Antioxidant Activities of the Ethanol Extract of Hamcho (Salicornia herbacea L.) Cake Prepared by Enzymatic Treatment

  • The antioxidant activities of water ($H_2O$) and ethanol (EtOH) extracts from hamcho (Salicornia herbacea L.) juice and cake prepared by enzymatic treatments were evaluated by in vitro assays against DPPH, superoxide, and hydroxyl radicals. Among the $H_2O$ and EtOH extracts from five different carbohydrases treated, the EtOH extract from viscozyme-treated hamcho cake had higher yield and phenolic content, and exhibited the strongest radical scavenging activity against DPPH ($IC_{50}=186.91\;{\mu}g/mL$), superoxide ($IC_{50}=87.54\;{\mu}g/mL$), and hydroxyl radicals ($IC_{50}=367.07\;{\mu}g/mL$). Antioxidant assay-guided fractionation and purification of the EtOH extract led to isolation and identification of five phenolic compounds, procatechuic, ferulic and caffeic acids, quercetin, and isorhamnetin. Most of these phenolic compounds exhibited considerable DPPH, superoxide, and hydroxyl radical scavenging activities, and in particular, caffeic and ferulic acids had stronger superoxide and hydroxyl radical scavenging activities than the well-known antioxidant radical scavenger, (+)-catechin (p<0.05). Quercetin and isorhamnetin were the primary compounds responsible for the strong antioxidant activity in the EtOH extract of the viscozyme-treated hamcho cake. Meanwhile, these five phenolic compounds were detected in the EtOH extract of the viscozyme-treated hamcho cake at the following levels (dry base of hamcho); procatechuic acid (1.54 mg%), caffeic acid (6.87 mg%), ferulic acid (8.45 mg%), quercetin (12.63 mg%), and isorhamnetin (6.65 mg%). However, three of these phenolic compounds (procatechuic, caffeic acid, and ferulic acids) were detectable in the $H_2O$ extract of viscozyme-treated hamcho juice. These results suggest that the EtOH extract of viscozyme-treated hamcho cake may be a potential source of natural antioxidants.

미강의 약리성분 감마-오리자놀의 정량 (Quantitative Analysis of ${\gamma}-oryzanol$ in Rice Bran)

  • 미강에 함유된 약리성분인 ${\gamma}-oryzanol$ 함량을 검정하여 용도 다양화 연구의 기초 자료를 제공코자 시험을 수행한 바 다음과 같은 결과를 얻었다. Soxhlet 추출장치에서 얻은 미강유에서 유리 sterol과 ferulic acid ester가 분리되었다. 유리 sterol은 GC-MS에 의하여 campesterol, ${\beta}-sitosterol$ 및 stigmasterol로 구성되어 있음을 알 수 있었다. ferulic acid ester는 알칼리 가수분해에 의하여 얻어지는 검화분해물을 분리용 TLC로 분리하여 triterpene과 sterol을 얻었다. sterol은 GC-MS 측정으로 유리 sterol과 같은 조성임을 알았으며 triterpene은 cycloartanol, cycloartenol, 24-methylenecycloartanol 및 미지의 triterpene로 구성되었다. 미강유나 pitch 중 ferulic acid ester의 흡광도를 기준으로 ${\gamma}-oryzanol$의 함량을 평가한다면 큰 오차는 없으리라 생각된다.

동백(冬柏)을 위시한 차나무과(科) 식물(植物) 종실(種實)에 함유(含有)된 항산화제(抗酸化劑)에 관한 연구(硏究) (Studies on the Antioxidative Substances in the Seeds of Some Theaceae Family)

  • Dried seeds of Camellia japonica and Thea sinensis were investigated to determine the nature of their antioxidative activity. Activity was measured by the induction period in the coupled oxidation of a substrate lard and extracts or isolates to be tested. 70% methanol and dichloromethane extracts were found to be antioxidative abilities. Their unsaponifiables revealed weak antioxidative activity, although hexane extracts did not show antioxidative effect on lard. Column chromatography for dlchloromethane extracts gave 4 fractions(only 2 fractions were potent). HPLC was used in isolating potent antioxidative components from the column fractions and the precolumn-passed methanol extracts. They were separated into 7 and 8 components, respectively. The column fractions obtained from both seeds comprised trans-p-coumaric acid. trans-p-ferulic acid and an unknown component with minor components such as chlorogenic acid and catechin. On the other hand, the most prominent components in the methanol extracts were an unidentified component. trans-pcoumaric acid, trans-p-ferulic acid, catechin and chlorogenic acid. The unknown compound isolated from the column fractions and methanol extracts was identified as epicatechin by $^1H-and\;^{13}C-NMR$. The antioxidative activities of these components were epicatechin > catechin > chlorogenic acid > trans-p-ferulic acid > trans-p-coumaric acid.

모링가 부위별 추출물의 항산화 활성 및 파이토케미컬 함량 (Antioxidant activity and phytochemical contents of the extracts from different parts of Moringa oleifera)

  • 열대 및 아열대 기후에서 자생하는 모링가가 기후변화로 인하여 국내에서도 재배가 되고 있으면서 관심이 증가하고 있다. 다양한 나라에서 식품 및 기능성제품으로 사용이 되면서 국내로 수입되는 모링가에 대한 분석이 필요하여 본 연구를 실시하게 되었다. 모링가의 부위별(잎, 가지 및 종자) 항산화 능력과 페놀 및 플라보노이드 함량은 잎과 가지에서 높았으며, 정의 상관관계를 나타내었다. 무기물 함량 역시 잎과 가지에서 풍부하였으며 특히 칼륨 이온이 가장 많았다. 또한 7종의 페놀산과 4종의 플라보노이드 성분이 검출이 되었으며 페놀산에서는 페룰릭산이, 플라보노이드에서는 쿼세틴 함량이 높게 나타났다. 이 연구결과는 향후 국내 모링가 재배농가의 비교자료로 유용하게 이용될 것으로 기대된다.

GC에 의한 인삼 중의 방향족 산성 성분 분석 (Analysis of Aromatic Acids in Panax Ginseng by Gas Chromatography)

  • Eight aromatic acids in Panax ginseng were determined by GC. Ultra-1 $(25\;m{\times}0.2\;mm{\times}0.33\;{\mu}M)$ capillary column was employed with temperature programming from $150^{\circ}C$ to $240^{\circ}C$ at a rate of $3^{\circ}C/min$. The mean contents of eight aromatic acids in 8 white ginseng samples were as follows; salicylic acid: 4.30 ppm, cinnamic acid: 18.2 ppm, vanillic acid: 4.22 ppm, gentisic acid: trace, syringic acid: 6.69 ppm, p-coumaric acid: 13.3 ppm, ferulic acid : 21.9 ppm, caffeic acid: 24.3 ppm, respectively.

에탄올 생산 향상을 위한 발효저해물질 제거와 리그닌 유래 발효저해물질이 에탄올 발효에 미치는 영향 (Enhancement of Ethanol Production by The Removal of Fermentation Inhibitors, and Effect of Lignin-derived Inhibitors on Fermentation)

  • 본 연구에서는 옥살산 전처리 바이오매스의 액상가수분해산물에 포함된 발효저해물질을 전기투석과 XAD 수지 처리하여 제거한 후 에탄올을 생산하였다. 전기투석 과정에서 아세트산은 대부분 제거되었으며(95.6%), 비이온성 발효저해물질(Total phenolic compound: TPC, 5-hydroxymethyl furfural: HMF, furfural)은 XAD 수지 처리에 의해 효과적으로 제거되었다. 전기투석과 XAD 수지 처리된 액상가수분해산물로 발효를 수행한 결과 XAD 수지의 침지시간이 짧을수록 에탄올 생산이 향상되었다. 최대 에탄올 생산은 발효 72시간 후 $6.16g/{\ell}$로 전기투석 후 액상가수분해산물을 XAD-4 수지에서 5분 침지하였을 때 나타났다. 리그닌 유래 발효저해물질 중 syringaldehyde는 저농도(1 mM, 2 mM)에서 에탄올 생산을 향상시켰으며 5 mM에서는 발효에 부정적인 영향을 주었다. 리그닌 유래 발효저해물질의 시너지 효과를 확인하고자 합성배지로 발효를 수행하였으며, Syringaldehyde (1 mM)와 ferulic acid (1 mM) 합성배지를 이용하여 발효를 수행한 결과 syringaldehyde보다 ferulic acid의 영향으로 에탄올 생산이 감소했다.

다양한 발효균주를 이용한 인삼패독산의 생물 전환 성분의 정량 분석 (Quantitative Analysis of Bioconversion Constituents of Insampeadock-san Using Various Fermented Bacteria)

  • Insampaedoksan (IS) is the decoction of medicinal herbs, which was commonly used for anti-inflammatory and anti-pyretic in the Korean traditional medicine (KTM). Several studies on improving efficiency or searching new efficiency by fermenting traditional herbal medicines are recently in progress. The bioconversion has been conducted on IS using various bacteria. Liquiritin, ferulic acid, hesperidin, liquiritigenin, and glycyrrhizin in IS before and after fermented IS were simultaneously analyzed. These compounds were qualitatively analyzed and quantitatively analyzed using the HPLC-DAD system. The identifications of liquiritin, ferulic acid, hesperidin, liquiritigenin and glycyrrhizin were achieved by comparing the HPLC retention time ($R_t$) and the UV absorption of five pure compounds in the IS. As a result, the increased constituents were identified to be liquiritin, liquiritigenin and glycyrrhizin, while the decreased constituent was ferulic acid and the constituent of hesperidin was similar to before and after fermentation. Insampeadock-san fermented by Lactobacillus plantarum KFRI 144 exhibited the most remarkable changes in all of fermentation.

Total polyphenol and ferulic acid analysis of a new variety of corn, Bandiburichodang, according to steaming time and roasting temperature

  • Bandiburichodang (BDC) is a new variety of Zea mays L. Total polyphenol content (TPC) assay and quantitative analysis of ferulic acid (FA) were performed to determine the steaming, roasting conditions of BDC kernels that lead to the highest content. TPC levels increased after roasting under all conditions. TPC levels in samples steamed at 115 ℃ for 25 min were 3.157 mg/g before roasted, and increased to 3.825 and 4.739 mg/g after roasting at 160 and 200 ℃, respectively. Whether BDC kernels were roasted was relevant with TPC content. BDC kernels were extracted to perform quantitative analysis of FA. Roasting temperature affected FA content: the higher the temperature, the lower the content. BDC kernels that were steamed at 115 ℃ for 25 min had 0.178 mg/g of FA content before roasting, and levels decreased to 0.132 and 0.115 mg/g after roasting. Under different roasting conditions, FA content decreased 15 to 50%. We hypothesize that this phenomenon is due to a breakdown of phenolic compounds or cell wall disruption.

발효처리가 인삼잎의 진세노사이드 및 페놀산 조성 변화와 생리활성에 미치는 영향 (Ginsenoside, Phenolic Acid Composition and Physiological Significances of Fermented Ginseng Leaf)

  • 본 연구에서는 인삼잎이 인삼뿌리보다 사포닌 함량이 높은 부위로서 식품 소재로 이용가치가 있을 것으로 생각되어 인삼잎을 이용하여 차 제품을 개발하기 위한 방안으로 인삼잎을 발효시켜 진세노사이드 조성 및 형태별 페놀산 조성의 변화를 분석하고 인삼잎을 침출시켜 침출액에 대한 전자공여능과 tyrosinase 저해활성을 측정하였다. 인삼잎에서 진세노사이드는 10종이 검출되었고 주된 진세노사이드는 ginsenoside-Rg1(26.0 mg/g), -Re(47.3 mg/g) 및 -Rd(23.9mg/g)이었고 발효에 의하여 ginsenoside-Rh2, -Rh1, -Rg2 및 -Rg3는 증가하였으며 특히 Rg3는 15배가 증가하였다. 인삼잎의 총 폴리페놀성 함량은 350.4 mg%이었고 발효인삼잎은 312.5 mg%으로 발효에 의해서는 약간 감소하였다. 인삼잎의 페놀산은 결합형은 검출되지 않았고, 유리형과 에스테르형이 각각 8 및 6종이 검출되었으며 그중에서 ferulic acid가 각각 12.6 및 50.7 mg%로 가장 많은 함량을 차지하고 있었다. 발효인삼잎에서는 ferulic acid는 상당량이 감소하였으나 protocatechuic acid, p-hydroxybenzoic acid, vanillic acid의 3종의 페놀산이 유리형, 에스테르형 및 결합형 모두에서 상당량 증가하여 총 함량이 각각 28배, 5배 및 7.8배 증가하였다. 인삼잎을 침출시킨 액을 이용하여 전자공여능과 tyrosinase 저해활성을 측정한 결과 전자공여능은 발효에 의하여 활성이 증가하지는 않았으나, tyrosinase 저해활성은 증가하여 $500\;{\mu}L/mL$ 농도로 첨가 시 46.5%를 나타내어 무발효인삼잎에 비하여 2배 이상 증가하여 시판녹차와 비슷한 결과를 보여주었다.

Role of NADPH Oxidase-Mediated Generation of Reactive Oxygen Species in the Mechanism of Apoptosis Induced by Phenolic Acids in HePG2 Human Hepatoma Cells

  • Although plant-derived phenolic acids have been reported to have anti-cancer activity, the exact mechanism is not completely understood. In this study, we investigated the role for reactive oxygen species (ROS) as a mediator of the apoptosis induced by caffeic acid (CA) and ferulic acid (FA), common phenolic acids in plants in HepG2 human hepatoma cells. CA and FA reduced cell viability, and induced apoptotic cell death in a dose-dependent manner. In addition, they evoked a dose-related elevation of intracellular ROS. Treatment with various inhibitors of NADPH oxidase (diphenylene iodonium, apocynin, neopterine) significantly blunted both the generation of ROS and the induction of apoptosis induced CA and FA. These results suggest that ROS generated through activation of NADPH oxidase may play an essential role in the apoptosis induced by CA and FA in HepG2 cells. These results further suggest that CA and FA may be valuable for the therapeutic management of human hepatomas.
