The objectives of this study were to identify the period of initiation of floral primordia in black locust (Robinia pseudoacacia L.) and subsequent development of floral buds until following spring. Four mature trees of black locust located in Suwon, Korea were selected. Bud samples were collected from the current-year shoots, starting from mid June to July every week, from August to October and from February to April every month. The buds were fixed in FAA solution, dehydrated, and imbedded in paraffin for microscopic observation. Buds collected on June 16, and 23, 1997, contained primitive primordia that might be interpreted as early floral primordia. By June 30, a bud showed a positive indication of inflorescence primordium with a well-formed shoot apex. All the inflorescence primordia observed throughout the collection periods were always associated with unique hairy appendages around the primordium and enclosed within a sclerenchymatous chamber. By July 7 and 15, a floral apex had early bud scales. By July 22, primitive inflorescence developed into visible arrangement of individual floral primordial By July 29, the inflorescence developed into whirl arrangement of individual floral primordia in a transverse section, but showed little further development until October 15. The inflorescence primordia seemed to over-winter at this stage. Buds collected from February 15 and March 24 the following year also showed no further development of inflorescence primordia. By April 7 the inflorescence started to show further development with elongated axis. At this time individual flowers were easily recognized.
To define the relations between endogenous GA levels and growth and flowering in short-day plant sorghum, growth retardant BX-112 was applied to two sorghum genotypes, wild-type and phytochrome B mutant (phyB-1), which grows faster and flowers earlier than the wild-type. BX-112 and $GA_3$ were applied as a soil drench, and plant height, culm length, and date to floral initiation were investigated. Endogenous GAs contents were measured with GC-MS-SIM. BX-112 treatments inhibited shoot growth in both genotypes and drastically reduced $GA_1$ and $GA_8$ levels. With increasing BX-112 concentrations, $GA_1$ concentrations declined linearly, but caused the accumulation of intermediates from $GA_12$ to $GA_20$. This result implies that $GA_1$ is the major active endogenous GA in shoot elongation in a short day plant sorghum. The inhibition of plant growth in both of wild type and phyB-1 by BX-112 was very similar, while BX-112 effects on floral initiation in two types of plants differed significantly. Floral initiation of phyB-1 was not affected by BX-1l2, but that of wild-type was delayed as BX-1l2 concentration increased. Because BX-112 treatment causes accumulation of biosynthetic intermediates between synthetic pathway from $GA_12$ to $GA_20$ and because phyB-1 is altered in GA metabolism in this same region of the early C13-hydroxylation pathway, BX-112 may fail to block flowering of phyB-1.
This experiment was conducted to extend utility of in-vitro culture material stalk and to mass produce uniform plants in Phalaenopsis. The optimal concentration of hyponex medium and plant hormone were examined. The most effective concentration of the hyponex in the flower stalk tissue culture of Phalaenopsis was $4mg{\cdot}L^{-1}$. The most effective concentration of TDZ and BA on the formation of multiple shoots in Phalaenopsis flower stalk culture was $0.3mg{\cdot}L^{-1}$ and $5mg{\cdot}L^{-1}$, respectively. TDZ was more effective for formation of the shoots and multiple shoots than BA at the basic medium with hyponex $4g{\cdot}L^{-1}$.
This study was carried out to establish the micropropagation system of Bupleurum latissimum Nakai that is a Korean native endangered species. Callus were induced from the leaf, petiole and floral bud and the percentage of callus formation was highest in the floral bud on the MS medium containing 2.0 mg ${\cdot}$$L^{-1}$ 2,4-D. Especially, callus induced from floral bud was formed 77.8% and the percentage of shoot formation was 42.6% on the MS medium containing 2.0 mg ${\cdot}$$L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$$L^{-1}$ TDZ. For simultaneously callus formation and shoot regeneration, 1/2 MS medium was more effective than MS medium. The percentage callus formation, shoot regeneration and rooting were 46.3%, 13.0%, 13.0% in 1/2 MS medium, respectively. Soot regeneration from callus was good in 1/2 MS medium supplemented with 2.0 mg ${\cdot}$$L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$$L^{-1}$ BA where percentage of shoot regeneration was 74.1 %, and the number of shoot per explant was 2.4. The percentage of rooting was lowest (57.8%) in control while it was highest (97.8%) in 1.5 mg ${\cdot}$$L^{-1}$ NAA. In acclimatization of regenerated plantlets, the percentage of survived plantlets was highest (86.1%), and plant height, root length and fresh weight were good in the soil for horticulture.

BACKGROUND: Kiwifruit, which was introduced to Korea in late 1970s, is a warm-temperate fruit tree, whose leaves are easily damaged by wind because of their large size. To produce high quality fruits, efficient windbreak is necessary to protect leaves until harvest. In Korea, typhoons from July onwards usually influence the production of kiwifruit. Damages from typhoons include low fruit quality in the current year and low flowering ratio the following year. This study was conducted to investigate the effect of early defoliation of kiwifruit vines from July to October on the regrowth of shoot axillary buds the current year and bud break and flowering the following year. METHODS AND RESULTS: Scions of kiwifruit cultivar 'Goldrush' were veneer grafted onto five-year-old Actinidia deliciosa rootstocks, planted in Wagner pots (13L) and grown in a rain shelter. Kiwifruit leaves in the proximity of leaf stalk were cut by lopping shears to simulate mechanical damage from typhoon since only leaf stalks were left when kiwifruit vines were damaged by typhoons. Kiwifruit vines were defoliated from July 15 to October 14 with one monthintervals and degrees of defoliation were 0, 25, 50, 75 and 100%. All experiments were conducted in the rain shelter and replicated at least five times. Defoliation in July 15 resulted in a high regrowth ratio of 20-40% regardless of degree of defoliation but that in August 16 showed only 5.8% of regrowth ratio in the no defoliation treatment; however, more than 25% of defoliation in August 16 showed 17-23% of regrowth ratio. In September 15, regrowth ratio decreased further to less than 10% in all treatments and no regrowth was observed in October 14. Percent bud break of all defoliation treatments were not significant in comparison to 64.7% in no defoliation except for 42.1% and 42.9% in 100% defoliation in July 15 and August 16, respectively. Floral shoot in the no defoliation treatment was 70.2% and defoliation of 50% or less resulted in the same or increased floral shoot ratio in July 15, August 16, and September 15; however, defoliation in October 14 showed no difference in all treatments. In flower number per floral shoot, 2-3 flowers appeared in no defoliation and only 1 flower was observed when the vines were defoliated more than 50% in July 15 and September 15. In October 14, contrary to the floral shoot ratio, flower number decreased with increased defoliation. CONCLUSION(S): Therefore, it is suggested that dormancy of 'Goldrush' axillary buds, was started in August and completed in October. The effect of defoliation on bud break of axillary buds the following year was insignificant, except for 100% defoliation in July 15 and August 16. From July 15 to September 15, floral bud ratio was significantly reduced when more than 50% of leaves were defoliated compared to no defoliation. Also, the number of flowers per flower-bearing shoot the following year decreased by less than 50% when compared to no defoliation, and this decrease was more prominent in September 15 than July 15 and August 16.
본 연구는 수선화 구근의 각 조직으로부터 기내배양시 캘러스형성과 신초재분화를 위한 성장조절제의 효과를 측정하기 써하여 시행되었다. 신초형성과 callus의 형성은 기저부를 포함하는 floral axis에서 $50\%$의 신초발아율을 관찰하였고 scale 에서는 신초형성이 거의 관찰되지 않았다. 그리고 floral axis만을 치상한 경우는 아주 저조한 신초형성율이 관찰되었다. 지저부를 포함한 floral axis의 신초형성은 callus의 형성과 같은 NAA 0.5 mg/L과 BA 1.0 mg/L을 포함하는 MS 배지에서 만족할만한 결과를 얻었다. 신초형성으로부터 모든 신초가 형성되기까지는 약 140일이 소요되었다. 신초가 형성된 구근조직을 NAA 5.0 mg/L과 TDZ 0.02 mg/L을 포함하는 뿌리유도 MS배지에서 뿌리가 유도되는 결과를 얻었다. 본 실험에서는 수선화의 재 분화에 있어서 구근의 조직에 따라 신초형성이 다르게 나타남을 발견 할 수 있었다.
In order to investigate the effects of low temperature pretreatment of floral bud and plant growth regulators on anther-derived callus and shoot differentiation, anthers were cultured on 1/2 MS medium supplemented with 2,4-D, NAA, BA and TDZ. This plant depends on the plant growth regulators, for these anthers couldn't respond on 1/2 MS medium without plant growth regulators. 2,4-D was a prerequisite substance in this experiment, especially 52.6% of callus formation on MS medium with 2.0mg/L 2,4-D alone. However, the optimum medium was on 1/2 MS medium with 0.1 mg/L 2,4-D and 1.0mg/L BA for continuous growth and shoot differentiation from the anther. Calli derived from on MS medium with 2.0mg/L 2,4-D transferred to the 1/2MS medium with TDZ and BA. TDZ were less superior to BA, only one anther could produce shoot on MS media with 1.0mg/L TDZ. On the other hand, when the calli transferred to the medium with 3.0mg/L BA, adventitious shoots were proliferated, subsequently, regenerated shoots elongated from the embryogenic calli. After floral buds of one week before anthesis were incubated at $5^{\circ}C$ refrigerator for eight or fifteen days, anthers seperated from floral buds were cultured on 1/2MS medium supplemented with 0.1mg/L 2,4-D and 1.0mg/L BA. Callusing and shoot differentiation on anthers from treated at $5^{\circ}C$ for eight days were more effective than those of fifteen days or control.
This study was conducted to correlate changes in plant growth and flowering behavior with inhibition of gibberellin synthesis following application of GA biosynthesis inhibitor. Two sorghum genotypes, wild-type and phyB-1(phytochrome B mutant) which grow fast and flowers early relative to the wild-type, were used. Both growth and floral initiation of these two genotypes were greatly affected by ancymidol concentration increased. However, these growth inhibition and delayed flowering are almost completely overcome by simultaneous applications of 31.6ppm $GA_3$. The ability of $GA_3$ to reverse the effect of the inhibition on both growth and floral initiation in sorghum suggests a role for native GAs in sorghum flowering. This result was contrast to the fact that in some long day plants GA biosynthesis inhibitors will inhibit shoot elongation but not floral initiation. In sorghum, inhibition of vegetative growth by GA biosynthesis inhibitor is accompanied by a delay in flowering. Ten ppm of ancymidol treatments drastically reduced all early-13-hydroxylation pathway GAs($GA_{12}$, $GA_{53}$, $GA_{19}$, $GA_{20}$, $GA_3$, and $GA_8$) levels.
Evolutionary studies conducted on NAC (NAM, ATAF1&2, and CUC2) genes for all major groups of land plants, indicate the presence of the NAC subfamilies, even in the early land plants. The varied roles played by NAC proteins in plant growth and development range from the formation of shoot apical meristem, floral organ development, reproduction, lateral shoot development, and defense responses to biotic and abiotic stresses. Considering the value and importance of solanaceous crops, the study of NAC proteins in these plants needs to be intensified. This will help to identify and functionally characterize their promoters, which will subsequently aid in engineering plants with improved performance under stressful conditions. In this review, the functionally characterized NAC transcription factors specific to tomato, potato, tobacco, chili pepper and eggplant (aubergine) are summarized, clearly indicating their biological functions in the defense mechanism of the plants, against biotic and abiotic stresses.

Among the various environmental conditions necessary for growing crops, light is closely related to the anthesis. This study aimed to determine the optimal photoperiod affecting floral differentiation in an edible flower, marigold, to efficiently cultivate the crops in a closed-type plant factory. The experiment was conducted with photoperiods of 4, 8, 12, and 16 hours. French marigold (Tagetes patula L.) 'Durango Red' seeds were sown in polyurethane sponges, and the photoperiod treatments were applied immediately. The extent of floral differentiation was examined at 2-3 day intervals, defined as the visible appearance of flower buds at least 2 mm in size. The growth parameters such as shoot fresh weight and dry weight, height, and leaf area were measured. The optimal photoperiod was determined based on the days when the floral differentiation had occurred in 50% of the total plants. In the 4-hour treatment, proper growth and flower buds did not appear. From the 8-hour treatment, the plant grew normally, and floral differentiation occurred, however, the 8-hour treatment showed the slowest floral differentiation compared to the 12 hours treatments or more. The 12- and 16-hour treatments didn't show significant differences in floral differentiation. While the 16-hour treatment exhibited the highest results in all growth parameters, it was not significantly different from the 12-hour treatment except for shoot dry weight and leaf area. According to the results, 8 hours of photoperiod induced floral differentiation. However, more time was required for flower bud formation, and plant growth was significantly lower compared to photoperiods of 12 hours or more. Considering the energy consumption and its growth, the optimal photoperiod for marigold was 12 hours.
