모델과 비교해 보았다. 또한 얻어진 모델을 이용하여 지구 내부의 원인인 지구자기장의 급작스런 변화를 일컫는 지구자기장 '급변'을 찾아보고 이에 대한 발생 시기에 대해 논의하였다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.
