맥동유동하에 있는 유연성 있는 평판 사이의 벽면전단응력: 벽면운동과 임피던스 페이즈 앵글과 비뉴턴유체의 영향 (Wall Shear Stress Between Compliant Plates Under Oscillatory Flow Conditions: Influence of Wall Motion, Impedance Phase Angle and Non-Newtonian Fluid)

  • The present study investigates flow dynamics between two dimensional compliant plates under sinusoidal flow conditions in order to understand influence of wall motion, impedance phase angle (time delay between pressure and flow waveforms), and non-Newtonian fluid on wall shear stress using computational fluid dynamics. The results showed that wall motion induced additional terms in the streamwise velocity profile and the pressure gradient. These additional terms due to wall motion reduced the amplitude of wall shear stress and also changed the mean wall shear stress. The trend of the changes was very different depending on the impedance phase angle. As the impedance phase angle was changed to more negative values, the mean wall shear stress decreased while the amplitude of wall shear stress increased. As the phase angle was reduced from 0°to -90°under $\pm$4% wall motion, the mean wall shear stress decreased by 12% and the amplitude of wall shear stress increased by 9%. Therefore, for hypertensive patients who have large negative phase angles, the ratio of amplitude and mean of the wall shear stress is raised resulting in a more vulnerable state to atherosclerosis according to the low and oscillatory shear stress theory. We also found that non-Newtonian characteristics of the blood protect atherosclerosis by decreasing the oscillatory shear index.

Laminar Flow Inhibits ER Stress-Induced Endothelial Apoptosis through PI3K/Akt-Dependent Signaling Pathway

  • Atherosclerosis preferentially involves in prone area of low and disturbed blood flow while steady and high levels of laminar blood flow are relatively protected from atherosclerosis. Disturbed flow induces endoplasmic reticulum (ER) stress and the unfolded protein response (UPR). ER stress is caused under stress that disturbs the processing and folding of proteins resulting in the accumulation of misfolded proteins in the ER and activation of the UPR. Prolonged or severe UPR leads to activate apoptotic signaling. Recent studies have indicated that disturbed flow significantly up-regulated $p-ATF6{\alpha}$, $p-IRE1{\alpha}$, and its target spliced XBP-1. However, the role of laminar flow in ER stress-mediated endothelial apoptosis has not been reported yet. The present study thus investigated the role of laminar flow in ER stress-dependent endothelial cell death. The results demonstrated that laminar flow protects ER stress-induced cleavage forms of PARP-1 and caspase-3. Also, laminar flow inhibits ER stress-induced $p-eIF2{\alpha}$, ATF4, CHOP, spliced XBP-1, ATF6 and JNK pathway; these effects are abrogated by pharmacological inhibition of PI3K with wortmannin. Finally, nitric oxide affects thapsigargin-induced cell death in response to laminar flow but not UPR. Taken together, these findings indicate that laminar flow inhibits UPR and ER stress-induced endothelial cell death via PI3K/Akt pathway.

  • The present study has numerically predicted both the flow -induced and thermally-induced residual stresses and birefringence in injection o. injection/compression molded center -gated disks. Analysis system for entire molding process was developed based on an ap propriate physical modeling including a nonlinear viscoelastic fluid model, stress-optical law, a linear viscoelastic solid model, free volume theory for density relaxation phenomena and a photoviscoelasticity and so on. Part I presents physical modeling a nd typical numerical analysis results of residual stresses and birefringence in the injection molded center-gated disk. Thermal residual stress was found to be extensional near the center, compressive near the surface and tend to become toward tensional at the surface. A double-hump profile was obtained across the thickness in birefringence distribution: nonzero birefringence is found to be thermally induced, the outer peak is due to the shear flow and subsequent stress relaxation during the filling stage a nd the inner peak is due to the additional shear flow and stress relaxation during the packing stage. Predicted birefringence including both the flow -induced and thermally-induced one becomes quite similar to the experimental one.

  • Intracellular gradients of the free calcium concentration are thought to be critical for the localization of functional responses within a cell. The mechanism of mechanotransduction may be associated with the localized accumulation of calcium stores for shear stress-exposed endothelial cells. The distribution of the calcium stores and the formation of the stress fibers were investigated by the indirect double immunofluorescent staining method with the calreticulin antibody and rhodamine phalloidin under flow condition. The shear stress of steady flow reorganized the cytoskeleton structure including the bundling and translocation to focal contacts. The calcium stores translocated from the cytoplasm to the focal contacting area. Consequently. accumulation of the calcium stores may participate in the shear stress-induced cytoskeleton organization of endothelial cells.

Assessment of flow-accelerated corrosion-induced wall thinning in SA106 pipes with elbow sections

  • A combination of flow-accelerated corrosion (FAC) tests and corresponding computational fluid dynamics (CFD) tests were performed to determine the hydrodynamic parameters that could help predict the highly susceptible location to FAC in the elbow section. The accelerated FAC tests were performed on a specimen containing elbow sections fabricated using commercial 2-inch carbon steel pipe. The tests were conducted at flow rates of 9 m/s under the following conditions: water temperature of 150 ℃, dissolved oxygen <5 ppb, and pH 7. Thickness reduction of the specimen pipe due to FAC was measured using ultrasonic testing. CFD was conducted on the FAC test specimen, and the turbulence intensity, and shear stress were analyzed. Notably, the location of the maximum hydrodynamic parameters, that is, the wall shear stress and turbulent intensity, is also the same location with maximum FAC rate. Therefore, the shear stress and turbulence intensity can be used as hydrodynamic parameters that help predict the FAC-induced wall-thinning rate. The results provide a method to identify locations susceptible to FAC and can be useful for determining inspection priority in piping systems.

  • We introduced mechanical stimuli and micropatterned substrate with micro fibers to investigate the effects of those on neurite outgrowth along with nerve growth factor (NGF) in vitro. Microfiber substrates were fabricated using an electrospinning process. And PC-12 cells cultured on substrates were simulated with nerver growth factor and laminar flow shear stress in a fluid flow system The results suggest that microfiber substrates and fluid-induced shear stress are promising for simulating neuronal regeneration in a desired direction.

  • When exposed to uniform magnetic fields externally applied, paramagnetic particles acquire dipole moments and the induced moments interacting with each other lead to the formation of chainlike structures or clusters of particles aligned with the field direction. A direct simulation method, based on the Maxwell stress tensor and a fictitious domain method, is applied to solve flows with magnetic chains in simple shear flow. We assumed that the particles constituting the chains are paramagnetic, and inertia of both flow and magnetic particles is negligible. The numerical scheme enables us to take into account both hydrodynamic and magnetic interactions between particles in a fully coupled manner, enabling us to numerically visualize breakup and reformation of the chains by the combined effect of the external field and the shear flow. Simple shear flow with suspended magnetic chains is solved in a periodic domain for a given magnetic field. Dynamics of interacting magnetic chains is found to be significantly affected by a dimensionless parameter called the Mason number, the ratio of the viscous force to the magnetic force in the shear flow. The effect of particle area fraction on the chain dynamics is investigated as well.

전교통동맥류 내부 유동 전산해석을 통한 낮은 벽면 전단 응력 영역 발달 분석 (Evolution of Low Wall-Shear Stress Area in Anterior Communicating Artery Aneurysm)

  • We analyzed the low wall-shear stress area in the intracranial aneurysm that occurred at an anterior communicating artery with a special emphasis on vortical structures close to the wall. We reconstructed the aneurysm model from patient CTA data. We assumed blood as an incompressible Newtonian fluid and treated the blood vessel as a solid wall. The pulsatile boundary condition was applied at the inlet of the anterior cerebral artery. From the instantaneous flow field, we computed the histogram of the wall-shear stress over the aneurysm wall and found the low wall-shear stress event (< 0.4 Pa). This extreme event was due to the low wall-shear stress area that occurred at the daughter sac. We found that the merging of two vortices induced the low wall-shear stress area; one arises from the morphological characteristics of the daughter sac, and the other is formed by a jet flow into the aneurysm sac. The latter approaches the daughter sac, which ultimately leads to the strong ejection event near the daughter sac.

유동 전단응력에 의한 선박 유기도막의 열화거동 연구 (A Study on the Aging Behavior of Ship Organic Coating by the Flow Induced Shear Stress)

  • 유기도막의 방식성능은 도막의 수지성분과 안료의 화학적 특성에 의존한다 전자는 부식인자의 침투를 차단 및 지연시키는 역할을 하며, 후자는 침투된 부식인자들에 의해 일어나는 부식반응을 억제하는 기능을 갖고 있다. 또한 도막 자체의 영향 외에, 외부 환경에 의해서 다르게 나타난다. 본 연구에서는 교류 임피던스 법을 이용하여 유속과 그에 따른 유동 전단응력에 의한 도막의 열화거동을 조사하였다. 실험기 사용된 도막의 두께는 $70{\mu}m$에서 $140{\mu}m$까지 변화시켰다. 두꺼운 도막($140{\mu}m$) 에서는 유속이 증가함에 따라 물 흡수량의 감소와 높은 임피던스 특성을 나타내었다. 그러나 얇은 도막($90{\mu}m$) 에서는 유속이 증가함에 따라 도막의 파괴가 진행되고 있음을 확인할 수 있었다. 도막에 가해지는 전단응력이 증가할수록, 즉 선박의 운항속도가 증대될수록 도막의 열화에 의하여 방식성능이 떨어짐을 확인할 수 있었다.

Ursodeoxycholic Acid (UDCA) Exerts Anti- Atherogenic Effects by Inhibiting Endoplasmic Reticulum (ER) Stress Induced by Disturbed Flow

  • Disturbed blood flow with low-oscillatory shear stress (OSS) is a predominant atherogenic factor leading to dysfunctional endothelial cells (ECs). Recently, it was found that disturbed flow can directly induce endoplasmic reticulum (ER) stress in ECs, thereby playing a critical role in the development and progression of atherosclerosis. Ursodeoxycholic acid (UDCA), a naturally occurring bile acid, has long been used to treat chronic cholestatic liver disease and is known to alleviate endoplasmic reticulum (ER) stress at the cellular level. However, its role in atherosclerosis remains unexplored. In this study, we demonstrated the anti-atherogenic activity of UDCA via inhibition of disturbed flow-induced ER stress in atherosclerosis. UDCA effectively reduced ER stress, resulting in a reduction in expression of X-box binding protein-1 (XBP-1) and CEBP-homologous protein (CHOP) in ECs. UDCA also inhibits the disturbed flow-induced inflammatory responses such as increases in adhesion molecules, monocyte adhesion to ECs, and apoptosis of ECs. In a mouse model of disturbed flow-induced atherosclerosis, UDCA inhibits atheromatous plaque formation through the alleviation of ER stress and a decrease in adhesion molecules. Taken together, our results revealed that UDCA exerts anti-atherogenic activity in disturbed flow-induced atherosclerosis by inhibiting ER stress and the inflammatory response. This study suggests that UDCA may be a therapeutic agent for prevention or treatment of atherosclerosis.
