Antioxidant Activity of Asteraceae Plant Seed Extracts (14종 국화과(Asteraceae) 식물 종자 추출물의 항산화 활성)

  • Approximately 10% of all angiosperms belong to the Asteraceae family. Plant species belonging to this family have traditionally been used as medicinal plants in the Korean Peninsula. We investigated the antioxidant activity of seed extracts from 14 species belonging to the Asteraceae family. Seeds with ≥ 90% percentage of filled seed and ≥ 50% final germination were used. The total phenolic content was the highest in Dendranthema zawadskii var. tenuisectum (13.5 mg of gallic acid equivalents (GAEs)/g seeds), followed by Dendranthema zawadskii var. latilobum (11.8 mg of GAEs/g seeds), and Callistephus chinensis (11.0 mg of GAEs/g seeds). The total flavonoid content was highest in C. chinensis (9.8 mg of quercetin equivalents (QEs)/g seeds), followed by D. zawadskii var. tenuisectum (7.2 mg of QEs/g seeds) and Taraxacum officinale (6.3 mg of QEs/g seeds). Our results showed that 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) scavenging activity was highest in D. zawadskii var. tenuisectum (57.4 ㎍/ml), followed by T. officinale (59.1 ㎍/ml) and D. zawadskii var. latilobum (65.0 ㎍/ml), with a half maximal inhibitory concentration (IC50) of DPPH scavenging activity. Furthermore, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity was highest in C. chinensis (26.2 ㎍/ml), followed by D. zawadskii var. tenuisectum (38.4 ㎍/ml), T. officinale (40.2 ㎍/ml), with a half maximal inhibitory concentration (IC50) of ABTS scavenging activity. Based on a cluster analysis according to the antioxidant activity, the 14 species were classified into five groups, with group 4 having the highest antioxidant activity and group 0 having the lowest antioxidant activity. D. zawadskii var. latilobum, D. zawadskii var. tenuisectum, T. officinale, and C. chinensis belonging to groups 3 and 4, exhibited high phenolic content and antioxidant activity and can be considered potent plant-derived natural antioxidants.

Cone and Seed Development after Self-pollination in Pinus rigida Mill. (리기다소나무에 있어서 자배(自配) 후(後) 구과(毬果) 및 종자(種子)의 발달(發達))

  • Cone and seed development after self-pollination in 14-year-old Pinus rigida Mill. was investigated in an $F_1$-hybrid seed orchard that consists of Pinus rigida Mill. and Pinus taeda L. in alternative rows. On an average, 83.8% of the self-pollinated receptive female strobili developed into cones. This value appears to be normal compared to cone development after open-pollination in other pine species such as in Pinus sylvestris L. However, there was a great variation in filled seed development after self-pollination among the investigated individual trees (Table 1-1). The 34.5%, a ratio of filled seeds after self-pollination was significantly lower than 91.7%, that by the open-pollination. Some of the investigated trees developed 70.0%-83.5% of filled seeds after self-pollination. Thus, those are not desirable as trees for seed parent in establishing $F_1$-hybrid seed orchards in future. A further study on the rate of filled seed development by various pollination systems consisting of self-pollination, self-and cross-pollination with intraspecific, mixed (tree's own and foreign pollens) pollens and cross-pollination with interspecific, mixed pollens of the two parental tree species is necessary to determine the quality of seed parent trees in establishing $F_1$-hybrid seed orchards.

Healing after Implantation of Bone Substitutes and Safflower Seeds Feeding in Rat Calvarial Defects (백서 두개골 결손부의 골 대체물 이식과 홍화씨 섭취 후의 치유양상)

  • Many synthetic bone materials have been studied for their potential of regenerative effects in periodontal tissue. Safflower seeds have been traditionally used as a drug for the treatment of fracture and blood stasis in oriental medicines. The purpose of this study was to assess and compare the osseous responses in rat calvarial defects between bone substitutes such as calcium carbonate and bovine-derived hydroxyapatite and feeding of safflower seeds. The calvarial defects were made with 8 mm trephine bur in 24 Sprague-Dawley rats. Two graft materials were implanted in each experimental groups, whereas the control and safflower seed feeding groups were sutured without any other treatment. And then the rats of safflower seed feeding group were supplied with 3 g/day of safflower seeds. Each group was sacrificed at 4 weeks and 8 weeks. To study a histopathology related to bone healing and regeneration, Goldner's Masson Trichrome stain was done at each weeks. The tissue response was evaluated under light microscope. There were more osteoblastic activity, new bone formation, dense bony connective tissues in bovine-derived hydroxyapatite group compared to other groups at 8 weeks. The osseous defect area of safflower seed feeding group was filled with prominent fibrous tissues, where less inflammatory infiltration and new capillary proliferation. In the early phase of bone healing, safflower seed feeding reduces the inflammatory response and promotes the proliferation of connective tissue. These results suggest that natural bovine-derived HA and safflower seed feeding could enhance the regenerative potential in periodontal defects.

Development of Raising Seedling Technology for Veronica pyrethrina Nakai Using Plug Trays

  • Background and objective: This study was carried out to develop an effective technique for raising seedlings of Veronica pyrethrina Nakai, a native plant species in the Korean Peninsula, in plug trays. Methods: To investigate the optimum plug cell size and sowing media, we sowed seed in to plug trays with 34, 21, and 10 mL cells and filled with a commercial horticultural substrate and mixtures of peatmoss and perlite in 1:1, 3:1, and 4:1 ratios. Fertilization levels were set at 0, 500, 1000 and 2000 mg·L-1. Results: Plug cell size did not significantly influence the seedling growth of V. pyrethrina. By substrate type, the growth rate was highest in the horticultural substrate, followed by 4:1, 3:1, and 1:1. Growth by fertilization level was higher in all fertilized treatment groups than in the control group, and there was no difference among 500, 1000, and 2000 mg·L-1. Conclusion: The results of this study proved that it is most suitable for raising seedlings of V. pyrethrina to sow the seeds in a 10 mL cell plug tray filled with horticultural substrates, and apply fertilizers with less than 500 mg·L-1 concentration.

Variation in Seed and Cone Characteristics of Loblolly Pine (Pinus taeda L.) Families in Southern Part of Korea

  • Loblolly pine (Pinus taeda L.) is widely distributed in the southern part of the United States and it has been used as a major economic species in the region due to its excellent growth and stem straightness. The tree also grows only in the southern part of Korea because it is susceptible to cold. Recently climate changes have had widespread impacts on forest trees. Thus, the use of good quality seeds is prerequisite for assessing assisted migration adaptation trial. In this study, we conducted to investigate its cone and seed characteristics of each family, which is expected to improve seeds productivity for planting valuable timber trees. A total of 14 families were selected from the experimental forest in Boseong, Jeonnam province in 1981. The seed production capacity was estimated to range from 87.2 to 129.4 among families and the average was 111.3. The number of aborted ovules was investigated in the range of 11.4 to 29.5 for the first test and 7.4 to 22.2 for the second test. The average number of empty and filled seeds was 1.4 and 79.2 per cone, respectively. Based on the results, we can conclude that there is a strong correlation between the number of fertile scale and the seed production ability.

Reproduction and Parasitization Capacity of an Insect Parasitic Mite, Pyemotes moseri Yu et Liang (Acarina: Pyemotidae) New to Korea (국내 미기록 곤충기생성 응애, Pyemotes moseri Yu et Liang (Acarina: Pyemotidae)의 생식 및 기생 능력)

  • An insect parasitic mite was found on a larva from Japanese apricot seed. The mite was identified as Pyemotes moseri Yu et Liang (Acarina: Pyemotidae) new to Korea. The host larva was identified as Eurytoma maslovskii using mitochondrial DNA sequencing analysis. We conducted preliminary study on its reproduction and parasitization capacity in laboratory condition. A mated female mite reared on Eurytoma maslovskii larva. We counted and sexed newborn progenies and then eliminated them during periodical observations. To test parasitization capacity, a PCR tube containing mass reared P. moseri and Japanese apricot seeds (assumed bear larva of E. maslovskii) placed in a stainless bath filled with potting soil. One month later, we surveyed the seeds whether the E. maslovskii larva parasitized by mite or not. We repeated this experiment three times with five replications each. Average life span (days from parasitization to the end of reproduction) of gravid females was 24.4 days (n=8). A gravid female reproduced 104.0 female progenies (n=8). Although there were more than seven Japanese apricot seeds per bath containing larva or pupa, we found parasitization only in two seeds.

Cone and Seed Characteristics among Different Cone Abundance Classes in a Seed Orchard of Pinus koraiensis (잣나무 채종원에서 구과 착과수에 따른 구과 및 종자특성)

  • This study was conducted to examine the relationship between cone abundance and cone/seed characteristics to improve seed quality and seed orchard management of Pinus koraiensis. The data was collected from P. koraiensis seed orchards at Chungju in 2007. That year was a rich year of cone harvest and the average number of cone per tree was 2.5 times much than that of normal year. We collected the cones from all trees in the sample plot of the seed orchard and investigated morphological characters such as cone length, cone width and cone weight. According to cone abundance per tree, the samples were divided into eight classes and analysed by cone analysis method. There is a decreasing tendency of number of fertile scale, seed potential, percent of filled seeds, seed weight per cone and seed efficiency, when the cone number per tree is increased. Additionally, the implications of the results to seed orchards management of P. koraiensis were discussed.

Effect of Apical Ear Removal on Characters of Subapical Ear in Corn Inbreds (옥수수 백식계통의 상부이삭 제거가 하부이삭의 형질에 미치는 영향)

  • Removal of apical ear after silking is a method to increase seed production in corn inbreds. Due to the phenomenon of apical dominance, the subapical ear would develope and produce seeds following the removal of the apical ear. This experiment was conducted to investigate the varietal difference in subapical ear development and seed production. Days to silking of subapical ears were delayed by about 4 days compared to normal apical ears. Cob length and filled ear length of subapical ears across seven inbreds tested were reduced by 21% and 36%, respectively. Those of inbred INV302 decreased less and subapical ear of inbred NC246 were barren, which incicated the varietal defferences. Number of kernel rows, kernels per row, and total kernels per ear also decreased. The mean reduction rates for those characters were 28%, 37%, and 47%, respectively. Weights of ears and kernels were reduced and showed different response among inbred lines. Coefficients of variations for cob and filled ear lengths, number of kernel rows and kernels were greater in subapical ears than in apical ears. Seed production of subapical ears was 40 ~ 80% of apical ears, depending on inbred lines when the apical ear is removed immediately after silking.

Effect of Substrate Amount of Perlite on the Growth and Fruit Yield of Hydroponically Grown Cucumber (Cucumis sativus L.) Plants (펄라이트 배지량이 양액재배 오이의 생장과 수량에 미치는 영향)

  • This experiment was carried out to investigate the effect of container size and substrate volume on the growth and fruit yield of hydroponically grown cucumber(Cucumis sativus L.) plants. Seeds were sown in plug tray filled with coir dust on Feb. 13, 1998. Seedlings with 5 to 6 true leaves were transplanted in $2{\ell},\;4{\ell},\;6{\ell},\;8{\ell}\;and\;10{\ell}$ plastic pots filled with perlite. Cucumber fruits were harvested with 1 to 2 days interval, and fresh weight, number of normal and malformed fruit were recorded. Plant height, stem diameter, number of leaves and leaf area were highly depended on the container size. Total fresh weight and the number of fruit were increased with increasing container size. NAR(net assimilation rate) and CGR(crop growth rate) increased also with increasing the container size. Optimum container size for hydroponically grown cucumber plant using perlite was recommended as $8{\ell}$ per plant.

A New Grafted Cactus with Bright Yellow Color, 'Hwangseon'

  • A new grafted cactus cultivar, called 'Hwangseon', Gymnocalycium mihanovichii, is a hybrid crossed between two orange color cultivars 'Geoseong' and 'Huhwang'. Mating was performed in the greenhouse of National Institute of Horticultural & Herbal Science (NIHHS) in 2005. The harvested seeds were sown at 100 mL volumetric flasks filled with 'Kyoto' medium. Seedling had been grafted into a young rootstock, Hylocereus trigonus, and cultured in vitro in 100 mL test tube for more than six months. After re-grafting and planting in the green house in 2006, their characteristics were evaluated three times until 2009. The new cultivar has a flat round shape and bright yellow skin color. In addition, it has 7 to 9 ribs with gray and straight spines on the body and produces an average of 12.3 offsets.
