Three Predictive Tests Using Mice for the Identification of Contact Sensitizer

  • Predictive tests for the identification of contact sensitizing chemicals have been developed. We measured the sensitization potential with three predictive tests, the in vitro and the in vivo Local Lymph Node Assay(LLNA), ELISA to detect interferon-gamma(IFN-${\gamma}$) from supernatant and flow cytometry to detect change of cell surface proteins, using draining lymph nodes of mice. BALB/c mice were exposed to various chemicals or vehicles on the ears daily for 3 consecutive days in all experiments. With some exceptions of propyl paraben, neomycin sulfate, the in vivo LLNA was able to detect the sensitizing capacity of test chemicals and was more sensitive than the in vitro LLNA for chemicals used in the present study. In another experiment, contact sensitivity was assessed by the ELISA to detect IFN-Υ from the supernatants of the cultured LNCs after sensitization with chemicals. There was a good correlation between the LLNA and the IFN-Υ production for test chemicals. We also examined the change of cell surface proteins on LNCs after sensitization by flow cytometry for some cell adhesion molecules(ICAM-1, E-cadherine, B7 molecule), T cell markers(CD3, CD4, CD8, T$\alpha$$\beta$,T${\gamma}$$\delta$) and B cell markers(LR1, CD45R, I-Ad). The number of ICAM-1 positive cells and B cells in LNCs were increased after sensitization with DNCB, TNCB, isoeugenol and 25%, 50% cinnamic aldehyde compared with that of vehicle as a control. In conclusion, the in vivo LLNA could provide more sensitive screening test for moderate to strong sensitizers and some weak sensitizers including cosmetic raw materials than the in vitro LLNA. The production of IFN-Υ by allergen-activated LNCs might be a values indicators without radioisotopes for the identification of contact allergens. Detection of allergens by testing the increase of ICAM-1 positive cells and B cells in LNCs by flow cytometry might be used as a test method to detect allergens.

Determination of Cadmium(II) and Copper(II) by Flame Atomic Absorption Spectrometry after Preconcentration on Column with Pulverized Amberlite XAD-4 with Bismuthiol I

  • A column preconcentration method with pulverized Amberlite XAD-4 loaded with bismuthiol I (BI) has been developed for the determination of trace Cd(II) and Cu(II) in various real samples by flame atomic absorption spectrophotometry. Various experimental conditions, such as the size of XAD-4, adsorption flow rate, amount of bismuthiol I, stirring time for adsorbing bismuthiol I on XAD-4, pH of sample solution, amount of XAD-4- BI, desorption solvent, and desorption flow rate, were optimized. Also, the adsorption capacity and the adsorption rate of Cd(II) and Cu(II) on XAD-4-BI were investigated. The interfering effects of various concomitant ions were investigated, Bi(III), Sn(II) and Fe(III) were found to affect the determination. But the interference by these ions was completely eliminated by adjusting the amount of XAD-4-BI resin to 0.70 g, although the adsorption flow rate was slower. For Cd(II) our proposed technique obtained a dynamic range of 0.5-40 ng mL-1, a correlation coefficient (R2) of 0.9913, and a detection limit of 0.3 ng mL-1. For Cu(II), the corresponding values were 2.0-120 ng mL-1, 0.9921 and 1.02 ng mL-1. To validate this proposed technique, the aqueous samples (stream water, reservoir water, tap water and wastewater), the diluted brass sample and the plastic sample, as real samples, were used. Recovery yields of 91-103% were obtained. These measured data were not different from ICP-MS data at 95% confidence level. Our proposed method was also validated using rice flour CRM (normal, fortified) samples. From the results of our experiment, we found that the technique we present here can be applied to the determination of Cd(II) and Cu(II) in various real samples.

Determination of Glucose in Whole Blood by Chemiluminescence Method (화학발광법에 의한 전혈 중의 당 정량)

  • A method for the determination of glucose in human whole blood by chemiluminescence method using a stopped flow injection system has been studied. The method is based on the differences in the chemiluminescence intensities of luminol due to the different amounts of hydrogen peroxide produced from the glucose oxidase catalyzed reaction. The enzyme reactor was prepared by immobilization of glucose oxidase on aminopropyl glass beads and the chemiluminescence from a flow cell was measured by means of an optical fiber bundle. In order to obtain the optimum experimental conditions, effects of pH for the chemiluminogenic solution and enzyme reactor, flow rate and temperature on the chemiluminescence intensity were investigated. The calibration curve obtained under optimum experimental conditions was linear over the range from $1.0{\times}10^{-1}$ mM to 7.0 mM and the detection limit was $6.0{\times}10^{-2}$ mM. The proposed method was applied to the determination of glucose in whole human blood sample and the results were compared with those obtained by an official method. The present method was also evaluated by the results of recovery experiments.

  • An analytical method for the spectrometric simultaneous determination of the individual ions in the mix-tures of $Fe^{2+}$ and $Fe^{2+}$ ions utilizing a technique of flow injection analysis has been developed. The method was based on the oxidation reaction between $Fe^{2+}$ ion and $H_2O_2$ in an acidic medium and the subsequent formation of a red Fe$(SCN)^{3-x}_x$ ion by the complexation reaction between $Fe^{2+}$ ion and $SCN^-$ ion. Unlike the conventional methods which require separate processes for the pre-treatment of the sample solution, the current method uses the same FIA system for the pre-treatment and the analysis of the sample. The detection limit for the determination of $Fe^{2+}$ ion was found to be 6.00${\times}10^{-7}$M.

  • In this paper, we propose the NCP (Network Control Platform)-based defense mechanism against DDoS (Distributed Denial of Service) attacks in order to guarantee the transmission of normal traffic and prevent the flood of abnormal traffic. We also define defense modules, the threshold and packet drop-rate used for the response against DDoS attacks. NCP analyzes whether DDoS attacks are occurred or not based on the flow and queue information collected from SR (Source Router) and VR (Victim Router). Attack packets are dopped according to drop rate decided from NCP. The performance is simulated using OPNET and evaluated in terms of the queue size of both SR and VR, the transmitted volumes of legitimate and attack packets at SR.

  • Pulmonary function tests are widely used to diagnose and determine patients' therapy in clinic. And it was also applied in the research of the physiology and dynamics for lung disease. Among the pulmonary function tests, spirometry is the most easy and economic test. Spirometers are medical instruments that measure the instantaneous rate of volume flow of respired Bas. The mechanical spirometer was mostly used in the past. Up to the present, the most popular method of spirometer is the differential pressure technique with which change in the volume of flow are transferred to change in pressure. This kind of instrument suffers from several limitations, pressure drop, difficulty in maintenance and short period of calibration. Therefore, this study has begun to implement ultrasound spirometer, which is free of pressure loss and has wide range, focusing on the flow measurement technique and diagnostic algorithm.

Unsupervised Motion Pattern Mining for Crowded Scenes Analysis

  • Crowded scenes analysis is a challenging topic in computer vision field. How to detect diverse motion patterns in crowded scenarios from videos is the critical yet hard part of this problem. In this paper, we propose a novel approach to mining motion patterns by utilizing motion information during both long-term period and short interval simultaneously. To capture long-term motions effectively, we introduce Motion History Image (MHI) representation to access to the global perspective about the crowd motion. The combination of MHI and optical flow, which is used to get instant motion information, gives rise to discriminative spatial-temporal motion features. Benefitting from the robustness and efficiency of the novel motion representation, the following motion pattern mining is implemented in a completely unsupervised way. The motion vectors are clustered hierarchically through automatic hierarchical clustering algorithm building on the basis of graphic model. This method overcomes the instability of optical flow in dealing with time continuity in crowded scenes. The results of clustering reveal the situations of motion pattern distribution in current crowded videos. To validate the performance of the proposed approach, we conduct experimental evaluations on some challenging videos including vehicles and pedestrians. The reliable detection results demonstrate the effectiveness of our approach.

  • Successful early diagnosis of cancer diseases such as lung, prostate, liver and adrenocortical carcinoma is a key step in determining the cost of treatment, survival rate, and cure rate. Most of current cancer diagnosis systems including biopsy, computed tomography (CT), positron emission tomography (PET)-CT, magnetic resonance imaging (MRI), ultrasonography, etc., require expensive and complicated equipment with highly trained human resources. Global medical and scientific communities have thus made numerous efforts on developing effective but rapid disease management system via introducing a wide spectrum of point-of-care medical diagnosis devices. Among them, a lateral flow immunoassay strip technique has gained a great attention due to many advantages such as cost-effectiveness, short inspection time, and user friendly accessibility. In this mini-review, we will highlight recent research trend on the development of colorimetry based LFIA strips for cancer diagnosis and discuss the future research direction and potential applications.

Production of Spirometer 'The Spirokit' and Performance Verification through ATS 24/26 Waveform (휴대형 폐기능 검사기 'The Spirokit'의 제작 및 ATS 24/26파형을 통한 성능검증)

  • Background: This study aims to examine the useful- ness of the portable spirometer "The Spirokit" as a clinical diagnostic device through technology introduction, precision test, and correction. Design: Technical note Methods: "The Spirokit" was developed using a propeller-type flow rate and flow rate measurement method using infrared and light detection sensors. The level of agreement between the Pulmonary Waveform Generator and the measured values was checked to determine the precision of "The Spirokit", and the correction equation was included using the Pulmonary Waveform Generator software to correct the error range. The analysis was requested using the ATS 24/26 waveform recognized by the Ministry of Food and Drug Safety and the American Thoracic Society for the values of Forced Voluntary Capacity (FVC), Forced Expiratory Volume in 1second (FEV1), and Peak Expiratory Flow (PEF), which are used as major indicators for pulmonary function tests. All tests were repeated five times to derive an average value, and FVC and FEV1 presented accuracy and PEF presented accuracy as the result values. Results: FVC and FEV1 of 'The Spirokit' developed in this study showed accuracy within ± 3% of the error level in the ATS 24 waveform. The PEF value of 'The Spirokit' showed accuracy within the error level ± 12% of the ATS 26 waveform. Conclusion: Through the results of this study, the precision of 'The Spirokit' as a clinical diagnosis device was identified, and it was confirmed that it can be used as a portable pulmonary function test that can replace a spirometer.

A Study on the Relationship between Halitosis Developments and Oral Environmental (구취발생과 구강환경의 연관성에 관한 연구)

  • The purpose of this study was to analysis know the important oral environmental factors which affect halitosis components of the adult in order to provide basic data for halitosis prevention and establish a device to eliminate halitosis efficiently. The 97 adults who visited at the Dental Clinic in Metropolis (M=68, F=30) participated in this study that performed from March in 2009 to in 2010. The obtained results through items as caries status, periodontal status, salivary flow, the viscosity, pH, Snyder test, plaque deposit, tongue plaque and halitosis check were as followings. The average shame of halitosis components appeared at hydrogen sulfide 36.71 ppb methyl mercaptan 31.46ppb dimethyl sulfide 54.33 ppb and Ammonia 22.60 ppm. The normality and the detection comparative result dimethyl sulfide above reverse appeared highly at 46.9%, ammonia appeared highly at 52%. According to the Hydrogen sulfide level was a high relationship among age, CPI, tongue coat status, DMFT index which were statistically significant (p<0.05). According to the quantity of hydrogen sulfide level there was relationship where tongue coat status Saliva flow rate considers statistically(p<0.05). The quantity of methyl mercaptan level there was relationship where Dimethyl sulfide level, tongue coat status, Saliva flow rate considers statistically(p<0.05). The quantity of Dimethyl sulfide level there was relationship where Hydrogen sulfide level, ammonia level, tongue coat status, Saliva pH and Saliva flow rate considers statistically(p<0.05). Ammonia level there was relationship where Methyl mercaptan level, CPI, and Saliva flow rate considers statistically(p<0.05).
