Effects of PTTG Down-regulation on Proliferation and Metastasis of the SCL-1 Cutaneous Squamous Cell Carcinoma Cell Line

  • Aims: To study effects of down-regulation of pituitary tumor-transforming gene (PTTG) on proliferation and metastasis ability of the SCL-1 cutaneous squamous cell carcinoma (CSCC) cell line and explore related mechanisms. Methods: SCL-1 cells were divided into 3 groups (untreated, siRNA control and PTTG siRNA). Cell proliferation assays were performed using a CCK-8 kit and proliferation and metastasis ability were analyzed using Boyden chambers. In addition, expression of MMP-2 and MMP-9 was detected by r-time qPCR and Western blotting. Results: Down-regulation of PTTG could markedly inhibit cell proliferation in SCL-1 cells, compared to untreated and control siRNA groups (P < 0.05). Real-time qPCR demonstrated that expression levels of PTTG, MMP-2 and MMP-9 in the PTTG siRNA group were 0.8%, 23.2% and 21.3% of untreated levels. Western blotting revealed that expression of PTTG, MMP-2 and MMP-9 proteins in the PTTG siRNA group was obviously down-regulated. The numbers of migrating cells ($51.38{\pm}4.71$) in the PTTG siRNA group was obviously lower than that in untreated group ($131.33{\pm}6.12$) and the control siRNA group ($127.72{\pm}5.20$) (P < 0.05), suggesting that decrease of proliferation and metastasis ability mediated by PTTG knock-down may be closely correlated with down-regulation of MMP-2 and MMP-9 expression. Conclusion: Inhibition of PTTG expression may be a new target for therapy of CSCC.

Effects of Tissue Factor, PAR-2 and MMP-9 Expression on Human Breast Cancer Cell Line MCF-7 Invasion

  • Objective: This study aimed to explore the expression of tissue factor (TF), protease activated receptor-2 (PAR-2), and matrix metalloproteinase-9 (MMP-9) in the MCF-7 breast cancer cell line and influence on invasiveness. Methods: Stable MCF-7 cells transfected with TF cDNA and with TF ShRNA were established. TF, PAR-2, and MMP-9 protein expression was analyzed using indirect immunofluorescence and invasiveness was evaluated using a cell invasion test. Effects of an exogenous PAR-2 agonist were also examined. Results: TF protein expression significantly differed between the TF cDNA and TF ShRNA groups. MMP-9 protein expression was significantly correlated with TF protein expression, but PAR-2 protein expression was unaffected. The PAR-2 agonist significantly enhanced MMP-9 expression and slightly increased TF and PAR-2 expression in the TF ShRNA group, but did not significantly affect protein expression in MCF-7 cells transfected with TF cDNA. TF and MMP-9 expression was positively correlated with the invasiveness of tumor cells. Conclusion: TF, PAR-2, and MMP-9 affect invasiveness of MCF-7 cells. TF may increase MMP-9 expression by activating PAR-2.

Construction of a artificial levee line in river zones using LiDAR Data (라이다 자료를 이용한 하천지역 인공 제방선 추출)

  • Mapping of artificial levee lines, one of major tasks in river zone mapping, is critical to prevention of river flood, protection of environments and eco systems in river zones. Thus, mapping of artificial levee lines is essential for management and development of river zones. Coastal mapping including river zone mapping has been historically carried out using surveying technologies. Photogrammetry, one of the surveying technologies, is recently used technology for national river zone mapping in Korea. Airborne laser scanning has been used in most advanced countries for coastal mapping due to its ability to penetrate shallow water and its high vertical accuracy. Due to these advantages, use of LiDAR data in coastal mapping is efficient for monitoring and predicting significant topographic change in river zones. This paper introduces a method for construction of a 3D artificial levee line using a set of LiDAR points that uses normal vectors. Multiple steps are involved in this method. First, a 2.5-dimensional Delaunay triangle mesh is generated based on three nearest-neighbor points in the LiDAR data. Second, a median filtering is applied to minimize noise. Third, edge selection algorithms are applied to extract break edges from a Delaunay triangle mesh using two normal vectors. In this research, two methods for edge selection algorithms using hypothesis testing are used to extract break edges. Fourth, intersection edges which are extracted using both methods at the same range are selected as the intersection edge group. Fifth, among intersection edge group, some linear feature edges which are not suitable to compose a levee line are removed as much as possible considering vertical distance, slope and connectivity of an edge. Sixth, with all line segments which are suitable to constitute a levee line, one river levee line segment is connected to another river levee line segment with the end points of both river levee line segments located nearest horizontally and vertically to each other. After linkage of all the river levee line segments, the initial river levee line is generated. Since the initial river levee line consists of the LiDAR points, the pattern of the initial river levee line is being zigzag along the river levee. Thus, for the last step, a algorithm for smoothing the initial river levee line is applied to fit the initial river levee line into the reference line, and the final 3D river levee line is constructed. After the algorithm is completed, the proposed algorithm is applied to construct the 3D river levee line in Zng-San levee nearby Ham-Ahn Bo in Nak-Dong river. Statistical results show that the constructed river levee line generated using a proposed method has high accuracy in comparison to the ground truth. This paper shows that use of LiDAR data for construction of the 3D river levee line for river zone mapping is useful and efficient; and, as a result, it can be replaced with ground surveying method for construction of the 3D river levee line.

The Response of the Solar Chromosphere and Transition Region to a Coronal Rain Event

  • We report that a strong downflow event caused three-minute oscillations in the solar atmosphere. Our observations were carried out by using the Fast Imaging Solar Spectrograph (FISS) of the 1.6 meter New Solar Telescope (NST) and the Interface Region Imaging Spectrograph (IRIS). Our main findings are as follows: (1) The strong downflow was seen at the $H{\alpha}$ absorption line at first, and then appeared at the Si IV and C II emission lines. It seems that the characteristics of the downflow are consistent with a coronal rain event. (2) After the event, oscillations of velocity were identified in the chromospheric lines and transition region lines. (3) The amplitudes of oscillations were 2km/s at Mg II line and 3km/s at C II and Si IV lines and decreased with time. (4) The period of the oscillation was 2.67 minutes at first, but gradually increased with time. Our findings are in agreement with Chae & Goode (2015)'s theory that of acoustic waves generated by a disturbance in a gravitationally-stratified medium.

  • The first line of antioxidant defense against reactive oxygen species includes the enzymatic activity of the superoxide dismutase (SOD) that catalyzes the disproportionation of superoxide to hydrogen peroxide and water. The SOD mainly removes highly toxic $O_2$$^{[-10]}$ and also prevents $O_2$$^{[-10]}$ mediated reduction of iron and subsequent OH$^{[-10]}$ generation. Along with an interest in SOD as a first line of defense against damage mediated by the superoxide anion, the SOD1 enzyme has been subjected to investigation in the molecular and cellular level.   • 345kV youngheung tansmission line which has been constructed in the western sea is the first and the largest marine project in the world crossing the youngheung island and the sihwa bay. It helps the stabilization of power supply greatly to cope with the rapid growth of the electric power consumption in the metropolitan areas and Gyeonggi province. Haying constructed the project successfully with the purely domestic-developed technologies, KEPCO is able to not only accumulate more advanced technologies of the marine project but also establish a bridgehead toward overseas market. It is very meaningful that we completed project successfully in spite of many difficulties including the uncertainty of the first project in the sea and strong objections from the environment groups and local residents. In this paper I will briefly introduce the project, newly developed technologies and materials.

Update on the Evidence Regarding Maintenance Therapy

  • Maintenance therapy has emerged as a novel therapeutic paradigm for advanced non-small-cell lung cancer (NSCLC). Maintenance therapy that aims to sustain a clinically favorable state after first-line chemotherapy has two strategies. Switch maintenance therapy entails switching to a new and non-cross-resistant agent in an alternating or sequential manner, on completion of first-line chemotherapy. Continuous maintenance therapy keeps ongoing administration of a component of the current regimen after four to six cycles of chemotherapy, if there is a stable disease, or better response. Both maintenance therapies can be continued, until disease progression. The potential evidence regarding maintenance therapy includes providing the opportunity to receive additional treatment, through sustaining tumor shrinkage, and delayed emergence of tumor-related symptom. Thus far, debates over the parameters used to predict the effectiveness of maintenance therapy, financial burden, and uncertainty of improving the quality of life exist. Despite many debates, maintenance therapy, which is currently recommended, has been disclosed to be beneficial.

Core Formation in a Turbulent Molecular Cloud

  • The two competing theories of star formation are based on turbulence and ambipoar diffusion. I will first briefly explain the two theories. There have been analytical (or semi-analytic) models, which estimate star formation rates in a turbulent cloud. Most of them are based on the log-normal density PDF (probability density function) of the turbulent cloud without self-gravity. I will first show that the core (star) formation rate can be increased significantly once self-gravity of a turbulence cloud is taken into account. I will then present the evolution of molecular line profiles of HCO+ and C18O toward a dense core that is forming inside a magnetized turbulent molecular cloud. Features of the profiles can be affected more significantly by coupled velocity and abundance structures in the outer region than those in the inner dense part of the core. During the evolution of the core, the asymmetry of line profiles easily changes from blue to red, and vice versa. Finally, I will introduce a method for incorporating ambipolar diffusion in the strong coupling approximation into a multidimensional magnetohydrodynamic code.

  • Among the various types of waiting line systems, the elevator servicing system is quite different from the usual queueing system in view of the service characteristics. For example, the FIFO discipline is not always valid depending upon the situation when the direction of first-come customer's is opposite of the operating elevator direction and at that time a later-arrived one has a luck to be served first. In this paper, a simulation model is constructed and tested by the sample data and the results have turned out to be fairly adequate. This model, therefore, will provide a good guide to anyone who is interested in the decision of optimal location selection of no-passenger elevator in high buildings whatsoever. This model is also available, with slight modification, to the problem of city bus dispatching or any other waiting line problems where the servicing equipments are moving.

  • In this paper, we propose a new switch element of buffered Banyan network and analysis it. The proposed switch element consists of CASO(Content ASsociated Output) buffers, its controller and 2*2 crossbar switch. This switch element increase the performance of buffered Banyan network by removing HOL blocking. Also, we analyze the proposed switch element by mathematical modelling method based on MY analysis model which is one of earier proposed models.
