Determination of the dietary lysine requirement by measuring plasma free lysine concentrations in rainbow trout Oncorhynchus mykiss after dorsal aorta cannulation

  • This study evaluated the dietary lysine requirement by measuring the plasma free lysine concentrations in rainbow trout, Oncorhynchus mykiss after dorsal aorta cannulation. A basal diet containing 36.6 % crude protein (29.6 % crystalline amino acids mixture, 5 % casein and 2 % gelatin) was formulated to one of the seven L-amino acid based diets containing graded levels of lysine (0.72, 1.12, 1.52, 1.92, 2.32, 2.72 or 3.52 % dry diet). A total of 35 fish averaging $512{\pm}6.8g$ ($mean{\pm}SD$) were randomly distributed into seven groups with five fish in each group. After 48 h of feed deprivation, each group of fish was fed one of the experimental diets by intubation at 1 % body weight. Blood samples were taken at 0, 5 and 24 h after intubation. Post-prandial plasma free lysine concentrations (PPlys, 5 h after intubation) of fish fed diets containing ${\geq}2.32%$ lysine were higher than those of fish fed diets containing ${\leq}1.92%$ lysine. Post-absorptive free lysine concentrations (PAlys, 24 h after intubation) of fish fed diets containing 2.32 and 3.52 % lysine were higher than those of fish fed diets containing ${\leq}1.52%$ lysine. The brokenline regression analysis on the basis of PPlys and PAlys indicated that the lysine requirement of rainbow trout could be 2.34 and 2.20 % in diet. Therefore, these results strongly suggested that the dietary lysine requirement based on the broken-line model analyses of PPlys and PAlys could be greater than 2.20 % but less than 2.34 % (corresponding to be $6.01%{\leq},but{\leq}6.39%$ in dietary protein basis, respectively) in rainbow trout. Also, these results shown that the quantitative estimation of lysine requirement by using PPlys and PAlys could be an acceptable method in fish.

Effects of Three Dietary Growth Hormones on Growth Performance and Lysozyme Activity in Juvenile Olive Flounder, Paralichthys olivaceus

  • In this study, tests were conducted to investigate the effects of three dietary growth hormones, administered in various amounts, on the growth performance and lysozyme activity in juvenile olive flounder, Paralichthys olivaceus. Three dietary growth hormones, recombinant human growth hormone (rHGH), recombinant bovine somatotropin A (rBST A) and recombinant bovine somatotropin B (rBST B) were tested at three different supplemental levels (10, 20 or 40 mg/kg body weight per week) by a $3{\times}3$ factorial design and a complete randomized design in comparison to a control group. Fish were fed one of the ten experimental diets (control, $rHGH_{10}$, $rHGH_{20}$, $rHGH_{40}$, rBST $A_{10}$, rBST $A_{20}$, rBST $A_{40}$, rBST $B_{10}$, rBST $B_{20}$ and rBST $B_{40}$) for 6 weeks and afterward were analyzed for growth performance by measuring weight gain (WG), feed efficiency (FE), specific growth rate (SGR) and protein efficiency ratio (PER). Based on the factorial design analysis, fish fed rHGH diets demonstrated significantly higher growth performance than fish fed rBST A or rBST B diets. However there were no significant differences in WG, FE, SGR and PER between fish fed rBST A and rBST B diets. Neither hormone level nor the interaction between the different hormones and their various levels had a significant effect on WG, FE, SGR, PER, lysozyme activity or whole-body proximate composition. A complete randomized design analysis confirmed fish fed $rHGH_{10}$, $rHGH_{20}$, $rHGH_{40}$, rBST $A_{10}$, rBST $A_{20}$, rBST $A_{40}$, rBST $B_{20}$ and rBST $B_{40}$ diets for 6 weeks showed higher WG than fish fed the control diet (P<0.05). A higher FE was observed in fish fed $rHGH_{10}$, $rHGH_{20}$, $rHGH_{40}$, rBST $A_{20}$ and rBST $A_{40}$ diets in comparison to fish fed the control diet. Fish fed all graded rHGH, rBST A and rBST B supplemented diets showed a higher SGR than fish fed the control diet. Regarding PER, fish fed $rHGH_{10}$, $rHGH_{20}$, $rHGH_{40}$, rBST $A_{10}$, rBST $A_{20}$, rBST $A_{40}$ and rBST $B_{20}$ diets were higher than fish fed the control diet. Furthermore, the lysozyme activity of fish fed a diet of $rHGH_{20}$ was significantly higher than that of fish fed any other diet. The results measuring the growth and development of the fish clearly suggest the biopotency of dietary rHGH could be higher than those of both dietary rBST A and rBST B. Further implied is the probability that within the range of 10 to 40 mg/kg BW/week the dietary growth hormones could accelerate growth performance, and that 20 mg rHGH/kg BW/week could possibly enhance lysozyme activity in juvenile olive flounder, Paralichthys olivaceus.

뱀장어 치어의 사료내 에너지에 대한 적정 단백질 비 평가 (Evaluation of the Optimum Dietary Protein to Energy Ratio of Juvenile Japanese eel, Anguilla japonica)

  • 본 실험은 치어기 Japanese eel (A. japonica)의 에너지 수준에 대한 사료내 적정 단백질 비를 평가하기 위하여 갈색어분(Brownfish meal)과 혈분(Blood meal)을 사용하여 조단백질 함량은 에너지에 대해 각각 45%와 50%,그리고 총에너지 함량은 각각 3800, 4150, 4500 kcal/kg의 반정제사료(semipurified)를 제조하여 16주간 사육실험을 실시하였다. 본 연구는 사료내 에너지 수준에 따라 단백질 함량을 달리 조절하여 사육실험을 실시하여 에너지 수준에 따른 적정 단백질 함량을 규명하는데, %증체율(WG), 일간성장률(SGR) 등의 성장평가와 사료효율(FE), 단백질효율(PER) 등의 사료영양학적 평가 및 혈액성상학적 평가와 연관해서 수행하였으며 실험 종료 후 어체내 일반성분 및 혈청 내 생화학적 조성을 비교 조사하였다. WG과 SCR에 있어서 $_{100}P_{45}$실험구가 다른 실험구들에 비해서 유의적으로 높게 나타났다(P<0.05). FE에 있어서 $_{100}P_{45}$실험구가 $_{100}P_{45}$ 실험구를 제외한 다른 실험구들에 비해서 유의하게 높은 것으로 나타났으며(P<0.05), SGR 역시 $_{100}P_{45}$ 실험구가 다른 실험구들에 비하여 유의적으로 높게 나타는데 $_{100}P_{45}$ 실험구와는 유의적인 차이가 없었으며, 이들을 제외한 나머지 실험구들간에 있어서 유의한 차이는 나타나지 않았다(P<0.05). 따라서, 본 결과를 통하여 치어기 Japanese eel (A. japonica)에 있어서 사료내 단백질 함량 45%, 에너지 4500 kcal/g가 최적의 성장을 나타낼 것으로 예측되며, 이때 단백질과 에너지 비율(P/E ratio)은 100mg protein/kcal가 적합한 것으로 판단된다.

Field Study on the Mycotoxin Binding Effects of Clay in Oreochromis niloticus Feeds and Their Impacts on the Performance as Well as the Health Status throughout the Culture Season

  • Total aflatoxin and ochratoxin were detected in 3 naturally contaminated fish feed samples using immune-affinity method. The results revealed that the average levels of aflatoxins in the 3 examined samples were (15, 22 and $12\;{\mu}g/kg$) respectively while the average levels of ochratoxins were (15, 6 and $6\;{\mu}g/kg$). The results of determination of the effects of clay as a mycotoxin binder on the health status and performance of Oreochromis niloticus in comparing with a control group revealed that the survival rate in control group was 81% after the end of the culture season. The results also revealed that the survival rate in group 2 which received clay treated feed was 86%. The results of regular parasitological examination revealed the identification of trichodina as external protozoa in Oreochromis niloticus from both ponds but without manifestation of disease signs. The results of bacteriological examination revealed the isolation and identification of Pseudomonas flouresence from some moribund Oreochromis niloticus. Higher performance parameters were recorded in group 2 that received feeds treated with clay which reflected in the total production which reaches 1646.47 kg while in the control pond, the total production was1308.36 kg.

On-farm evaluation of dietary animal and plant proteins to replace fishmeal in sub-adult olive flounder Paralichthys olivaceus

  • Background: High demand and low supply of fishmeal due to overexploitation of fisheries resources have resulted in a dramatic increase in the price of this ingredient. Olive flounder (Paralichthys olivaceus) commercial feed contains approximately 60% fishmeal and limited success has been achieved in identifying sustainable alternative protein sources for this species. Methods: An on-farm feeding trial was conducted to compare a basal diet containing 65% as the control (CONT) with two experimental diets replacing 10% of fishmeal by animal protein (AP10) or 20% of fishmeal by animal and plant protein (APP20). Sub-adult olive flounder averaging 327 ± 9.3 g (mean±SD) were fed one of the three diets in triplicate groups for 16 weeks. Results: Weight gain, specific growth rate, feed efficiency, protein efficiency ratio, and survival were not significantly different among fish fed all the experimental diets (P > 0.05). Also, non-specific immune responses (superoxide dismutase and lysozyme activity), serum biochemical parameters, and intestinal villi length were not significantly different among fish fed all the experimental diets (P > 0.05). Conclusions: Therefore, based on growth performance, non-specific immune responses, serum biochemical parameters, and intestinal histology, dietary animal and plant protein mixtures could replace up to 20% of fishmeal in the diet of sub-adult olive flounder.

Effects of two different organic acid blands in olive flounder

  • This experiment was conducted to evaluate the effect of two different organic acid products as antibiotic replacement in olive flounder paralichthys olivaceus. Fish averaging $3.5{\pm}0.05$ g($mean{\pm}SD$) were fed one of the ten semi-purified diets : Diet 1 ; Control, Diet 2 ; add antibiotics - 50mg OTC/kg body weight/day(OTC), Diet 3 ; Add organic acid bland A(OABA) - 4g/kg diet, Diet 4 ; add organic acid bland B(OABB) - 4g/kg diet for 10 weeks. Total gut microflora counts were significantly higher in the control group compared to the OTC and organic acid groups(P<0.05). Fish fed OABA, OABB and OTC had lower gut Vibrio counts compared to the control, but were not significantly different. Results from the challenge study indicate that mortality in the different treatment groups (50%) was significantly lower than those observed for the control group (100%). There were no differences in mortality between the OTC and organic acid groups. Overall findings from this study indicate that the organic acid blends A and B were as effective as oxytetracycline, an antibiotic, in regulating total gut bacterial numbers, Vibrio counts and providing protection against a pathogen such as Edwardsiella tarda.

  • Song-Gang® bio-stone (SGS) is a microporous crystalline hydrated aluminosilicate which has found various applications because of their very unique physiochemical characteristics such as ion exchange and absorptive-desorptive properties. Significant progress has been made in recent years on applications of these inorganic adsorbents in different industries including agriculture, aquaculture, water and wastewater treatment. This review article intends to summarize the published reports on the applications of SGS in aquaculture industry. SGS application as a feed additive to enhance fish growth and promote their health and nutritional parameters is the most important discussed areas. According to the technical data that are discussed in this review, SGS should be considered as a material with tremendous potential for application in the aquaculture industry. Considerable amounts of research works are under way to explore other opportunities for application of SGS to benefit aquaculture industry.

  • The characterization of mixed microorganisms that have activities of protein- and lipid-degradation were studied to reduce marine pollution by fish wastes and to utilize them again as feeds or fertilizers. There was no mutual inhibition among the microorganisms. The pH and temperature for the mixed culture were found to be 7 and $32^{\circ}C$, respectively. The fish waste were started to degrade after one day, and were almost degraded after 4 days without any odor.

복합미생물 프로바이오틱을 이용한 환경친화적 넙치 순환여과양식시스템에서의 미생물군집 분석 (Microbial community analysis of an eco-friendly recirculating aquaculture system for olive flounder (Paralichthys olivaceus) using complex microbial probiotics)

  • 본 연구는 순환여과양식시스템(RAS)에 있어서 복합프로바이오틱스의 적용이 넙치의 성장과 병저항성에 미치는 영향과 이 프로바이오틱스를 RAS에 생물증강처리 시 미생물군집 구조 및 수질에 미치는 영향을 평가하고자 실시하였다. RAS 내에서 80미의 넙치치어($25.7{\pm}7.6g$; $15.2{\pm}1.7cm$)에 프로바이오틱스 CES-AQ1를 첨가하여 사료를 제조하여(CES 사료; $1{\times}10^9\;CFU/kg$) 8주일 동안 급이하였다. 이 경우 넙치의 증체율, 비성장속도, 사료효율, 및 단백질 전환효율은 비유수식 양식시스템에 있어서 CON, PI 및 OTC 사료를 처리한 경우에 비해 1.5~2.5배 정도 높게 나타났다. 1주일간 병원균 저항성 시험에 있어서 비유수식에서 항생제함유 사료(OTC)를 급이한 경우와 RAS에서 CES 사료를 처리한 경우간에는 별 차이가 나타나지 않았다. 따라서 이 CES 프로바이오틱스를 RAS에서 넙치를 양식하는데 있어서 항생제 대용으로 활용할 수 있을 것으로 판단되었다. RAS의 생물여과막에서는 가장 높은 미생물다양성이 나타났으며 암모니아의 산화 및 탈질능을 가진 미생물이 관찰되었고, 병원미생물의 성장억제도 관찰되었다. 더구나 RAS 운전 19일 경과 시 암모니아가 0.5 mg/L이하의 농도로 감소하여 양호한 RAS 수질의 유지에 있어서 프로바이오틱스 처리가 효과가 있음이 밝혀졌다. 사료에 프로바이오틱스(CES-AQ1)를 첨가하여 넙치 장내 미생물이 안정화되고 또한 이 프로바이오틱스를 RAS 양식수에도 처리하여 RAS를 운전할 경우 건강한 넙치의 양식과 양호한 수질을 유지할 수 있어서 경제적이고 환경친화적인 넙치양식이 가능할 것으로 판단되었다.

Nitrogen allocation of Gracilaria tikvahiae grown in urbanized estuaries of Long Island Sound and New York City, USA: a preliminary evaluation of ocean farmed Gracilaria for alternative fish feeds

  • The red seaweed, Gracilaria tikvahiae McLachlan, was cultivated in open water farms in urbanized estuaries of Long Island Sound (26-30 psu of salinity) and New York City (20-25 psu), USA in 2011. Plants were harvested monthly from summer (August, $24^{\circ}C$) to fall (November, $13^{\circ}C$) and analyzed for total nitrogen, protein, and amino acid content. On a dry matter (DM) basis, nitrogen and protein significantly increased over the harvest period until October and then plateaued. Nitrogen increased from $22{\pm}1g\;kg^{-1}$ DM in August to $39{\pm}3g\;kg^{-1}$ DM in October (p < 0.001). Protein increased from $107{\pm}13g\;kg^{-1}$ DM in August to $196{\pm}5g\;kg^{-1}$ DM in November (p < 0.001). With two exceptions, amino acid concentrations expressed on a crude protein (CP) basis were similar over the harvest period. Essential amino acids accounted for $48{\pm}1%$ of all amino acids present with lysine and methionine averaging $56{\pm}2g\;kg^{-1}$ CP and $18{\pm}1g\;kg^{-1}$ CP, respectively. Histidine was underrepresented among essential amino acids and averaged $13{\pm}1g\;kg^{-1}$ CP. Taurine ranged from 2.1 to $3.2g\;kg^{-1}$ DM. With its moderate levels of lysine, methionine and taurine, ocean farmed G. tikvahiae has the potential of overcoming many nutrient deficiencies currently associated with terrestrial plant ingredients in alternative feeds for fish and shrimp.
