칡추출물이 알코올을 급여한 흰쥐의 뇌조직에 미치는 영향 (The Effect of Puerariae thubergiana Bentham Extract on Brain Tissue in Alcohol-Treated Rats)

  • 칡추출물이 알코올성 뇌손상에 미치는 영향 을 구명하기 위해 알코올을 투여한 흰쥐에게 갈화와 갈근을 수준별 (I;1.2 g/kg B.W., II;2.4 g/kg B.W.) 로 5주간 급여한 후 \ulcorner 열수추출물이 알코올 대사와 유리기 생성 및 제거효소 활성에 미치는 영향을 관찰하였다. ADH 활성응ㄴ 갈화 및 갈근추출물 급여시 에탄올만 투 여한 대조군에 비하여 유의적으로 감소한 반면, ALDH 활성은 갈근추출물 급여군에서 유의 적으로 증가하였는데 1수준군의 증가정도가 현저하였다. P450 함량과 AD, AH 활성은 대조 군에 비하여 칡 열수추출물 급여시 감소되는 경향이었는데 갈근 급여군의 감소효과가 현저 하게 나타났다. AO와 XO 활성은 갈화 및 갈근추출물 급여군이 대조군에 비하여 감소되었 는데 특히 갈근추출물 1수준군에서 현저하게 나타났다. SOD 활성은 칡 열수추출물 급여시 증가하였으며 CAT와 GSH-Px 활성은 에탄올 투여로 증가된 활성이 갈근 열수추출믈 I 수 준 급여시 유의적으로 증가되었다. 이상의 결과에서 갈화 및 갈근 열수추출물 급여는 뇌조 직 중의 에탄올 대사효소계의 활성을 촉진시켰으며, 유리기제거 효소의 활성을 억제하고 항 산화효소계를 활성화하여 에탄올 투여에 인한 뇌조직의 산화적 스트레스를 완화시킬 수 있 을 것으로 사료된다.

The Effects of Puerariae Flos on Stress-induced Deficits of Learning and Memory in Ovariectomized Female Rats

  • Puerariae flos (PF) is a traditional oriental medicinal plant and has clinically been prescribed for a long time. The purpose of the present study was to examine the effect of PF on repeated stress-induced alterations of learning and memory on a Morris water maze (MWM) test in ovariectomized (OVX) female rats. The changes in the reactivity of the cholinergic system were assessed by measuring the immunoreactive neurons of choline acetyltransferase (ChAT) in the hippocampus after behavioral testing. The female rats were randomly divided into four groups: the nonoperated and nonstressed group (normal), the sham-operated and stressed group (control), the ovariectomized and stressed group (OS), and the ovariectomized, stressed and PF treated group (OSF). Rats were exposed to immobilization stress (IMO) for 14 d (2 h/d), and PF (400 mg/kg, p.o.) was administered 30 min before IMO stress. Results showed that treatments with PF caused significant reversals of the stress-induced deficits in learning and memory on a spatial memory task, and also increased the ChA T immunoreactivities. In conclusion, administration of PF improved spatial learning and memory in OVX rats, and PF may be useful for the treatment of postmenopausal-related dementia.

갈화(葛花)와 진피(陳皮) 추출물로 이루어진 복합물의 에스트로겐 활성과 파골세포 분화억제효과 (The effects of a Mixture of Puerariae Flos Extract and Citri Unshius Pericarpium Extract on Estrogenic Activities and Osteoclastogenesis)

  • Objectives : In this study, we examined the estrogenic activities and anti-osteo clastogenesis effects of PCE17, a mixture of PE (an extract of Puerariae Flos), and CE (an extract of Citri Unshius Pericarpium). Methods : The estrogenic effect of PCE17, PE and CE were examined by ER-β/ERE reporter gene assay and proliferation assay in 293 T and MCF-7 cells. The expression of estrogen-responsive gene and protein were checked by Real Time-PCR (RT-PCR) and Western blotting in MCF-7 cells. Inhibitory effect of PCE17, PE and CE on RANKL-induced osteoclast differentiation were evaluated by TRAP staining and RT-PCR in primary osteoclast precursors from rat bone marrow cells. Results : PCE17 and PE bind to ERs (estrogen receptors) and show estrogenic activities in 293T cells. They also stimulated the proliferation of MCF-7 cells and increased the expression of ER response gene, pS2. Tectorigenin, an active ingredient of PE, shows similar estrogenic activities in MCF-7 cells. PCE17 and CE inhibited RANKL-induced osteoclastogenesis in rat primary osteoclast precursor cells and down-regulated the osteoclast-specific genes of Nfatc1, Ctsk, and Acp5. Conclusions : In conclusion, PCE17 may have therapeutic potential in cases of menopause and osteoporosis.

장내 유용세균 Clostridium butyricum KCTC 1785의 성장을 촉진시키는 항산화 천연산물의 조합구성 (The Growth Promoting Effect of Useful Entevobacteria Clostridium butyricum KCTC 1785 by Combination of Natural Products Bearing Antioxidative Capacity)

  • C. butyricum은 선택되어진 천연산물의 한 종류 단독으로서도 성장이 촉진되었으며, 또한 두 종류, 세 종류 및 네 종류의 조합에 의하여 특징적으로 성장이 촉진되었다 이러한 결과로부터 항산화력이 강한 천연산물들을 사용하여 free radical의 제거와 동시에 C. butyricum의 성장 촉진물질로 사용할 수 있을 뿐 아니라, 생체의 기능을 향상시킬 수 있는 기능성 제제로의 응용 가능성을 보였다. C. butyricum을 CAB Medium에서 배양하였을 때, 구기자, 괴화, 백굴채, 백작약, 백출, 음양곽, 석창포, 진피, 당귀, 황금 및 오가피 등의 한가지의 천연산물에 의하여 성장이 촉진 되었고, 두 종류 천연산물의 조합인 백작약과 음양곽, 백작약과 진피, 백작약과 갈근, 백작약과 당귀, 백작약과 황금, 백작약과 괴화, 백작약과 녹차, 음양곽과 진피, 음양곽과 갈근, 음양곽과 황금, 음양곽과 괴화 등의 조합에 의하여 성장이 촉진되었으며, 백작약과 음양곽, 백작약과 진피, 백작약과 당귀, 음양곽과 갈근, 음양곽과 진피, 음양곽과 괴화의 조합 등은 대조군보다 2.0배 이상의 성장이 촉진된 것으로 나타났다. 이는 천연산물의 성분이 membrane의 구성 성분에 영향을 주기 때문으로 추정된다. 그리고 세 종류의 천연산물 조합인 음양곽과 갈근 및 괴화, 음양곽과 갈근 및 백작약, 음양곽과 갈근 및 당귀, 음양곽과 갈근 및 연자육, 음양곽과 갈근 및 황금 등의 조합에 의하여 성장이 촉진되었고, 음양곽과 갈근 및 연자육의 조합은 대조군 보다 1.7배의 성장 촉진 효과를 보였다. 4 종류 천연산물 조합인 음양곽, 갈근, 연자육 및 괴화의 조합, 음양곽, 갈근, 연자육 및 당귀의 조합은 대조군의 2.2배, 연자육, 백작약, 괴화 및 당귀의 조합, 갈근, 연자육, 백작약 및 괴화의 조합은 2.3배, 갈근, 백작약, 괴화 및 당귀의 조합은 2.0배의 성장 촉진 효과를 보여, 천연잔물의 조합에 의하여 생체에 probiotics의 역할을 하는 대부분의 유용균의 성장 향상을 위한 조합의 구성이 가능할 것으로 사료되고, 이들의 응용은 생체방어의 새로운 소재가 될 수 있을 것으로 판단된다 4 종류의 조합의 항산화력을 용존산소 분석법에 의하여 분석한 결과 연자육, 백작약, 괴화 및 당귀의 조합의 AUC가 631.45로써 가장 높았으며 이때의 성장촉진 효과는 2.3배로 나타났다. C. butyricum의 성장을 촉진하는 4 종류의 조합에 대한 hydroxyl radical의 소거활성은 50, 100, 200 및 300 배로 희석하여 검토하였을 때 음양곽, 갈근, 연자육 및 당귀의 조합이 200 배의 희석율에서 94.63%의 소거율을 보였고, 4 종류의 조합 모두 높은 희석율에도 90% 이상의 높은 소거율을 보여 높은 희석률을 필요로 하는 음료 제제로서의 탁월함을 보였으며, 또한, 4종류의 각 조합에 따른 방향족의 함량 및 총 phenol도 가공된 차보다 함량이 높은 것으로 나타났다.

갈화의 Prolyl Endopeptidase 저해 활성 Isoflavonoid 및 이들의 $^{13}C-NMR$ Assignment (Prolyl Endopeptidase-inhibiting Isoflavonoids from Puerariae Flos and Some Revision of their $^{13}C-NMR$ Assignment)

  • 갈화로부터 항치매 효과를 기대할 수 있는 prolyl endopeptidase(PEP) 저해제를 분리하기 위하여 갈화의 methanol 추출물을 chloroform 및 ethyl acetate로 분배추출 후 chloroform 가용성 분획에 대하여 silica gel, Sephadex LH-20 column chromatography 및 RP-HPLC를 행한 결과, $FeCl_3$,에 양성 반음을 나타내는 4종의 화합물을 얻었다. 이들을 $^1H-$, $^{13}C-$, $^2D-NMR$ 및 MS 등을 이용하여 분석한 결과, 각각 tectorigenin, genistein, 5,7-dihydroxy-4',6-dimethoxyisoflavone, 5-hydroxy-6,7,4'-trimethoxyisoflavone으로 동정하었으며 HMBC, HMQC를 통하여 $^{13}C-NMR$ signal들을 assign함으로써 기 보고된 data들의 오류를 바로잡았다. PEP에 대한 이 화합물들의 $IC_{50}$값은 각각 5.30 ppm$(17.7\;{\mu}M)$, 10.39 ppm$(38.5\;{\mu}M)$, 13.92 ppm$(44.3\;{\mu}M)$, 20.61 ppm$(62.8\;{\mu}M)$이었다.

Protective Effect of Kakkalide from Puerarie Flos on Ethanol-Induced Lethality and Hepatic Injury Is Expressed by Human Intestinal Microflora

  • The inhibitory effect of kakkalide isolated from Puerariae flos on ethanol-induced lethality and hepatic injury were investigated. Intraperitoneally treated Kakkalide was weakly reduced the mortality associated with administration of ethanol and did not reduce alcohol hepatotoxicity. However, orally administered kakkalide and intraperitoneally administered irisolidone significantly reduced serum ALT and AST activities on liver-injured mice by ethanol. (omitted)

지갈탕(枳葛湯)이 C57BL/6형 생쥐의 알코올섭취량에 미치는 영향 (Influence of Gigaltang on Alcohol Intake Amount in C57BL/6 mice)

  • Objective : This study was designed to test the anti-craving effect of Gigaltang(extracts of Oriental medicinal herbs ; Hovenia dulcis and Puerariae flos) in C57BL/6 mice. Anti-craving effect was substituted by alcohol intake amount of C57BL/6 mice. Method : 60 C57BL/6 mice was randomized into vehicle(water) group and Gigaltang group. Only 39(vehicle group(21) and Gigaltang group(18)) was eligible for this study, and 21 C57BL/6 mice was dropped out. The procedure was composed of 5 days of adjustment period and 32 days of alcohol exposure and withdrawal period with limited access paradigm and 10 days of treatment period. Alcohol was offered only for 2 hours a day from 2 to 4PM and water was offered for 22 hours left. Food was offered for 24 hours a day. 1140mg/Kg/day of Gigaltang was administrated to 18 mice of Gigaltang group for 10 days whereas water to 21 mice of vehicle group. The amount of alcohol intake, water intake, food intake, body weight were measured every other day. Result : There weren't significant differences in 2 hours of alcohol intake, 22 hours of water intake, 24 hours of food intake and body weight for ten days between Gigaltang and vehicle group. Conclusion : Anti-craving effect of Gigaltang on C57BL/6 mice wasn't verified. To verify the anti-craving effect, further study with advanced animal model, various dose of Gigaltang and diverse period of drug administration should be taken.

Quantitative Analysis of the Isoflavone Content in the Flower and the Root of Pueraria thunbergiana before and after Acid Hydrolysis

  • The contents of isoflavones and 1,1-diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging activities were determined in flowers (Puerariae Flos) and roots (Puerariae Radix) of Pueraria thunbergiana to differentiate both pharmacochemical or pharmacognostical usage of both oriental medicinal drugs. The tectorigenin contents in flower before and after acid hydrolysis were shown to be 17.10 ${\mu}mole\;g^{-1}$ and 49.58 ${\mu}mole\;g^{-1}$, respectively, analyzed by HPLC. However, the root displayed much less content of tectorigenin even after acid hydrolysis (6.56 ${\mu}mole\;g^{-1}$) than in the flower. Acid hydrolysis made glycitin and tectoridin of the isoflavone glycosides almost disappear in the flower and root. DPPH assay results demonstrated that acid hydrolysis of both extracts or both glycosides should increase free radical-scavenging activities due to the increase of isoflavone contents. It is also suggested that the flower had much higher concentration of isoflavonoids than in the root.

천연물로부터 알코올 탈수소효소 저해제 검색 (Screening of Alcohol Dehydrogenase Inhibitors from Natural Products)

  • Excessive or long term ingestion of alcohol may cause hepatitis, cirrhosis, hepatic tumor and so on. Aldehyde and active form of free oxygen that are metabolites of alcohol in liver are the cause of liver cell damage. The main system of alcohol metabolism is composed of alcohol dehydrogenase (ADH), aldehyde dehydrogenase (ALDH) and cytochrome P450. In connection with in vivo alcohol metabolism, more than one hundred natural products were screened for inhibition or activation of alcohol dehydrogenase. As a results, we found significant inhibition ($IC_50$) of ADH by methanolic extracts of Puerariae Radix ($61.2{\;}\mu\textrm{g}/ml$), Glycyrrhizae Radix ($105.0{\;}\mu\textrm{g}/ml$), Cinnamomi Ramulus ($7.0{\;}\mu\textrm{g}/ml$), Rhei Rhizoma ($36.7{\;}\mu\textrm{g}/ml$), Mori Cortex Radicis ($106.2{\;}\mu\textrm{g}/ml$), Chrysanthemi Flos ($112.2{\;}\mu\textrm{g}/ml$), Erycibes Caulis ($36.7{\;}\mu\textrm{g}/ml$), and Scutellariae Radix ($122.5{\;}\mu\textrm{g}/ml$)

Effect of Tectorigenin obtained from Pueraria thunbergiana Flowers on Phase I and -II Enzymes and Tissue Factor in the Streptozotocin-induced Diabetic Rat

  • We investigated the effect of tectorigenin (1) with hypoglycemic and hypolipidemic effects on Phase I and II enzymes and TF activity to elucidate the action of an immunosuppressive compound (1) in the diabetic rat. Compound 1 was obtained from the hydrolysis of tectoridin easily isolated from the flower of Pueraria thunbergiana(Leguminosae). Puerariae Flos has been used as therapeutics for diabetes mellitus in traditional medicine of Korea. (omitted)
