Effect of the Domestic Tall Fescue (Festuca arundinacea Schreb.) Varieties on Seed Production (국내육성 톨 페스큐 품종간 생육특성 및 종자생산성)

  • A new tall fescue variety (Festuca arundinacea Schreb.) named Greenmaster and Purumi were. In this study, we evaluated the seed production of new tall fescue domestic varieties were cultivated in Korea. Agronomic aspects of tall fescue domestic varieties were determined using seed production and forage nutritive value of straw at the National Institute of Animal Science from 2013 to 2014. The heading date according to the varieties displayed differences and domestic varieties showed enhanced disease resistance. The harvested seed yields of Greenmaster was 35% higher as 1,664 kg/ha and it's also yield of straw higher than that of Fawn. However, the nutritive value of both varieties was similar. The dry matter yield of Pumumi was 25% higher as 11,080 kg/ha. Accordingly, developed a new variety of domestic tall fescue with excellent environmental adaptability, aiming to make contribution to the vitalization of the Korean grassland industry.

  • To find out the optimum level of nitrogen fertilization and better distribution of application time for seed production of Festuca arundianacea S. (var. Alta), this filed experiment was performed at Livestock Experiment Station in Suweon, during 1979 to 1981. The treatments used in this study were three nitrogen fertilizing levels (120, 180 and 240kg/ha) and four different methods of nitrogen distribution (i. single application of whole amount in early Spring, ii. 50 percent each in Autumn and early Spring, iii. 50 percent each in early Spring and at begin of heading stage and iv. 50 percent in Autumn and 25 percent in early Spring and at begin of heading stage respectively). The experiment was arranged as a split-plot design with 4 replications and performed at the experimental field of the Livestock Experiment Station in Suweon, during 1979 to 1981. The results obtained are summarized as follows: 1. Date of heading stage of Festuca arundinacea was about May 21 and that of flowering stage was June 1 to 3. The optimum stage for the seed harvesting of Festuca arundinacea (var. Alta) was June 25 about 22 to 23 days after full flowering stage. Average plant height was about 127cm and the panicle length, 24cm. 2. 1000 grain weight was 2.72g and the number of panicles were 85 to 107 per square meter. 3. The mean seed wield for two years was 678.8kg/ha with the average of whole treatments and 781.9kg/ha with the best treatment (50 percent in Autumn and 25 percent in early Spring and at begin of heading stage with 240kg/ha respectively). 4. The average germination rate of harvesting seeds was 87.0 percent and it was increasing trend according to frequent application of fertilizer. 5. The average DM yield of aftermath seed harvesting was 6155kg/ha with two cut, and it was the largest DM yield from the higher nitrogen level and also from the single application in early Spring.

Production of New Regenerated Plants by Anther Culture of the Hybrids of Italian ryegrass X Tall fescue (이탈리안 라이그라스 X 톨페스큐 속간 교잡종의 약배양 기법을 이용한 식물체 생산)

  • Several regenerated plants were obtained from anther culture of the hybrid lines of Italian ryegrass (Lolium multiflorum Lam.) X Tall fescue (Festuca arundinacea Schreb.). When the anthers of the hybrid plants were incubated for 20 days on callus induction medium, MS medium containing 30 g/$\ell$ of sucrose, 2 mg/$\ell$ of NAA and 1 mg/$\ell$ of kinetin, their calli were induced. The mean ratio of callus induction was 11.6 percent, and the mean of callus weight was 9.1 mg/callus/anther. When the calli of the hybrid plants were incubated for 50 days on plant regeneration medium, MS medium containing 30 g/$\ell$ of sucrose, 1 mg/$\ell$ of NAA and 2 mg/$\ell$ of kinetin, the hybrid plants were regenerated. The mean ratio of plant regeneration was 27.1 percent and line Hyb-1 showed highest regenerabillity with the frequency of 30.2 percent.

Efficient Callus Culture and Plant Regeneration from Mature Seed of Tall Fescue (Festuca arundinacea Schreb.) (톨 페스큐의 성숙종자로부터 효율적인 캘러스 배양 및 식물체 재분화)

  • In an effort to optimize tissue culture conditions for genetic transformation of tall fescue (Festuca arundinacea Schreb.), an efficient plant regeneration system from seed-derived calli was established. MS medium containing 6 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D) and 0.1 mg/L benzyladenine (BA) were optimal for embryogenic callus formation from mature seed and had a strong effect on successive plant regeneration. The plant regeneration frequency above 50% was observed when embryogenic calli induced in this medium were transferred to N6 medium supplemented with 1 mg/L 2,4-D and 3 mg/L BA. Among several basic media, MS and N6 medium were optimal for callus induction and plant regeneration, respectively. 'Kentucky-31' showed to have high frequencies of embryogenic callus induction and plant regeneration up to 58.3 and 50%, respectively. Addition of sucrose to the regeneration medium as a carbon source increased regeneration frequency up to 55%. A short tissue culture period and high-frequency regeneration system established in this study will be useful for molecular breeding of tall fescue through genetic transformation.

Effect of Root Zone Cooling on Growth and Mineral Contents of Turfgrasses in Simulated Athletic Field during Summer Season (여름철 근권부의 냉온처리가 경기장 잔디의 생육 및 무기성분 함량에 미치는 영향)

  • This study was conducted to determine the effect of root zone cooling on growth and quality of turfgrasses including Kentucky bluegrass (Poa pratensis L.‘Nuglade’), perennial ryegrass (Lolium perenne L.‘Accent’), tall fescue (Festuca arundinacea Schreb.‘Pixie’), and Japanese lawngrass (Zoysia japonica Steud.) in simulated athletic field during summer season in Korea. Mineral contents in clippings of turfgrasses grown at different soil mixtures and temperatures were also analyzed. Root zone cooling (approximately 4~6$^{\circ}C$ lower than that of untreated-control) resulted in good uniformity, little disease incidence and higher level of chlorophyll contents in cool-season turfgrasses. The effectiveness of root zone cooling in protecting disease incidence from high temperature stress was the most manifest in perennial ryegrass compared to others. Fresh clipping weight in treatment of root zone cooling was increased approximately 2 times in Kentucky bluegrass and perennial ryegrass, and 2.5 times in tall fescue compared to those of control. There was higher growth rate in a soil mixture composed of 80% peat moss +10% sand +10% soil (v/v/v) than in that of 80% pea moss +20% sand (v/v), Mineral contents of N, P, K, Ca, and Mg in clippings of three species of cool-season turfgrasses were significantly increased in treat-ment of root zone cooling but this was not found in Japanese lawngrass. Results showed that root zone cooling has a benefit in keeping good quality and growth of cool-season turfgrasses in sports field under supraoptimal ambient temperature during summer season.

  • This experiment was to study the effects of soil compaction and artificial pore space on the shoot density of tall fescue (Festuca arundinacea Schreb.). Tall fescue subjected to compaction treatments with control, 10, 20 and 40 kg power roller, used for two times bi-weekly during six months. Artificial pore space treatments were control, 13.5, 37.5 and 84.5% at 0-lOcm depth, respectively. 1. Soil compaction increased soil hardness and soil bulk density.2. Compaction level of lOkg (soil hardness 2.5kg/$cm^3$) showed the highest shoot density than that of other treatments. 3. Artificial pore space was positive significant correlated (p<0.01) with shoot density. 4. When over the 37.5% of total pore space could be mainternance for high shoot density after the soil compacted.

Occurrence of Brown Patch Disease and Turfgrass Survival of Cool Season Turfgras Cultivar in Species during Summer Season (여름철 한지형 잔디의 갈색마름병 발생과 잔디 생존)

  • Turfgrass survival and ocurrence of brown patch disease during summer season were investigated the forty six cultivars of cool season grasses in 6 species, popularly used in golf courses in Korea. Ocurrence of brown patch disease was evaluated two times in July. The percentage of turfgrass survival was investigated on Aug. 20 and on Sep. 20 in 2010. There were significant difference for the percentage of turfgrass survival and brown patch disease occurrence among cultivars. Brown patch disease caused by Rhizoctonia spp. and Pythium aphanidermatum with rDNA ITS were showed most susceptible in creeping bentgrass (Agrostis palustris Huds) cultivars and chewings fescue (Festuca rubra var. commutata Gaud) cultivars. The percentage of turfgrass survival during August to September was showed lower creeping bentgrass cultivars and chewings fescue cultivars than another cultivars. Among of 20 cultivars in Kentucky bluegrass (Poa pratensis L.) species were showed highest survival in cultivars of "Nuglade", "Cabernet", "Midnight II" and "Beyond". 6 cultivars of Tall fescue (Festuca arundinacea Schreb) were showed highest survival in cultivars of "Double sentry", "Inferno", and cultivars of "Accent II" in Perennial ryegrass (Lolium parenne L.) species.

Comparison of Early Germinating Vigor, Germination Speed and Germination Rate of Varieties in Poa pratensis L., Lolium perenne L. and Festuca arundinacea Schreb. Grown Under Different Growing Conditions (생육환경에 따른 Poa pratensis L., Lolium perenne L. 및 Festuca arundinacea Schreb.의 초종 및 품종별 발아세, 발아속도 및 발아율 비교)

  • Research was Initiated to investigate germination characteristics of cool-season grasses (CSG). Several turfgrasses were tested in different experiments. Experiments I and III were conducted under a room temperature condition of 16$^{\circ}C$ to 23 $^{\circ}C$ and under a constant light condition at 25 $^{\circ}C$, respectively. An alternative environment condition that is a requirement for a CSG germination test by International Seed Testing Association (ISTA) was applied in the Experiment II, consisting of 8-hr light at 25 $^{\circ}C$ and 16-hr dark at 15 $^{\circ}C$. In each experiment, data such as early germinating vigor, germination speed and germination rate were evaluated. Six turfgrass entries were comprised of two varieties each from Kentucky bluegrass (KB, Poa pratensis L.), perennial ryegrass (PR, Lolium perenne L.), and tall fescue (TF, Festuca arundinacea Schreb.), respectively. Significant differences were observed in early germinating vigor, germination speed and germination rate. Early germinating vigor as measured by days to 70% seed germination was variable according to environment conditions, turfgrasses and varieties. It was less than 6 days in PR and 6 to 9 days in TF. However, KB resulted in 11 to 13 days under an alternative condition and 11 to 28 days under a room temperature condition. The germination speed was fastest in PR of 7 to 10 days and slowest in KB of 14 to 21 days. However, intermediate speed of 10 to 14 days was associated with TF. There were considerable variations in germination rate among turfgrasses according to different conditions. Generally, PR and TF germinated well, regardless of environment conditions. However, a great difference was observed among KB varieties, when compared with others. Under a room temperature condition, total germination rate was 71.0% in Midnight and 77.7% in Award. And it increased under an alternative condition, which was 81.7% and 91.7% in Award and Midnight, respectively. However, the poorest rate was found under a constant temperature condition, resulting in 18.0% in Award and 15.3% in Midnight. These results suggest that an intensive germination test required by ISTA be needed prior to the decision of seeding rate, including early germinating vigor and germination speed as well as total germination rate. KB is very sensitive to environment conditions and thus its variety selection should be based on a careful expertise.

Comparisons of Foliar Nutrient Concentrations and Soil Nutrient Stocks Between Restoration Areas and Adjacent Red Pine Stands in Opencast Kaolinite Mines in Sancheong-gun (산청군 고령토 광산 복원지와 인접 소나무 임분의 토양양분 저장량 및 잎 양분 농도 비교)

  • We performed this study to determine the foliar nutrient concentration and the nutrient stocks of restoration areas and adjacent Pinus densiflora S. et. Z. (red pine) stands in opencast kaolinite mines in Sancheong-gun, Gyeongsangnam-do, southern Korea. We chose six sites to determine foliage nutrient concentrations and the nutrient stocks of soils (0-10 cm depth). The dominant vegetation planted in restoration areas comprised Quercus acutissima Carruth., P. koraiensis S. et. Z., Festuca arundinacea Schreb., and Lespedeza cuneata G. Don. Invading vegetation in the restoration areas comprised Alnus incana (L.) Medik., Robinia pseudoacacia L., and Lespedeza spp., among others. The carbon and nitrogen stocks at 10 cm soil depth were significantly higher in the red pine stands than those in the restoration areas, whereas those of phosphorus, potassium, and magnesium were not significantly different between the two areas. However, calcium stocks were significantly higher in the restoration areas than in the red pine stands. Nitrogen concentration in foliage was higher in L. cuneata (20.28 mg N g-1) than that in F. arundinacea (5.67 mg N g-1), whereas potassium concentration was twice as high in F. arundinacea (18.8 mg K g-1) as that in L. cuneata (9.07 mg K g-1). Foliar nitrogen concentrations in invasive vegetation such as A. incana, R. pseudoacacia, and Lespedeza spp. were twice or four times higher than those of Q. acutissima and P. koraiensis. Our results indicate the development of suitable vegetation and soil amendment treatments to improve poor soil environmental conditions in restoration areas are necessary following opencast kaolinite mining.

Transgenic Expression of MsHsp23 Confers Enhanced Tolerance to Abiotic Stresses in Tall Fescue

  • Tall fescue (Festuca arundinacea Schreb.) is an important cool season forage plant that is not well suited to extreme heat, salts, or heavy metals. To develop transgenic tall fescue plants with enhanced tolerance to abiotic stress, we introduced an alfalfa Hsp23 gene expression vector construct through Agrobacterium-mediated transformation. Integration and expression of the transgene were confirmed by polymerase chain reaction, northern blot, and western blot analyses. Under normal growth conditions, there was no significant difference in the growth of the transgenic plants and the non-transgenic controls. However, when exposed to various stresses such as salt or arsenic, transgenic plants showed a significantly lower accumulation of hydrogen peroxide and thiobarbituric acid reactive substances than control plants. The reduced accumulation of thiobarbituric acid reactive substances indicates that the transgenic plants possessed a more efficient reactive oxygen species-scavenging system. We speculate that the high levels of MsHsp23 proteins in the transgenic plants protect leaves from oxidative damage through chaperon and antioxidant activities. These results suggest that MsHsp23 confers abiotic stress tolerance in transgenic tall fescue and may be useful in developing stress tolerance in other crops.
