An FMN-containing NADH-quinone reductase from streptomyces sp (An FMN-Containing NADH-Quinone Reductase from Streptomyces sp.)

  • NADH-quinone reductase was purified 22-fold from the cytosolic fraction of Streptomyces sp. Imsnu-1 to apparent hemogenity, with an overall yield of 9%, by the purification procedure consisting of ammonium, sulfate precipitation and DEAE Sephacryl S-200 and DEAE 5 PW chromatographies. Thes molecular mass of the enzyme determined by gel filtration chromatography was found to be 110 kDa. SDS-PAGE revealed that the enzyme consists of two sugunits with a molecular mass of 54 kDa. The enzyme contained 1 mol of FMN per subunit as a cofactor. The $A_{272}$ A$_{457}$ ratio was 6.14 and the molar extinction coefficients were calculated to be 20, 800 and 25, 400M$^{-1}$ $cm^{-1}$ / AT 349 AND 457 nm, respectively. The N-terminal sequence of the enzyme contained the highly conserved fingerprint of ADP-binding domain. The enzyme used NADH as an electron donor and various quinones as electron acceptors. Cytochrome c was practically inactive. Air-stable flavin semiquinone was produced by the addition of NADH to the enzyme. Also, naphthosemiquinone was detected in the reaction mixture containing the enzyme.

  • A polysaccharide with antithrombin activity in Manda$^{(R)}& (PAM) was purified via procedures comprising three major steps, i.e. fractional precipitation with ethanol, anion exchange chromatography, and gel permeation chromatography. PAM showed a symmetrical peak on size exclusion HPLC, as assessed by refractive index, and behaved as a single band on cellulose acetate electrophoresis. The average molecular mass was estimated to be 222 kDa by gel filtration. PAM was found to be a sulfated heteropolysaccharide that contains sulfate group (20.5%, w/w) and uronic acid moiety (7.1 %, w/w) in addition to neutral sugar consisting of fucose, xylose, mannose, galactose, and glucose in a molar ratio of 1.00 : 0.35 : 0.28: 0.22 : 0.15. This polysaccharide appeared to inhibit blood coagulation via the intrinsic pathway in a dose-dependent pattern. The clotting of fibrinogen by thrombin was also significantly mitigated by the presence of PAM.

Analysis of sewage treatment process for the improvement of T-N removal process (T-N 제거공정 개선을 위한 하수처리장 공정 분석)

  • In order to design the improvement process for T-N removal, the treatment process of Suyoung, Gangbyeon, and Noxan sewage treatment plants (STP) in Busan was anlayzed. Suyoung STP shows a T-N removal efficiency of about 69.8% with MLE(Modified Ludzack ettinger) and A2O+MBR. However, it is necessary to improve the process to maintain over DO of 1 mg/L and is required to install a flow control tank to minimize the rainfall effect. Gangbyun STP shows a about 70.2% T-N removal efficiency with A2O+GFF(gravity fiber filtration). However, in order to improve T-N removal efficiency, it is needed to install MLE process to treat recycle water. Noksan STP shows a T-N removal efficiency of about 71.0% with MLE+Chemical treatment and shows stable T-N concentration in effluent. However, it is required a toxic chemical management process because bad wastewater flows into the STP, also is necessary a process improvement in order to increase internal recycling ratio. Especially, it is required a process improvement to increase HRT of nitrification tank because Suyoung and Gangbyeon STPs shows low nitrification efficiency during winter season.

Polymer Base Bored Pile in Bangkok Subsoils

  • The bentonite slurry has been used as the stabilize suspension for wet process bored pile construction in Thailand. The bentonite suspension has benefit on filtration in the sand layer, but it creates thick cake film along pile shaft and loose sedimentation at pile toe. The base grouting technique was widely used to rectify the soft base or loose sedimentation problem of bored pile. The base grouting technique was not increased only end bearing capacity, but was also more increase in skin friction capacity of the bored piles. The comprehensive researches on base grouting was carried out by installing PVC casing inside the shaft to allow the drilling through the pile base in order to collect the soil sample below the pile tip. The polymer based slurry recently was used to replace the bentonite slurry to overcome the thick cake film along pile shaft as well as loose sedimentation at pile toe. The extent research on polymer slurry by physical model was performed to verify the real behavior of polymer. The appropriate mixing ratio of polymer was proposed. The design skin friction coefficient, $\beta$ and end bearing coefficient, Nq, for sand layer base on fully instrumented tested pile were proposed. The application on remedial of the lose capacity bored pile with large displacement in Bangladesh was proposed and discussed.

  • Using $6wt%Y_2O_3-2wt%Al_2O_3$ as sintering additives and Si as a raw powder, the continuously porous in-situ $Si_2N_2O-Si_3N_4$ bodies were fabricated by multi-pass extrusion process and their microstructures were investigated depending on the addition of carbon (0-9wt%) in the mixture powder. The introduction of $Si_2N_2O$ fibers observed in the unidirectional continuous pores as well as in the pore-frame regions of the nitrided bodies can be an effective method in increasing the filtration efficiency. In the case of no carbon addition, the network type $Si_2N_2O$ fibers with high aspect ratio appeared in the continuous pores with diameters of 150-200 nm. However, in the case of 9wt% C addition, the fibers were found without any network type and had diameters of 200-250 nm.

Pilot Scale Test of Non-woven Fabric Filter Separation Activated Sludge Process for Practical Application on Domestic Wastewater Reclamation (파일럿 규모의 침지식 부직포 여재 활성슬러지 공정의 시스템 처리 특성에 관한 연구)

  • A pilot scale non-woven fabric filter separation activated sludge system was investigated for practical application on domestic wastewater reclamation and reuse. The system was operated in A/O (Anaerobic/Oxic) process with submerged filter module in the aerobic compartment. In the test of two types of filter materials ($70g/m^2$ and $35g/m^2$), the initial flux (0.42m/d) could be maintained for about three months by regular air backwashing of $70g/m^2$ filter at 0.3m water head. The removal efficiency of organic matter by the system was BOD 93.3%, CODcr 96.3%, SS 96.7%. The effluent quality was 7.8mg/L, 12mg/L and 5mg/L for BOD, CODcr and SS, respectively. The water quality was enough to meet a standard for domestic reuse without human contact. T-N removal efficiency was 49.9% at internal recycle rate 2Q and C/N ratio 3.3. The removal efficiency of T-P was 50% with average effluent concentration, 2.6mg/L.

Partial Purification and Characterization of a Cysteine Protease Inhibitor from the Plerocercoid of Spirometra erinacei

  • Helminthic cysteine proteases are well known to play critical roles in tissue invasion, nutrient uptake, and immune evasion of the parasites. In the same manner, the sparganum, the plerocercoid of Spirometra mansoni, is also known to secrete a large amount of cysteine proteases. However, cysteine protease inhibitors regulating the proteolytic activities of the cysteine protease are poorly illustrated. In this regard, we partially purified an endogenous cysteine protease inhibitor from spargana and characterized its biochemical properties. The cysteine protease inhibitor was purified by sequential chromatographies using Resource Q anion exchanger and Superdex 200 HR gel filtration from crude extracts of spargana. The molecular weight of the purified protein was estimated to be about 11 kD on SDS-PAGE. It was able to inhibit papain and 27 kDa cysteine protease of spargana with the ratio of 25.7% and 49.1%, respectively, while did not inhibit chymotrypsin. This finding suggests that the cysteine protease inhibitor of spargana may be involved in regulation of endogenous cysteine proteases of the parasite, rather than interact with cysteine proteases from their hosts.

Effect of Chlorella intake on Cadmium metabolism in rats

  • This study was performed to investigate the effect of chlorella on cadmium (Cd) toxicity in Cd-administered rats. Sixty male Sprague-Dawley rats (14 week-old) were blocked into 6 groups. Cadmium chloride was given at levels of 0 or 325 mg (Cd: 0, 160 ppm), and chlorella powder at levels of 0, 3 and 5%. Cadmium was accumulated in blood and tissues (liver, kidney and small intestine) in the Cd-exposed groups, while the accumulation of Cd was decreased in the Cd-exposed chlorella groups. Fecal and urinary Cd excretions were remarkably increased in Cd-exposed chlorella groups. Thus, cadmium retention ratio and absorption rate were decreased in the Cd exposed chlorella groups. Urinary and serum creatinine, and creatinine clearance were not changed in experimental animals. In addition, metallothionein (MT) synthesis in tissues was increased by Cd administration. The Cd-exposed chlorella groups indicated lower MT concentration compared to the Cd-exposed groups. Moreover, glomerular filtration rate (GFR) was not changed by dietary chI orella and Cd administration. According to the results above, this study could suggest that Cd toxicity can be alleviated by increasing Cd excretion through feces. Therefore, when exposed to Cd, chlorella is an appropriate source which counteracts heavy metal poisoning, to decrease the damage of tissues by decreasing cadmium absorption.

  • The durg-macromolecule conjugates i.e. 5-fluorouracil-1-acetyl-human serum albumin(FU-AA-HSA, 8) and 5-fluorouracil-1-acetyl-poly-L-lysine(FU-AA-polylys, 9) have been synthesized and purified by sephadex G-25 gel filtration with 0.05M phosphate buffer(pH 7.5). The analyses of conjugates gave the molar ratio of FU-AA : HSA of 70-100:1 and FU-AA: poly-L-lysine of 109:1. The weight percent of FU-AA(as $FU-CH_2CO-$) in FU-AA-HSA conjugate was 16-22% and the one in FU-AA-polylys was 22.4%.

  • Recently, as the fine dust is increased and the emission regulations of diesel engines are strengthened, interest in diesel soot filtration devices is rapidly increased. In particular, there is a demand for technology development for higher efficiency of diesel exhaust gas after-treatment devices. As part of this, many studies conducted to increase the exhaust gas treatment efficiency by improving the flow uniformity of the exhaust gas in the DPF and reducing the pressure drop between the inlet and outlet of disel particle filter (DPF). In this study, computational fluid dynamics (CFD) simulation was performed when exhaust gas flows into the canning reduction device equipped with a 13" asymmetric DPF in order to maintain the flow uniformity in the diesel exhaust system and reduce the pressure. In particular, a study was conducted to find the geometry with the smallest pressure drop and the highest flow uniformity by simulating the DPF I/O ratio, exhaust gas temperature, inlet-outlet pressure and flow uniformity according to the geometry and hole size of distributor.
