PURIFICATION AND PROPERTIES OF EXTRACELLULAR NUCLEASE(S) FROM RUMEN CONTENTS OF BUBALUS BUBALIS

  • Extracellular nuclease(s) in buffalo rumen fluid were purified from strained rumen fluid by a procedure involving Seitz filtration, acetone fractionation and gel filtration on Sephadex G-100. The enzyme resolved into two peaks exhibiting both DNase and RNase activities. The molecular weight of enzyme corresponding to peaks I and II were approximately 30,000 and 12,000 respectively. The properties of enzymes from the two peaks, however, were same. Optimum temperature for both DNase and RNase activities was at $50^{\circ}C$. Whereas DNase activity was stable upto $60^{\circ}C$, RNase activity was stable only up to $50^{\circ}C$. DNase activity recorded two pH optima, one at pH 5.5 and the other at pH 7.0. RNase activity recorded a broad pH optimum between pH 6.0-8.0. pH stability of the enzyme coincided with pH optima for both the activities. DNase activity was stimulated by $Mg^{2+}$ and $Mn^{2+}$ and inhibited by $Fe^{2+}$, $Zn^{2+}$, $Hg^{2+}$ and $Ag^+$. RNase activity was also stimulated by $Mg^{2+}$ and $Mn^{2+}$ and inhibited by $Cu^{2+}$, $Fe^{2+}$, $Zn^{2+}$, $Hg^{2+}$ and $Ag^+$. Reducing agents stimulated both the activities.

Characteristics of Bentonite Filter Cake on Vertical Cutoff Walls Evaluated by Modified Fluid Loss Test (수정 fluid loss 시험을 이용한 연직 차수벽에 생성된 벤토나이트 필터케익 특성 평가)

  • During the construction of vertical cutoff walls, filtration of bentonite slurry into the adjacent soil formation fabricates a layer of bentonite filter cake on the wall surface. The bentonite filter cake possesses much lower hydraulic conductivity compared to that of backfill materials in the cutoff wall. Hydraulic conductivity of bentonite filter cakes formed with three different types of bentonites has been measured by performing the modified fluid loss test under various pressure levels. Three different mixture ratios, 4, 6, and 8%, were selected for fabricating bentonite filter cakes to represent common field conditions. Two analysis methods for interpreting the experimental data from the modified fluid loss tests were employed to estimate hydraulic conductivity of the bentonite cakes. The range of hydraulic conductivities of the three bentonite cakes is between $2.15{\times}10^{-11}\;m/s$ and $2.88{\times}10^{-10}\;m/s$ which is 1 to 2.7 orders of magnitude lower than that of the design cutoff wall backfill. The stress distribution and thickness of the bentonite cakes were also evaluated in this paper.

Meta-analysis of Inline Filtration Effects on Post-infusion Phlebitis Caused by Particulate Contamination of Intravenous Administration

  • The particulate contamination of intravenously administered fluid has been of major concern. One of the most common complications associated with long term i.v. therapy is post-infusion phlebitis (PIP). We undertook a systematic review and meta-analysis of the effect of inline filters on PIP. An electronic search of Medline, KoreaMed, and KRIST was conducted to identify randomized controlled trials evaluating the effect of inline filters. Meta-analysis was undertaken using STATA 10. A total of 62 literatures were retrieved, of which 7 were included in meta-analysis. Inline filtration for intravenous infusion significantly reduced by 39% of the incidence of phlebitis, with a relative risk of 0.61 (95% CI 0.41-0.90, p=0.012). Therefore, inline filtration is a highly effective means of decreasing the incidence of infusion phlebitis and should be considered as a part of intravenous therapy.

Effect of Epididymal Fluid Fractionated by Chromatography on In Vitro Maturation of Porcine Follicular Oocytes

  • The aim of this study was to investigate what protein(s) of porcine epididymal fluid (pEF) are able to enhance the nuclear maturation of porcine germinal vesicle (GV) oocytes in vitro. Proteins of pEF were fractionated by affinity, ion exchange, and gel filtration chromatography. Porcine cumulus-oocytes complexes (COC) from follicles were cultured in tissue culture medium (TCM 199) containing various fractions obtained by chromatography. Porcine COCs were also cultured in TCM 199 containing various meiosis inhibitors and pEF. After 24 or 48 h culture, oocytes were examined for evidence of GV breakdown, metaphase I, anaphase-telophase I, and metaphase II. When porcine COCs were cultured in the medium with meiosis inhibitor such as, dibutyryl cAMP (dbcAMP) and forskolin (Fo), more than 80% of oocytes were unable to resume meiosis. However, porcine COCs supplemented with pEF were able to overcome the inhibitory effect of dbcAMP and Fo. Maturation rate of oocytes was significantly (p<0.05) increased in the media supplemented with cationic protein(s) during in vitro maturation than in those with anionic protein(s) (44.1% vs 20.0%). When oocytes were cultured in the TCM 199 with fractions obtained by gel filtration, the maturation rate of oocytes was significantly (p<0.05) higher in fraction 11 containing 18 kDa than other fractions. The present study suggests that 1) dbcAMP and Fo prevent the spontaneous maturation of oocyte after isolation from follicles, and that pEF contain a substance(s) that improves meiosis resumption in vitro of porcine COCs, 2) cationic 18 kDa protein(s) are responsible for promotion of Mil stage.

Isolation of an Oocyte Stimulatory Peptide from the Ovarian Follicular Fluid of Water Buffalo (Bubalus bubalis)

  • Ovarian follicular fluid contains both stimulatory and inhibitory agents that influence the growth and maturation of oocyte. In the present study, an attempt was made to isolate and study the biological properties of ovarian follicular fluid peptide(s) in buffaloes. Bubaline ovarian follicular was made steroid- and cell-free. A protein fraction was obtained by saturation (30-35% level) of the follicular fluid with ammonium sulfate. The protein fraction was purified with Sephadex-G 50 gel filtration chromatography and a single peak was obtained in the eluant volume, which was lyophilized. SDS-PAGE of the lyophilized fraction revealed a single band and the molecular weight of the peptide was 26.6 kDa. The peptide stimulated the cumulus cell expansion and in vitro maturation rate of oocytes in buffaloes in a dose dependent manner when it was incorporated at different dose levels (0, 10, 25, 50, 100 and 1,000 ng $ml^{-1}$ of maturation medium). The basic culture medium consisted of TCM 199 with Bovine serum albumin (0.3%). The in vitro maturation rates were comparable to those obtained with a positive control medium (TCM 199+20 ng EGF $ml^{-1}$+steer serum (20%)). Further purification and biological assays may throw more light on the nature and functions of this peptide.

In addition, this means selecting a emulsion on the basis of quality criteria processing method, the material of the material, cutting depth, cutting speed, Djourou fence Liang, and surface roughness, cutting oil, the shape of the device based on the emulsion, I will be the structure of the tank, filtration equipment also changes. In particular, acting bacteria is now breeding in response to the passage of time due to metal ion degradation due to heat generated hydraulic fluid leakage, humidity tung, during processing, seep from processing material at the time of processing the water-soluble cutting oil for generating the malodor by dropping significantly the performance of the cutting oil to corruption from, sometimes by introducing various additives to suppress spoilage in advance. In this study, we expect the effect of the cost reduction in the extension of fluid replacement cycle through the application of the management apparatus and deep understanding in the management of cutting fluid, the working environment through the understanding and interest of workers in the production site more than anything I try to become useful for the improvement.

  • This study aimed to evaluate the validity of an exhausted injector design for filtration system and the performance characteristics thereof. The evaluation was intended through computational fluid dynamics(CFD) analysis based on computer simulation rather than through prototype fabrication and testing. Furthermore, the design of experiment was used to create an experimental design table by which the reaction characteristics of response factors were analyzed for design parameters. All experiments were substituted with computer simulations. Lastly, an optimal design model for the injection tubes was determined based on response surface method algorithms.

  • In order to understand the flow and filtration characteristics in a wall-flow type DPF(Diesel Particulate Filter), 0-D, 1-D, and 3-D simulations are preformed. In this paper, three model are explained and validated with each other. Based on the comparisons with 1-D and 3-D results for the steady state solution, 3-D CFD analysis is preferable to 1-D for the prediction of wall velocity at the inlet and exit plane. Because PM loading process is transient state phenomena, the combination of full 3-D and time dependent simulation is crucial for the configuration of wall channels.   • The action of serotonin (5-Hydroxytryptamine) on the excretory function of the kidney was investigated in the dog, utilizing the clearance method and the stop-flow technique. It was shown that serotonin, $10{\mu}g/kg/min$, i. v., exerts a marked antidiuretic effects and elicits a marked hemodynamic changes in the kidney: a highly significant decrease of the glomerular filtration rate and a tendency of decrement in the renal plasma flow. Little change in the systemic blood pressure was noted, and the participation of the antidiuretic hormone in the antidiuretic action was ruled out by adding vasopressin to the infusion fluid. The stop-flow analysis showed that there is no evidence of altered activity in the tubules by serotonin. It was thus concluded that serotonin elicits anti diuresis in the dog by decreasing glomerular filtration rate, which results from the constriction of Vas afferens in the glomeruli.

The Comparison of Methods Processing Cells Recovered by Bronchoalveloar Lavage (기관지폐포세척액에서 세포수 측정을 위한 처리방법의 비교)

  • Background: The total and differential cell count of bronchoalveolar lavage(BAL) fluid are useful assessing activity, prognosis and response to therapy in diffuse interstitial lung disease. But controversy exist as to the appropriate method in processing BAL fluid. Therefore we investigated the effect of gauze filtration, centrifugation and different storage time of BAL fluid on the total and differential cell count. Methods: We obtained BAL fluid from 6 persons with no active lung lesion and divided pooled BAL fluid into several siliconized glass tubes and filtered through 0, 1, 2, 4 folds of cotton guaze(pore size: 1mm), and compared total cell count using hemocytometer after trypan blue staining and differential cell count after Wright-Giemsa staining of cytocentrifuged preparations. And we also counted total and differential cell count after centrifugation(400g for 30 min) and various storage time(2hr, 24hr, and 48hr). Results: There was no difference in total and differential cell count according to folds of gauze filtraion. But without gauze filtration, mucus threads that hampered total and differential cell count were found in 2 cases (33%). Centrifugation resulted in loss of total cell count($24{\pm}18%$) without change in differential cell count. There was no change in total cell count after 2hr storage but significant cell loss was found after 24hr storage time(24hr : $28{\pm}21%$, 48hr : $41{\pm}24%$). However there was no change in differential cell count with 48hr storage time. Conclusion: Total and differential cell count of BAL fluid may be best performed after cotton gauze filtration without centrifugation and within 2 hours.
