  • Objectives: Wastewater treatment plants typically produce a large volume of waste sludge. In this study, the conditioning and dewatering properties of a digested and thickened sludge from an industrial wastewater treatment plant were investigated in order to improve the dewaterbility of the sludge. Methods: Jar-tests and Buchner funnel tests were carried out to assess the conditioning and dewatering properties of a waste sludge. TTF (Time to Filter Test) and SRF (Specific Resistance to Filtration) were adopted as the indices of sludge dewaterbility. Results: The valuation indices influencing the dewaterbility of the waste sludge, including TTF, SRF, water contents, VS/TS ratio and turbidity, were measured. The TTF and SRF of the digested and thickened sludge were decreased to 40 sec, $3.43{\times}10^{12}$ m/kg, and 39 sec, $1.09{\times}10^{12}$ m/kg, respectively. Conclusions: The conditioner composed of natural inorganic materials turned out to be effective in the reduction of sludge water contents.

Preparation and characterization of PVDF/TiO2 composite ultrafiltration membranes using mixed solvents

  • To study the effect of titanium dioxide ($TiO_2$) nanoparticles on membrane performance and structure and to explore possible improvement of using mixed solvents in the casting solution, composite polyvinylidene fluoride (PVDF) ultrafiltration membranes were prepared via immersion precipitation method using a mixture of two solvents triethyl phosphate (TEP) and dimethylacetamide (DMAc) and addition of $TiO_2$ nanoparticles. Properties of the neat and composite membranes were characterized using scanning electron microscope (SEM), energy dispersive X-ray spectroscopy (EDS), Atomic force microscopy (AFM) and contact angle and membrane porosity measurements. The neat and composite membranes were further investigated in terms of BSA rejection and flux decline in cross flow filtration experiments. Following hydrophilicity improvement of the PVDF membrane by addition of 0.25 wt.% $TiO_2$, (from $70.53^{\circ}$ to $60.5^{\circ}$) degree of flux decline due to irreversible fouling resistance of the composite membrane reduced significantly and the flux recovery ratio (FRR) of 96.85% was obtained. The results showed that using mixed solvents (DMAc/TEP) with lower content of $TiO_2$ nanoparticles (0.25 wt.%) affected the sedimentation rate of nanoparticles and consequently the distribution of nanoparticles in the casting solution and membrane formation which influenced the properties of the ultimate composite membranes.

Characterization of ${\beta}-Galactosidase$ from a Bacillus sp. with High Catalytic Efficiency for Transgalactosylation

  • A ${\beta}$-galactosidase with high transgalactosylic activity was purified from a Bacillus species, registered as KFCC10855. The enzyme preparation showed a single protein band corresponding to a molecular mass of 150 kDa on SDS-PAGE and gave a single peak with the estimated molecular mass of 250 kDa on Sephacryl S-300 gel filtration, suggesting that the enzyme is a homodimeric protein. The amino acid and sugar analyses revealed that the enzyme is a glycoprotein, containing 19.2 weight percent of sugar moieties, and is much more abundant in hydrophilic amino acid residues than in hydrophobic residues, the mole ratio being about 2:1. The pI and optimum pH were determined to be 5.0 and 6.0, respectively. Having a temperature optimum at $70^{\circ}C$ for the hydrolysis of lactose, the enzyme showed good thermal stability. The activity of the enzyme preparation was markedly increased by the presence of exogenous Mg (II) and was decreased by the addition of EDTA. Among the metal ions examined, the most severely inhibitory effect was seen with Ag (I) and Hg (II). Further, results of protein modification by various chemical reagents implied that 1 cysteine, 1 histidine, and 2 methionine residues occur in certain critical sites of the enzyme, most likely including the active site. Enzyme kinetic parameters, measured for both hydrolysis and transgalactosylation of lactose, indicated that the enzyme has an excellent catalytic efficiency for formation of the transgalactosylic products in reaction mixtures containing high concentrations of the substrate.

Aspergillus sp. LAM 94-142가 생산하는 세포응집물질의 특성 (Characterization of a Cell Aggregation Factor from Aspergillus sp.LAM 94-142)

  • A cell aggregation factor produced by Aspergillus sp. LAM 94-142 was purified and partially characterized. The factor was purified about 15 folds from culture broth by IRA 420 and IRC 120 treatment, 1% NaCl added acetone precipitation, and Sepharose 4B column chromatography with overall yield of 48%. It was heteropolysaccharide consisted of mannose, arabinose, and glucose with a molar ratio, 31:17:2, and its molecular weight was estimated to be about 900,000 daltons by Sepharodse 4B gel filtration method. The optimum pH and temperature was 8 and 40$\circ$C, respectively. The factor was stable in pH range of 3-9 and at 100$\circ$C for 90 min. The cell aggregation activity of the factor was inhibited by the addition of Hg$^{2+}$, Fe$^{2+}$, Cu$^{2+}$, and some polypeptides such as milk casein or hemoglobin. The factor aggregated Bacillus subtilis, B. macerans, B. turingiensis, E. coli, Peudomonas aeruginosa, P. fluorescens, P. malophilia, and weakly aggregated Staphylococcus sp., Sarcina lutea, P. putida and Cryptococcus neoformnans, but it didn't aggregate various strains of Candida sp. and Saccharomyces sp.

골다공증 모델 흰쥐에서 고수준의 단백질의 칼슘 섭취가 칼슘대사 및 신장기능에 미치는 영향 (Effects of High Protein and Calcium Intakes on Calcium Metabolism and Renal Function in Ovariectomized Osteporosis Rat Model)

  • This study was to investigate interaction between dietary protein and Ca levels in Ca metabolism and renal function in osteporosis rats. Five week-old female rats were fed a low Ca diet for 4 weeks after ovariectomy operation to establish rat models of osteoporosis. The ovariectomized osteoporosis rats were divided into six groups and were fed experimental diets which contained two levels of protein, normal (20%) and high(40%) , and three levels of Ca, low (0.06%), normal (0.47%) and high(0.94%) for 4 weeks , respectively. The ovaricetmized rat model of osteoporosis showed a remarkable decrease in serum Ca concentration, fresh weight and breaking force of femur, Ca and P contents of femur, and apparent absorption and retention of Ca. The supplementations of Ca and P contents of femur, and apparent absorption and retention of Ca. The supplementations of Ca at the dietary levels of normal and high levels significantly enhanced Ca bioavailability shown in the above experimental rat models of osteoporosis, regardless of dietary protein levels ; whereas the rats which were fed the low Ca diet demonstrated rather a decrease in its bioavailability. Irrespectively of the dietary Ca levels, the rats which were fed high protein diet exhibited an increase in kidney weight, urinary Ca, volume and hydroxyproline, and glomerular filtration ratio(GFR). The results show that dietary protein and calcium levels affect the renal function and Ca metabolism independently, while the interaction between protein and calcium have not been shown.

응집슬러지를 이용한 하수슬러지의 탈수성 및 침강성 개선 (Improvement of dewaterability and settleability of sewage sludge using coagulation sludge)

  • This study aimed to investigate the feasibility of improving dewaterability and settleability of sewage sludge using coagulation sludge. When mixed with sewage sludge and coagulation sludge at 1:1 ratio, capillary suction time(CST) and specific resistance to filtration(SRF) decreased by about 56% and 68%, respectively. It is found that total solids(TS) and volatile solids(VS) of mixing sludge are increased by about 59% and 53%, respectively. Also, the turbidity of the mixing sludge supernatant was reduced from 99 to 16 NTU. It is observed that the mixing of sewage sludge and coagulation sludge at 1:1 showed better effect than using poly-aluminum chloride(PAC) coagulant at 25 mg/L.

Membrane Concentrate Thickening by Hollow-fiber Microfilter in Drinkin Water Treatment Processes

  • A novel system to thicken the concentrated colloidal solution from membrane water treat-ment processes was developed. A hollow-fiber microfilter(hydrophilic polyethylene nominal pore size 0.1 μm total surface area 0.42 m2) was installed in an acrylic housing that has an aeration port 5 cm below the membrane and a clarifier in the bottom. The concentrate was uniformly supplied from the top of the housing. Bacuum filtration caused downward flow of concentrate and as a result thickening interface. The addition of poly-aluminum chloride (PAC) resulted in rapid increase of trans-membrane pressure (TMP) and in no improvement of the filtered water turbidity and thickening process. Two types of con-centrate and concentrate turbidity had little effect on the increase of TMP and concentrate thickening. It was observed that for the same height of membrane housing membrane surface area to housing volume (A/V) ratio had significant effect on the increase of TMP. When the housing volume was increased ten times the increasing rate of TMP was three times faster as compared to the original housing. A hydraulic model successfully simulated the formation and sedimentation of thickening interface.

Novel high performanced and fouling resistant PSf/ZnO membranes for water treatment

  • Antibacterial effective, high performanced, novel ZnO embedded composite membranes were obtained by blendig ZnO nanoparticles with polysulfone. IR, TG/DTG, XRD and SEM analysis were performed to characterize structure and morphology of ZnO nanoparticles and composite membranes. Contact angle, EWC, porosity and pore structure properties of composite membranes were investigated. Cross-flow filtration studies were performed to investigation of performances of prepared membranes. It was found from the cross section SEM images that ZnO nanoparticles dispersed homogenously up to additive amount of 2% and the membrane skin layer thicknesses increased in the presence of ZnO. Contact angle of pure PSf membranes were reduced from $70^{\circ}$ to $55^{\circ}$ after addition of 4% ZnO. Porosity of composite membrane contains 1% ZnO was higher about 22% than pure PSf membrane. BSA rejection ratio and PWF of 0.5% ZnO embedded composite membrane became 2.2 and 2.3 times higher than pure PSf membrane. It was determined from flux recovery ratios that ZnO additive increased the fouling resistance of composite membranes. Also, the bacterial killing ability of ZnO is well known and there are many researches related to this in the literature. Therefore, it is expected that prepared composite membranes will show antibacterial effect.

식이 단백질과 Ca 수준이 흰쥐의 Cd 중독과정중 Metallothionein 과 조직의 형태변화에 미치는 영향 (The Effect of Dietary Protein and Calcium Levels on Metallothionein and Histopathological Changes during Cadmium Intoxication in Rats)

  • This study was performed to investigate effect of dietary protein and calcium levels on cad-mium intoxication in rats. Adult Sprague-Dawley male rate(245$\pm$21g) were blocked into 18 groups of 7 animals according to body weight Nine experimental diets different with protein(40%, 15%, 7%) and calcium (1.3%, 0.6%, 0.1%) levels were prepared. Nine groups of animals were fed each diet with 50ppm cadmium in drinking water and the other 9 groups without cadmium for 30days. Results were summarized as follows: 1) Body weight gain F. E. R(Food Efficiency Ratio) and weights of liver kidney and femur were higher in high protein groups among cadmium exposed groups. 2) Cadmium contents in liver and intestine were higher in rats fed high protein diet or low calcium diet among cadmium exposed groups. Fecal cadmium excretion was highest in high protein-high calcium diet group among cadmium exposed animals. Metallothionein contents in liver kidney and intestine were higher in animals exposed to cadmium and fed high protein diets. 3) Gel filtration chromatography of cytosolic solution showed that the higher dietary protein and calcium levels were the more cadmium was found in metallothionein fractions. 4) No gross histopathological change was seen in liver kidney and intestine of cadmium exposed rats. However a significant increase of smooth endoplasmic reticulum which was alleveated by high protein-high calcium diet was observed. Results obtained indicated that not only high protein diet but also high calcium diet showed preventive effect on cadmium intoxication by increasing the induction of metallothionein syn-thesis and decreasing the cadmium absorption.

전오존과 중공사형 한외여과막을 이용한 간이정수처리시스템에 관한 연구 (A Study on the Potable Water Purification System with Pre-Ozone and Ultra Filtration Membrane of Hollow Fiber Type)

  • The objective of this study is to evaluate the possibility to apply pre-ozone and membrane system for drinking water. This system is improved in fouling control by pre-ozone system. It is composed of ultrafiltration hollow fiber type membrane and ozone reactor. The result of this study is that the flux is increased about 10.7% and decreased in TMP by the pre-ozone. Also, backwashing pressure decreased about 18%. The optimum concentration of residual ozone is 0.3~0.5 ppm. During the period, the recovery ratio of this system was turned out to be 90% as the flowrate of effluent is $67.1m^3/day$. When the TMP and backwashing pressure was $0.85kg/cm^2$ and $1.10kg/cm^2$, this system was stable without sudden fouling. Finally, the quality of effluent is satisfied the guidelines for potable water quality such as turbidity, color, E.coli, Mn, Al, Fe and so on.
