Hen's eggs have been regarded as one of the best animal bioreactors to produce biologically active peptides originated from many organisms including human. Despite the last decade's efforts to produce transgenic chicken for any commercial purposes, the results so far reported are very disappointing, indicating that hen's eggs are very difficult to crack for transgenesis. Comparatively large female gamete with enormous amount of yolk may be one of the major obstacles in achieving a similar feat to those of other vertebrate species including mouse, sheep, fish and frog. The delay or less efficiency evidenced may instruct to try an alternative way of gens transfer into chicken egg. Sperm-mediated gene transfer is one of them, and may require a great deal of understanding of mechanisms involved in early fertilization and embryonic development. In other animals where the technique was successful, basic mechanisms have been well studied and established only by painstaking efforts for decades. This paper discusses the accumulated knowledge on early fertilization mechanism in the chicken and how can this information be utilitzed to find the alternative gene transfer in making transgenic chicken.
In tobacco, in vitro pollination has been successfully applied to overcome interspecific incompatibility. The use of this technique will make it possible to introduce DNA into pollen tubes just before fertilization. In this study, we showed improvement of the efficiency of in vitro self-pollination and introduction of foreign genes into pollen tubes by the method of polycation. A plasmid harbouring the GUS gene was introduced into pollen grains and pollen tubes, which had incubated on pollen germination medium(PGM), by polyornithine method. Transient expression of the GUS in pollen grains and pollen tubes that were treated with 0, 2, 5 and $10{\mu}g/m\ell$ DNA was observed. In results, combination of the techniques of polyornithine and in vitro pollination was efficient new technique for genetic transformation through fertilization processes.
To promote the practical use of livestock manure compost (LC) for paddy rice cultivation, the fertilization efficiency of nutrients in LCs was investigated compared to that of chemical fertilizer. This experiment was conducted at rice field in Hwaseong, Korea, with 6 treatments by each of 3 kinds of tested LCs, cattle manure compost (CaC), swine manure compost (SwC) and chicken manure compost (ChC). The treatments consisted of 3 application levels of LCs and 3 chemical fertilizer treatments having the same application levels with LCs. $NH_4$-N content in soil became higher according to the increase in the urea application rate, while it became lower in LC plots than in urea plots, and statistically had no significant difference among LC plots. There was a close relationship between phosphate fertilization rate and the increment of soil available phosphate content after experiment resulting y = 0.1788x - 6.169 ($R^2=0.9425$) when applied fused superphosphate fertilizer, and y = 0.0662x - 2.689 ($R^2=0.9315$) when applied LC at the equivalent rates to phosphate input (x: phosphate application rate, kg $ha^{-1}$, y: increment in soil available phosphate content, mg $kg^{-1}$). And from these two equations, the correlation on the phosphate application rate between fused superphosphate fertilizer and LC could be obtained as y = 2.7056x - 52.492 (x: $P_2O_5$ application rate of fused superphosphate, kg $ha^{-1}$, y: $P_2O_5$ application rate of LC, kg $ha^{-1}$). Plant height, number of tillers, nutrients uptake by rice, and rice yield showed higher levels in N 100% and N 150% application plots of chemical fertilizers, while every LC plots exhibited lower values and no significant difference among them. Relative nitrogen fertilization efficiencies of LCs compared to urea was 12.3% for CaC, 8.8 for SwC and 24.6 for ChC, respectively.
Green house soils have been intensively cultivated with excessive application of compost and chemical fertilizer for vegetable growth. The objective of this study was to establish the reasonable fertilizer application system for rice cultivation in green house soil. Field experiment was carried out with rice cv. Geumo-byeo 1 in Jisan series soil (fine loamy, mixed, mesic family of Fluventic Haplaquepts) that was previously cropped with green pepper (Capsicum annuum L.) for the last 3 years. Treatment consisted of conventional fertilization $(N-P_2O_5-K_2O=11-4.5-5.7kg\;10a^{-1})$, no basal fertilization, 50% reduction of basal fertilization no top dressing, bulk blending fertilizer, and no fertilizer. The value of pH, available phosphate, and exchangeable potassium after experiment was lower than those before experiment while organic matter content was not difference in all treatment. The value of salt elusion was the highest in no basal fertilization plot. The amount of $NH_4-N$ in soil was higher in growth stage of rice as fertilizer amount increased in 1998. The changes of plant height and tiller were higher as fertilizer amount increased. Thousand-grain weight as yield component was higher in no basal fertilization plot all the year because of decreasing panicle. There was no significant difference in rice yield between treatments in 1998. However, conventional fertilization resulted in significantly increased rice yield in 1999. Nitrogen use efficiency was the highest in no basal fertilization plot in 1998 and in conventional fertilization plot in 1998. Our results suggest that no basal fertilization be best to increase salt elusion with slightly increased yield in first year for rice cropping after vegetable harvesting, which method improves fertilization efficiency. However, conventional fertilization was good for second rice cropping after vegetable harvesting in greenhouse.
To establish law-put fertilization technique and increase of fertilization efficiency during cultivation of vinyl mulching for plant, the improvement of soil properties, nutrition efficiency and yields by band spotty fertilization(BSF) using band spotty applicator was carried out at garlic(Alltuiti sativum L.) field in Honam Agricultural Research Institute from 2001 to 2002 for 2 years. The value of pH and the content of total nitrogen, organic matter, exchangeable potassium and calcium of soil after experiment were increased but the content of available phosphate was decreased than soil before experiment. Uptake amounts of nitrogen fertilized by plants were more than in BSF plots($89{\sim}111kg\;ha^{-1}$) compared to in CF(conventional fertilization) Plot ($76kg\;ha^{-1}$) and nitrogen use efficiency were high in BSF plots(42.9~58.2%) compared to in CF plot(34.9%). Also Uptake amounts of potassium fertilized by plants were more than in BSF plots($34{\sim}58kg\;ha^{-1}$) compared to in CF plot($33kg\;ha^{-1}$) and potassium use efficiency were high in BSF plots(21.6~41.2%) compared to in CF plot(19.4%). Residual amount of nitrogen fertilized on soil were more than in BSF plots($38{\sim}54kg\;ha^{-1}$) compared to in CF plot($22kg\;ha^{-1}$) while loss amount of nitrogen fertilized on soil were less than in BSF plots($32{\sim}53kg\;ha^{-1}$) compared to in CF plot($120kg\;ha^{-1}$). Also Residual amount of potassium fertilized on soil were more than in 100% BSF plot($109kg\;ha^{-1}$) compared to in CF plot($72kg\;ha^{-1}$) while loss amount of nitrogen fertilized on soil were less than in BSF plots($14{\sim}38kg\;ha^{-1}$) compared to in CF plot($113kg\;ha^{-1}$). The BSF plots were increased plant height, leaf number, leaf sheath diameter, bulb diameter and height compared to CF plot. The total yields of garlic were more increased 14~19% because of high large bulb rate, commercial yields in 70, 100% BSF plots compared to in CF plot($102.9Mg\;ha^{-1}$). It was found that 70% band spotty fertilization was more effective as fertilization method to reduce both environmental pollution and chemical nitrogen fertilizer in plastic film mulching cultivation.

This study examined the artificial fertilization efficiency of crossbred goats from a farmhouse using frozen semen. Electrostimulation was used to ejaculate and collect semen to assess the artificial fertilization efficiency of crossbred goats. The sperm concentration, vitality, and vitality after melting were investigated. The sperm volume was within 2.5~3 ml, and the concentration was 21~25 × 108/ml for each male crossbred goat. The melted semen had high vitality (≥90%). An IDEXX Rapid Visual Pregnancy Test kit was used for an earlier diagnosis of the pregnancy and to determine the pregnancy rate of fertilization using frozen-thawed semen. The reproductive performance of the artificially fertilized crossbred goats had the highest delivery rate (68%) from Farm C and the lowest delivery rate (45%) from farm A. The delivery rate through artificial fertilization was equal to the fertilization rate according to early pregnancy diagnostic kits. The artificial insemination efficiency was 45~68%. These findings can be used as the basis for improvement and breeding goats in goat farms and livestock research institutes.
To establish N fertilizer recommended scheme for the Chinese cabbage cultivation in green house based on the soil test of nitrate nitrogen, relationship among the content of soil nitrate and fertilizer effects and fertilizer N use efficiency were investigated from nine soils which differed amount of nitrate nitrogen from $14mg\;kg^{-1}$ to$226mg\;kg^{-1}$. The amount of nitrate nitrogen in soil showed a positive correlation with the dry weight of chinese cabbage in the plot of no fertilization. When the fertilizer effects were calculated by difference between the plots of fertilization and no fertilization in the dry weight and the amount of N uptake, a negative correlation was obtained between the amount of nitrate nitrogen in soils and the fertilizer effects. There was also a negative correlation between the amount of nitrate nitrogen in soils and fertilizer use efficiency. Recommendation of application rate of nitrogen fertilizer based on content of $NO_3-N$ in soils was evaluated by the regression equation among the content of soil nitrate, fertilizer effects and fertilizer N use efficiency. Incase the content of $NO_3-N$ nitrogen in soil is more than $200mg\;kg^{-1}$, No N fertilization is recommended; However, The standard N fertilization($320kg\;ha^{-1}$) is recommended for the soils with less than $50mg\;kg^{-1}$. For the soils ranged from $50mg\;kg^{-1}$ to $200mg\;kg^{-1}$ in the amount of nitrate nitrogen, an equation has been developed in order to calculate the recommended amount of fertilizer N.
The adoption of legume cover crops in no-tillage system can contribute to improve soil fertility by providing several benefits, including reduction in soil erosion, suppression of weed growth and N supply to subsequent crops. We conducted a field study to investigate the effect of cover crops and nitrogen fertilization rates on yield and nitrogen use efficiency of waxy corn (Zea mays L.) in no-tillage upland field. Two legume cover crops, hairy vetch (Vicia villosa Roth) and crimson clover (Trifolium incarnuturn L.) were mechanically terminated with roller in early June. For each cover crop treatment, nitrogen (N) fertilizer was applied at three different rates (145, 72.5 and $0kg\;N\;ha^{-1}$). The growth and yield characteristics of corn were significantly affected by the N fertilization rates in crimson clover plots, which suggest N mineralization from the cover crop residue was not sufficient. In contrast, N fertilization rates had no significant effect on growth and yield of corn in hairy vetch plots, indicating that the amount of N released from the cover crop is large enough to meet most of the N requirement of corn. However, the application of N fertilizer in hairy vetch cover plots resulted in slight increase of crop yield, though not statically significant, and high levels of N concentration in corn plant tissue possibly due to luxury consumption of N. Organic residues on the soil surface in hairy vetch cover plots had substantial amounts of N after harvest, ranging from 100 to $116kg\;N\;ha^{-1}$, which is presumably retained during winter season and released by microbial mineralization in subsequent year. The highest nitrogen yield efficiency was achieved in the plot with hairy vetch cover and no N fertilizer application, followed by the plot with hairy vetch cover and $72.5kg\;N\;ha^{-1}$ fertilization rate. In conclusion, hairy vetch showed better performance in corn productivity as compared with crimson clover. In addition, it was concluded that the application of N fertilizer between 0 and $72.5kg\;N\;ha^{-1}$ in combination with hairy vetch cover crop might be most efficient for corn yield under no-tillage system with climatic and soil characteristics similar to those of the experimental site.

This study was wnducte to investigate the effects of treatment level and seasons of sluny hm bovine feces on the productivity of rye, N efficiency and improvement of soil fertility under the Korean climate condition. The results obtained fiom this study summarized as follows ; 1. The highest dry matter yield of rye was obtained in the partial fertilization of sluny in spring or autumn. There is no differences of dry matter yield between spring and autumn application. 2. With increasing the amount of slurry-N, the dry matter yield of rye was signigicantly increased up to 100Kg sluny Nha. As the level of slurry-N rises above about 100Kg N/ha, the maximal yield of dry matter was unchanged or declined. 3. As the level of sluny fertilization rises, the crude protein content of rye increases significantly. However the contents of crude protein was less affected by the application seasons. 4. The amount of nitrogen which produced 6om rye is dependent upon the level of slurry-N. The highest nitrogen yield of rye was obtained by the partial fertilization of sluny-N. 5. The season or amount of slurry treatments did not affect the organic matter content in soil. N-content in soil was the lowest by the partial fertilization of slurry in spring or autumn. However, N-content was increased with the higher level of sluny-N.

This study was carried out to investigate the production efficiency of organic roughage for organic animal husbandry. Field experiment was conducted during 2000-2002 at National Livestock Research Institute in Suwon. Different set of production system tested such as: standard system(applied chemical fertilization), integrated system(50% reduced chemical fertilization and applied 50% organic fertilization) and organic system(applied only organic fertilization). The results were as fellows : o Average dry matter yield of standard production was higher than that of organic production system by 14%. The increment was slight compare to the forage corps. o There was no significant difference in Nutritive value and the percentage of grasses coverage affected by production system.
