Low frequency plasma disinfectant effect in seawater and three major fish bacterial disease pathogens (저온 대기압 플라즈마를 이용한 해수 및 어류 병원성 세균 3종에 대한 살균소독효과)

  • Fish bacterial diseases have spread and caused serious problem for cultured marine fish in Korea. The important bacterial disease affecting mariculture such as olive flounder (Paralichthys olivaceus) are caused by Edwardsiella tarda, Vibrio scophthalmi and Streptococcus parauberis. For the bacterial disease protection in aquaculture industry, the water treatment is needed in aquaculture system. During the last decades atmospheric pressure non-thermal plasma in contact with liquids have received a lot of attention of environmental and medical application. In this study, we determined the disinfectant effect in seawater and three major fish bacterial disease pathogens by using low frequency plasma treatment. Three fish bacteria (E. tarda, V. schophthalmi, S. parauberis) were not detected within 16 min, 150 min and 270 min of 20 L, 500 L and 1 ton seawater post low frequency plasma treatment, respectively. Three major fish bacterial disease pathogens were not detected within 2 min after the low frequency plasma treatment, suggesting that the low frequency plasma possess disinfectant effectiveness.

  • Database System of approved fish drug products was consisted of trade name, species, ingredients, sponsor, disease, dose forms, drug forms, indication, law and so on. Also in addition, we will develope the database system for fish disease information. So we propose to expand and practice for publishing a medical prescription and making an order of drug by Fish Disease Distinction System which is based on drug information system and disease information system.

  • The author introduces the preventive and therapeutic methods of diseases in Japanese flounder which have been conducted by the Fish Disease Laboratory, the Mikamewan Fishery Cooperative Union, Ehime Prefecture, since 1982. Prevention 1. Addition of a sand substrate in the culture pond was effective to prevent ulcer disease. 2. Bathing in 0.5ppm of copper ion was effective to prevent some parasites. 3. Low stocking density of the fish reduced an incidence of edwardsiellosis or gliding bacterial disease. 4. Removal of the diseased fish prevented thd spread of lyphoeystis. 5. So-called $\pi$-water was effective to prevent the fry from some diseases. 6. Immersion of the juvenile in a sodium nifrusylate solution during transportation was effective to prevent gliding bacterial disease. Therapy 1. Sodium nifrustylate or oxytetracycline was effective to cure gliding bacterial disease. 2. Bathing in formalin(150ppm) or freshwater was effective to cure scuticociliatidosis. 3. Erythromycin was effective to cure $\beta$-hemolytic Streptococcus sp. infection. 4. Bathing in a hydrogen peroxide solution(1.5%) was effective to cure white spot disease.

Studies on Lymphocystis Diseases in Sebastes schlegeli (조피볼낙(Sebastes schlegeli)에 유행(流行)한 Lymphocystis병에 대(對)하여)

  • There prevailed lymphocystis disease with 1 to 2cm in size in the skin and pins of cultured Sebastes schlegeli in fish farms in Tongyoung-gun, Sanyang-myeon, Kon-ri from summer, 1987 to fall, 1988. Though there were some difference between each fish farm, this disease prevailed widely from immature fish to mature fish, less than 10cm to 30cm in body length with about 8 through 80% of infection rate. Sebastes schlegeli with the lymphocystis disease showed combined nipple-shaped mass by doubly, triply propagated lymphocystis. But the author considered that lymphocystis disease was not a direct cause of death of Sebastes schlegeli, the fish showed marked inflammation by seconary infection of pathogenic organisms.

Development of an environment-friendly moving aquatic animal rendering equipment and evaluation of fertilizer value for recycling of fish waste (친환경 이동식 수산생물 폐사체 처리장치 개발 및 재활용을 위한 비료 가치 평가)

  • Although aquaculture production rates grown over the years, aquatic animal diseases occur every year which causes substantial economic losses. When an aquatic animal is infected with an aquatic animal pathogen it is either incinerated or buried according to the aquatic life disease control act. Although these methods prevent the spread of disease, it is not environment friendly. Here, we developed an aquatic animal rendering equipment for disposal of fish waste which is environment-friendly and efficient. Also, fertilizer components of fish waste were evaluated value for recycling. The mobile rendering equipment was designed for field operation and/or high temperature and pressure system, oil and water separator, and shredding drying apparatus. During the experiment (July-2016 to November-2016), a total of 53,824 kg fish waste was collected, and 29,216 kg compost of rendering by-product was made. Also, compost made from viral (Viral hemorrhagic septicemia virus) infected fish did not reflect any detectable pathogen. The concentration of nitrogen, phosphorus, and organic matter in the fish waste compost were 2.17%, 26.98%, and 92.44%, respectively. The results suggest that fish waste used in this study was decomposed efficiently as per the official standard for fertilizer product. This equipment can be useful for efficient inactivation of the aquatic animal pathogenic agents and recycling of the fish waste in an environment-friendly manner.

Prevalence of Zoonotic Trematode Metacercariae in Freshwater Fish from Gangwon-do, Korea

  • The infection status of zoonotic trematode metacercariae was investigated in a total of 2,293 freshwater fish collected from 11 rivers or streams in 9 administrative regions of Gangwon-do, Korea for 5 years (2009-2013). All fish were collected by netting methods and examined using the artificial digestion methods. Clonorchis sinensis metacercariae were detected in 4 fish species, i.e., Pungtungia herzi, Squalidus japonicus coreanus, Acheilognathus rhombeus, and Ladislabia taczanowskii, from only Hantangang in Cheorwon-gun. Metagonimus spp. metacercariae were found in 1,154 (50.3%) fish and their average number per infected fish was 55.8. Among the positive fish species, especially Tribolodon hakonensis from Namdaecheon in Yangyang-gun and Plecoglossus altivelis from Osipcheon in Samcheok-si were most heavily infected. Centrocestus armatus metacercariae were detected in 611 (26.7%) fish and the average metacercarial burden per infected fish was 1,032. Two chub species, Zacco platypus and Zacco temminckii were highly and heavily infected with C. armatus metacercariae in almost all regions surveyed. Echinostoma spp. metacercariae were also found in 24 fish from a few localities, but their numbers per fish infected were very low. From the above results, it is confirmed that the metacercariae of intestinal flukes, especially Metagonimus spp. and C. armatus, were heavily infected, while C. sinensis metacercariae were rarely found in fish from Gangwon-do, Korea.

Low-value Fish used as Feed is a Source of Disease in Farmed Fish

  • Low-value fish is the most commonly used feed in Asian fish farms despite the fact that its application is controversial in regard to the sustainability and biosecurity of aquaculture. In this study, the causal agent of a disease outbreak at a Korean rockfish Sebastes schlegeli farm was investigated to determine whether the low-value fish used at the farm was the source. Infected Korean rockfish and Pacific sand eel used as feed were sampled from the farm, and bacterial cultures recovered from the internal organs of all sampled rockfish were isolated as pure cultures and later identified as Vibrio harveyi. The causal agent of the disease was also isolated from the kidneys of some of the sampled Pacific sand eels. This study provides additional evidence that the low-value fish used as feed at fish farms can be a key source of infectious diseases.

Bacterial diseases of flounder, Paralichthys olivaceus (넙치의 세균성(細菌性) 질병(疾病))

  • Flounder culture has been developed mainly in the western parts of japan, and, to date, following six bacterial diseases have been reported. Bacterial white enteritis occurs in 16 to 30-day-old flounder larvae and often causes mass mortality in seed production. Bacterium named Vibrio sp. INFL invades and multiplies in the mucosae of posterier part of intestine, and causes desquamative enteritis. Gliding bacterial disease occurs mostly in juvenile stage and in spring to summer. Diseased signs are partial discoloration and erosion of skin and fins. Histologically, epidermis are removed, and the causative bacterium, Flexibacter maritimus, multiplies on the surface of demis and invades into the muscular tissue. Vibriosis caused by Vibrio anguillarum and related organisum is one of the well-known diseases among marine fish. Outbreaks of the disease in flounder culture are relatively few, but mass mortalities in fingerlings due to the disease were reported. An outbreak of nocardiosis in the autumn of 1984 has been reported, but since then the disease scarcely occurred. The disease is characterized by formation of abscesses under the skin and white nodes in the gill, heart, spleen and kidney. Streptococcicosis occurs frequently in recent years. Beta-hemolytic streptococcus is the causative bacterium, which possesses the same biochemical and serological characteristics as $\beta$-streptococci isolated from some marine and freshwater fish, and is seemed to related to Streptococcus iniae. Edwardsiellosis is the disease that causes most damage in flounder culture in Japan. Characteristic symptoms are swelling of abdomen and intestinal protrusion from the anus due to accumulation of ascites. Edwardsiella tarda, a well-known pathogen of freshwater fish, is the causative bacterium of the disease.

Image Augmentation of Paralichthys Olivaceus Disease Using SinGAN Deep Learning Model (SinGAN 딥러닝 모델을 이용한 넙치 질병 이미지 증강)

  • In modern aquaculture, mass mortality is a very important issue that determines the success of aquaculture business. If a fish disease is not detected at an early stage in the farm, the disease spreads quickly because the farm is a closed environment. Therefore, early detection of diseases is crucial to prevent mass mortality of fish raised in farms. Recently deep learning-based automatic identification of fish diseases has been widely used, but there are many difficulties in identifying objects due to insufficient images of fish diseases. Therefore, this paper suggests a method to generate a large number of fish disease images by synthesizing normal images and disease images using SinGAN deep learning model in order to to solve the lack of fish disease images. We generate images from the three most frequently occurring Paralichthys Olivaceus diseases such as Scuticociliatida, Vibriosis, and Lymphocytosis and compare them with the original image. In this study, a total of 330 sheets of scutica disease, 110 sheets of vibrioemia, and 110 sheets of limphosis were made by synthesizing 10 disease patterns with 11 normal halibut images, and 1,320 images were produced by quadrupling the images.
