Effect of Seed Priming on Quality Improvement of Maize Seeds in Different Genotypes

  • In Korea, production of super sweet corn has been economically feasible and is substituting for traditional sweet corn due to better flavor in recent years. Major limiting factors for super sweet corn production are low field emergence and low seedling vigor. The optimum water potential (WP) for the priming of normal and aged seeds of dent, sweet (su) and super sweet (sh2) corns was studied to improve low seed quality. Seeds were primed at 0, -0.3, -0.6, -0.9, and -1.2 MPa of polyethylene glycol (PEG) 8000 solution at $15^{\circ}C$ for 2 days. Priming effects differed depending on the type of corn, seed quality, and WP of PEG solution. Although WP of priming solution did not influence the emergence rate of extremely high quality normal dent corn seeds, it reduced time to $50\%$ emergence (T50) and increased plumule weight. In contrast, the emergence rate of aged field corn was improved by seed priming at 0 MPa and plumule weight and $\alpha-amylase$ activity was enhanced. The optimum WP for both normal and aged sweet and super sweet corn seeds was between -0.3 and -0.6 Mpa. At the optimum WP emergence rate, $\alpha-amylase$ activity, and content of DNA and soluble protein increased, while T50 and leakage of total sugars and electrolytes reduced.

Performance of Imported Sweet Corn Hybrids in Korea

  • The performance of 7 sugary (su) and 12 shrunken-2 (sh2) sweet com hybrids which are commercially grown in the United States was tested in Korea. The 100-seed weight of su hybrids (16.5-23.6 g) was much heavier compared to that of sh2 hybrids (10.9-17.5 g). The germination rate of su and sh2 hybrids at $25^{\circ}C$ ranged 93.3-100% and 86.7-98.9%, the emergence rate of su and sh2 hybrids in cold test ranged 78.9-97.8% and 62.2-97.8%, and field emergence rate of su and sh2 hybrids ranged 74.4-100.0% and 79.9-98.2%, respectively. In su hybrids, there was a significantly positive correlation between germination rate at $25^{\circ}C$ and emergence rate in cold test or early growth. In contrast, in sh2 hybrids seed weight was positively correlated with early plant growth, while not with the germination rate at $25^{\circ}C$ or emergence rate in cold test and field. Most sh2 hybrids produced larger and more marketable ears compared to su hybrids although there were significant differences among the hybrids in the same genotype. At harvest (24 days after pollination) soluble solids content of su hybrids (24.3-27.1 Brix %) was much higher than that of sh2 hybrids (13.8-18.0 Brix %), while total sugars of sh2 hybrids (21.4-28.6% on the dry weight basis) was much higher compared to su hybrids (2.4-15.9%). Considering germination and emergence rates, marketable ear production, and total sugar content, 'GCB 70' and 'Sweet Satin' in su hybrids and 'Ice Queen', 'Aspen', 'Sweet Magic', 'Bandit', 'Xtrasweet 82', 'Aspen', and 'Cambella 90' in sh2 hybrids performed better than other hybrids.

Priming Effect of Rice Seeds on Seedling Establishment under Adverse Soil Conditions

  • An experiment was carried out to find out the priming effects of rice seeds, Oryza sativa L. (cv. Ilpumbyeo) on. the seedling establishment and early emergence under excess soil moisture conditions. Seeds were primed by soaking in -0.6 MPa polyethylene glycol (PEG) solution at $25^{\circ}C$ for 4 days. The primed seeds were sown in soils with various soil moistures (60, 80, 100, 120, and 140% field capacity) at 17 and $25^{\circ}C$, respectively. Germination and emergence rates, plumule height, and radicle length of primed seeds were higher than those of untreated seeds at any soil moisture and temperature examined. The time from planting to 50% germination ($T_{50}$) of primed seeds was less than that of untreated seeds by 0.9~3.7 days. Germination rate, emergence rate, plumule height, and radicle length were highest at the soil moisture of 80% field capacity among the soil moistures. Priming effects of rice seeds on germination and emergence rates were more prominent under the unfavorable soil moistures (60, 100, 120, and 140% field capacity) than those under the optimum soil moisture condition (80% field capacity). However, priming effects on seedling growth were greater at near optimum soil moisture compared with too lower or higher soil moistures. Therefore, these findings suggest that priming of rice seeds may be a useful way for better seedling establishment under the adverse soil conditions.

발아 및 입묘율 향상을 위한 파종전 종자처리 모형설정에 대한 제언 (A Proposed Model for Prosowing Seed Treatments to Promote Germination and Seedling Emergence)

  • Various presowing seed treatments have been used to enhance the rates of germination and afterward seedling emergence. Seeds treated by them occasionally have not shown the same rate in indoor and field. The presowing treatments considering germination mechanism and factors affecting germination should be totally included in indoor experiments so that the results drawn could be reproduced in the field. Seed drying after the treatments and field conditions after sowing especially able to show Phytochromemediated responses related to light treatment must be determined prior to force the treatments to seeds. After set up these prerequisites to promote the rates of germination and emergence, many seed treatments proposed so far must be fixed how to be done; alone, sequential and simultaneous.

  • 실험실에서 실시한 검정으로 Lotus와 Red clover 종자의 포장출현율 예측에 효과적인 검정방법을 구명하기 위하여 상이한 조건에서 재배된 종자를 퇴화조절검정법, 인위노화검정법과 전기전도도 검정방법으로 검정하였다. 이들을 포장출현율과의 관련성을 구명 하였던 바 1. Lotus와 Red clover의 포장출현율을 예측할 때 퇴화조절검정에서는 (equation omitted)=12.786+0.65X($R^2$=0.687), 인위노화검정에서는 (equation omitted)=25.989+0.39X($R^2$=0.260), 전기전도도검정에서는 (equation omitted)=141.932-0.86X($R^2$=0.238)으로써 예측할 수 있었으며 2. 이들 세가지 검정에서의 결과와 포장출현율과는 다음과 같은 다중회귀방정식으로 나타낼수 있었다. (equation omitted)=-198.16+2.3493X$_1$-1.5564X$_2$+1.981X$_3$($R^2$=0.916) 여기서, X$_1$, 퇴화조절검정법 ; X$_2$, 인위노화검정법 ; X$_3$, 전기전도도검정임. 3. 이러한 결과는 단일검정법으로 실내에서 포장출현율을 예측하려면 인위노화검정법이나 전기전도검정법 보다도 퇴화조절검정법이 비교적 효율이 높았으며, 이들 세가지 방법을 병용할 때에는 단일검정법보다도 효율을 증가시킬수 있었다.

Germinability of Film-Coated Snap Bean Seed as Affected by Oxygen Diffusion Rate under Different Soil Moisture Contents

  • The film coated snap bean (Phaseolus vulgaris) seeds with five different coating materials treated with 3% increase in seed weight were planted at sandy loam soil controlled moisture content of 18, 19, 20 and 21 %. The oxygen diffusion rate (ODR) was calculated from the different moisture content soil. The number of normal seedlings, seedling vigor, and seedling capability in field (seed vigor x dry matter weight) were observed at 9 days after planting and compared to the changes of ODR. The germination rate and ODR were sharply decreased simultaneously in the seeds planted at 21 % soil moisture content. Seedling emergence did not occur at all as the soil moisture content increased above 22 %. Hence this value should be considered as the threshold of soil moisture content for seedling emergence. An ODR value under 20% did not influence the percent emergence significantly. The certain difference observing in the emergence at the same ODR was not related clearly to the condition of soil. So it can be assumed that the limit of soil moisture content for the emergence of snap bean was approximately 20%. The value of 18% soil moisture content may be considered as the optimum for snap bean emergence. There was close relationship between the mean value of ODR in different soil moisture contents and the emergence. The germination rates of the seeds coated with the different materials were quite different when the seeds were planted at 21 % soil moisture. Dry weight of the seedlings from film coated seeds was decreased slightly, but the germination rates were not much different from the non-treated control under relatively higher soil moisture content (21 %). Major factor lowering emergence rate was oxygen stress while film coating act as a minor constraint for snap bean sown in excess soil moisture condition.

파종기와 예취시기가 답리작 호밀의 생육 및 건물수량에 미치는 영향 (Effects of Sowing Date and Cutting Time on Growth and Dry Matter Yield of Rye in Paddy Field)

  • 충남 예산지방에서 호밀을 답리작으로 재배할 때 파종기가 생육, 수량 및 식물체 부위별 건물구 성비율에 미치는 영향을 요약하면 다음과 같다. 1. 예산지방에서 파종기가 11월 5일 이후로 늦어지면 호밀의 출현일수가 길어지고 출현율 및 월동율이 감소하며 출수기가 지연되었다. 2. 호밀의 생장속도는 파종기가 빠를수록 증가하였고 파종기가 11월 5일 이후로 지연됨에 따라 생육량이 크게 감소하였다. 3. 호밀의 청예 및 건물수량은 파종기가 빠를수록 증가하나 다른 지역에서보다 파종적기의 폭이 넓어서 10월 5일에서 10월 25일 사이의 파종기간 차가 크지 않았다. 청예수량은 개화기 예취시, 건물수량은 유숙기 예취시 가장 높았다. 4. 호밀의 식물체 부위별 건물구성비율은 파종기가 빠를수록 그리고 수확시기가 늦을수록 엽신과 엽초의 비율이 감소하고 줄기와 이삭의 비율은 증가하였다.

보리 피과성이 발아 및 출아율에 미치는 영향 (Effects of Hulled and Hulless Barley Isogenic Lines on Germination and Emergence Rate)

  • 본 실험은 과맥에서 문제가 되고 있는 낮은 출현율의 원인을 구명하기 위하여 피주성 isogenic line을 육성 공시하여 발아 및 출현에 저해요인이 될 수 있는 수확기의 강우, 탈곡기회전수, 파종심도, 온도등을 달리하여 비교 검토한 바 다음과 같은 결과를 얻었다. 1. 수확 전일 강우시는 피성에 비하여 과성의 발아율이 4∼6 % 낮았으며, 5 일 인공강수처리에서는 과성의 발아율은 현저히 저하되었다. 2. 탈곡기회전수가 600rpm에서 1,000rpm 으로 상승할 때 피성의 발아율이 100 %에서 90%로 저하된 반면, 과성은 98%에서 76%로 발아율이 크게 저하되었다. 3. 과성에서 발아율보다 출현율이 피성에 비하여 큰 차이가 있었으며 파종심도가 깊을수록 그 차이가 컸다. 4. 파종심도, 고탈곡회전수, 수확시의 강우 순으로 피성보다 과성의 출현율을 크게 저하시켰다. 따라서 과맥을 육종할 때 출현율 및 유묘활력 향상을 위한 육종적 노력이 필요할 것으로 생각된다.

온도에 따른 난지형 마늘 출현 모형 (A Simple Emergence Model of Southern Type Garlic Based on Temperature)

  • 일 평균기온을 이용하여 난지형 마늘의 출현시기와 출현율을 일별로 예측하는 동적 모형을 개발하였다. 난지형 마늘은 최적온도인 $12.7^{\circ}C$보다 증가하면서 출현율과 출현속도가 감소하였다. 일 평균기온을 입력하여 난지형 마늘의 일별 출현속도를 예측할 수 있는 베타함수와, 출현속도를 누적하여 마늘 출현율의 변화를 예측하는 비선형 모형을 개발하였다. 개발된 모형으로 생장상의 실험결과를 잘 설명할 수 있었다. 모형을 재배기간 온도조건이 다른 온도구배터널에 적용해본 결과 온도구배터널 재배 마늘의 출현시기와 출현율을 비교적 잘 예측할 수 있었다. 그러나 노지 포장에서는 모형에서는 출현시기가 실제 관측된 것보다 5일 정도 빠를 것으로 예측되어 토양수분 등 온도이외의 다른 요인을 반영할 수 있도록 모형의 개선이 필요하였다.

Optimum Harvest Time for High Quality Seed Production of Sweet and Super Sweet Corn Hybrids

  • The production of sweet (su) and super sweet corns (sh2) has been economically feasible in Korea in recent years. Major factors limiting super sweet corn production are low germination and low seedling vigor. Since seed quality is closely related to seed maturity, the optimum harvest time for the seed production of sweet and super sweet corns was studied and the quality of seeds with varying maturities was investigated in 2001 and 2002 cropping seasons. The parents of the sweet corn seeds were Hybrid Early Sunglow and 'Golden Cross Bantam 70' and those of super sweet corn were Xtrasweet 82 and 'For­tune'. Seeds were harvested at 21, 28, 35, 42, 49, and 56 days after silking (DAS). As the seeds developed, seed weight of sweet corn increased and the seed moisture content decreased faster than that of super sweet corn. Germination rates of sweet corn seeds harvested 21 and 28 DAS at $25^{\circ}C$ and emergence rates in the cold soil test were significantly lower than those of seeds harvested after 42 DAS in both years. Although the germination rates of super sweet corn seeds with varying maturities showed similar patterns as sweet corn seeds at $25^{\circ}C$, the emergence rate of super sweet corn seeds in cold soil test continuously increased with seed maturity. This suggests that seed quality of super sweet corn should be tested in a cold soil test to estimate field emergence. As the seeds developed, leakage of total sugars and electrolytes from the both sweet and super sweet corn seeds decreased up to 42 or 49 DAS. The $\alpha-amylase$ activities of both sweet and super sweet corn seeds increased with seed maturity from 21 to 35 or 49 DAS depending on genotype and year. The optimum harvest time for the seed production of sweet corn was 42 DAS and 49 DAS for super sweet corn considering emergence rate and plumule dry weight in the cold soil test, leakage of sugars and electrolytes from the seeds, and $\alpha-amylase$ activity.
