  • Ostrea denselamellosa and Eriocheir japonica samples were collected from the Seomjin River in 2019 as part of the "Research of Host-Associated Bacteria" research program. Almost 200 bacterial strains were isolated from the O. denselamellosa and E. japonica samples and subsequently identified by 16S rRNA gene sequencing. Among the bacterial isolates, ten strains possessed greater than 98.7% sequence similarity with published bacterial species that had not previously been recorded in Korea. These species were phylogenetically diverse, belonging to three phyla, four classes, seven orders, and eight genera. At the genus and class level, the previously unrecorded species belonged to Pseudoalteromonas, Aliivibrio, Rheinheimera, Leucothrix, and Shewanella of the class Gamma-proteobacteria, Olleya of the class Flavobacteriia, Algoriphagus of the class Cytophagia, and Lactococcus of the class Bacilli. The previously unrecorded species were further characterized by examining their Gram staining, colony and cell morphology, biochemical properties, and phylogenetic positions.

  • Various samples from island and coastal ecosystems in South Korea were investigated to discover unrecorded bacterial species. Soils from these areas, along with seawater samples, were plated on marine agar and R2A agar (containing 3% sea salt). From these samples, approximately 1,070 bacterial strains were isolated as single colonies and identified using 16S rRNA gene sequences. A total of 20 strains, which exhibited at least 98.7% similarity in their 16S rRNA gene sequences to those of validly published bacterial species not yet reported in Korea, were identified as unrecorded bacterial species. These strains belonged to three phyla, six classes, 10 orders, 14 families, and 16 genera. These were assigned as follows: Thioclava, Breoghania, Acidovorax, Erythrobacter, Paracoccus, Jiella, Aurantimonas, and Qipengyuania within the class Alphaproteobacteria; Pseudomonas, Cobetia, and Rheinheimera within the class Gammaproteobacteria; Aequorivita, Leeuwenhoekiella, and Polaribacter within the class Flavobacteriia; Algoriphagus within the class Cytophagia; and Microbacterium within the class Actinobacteria. The unreported species underwent further taxonomic characterization, which included assessments of Gram reaction, colony and cell morphology, biochemical and phylogenetic characteristics.

After aerobically incubating on marine agar and Reasoner's 2A agar, approximately 1,200 bacterial strains were isolated and identified using 16S rRNA gene sequences. A total of 36 strains showed ≥98.7% sequence similarity to previously published and validated bacterial species. However, these strains have not previously been reported in the Republic of Korea, indicating that they belong to Korean unrecorded bacterial species. The unrecorded bacterial species were assigned to the classes Actinomycetes, Bacilli, Bacteroidia, Flavobacteriia, Sphingobacteriia, Alphaproteobacteria, Betaproteobacteria, and Gammaproteobacteria. The information we obtained by examining the strains includes details of the Gram reactions, colony and cell morphology, biochemical characteristics, and phylogenetic positions of the unrecorded species.

  • Maribacter dokdonensis DSW-8 was isolated from the seawater off Dokdo in Korea. To investigate the genomic features of this marine bacterium, we sequenced its genome and analyzed the genomic features. After de novo assembly and gene prediction, 16 contigs totaling 4,434,543 bp (35.95% G+C content) in size were generated and 3,835 protein-coding sequences, 36 transfer RNAs, and 6 ribosomal RNAs were detected. In the genome of DSW-8, genes encoding the proteins associated with gliding motility, molybdenum cofactor biosynthesis, and utilization of several kinds of carbohydrates were identified. To analyze the genomic relationships among Maribacter species, we compared publically available Maribacter genomes, including that of M. dokdonensis DSW-8. A phylogenomic tree based on 1,772 genes conserved among the eight Maribacter strains showed that Maribacter speices isolated from seawater are distinguishable from species originating from algal blooms. Comparison of the gene contents using COG and subsystem databases demonstrated that the relative abundance of genes involved in carbohydrate metabolism are higher in seawater-originating strains than those of algal blooms. These results indicate that the genomic information of Maribacter species reflects the characteristics of their habitats and provides useful information for carbon utilization of marine flavobacteria.

  • As a result of the research project 'Survey and excavation of Korean indigenous species' to secure unrecorded species in Korea, a total of 14 unreported bacterial strains assigned to the phyla Deinococcus-Thermus and Bacteroidetes were isolated from various environmental habitats all around Korea. On the basis of 16S rRNA gene sequence similarity and affiliation to the clade with the closest species, it was concluded that the isolates represent deep groups of the phyla Deinococcus-Thermus and Bacteroidetes. There have been no records about these 14 unreported species in Korea; therefore 2 species of 2 genera in the class Deinococci within the phylum Deinococcus-Thermus, and 11 species of 9 genera in the class Flavobacteriia and one species in one genus in the class Sphigobacteriia within the phylum Bacteroidetes are described as unreported species found in Korea. Gram staining reaction, morphological and other biochemical characteristics are described in the species description section.

The unrecorded species were assigned to 18 different genera in five families: Flavobacterium, Epilithonimonas, Dokdonia, Gillisia, Flavicella, Chryseobacterium, Algibacter, Aquimarina, Lacinutrix, Gaetbulibacter, Cellulophaga, Tenacibaculum, and Maribacter of Flavobacteriaceae, Dyadobacter of Cytophagaceae, Draconibacterium of Draconibacterium_f, Sunxiuqinia of Prolixibacteraceae, and Fulvivirga of Fulvivirga_f. The selected isolates were subjected to further taxonomic characterization including analysis of Gram reaction, cellular and colonial morphology, biochemical activities, and phylogenetic trees. Descriptive information of the 28 unrecorded species is provided.
