Broccoli flower와 Broccoli sprout의 라디칼 소거능 및 산화적 스트레스 개선 효과 (Free radical scavenging effect and protective activity from oxidative stress of broccoli flowers and sprouts)

  • In this study, the antioxidative effect and protective potential against oxidative damage of extract and fractions from broccoli were investigated under in vitro and cellular system. The methanol (MeOH) extracts of broccoli flowers and sprouts were partitioned as dichloromethane, n-butanol (BuOH) and aqueous fractions. The comparison of antioxidative effect of broccoli flowers and sprouts showed that broccoli sprouts exerted the more effective protective activity from 2,2'-azobis (2-aminopropane) dihydrochloride (AAPH)-induced oxidative stress in LLC-$PK_1$ porcine renal epithelial cell. In addition, the extract and fractions from broccoli sprouts showed strong scavenging effect of 1,1-diphenyl-2-picrylhydrazyl radical and the BuOH fraction exerted the strongest activity. Therefore, the BuOH fraction was evaluated as the most active fraction with strong radical scavenging activity among the fractions of broccoli flowers and sprouts. The present study suggests the antioxidative potential against free radical-induced oxidative damage of flowers and sprouts of broccoli. In addition, the BuOH fraction of broccoli is considered as the active fraction with antioxidative effect.

조선시대 민화 <연화도> 콘텐츠를 활용한 문화상품 개발 (Development of Cultural Products Based on Minhwa of the Joseon Dynasty - Focusing on Lotus Flower Painting -)

  • This study aims to collect Minhwa, or folk painting, particularly the Lotus Flower Painting, to analyze its formative characteristics and related story, and to develop cultural fashion products by utilizing Korean traditional culture and modern flavor based on the results. Lotus flowers grow from mud, but are unstained, and they bloom beautiful flowers. Thus, they symbolize Gunja, a true gentleman who is very learned and proper in behavior. In Buddhism, lotus flowers are divine flowers that have the meaning of the creation of life and the eternal cycle of birth, death and rebirth. Lotus flowers also represent the love between man and woman as well as conjugal harmony and love. Lotus flower painting includes the paintings of the lotus flower alone as well as the paintings of the lotus flower with ducks, white heron, kingfisher, fish, butterfly, crab, or tortoise. Colors that are mostly used in lotus flower paintings is the compatible combination of red and blue (green). Based on these findings, fashion products such as bag accessories, sitting cushions, and kitchen utensils are developed using various designs such as realistically drawn lotus flower, schematized lotus flower, the lotus flower alone, or the lotus flower with kingfisher, crab, or dragonfly, that emphasizing the compatible color combination of red and blue.

절화류의 선도 유지를 위한 기능성 첨가제 및 포장 원지 개발 (Development of Functional Additives and Packaging Paper for Prolonging Freshness of Cut Flowers)

  • To prolong freshness and to reduce a decay rate of cut flowers during storage and distribution, a new packaging paper was developed with grapefruit seed extracts(GFSE) as a natural microorganism control agent. The GFSE was fractionated in order to identify antibiotic fractions by HPLC equipped with C18-reverse phase column chromatography. Among the active fractions, three ones were identified as 1-chloro-2-methyl-benzene (ο-toluene), N, N-dimethyl-benzenemethaneamine, and 1-〔2-(2-ethylethoxy)ethoxy〕-4-(1,1,3,3- tetra methyl)-benzene, while the other three remained unidentified. The GFSE-added paper displayed an effective inhibitory activity against putrefactive bacteria and fungi which were involved in the decay of flowers. Despite excellent antimicrobial acts of the GFSE-treated packaging paper, it was not possible to prevent the cut flowers from being dehydrated during storage, which led to the reduction of their fresh weight. However, additional treatment for giving water- repellency property to the GFSE-treated paper decreased a reduction rate of the fresh weight up to around 50% compared to the only GFSE-treated one.

Microbial Community Dysbiosis and Functional Gene Content Changes in Apple Flowers due to Fire Blight

  • Despite the plant microbiota plays an important role in plant health, little is known about the potential interactions of the flower microbiota with pathogens. In this study, we investigated the microbial community of apple blossoms when infected with Erwinia amylovora. The long-read sequencing technology, which significantly increased the genome sequence resolution, thus enabling the characterization of fire blight-induced changes in the flower microbial community. Each sample showed a unique microbial community at the species level. Pantoea agglomerans and P. allii were the most predominant bacteria in healthy flowers, whereas E. amylovora comprised more than 90% of the microbial population in diseased flowers. Furthermore, gene function analysis revealed that glucose and xylose metabolism were enriched in diseased flowers. Overall, our results showed that the microbiome of apple blossoms is rich in specific bacteria, and the nutritional composition of flowers is important for the incidence and spread of bacterial disease.

  • From the flowers of Ervatamia coronaria four indole alkaloids have been isolated and characterized as harmine, heyneanine, voacristine and apparicine with the help of various spectral data. The alkaloids Harmine and Heyneanine are reported first time from the flowers of Ervatamia coronaria.

COUNTING OF FLOWERS BASED ON K-MEANS CLUSTERING AND WATERSHED SEGMENTATION

  • This paper proposes a hybrid algorithm combining K-means clustering and watershed algorithms for flower segmentation and counting. We use the K-means clustering algorithm to obtain the main colors in a complex background according to the cluster centers and then take a color space transformation to extract pixel values for the hue, saturation, and value of flower color. Next, we apply the threshold segmentation technique to segment flowers precisely and obtain the binary image of flowers. Based on this, we take the Euclidean distance transformation to obtain the distance map and apply it to find the local maxima of the connected components. Afterward, the proposed algorithm adaptively determines a minimum distance between each peak and apply it to label connected components using the watershed segmentation with eight-connectivity. On a dataset of 30 images, the test results reveal that the proposed method is more efficient and precise for the counting of overlapped flowers ignoring the degree of overlap, number of overlap, and relatively irregular shape.

인삼 지상부의 채취시기에 따른 사포닌조성 비교 (Changes of Saponin Contents of Leaves, Stems and Flower-buds of Panax ginseng C. A. Meyer by Harvesting Days)

  • The parts of leaves, flowers and stems in ginseng were obtained for analyzing the component of saponin on 15th April, 25th April, 5th May, 25th May, which were considered as ginseng foliation stage. The total saponin content of the leaves were 97.29, 66.42, 67.61, 36.24 mg/g, respectively, in which the content of Re, $Rb_1$ and Rd were more than 2/3 amount of total saponin. Especially, the saponin content of leaves decreased according to the sequential collection days, in which the similar results were observed from the flowers and stems of ginseng. The total saponin content of the flowers and stems were 141.09,143.84,139.25,133.47 and 13.32, 9.85, 8.00, 4.65 mg/g, respectively. Among them, the content of Re, Rd and $Rb_2$ in flowers were more than 2/3 while the content of Re, $Rg_1$ and Rd in stems showed more than 9/10 amount of total saponin. The total saponin content of individual leaf were 19.46, 28.56, 58.82 and 169.24 mg/plant, 2.53, 2.76, 5.20 and 12.32 mg/plant in stems, and 14.11, 30.21, 37.60 and 73.41 mg/plant in flowers. Therefore, the total saponin content of aboveground parts in ginseng were leaves > flowers > stems.

싸리꽃 추출물의 tyrosinase 활성 및 멜라닌합성 억제효과 (Inhibitory effects of Flowers of Lespedeza bicolor on Tyrosinase Activity and Melanin Synthesis)

  • In this study, we investigated the effects of the flowers of Lespedeza bicolor on melanogenesis in a mouse melanocyte cell line, B16/F10 cells. Our results show that the flowers of Lespedeza bicolor significantly inhibits melanin synthesis in concentration-dependent manner. In addition, it was also found to inhibit the activity of tyrosinase, the rate-limiting melanogenic enzyme. While the flowers of Lespedeza bicolor have no direct inhibitory effect on tyrosinase activity in cell free assay system using mushroom tyrosinase. Moreover, the flowers of Lespedeza bicolor effectively suppressed the ${\alpha}-MSH-stimulated$ melanin formation, tyrosinase activity and dendrite outgrowth. These results suggest that the flowers of Lespedeza bicolor is a potent depigmetation agent.

싸리꽃 에탄올 추출물의 피부(皮膚) 생리활성(生理活性) 효과 (Dermal Bioactive Properties of the Ethanol Extract from Flowers of Lespedeza bicolor)

  • Objective : In this study, the ethanol extract from flowers of Lespedeza bicolor was investigated for their dermal bioactive properties related to cosmeceuticals such as depigmentation and radical scavenging effect. Results : The ethanol extract from flowers of Lespedeza bicolor showed considerable radical scavenging activity ($SC_{50}:\;10\;{\mu}g/ml$) and inhibited the production of nitric oxide (NO) in Raw 264.7 macrophages activated with the endotoxin lipopolysaccharide (LPS). Although the proliferation of B16/F10 cells was slightly decreased by the ethanol extract from flowers of Lespedeza bicolor at the concentration of $100\;{\mu}g/ml$, it did not appear necrosis. The ethanol extract from flowers of Lespedeza bicolor down-regulated melanin formation effectively. Methods : The free radical scavenging activity of Lespedeza bicolor was assayed in cell free systems using a stable free radical, 1,l-diphenyl-2-picrylhydrazyl (DPPH). Nitrite accumulated in culture medium was measured as an indicator of NO production using a Griess reaction. Cell viability was measured by MTT assay and melanin content was assessed using the method of Hosei with some modifications. Conclusions : These results suggest that the ethanol extract from flowers of Lespedeza bicolor is a potent depigmetation agent and it may be a candidate for antioxidant and anti-inflammatory agent.

Gene expression analysis related to ethylene induced female flowers of cucumber (Cucumis sativus L.) at different photoperiod

  • Photoperiod is one of the factors affecting productivity of cucumber plant by inducing ethylene hormone production and so triggering flower sex differentiation into female flower. However, only few studies have been perfomed in order to reveal the effect of photoperiod in molecular level in relation to the flower differentiation. Therefore, in this study, Mercy cultivar of cucumber (andromonoecious) was treated with photoperiod of 8, 12, 16 hours of light, while control received no treatment of additional light. Photoperiod of 8 hours was achieved by blocking the sunlight with shade net and 16 hours by giving longer light exposure using white LEDs. Cucumber's flowers were quantified and the apical and lateral shoots were extracted to evaluate the gene profile related to the photoperiod, ethylene production, and female flower differentiation, which were CsACS2, CsETR1, CsCaN, and CsPIF4 using PCR method. Photoperiod of 8 hours affected the production of female flower with average number of 6.7 flowers in main stem and 8.0 flowers in lateral stem, compared to photoperiod of 12 and 16 hours which produced 3.7 and 2.0 flowers in main stem with 7.0 and 11.3 in lateral stem, respectively. In silico studies in this experiment resulted in proposed model of signal transduction that showed the connection between ethylene production and flower differentiation. PCR analysis confirmed the expression of CsACS2, CsETR1, and CsCaN, that was positively correlated with numbers of female flowers in cucumber, but the expression of CsPIF4 that represent photoperiod haven't been confirmed correlated with the ethylene production and flower differentiation.
