Comparison of Aerodynamic Loads for Horizontal Axis Wind Turbine (I): with and without Turbulent Inflow (수평축 풍력터빈의 공력 하중 비교 (I): 난류 유입 유·무)

  • This study focused on the aerodynamic loads of the horizontal axis wind turbine blade due to the normal turbulence inflow condition. Normal turbulence model (NTM) includes the variations of wind speed and direction, and it is characterized by turbulence intensity and standard deviation of flow fluctuation. IEC61400-1 recommends the fatigue analysis for the NTM and the normal wind profile (NWP) conditions. The aerodynamic loads are obtained at the blade hub and the low speed drive shaft for MW class horizontal axis wind turbine which is designed by using aerodynamically optimized procedure. The 6-components of aerodynamic loads are investigated between numerical results and load components analysis. From the calculated results the maximum amplitudes of oscillated thrust and torque for LSS with turbulent inflow condition are about 5~8 times larger than those with no turbulent inflow condition. It turns out that the aerodynamic load analysis with normal turbulence model is essential for structural design of the wind turbine blade.

Simultaneous Determination of Six Components in the Traditional Herbal Medicine 'Oryeongsan' by HPLC-DAD and LC-MS/MS

  • A simple high performance liquid chromatography - diode array detector (HPLC-DAD) method has been developed and validated for simultaneous determination of the six components (cinnamic acid, cinnamaldehyde, eugenol, atractylenolide I, atractylenolide III, and ergosterol) in Oryeongsan. In addition, identification of six marker compounds was conducted by a LC-MS/MS analysis. The six compounds in Oryeongsan were separated on Shishedo $C_{18}$ column (5 ${\mu}m$, $4.6{\times}250$ mm) at a column temperature of $30^{\circ}C$. The mobile phase was a mixture of 0.1% trifluoroacetic acid (TFA) water and acetonitrile employing gradient elution at a flow rate of 1.0 mL/min. The detection wavelength was set at 205, 250, 280, and 330 nm. The developed method had good linearity ($R^2$ > 0.9997) and the limit of detection (LOD) and limit of quantification (LOQ) were observed within the ranges 0.01~0.15 and 0.05~0.45 ${\mu}g/mL$, respectively. The relative standard deviation (RSD) values of intra- and inter-day testing were indicated that less than 3% and 90.31~103.31% of accuracy. The results of recovery test were 90.56~106.72% and RSD range was measured from 0.84 to 2.95%. In conclusion, this HPLC-DAD method has been successfully applied to the simultaneous determination of six compounds in Oryeongsan samples.

  • The Large-scale structure of the circular jet in the transition region, which influences the subsequent flow in the turbulent region, was studied experimentally. Measuring equipments are composed of the two channel hot-wire anemometer, the computer controlled two-directional traverse mechanism, the data acquisition system, and FFT-analyzer. The circular jet has 50mm diameter. The mean velocity distribution, the velocity fluctuation, the auto 'cross correlations and the power spectra were acquired at moderate Reynolds number of 10$^{4}$. And the VITA method was used to measure the convection velocity of Large-scale eddy. The phase of u'is in advance of that of v'in all regions. .over bar. $R_{u}$(.tau.=0) is approximately zero in the potential core region, but a small regular deviation is observed. At a position in the mixing layer region the convection velocity is different along the part of the eddy, and in this experiment the convection velocity of the inner region is larger than the outer region. The averge convection velocity of the eddy along y/D=0 was approximately constant in the transition region.D=0 was approximately constant in the transition region.

Genetic Diversity and Population Structure of Chimaphila japonica in Southern Part of Korea (한국 남부지역의 매화노루발의 유전적 다양성과 집단구조)

  • Enzyme electrophoresis was used to estimate genetic diversity and population structure of Chimaphila japonica Miq. in Korea. The percent of polymorphic loci within the enzymes was 48.7%. Genetic diversity at the species level and at the population level was high (Hes=0.278 ; Hep=0.222, respectively), whereas the extent of the population divergence was relatively low ( $G_{ST}$ =0.079). $F_{IS}$ , a measure of the deviation from random mating within the 7 populations, was 0.355. An indirect estimate of the number of migrants per generation (Nm=2.61) indicates that gene flow is high among Korean populations of the species. In addition, analysis of fixation indices revealed a substantial heterozygosity deficiency in some populations and at some loci. Factors contributing to the high levels of genetic dive-rsity found in the entire species of C. japonica include wide distribution, long-lived perennials, ability to regenerate due to rhizomatous spread, outcrossing induced by animal vectors, and occasional pollen dispersal by wind.

  • Soil erosion by outflow of water or rainfall has caused many environmental problems as declining agricultural productivity, damaging pasture and preventing flow of water. As the interest in environment is increasing lately, soil erosion is considered as a serious problem, whereas the systematic regulation and analysis for that have not established yet. This research shows the method of extracting factor entered model which expects soil erosion by GSIS. There are several erosion model such as ANSWER, WEPP, RUSLE. The research used RUSLE erosion model which could expect general soil erosion connected easily with GSIS data. RUSLE's input factors are composed of rainfall runoff factor(R). soil erodibility factor(K), slope length factor(L), slope steepness factor(S), cover management factor(C) and support practice factor(P). The general equation used to extract L, S factor on the RUSLE to be oriented for agricultural area has some limitation to apply whole watershed. So, on this study we used a revised empirical equation applicable to the watershed by grid on the GSIS. Also, we analyzed RUSLE factors by watershed being analyzed with watershed extraction algorithm. Then we could calculate the minimum, maximum. mean and standard deviation of RUSLE factors by watershed.

Direct Determination of Tellurium in Simulated Nuclear Spent Fuels by Hydride Generation-Inductively Coupled Plasma Atomic Emission Spectrometry (수소화물 생성-유도결합플라스마 원자방출분광법을 이용한 모의사용후 핵연료 중의 텔루르 분석)

  • Tellurium in simulated nuclear spent fuels (SIMFUEL) has been determined by hydride generation-inductively coupled plasma atomic emission spectrometry (HG-ICP-AES). Parameters such as concentrations of HCl and $NaBH_4$, flow rate of HCl and $NaBH_4$ were optimized and then the effects of U, Mo, Pd, Rh and Ru on the Te intensity were investigated. A thiourea as a masking agent was used to eliminate or minimize such interferences specially caused by palladium. Tellurium was measured by HG-ICP-AES and ICP-MS after separation of tellurium from SIMFUEL with cation exchange chromatography. The relative deviation between direct measurement and separation method was less than 6% based on the data by ICP-MS.

A Preliminary Trophic Flow Model for Gwangyang Bay, Korea (광양만 예비 영양류 모형)

  • A preliminary quantitative model of the trophic structure in Gwangyang bay, Korea was obtained using ECOPATH and data from relevant studies to date in the region. The model integrates and analyzes biomass, food spectrum, trophic interactions and the key trophic pathways of the system. The bay model comprises 9 groups of benthic primary producer, phytoplankton, zooplankton, benthos, bivalve, pelagic fish, demersal fish and piscivorous fish. The total system throughput was estimated at $2.4\;kgWW/m^2/yr$, including a consumption of $41\%$, exports of $9\%$, respiratory flows of $24\%$ and flows into detritus of $26\%$. All of which originate from primary producers measured at $52\%$ and detritus of $48\%$. The total biomass was seen to be high compared to the levels of Somme, Delaware, Chesapeake Bays and Seine Estuary. This seems to be possibly due to artificial bivalve aquaculture and overestimation of benthos and benthic primary producer groups. The deviation can be calibrated by neglecting aquaculture and decreasing the habitat area for the groups. The trophic network of the bay shows a low level of recycling and organization as indicated by Finn's cycling index $3.3\%$, Ascendancy $3.1\;kgC/m^2/yr$ bits, Capacity $5.1\;kgC/m^2/yr$ bits and Redundancy $2.2\;kgC/m^2/yr$ bits. A high relative ascendancy of $62\%$ and a low internal relative ascendancy of $18\%$ indicate the system is not fully organized and stable towards disturbances, depending upon external connections. Although the model should be continuously provided with field data and calibrated further in depth, this study is the first trophic model applied to the region. The model can be a useful tool to understand the ecosystem in a quantitative manner.

Simultaneous Determination of Baicalein, Baicalin, Wogonin, and Wogonoside in Rat Plasma by LC-MS/MS for Studying the Pharmacokinetics of the Standardized Extract of Scutellariae Radix

  • A new composition of standardized Scutellariae Radix extract (HPO12) was developed for treatment of Alzheimer's disease. For the preclinical pharmacokinetic study of HPO12, a rapid, sensitive, and selective LCMS/MS method was developed and validated for the simultaneous determination of 4 bioactive compounds, baicalein, baicalin, wogonin, and wogonoside. After extraction with ethylacetate, chromatographic analysis was performed on a Thermo $C_{18}$ column ($150mm{\times}2.1mm$, $3{\mu}m$) with a mobile phase consisting of 0.1% formic acid (A) and 0.1% formic acid in 95% acetonitrile (B) by using gradient elution at a flow rate of $250{\mu}L/min$. Analytes introduced to a mass spectrometer were monitored by multiple reaction monitoring (MRM) in positive ion mode. Using $25{\mu}L$ of plasma sample, the method was validated over the following concentration ranges: 25-5000 ng/mL for baicalein, 20-40000 ng/mL for baicalin, 1-1000 ng/mL for wogonin, and 5-10000 ng/mL for wogonoside. The intra- and inter-day precision and accuracy of the quality control samples at the 4 concentrations showed $\leq$ 13.7% relative standard deviation (RSD) and 86.6-105.5% accuracy. The method was successfully applied to determine the concentrations of baicalein, baicalin, wogonin, and wogonoside in rat plasma after intraperitoneal and oral administrations of HPO12.

Quantitative Analysis of Lovastatin in Human Plasma and urine by Reversed-Phase High-Performance Liquid Chromatography (역상 고속액체크로마토그라프법을 이용한 혈장 및 뇨 중 로바스타틴의 정량)

  • Lovastatin (LOVA), a fungal metabolite isolated from cultures of Aspergillus terreus, is a competitive HMG-CoA reductase inhibitor used for the treatment of primary hyper cholesterolemia, and has also been shown to suppress growth in a variety of non-glioma tumor cell lines. A sensitive reversed-phase high-perfonnance liquid chromatographic method with ultraviolet (UV) absorbance detection has been developed to quantitate LOVA in human plasma and urine samples using liquid-liquid extraction procedure. Baseline separation of LOVA and internal standard, simvastatin was achieved on a Novapak $C_{18}$ analytical column with a mobile phase containing 0.025M $NaH_2PO_4$: CAN (35:65, v/v%), adjusted pH to 4.5. The flow rate was set at 1.5ml/min, and the column effluent was monitored by a UV detection at 238nm. The limit of quantification was determined to be 0.5${\mu}$g/ml while extraction efficiency of LOVA ranged from 73.4-82.9% at LOVA concentrations of 0.5 to 10${\mu}$g/ml. Good linearity with correlation coefficients greater than 0.999 was obtained in the range of LOVA concentrations from 0.5 to 10${\mu}$g/ml. The accuracy and the precision were proven excellent with relative standard deviation (RSD, %) and relative error (RE, %) of less than 4.2 and 4.0, respectively. Intraday precision, evaluated at five LOVA concentrations (0.5, 1, 2, 5, 10${\mu}$g/ml) and expressed as RSD ranged from 0-1.82% while the interday precision at the same concentrations ranged from 0.7-10.5%. The analytical method described was then successfully employed for the determination of LOVA concentrations in plasma samples obtained during a phase II clinical trial using high doses of LOVA (30-40mg/kg/day). This method could be further utilized for the ongoing pharmacolkinetic studies and therapeutic drug monitoring of the high-dose LOVA therapy in adenocarcinoma patients.

Determination of Uric Acid by Chemiluminescence Measurement Using Tris(2,2'-bipyridine)ruthenium(II)-Octylphenylpolyglycol Ether System

  • The determination of uric acid in urine samples was studied by a chemiluminescence measurement using tris (2,2'-bipyridine)ruthenium(II)-octylphenylpolyglycolether [Ru$(bpy)_3^{2+}$ -OPE] system. The oxidized uric acid by Ce(IV) excited Ru$(bpy)_3^{2+}$ to emit a chemiluminescence in this system so that the intensity was stoichio-metrically dependent upon the concentration of uric acid. In a reaction cell, a luminescent reagent, oxidant, surfactant and sulfuric acid were flowed into and mixed with a taken sample. Experimental conditions were optimized to obtain the maximum intensity of chemiluminescence. Each reactant solution of more than the following concentration gave a good result: $2\;{\times}\;10^{?4}$ M Ru$(bpy)_3^{2+}$ , 0.01 M Ce(IV), 6% OPE, and 0.33 M $H_2SO_4$. Any interferences were not shown in this process by the investigation of concomitant constitutes such as albumin, creatine, lactic acid, glucose, urea, $Cl^?,\; Mg^{2+},\;Ca^{2+}$ and so on. The linearity of a calibration curve was good with r = 0.998, the relative standard deviation of the slope was 3.3% and the detection limit was 5.6ng/mL. The recoveries of 80 to 91% were obtained from the standard spiked samples. The values were little bit low, but this procedure could be considered to be reliable for the determination of trace uric acid in urine samples.
