발효쪽 추출물의 생리적 기능과 염색특성(제1보) (A Study on the Physiological Effects and Dyeing Properties of the Extract of Fermented (Part I))

  • The purpose of this study was to investigate the antimicrobial activity, antimutagenic and anticancer effects and dyeing properties of the fermented indigo extract. The physiological effects of natural color extracts from colorant plants(gardenia, beet and indigo) were studied. The methanol extract of indigo showed an inhibitory effect on the growth of E. coli and Staph. aureus, and also showed a strong antimicrobial effect on Trich. mentagrophytes compared to others. The methanol extract of indigo showed antimutagenic activities against aflatoxin B1(AFB1) in the Ames test using Salmonella typhimurium TA 100. The proliferation of Clone M-3 mouse melanoma cells and A431 human epidermoid carcinoma cells was inhibited by the methanol extract of indigo. So we decided to use natural indigo for dyeing the fabrics because of those effects. Dried indigo leaves were fermented at variouss temperature and the fermented indigo was reduced by using alkaline(NaOH, Ca(OH)2) and glucose to dye the fabrics. The values of K/S fermented indigo showed the highest value when it was fermented at 3$0^{\circ}C$. The indigo fermented at 3$0^{\circ}C$ had the greatest number of total bacterial counts and we identified one of the main microorganisms as Aspergillus niger. This microorganism was responsible for the indigo fermentation and accelerated indigo fermentation. So it can be supposed to reduce the fermentation period of indigo by inoculating Aspergillus niger into the indigo leaves at 3$0^{\circ}C$.

  • The purpose of this study was to investigate the antimicrobial activity, antimutagenic and anticancer effects and dyeing properties of the fermented indigo extract. The methanol extract of fermented indigo showed a strong inhibition effect on Trich. mentagrophytes and antimutagenic activities against aflatoxin B$_1$(AFB$_1$) in the Ames test using Salmonella typhimurium TA 100. We also found in vitro anticancer effects of the methanol extract of fermented indigo and it was evaluated by using Clone M-3 mouse melanoma cells and A431 human epidermoid carcinoma cells and exerted little cytotoxity against 3T3-L1 embryo fibroblast cell. In the relationship between the K/S values of cotton and silk fabrics dyed with fermented indigo and dyeing repeating times, the K/S values became higher as the repeating times were increased. The K/S values were high when the fabrics were dyed at low temperture. The K/S values of cotton fabrics were higher than those of silk fabrics. Changes of surface color of silk fabrics were higher than those of cotton fabrics after water fastness test, laundering, irradiation and treatment of acidic and alkaline perspiration.

발효쪽 분말염료로 염색한 견직물의 색소 함량이 직물의 색상에 미치는 영향 (Effect of the Amount of Pigment on the Color of Silk Dyed with Fermented Indigo Powder Dye)

  • This research investigated the relationship between the amount of fixed pigment and the color of silk dyed with three types of fermented indigo powder dye under different temperatures and pH, by reduction or nonreduction method. Amount of fixed pigment was analyzed using the Ultimate 3000 HPLC-DAD instrument and the color of dyed silk was measured using the X-rite spectrocolorimeter. All silk samples dyed by reduction method showed PB color. The amount of indigotin fixation was dependent on the dyeing temperature and pH regardless of the indigotin composition in the dye. Indirubin was less dependent upon the dyeing condition in the reduction dyeing and its fixation was minimum level. Dyeing conditions which can maximize the indigotin fixation were $50^{\circ}C/pH$ 11 and $70^{\circ}C/pH$ 7 conditions in reduction dyeing. Color of silk showed more redness ($a^*$) thus higher PB color when the indigotin fixation was low and indirubin fixation was relatively high. Indirubin fixation was very low with slightly better fixation by nonreduction method. More reddish color was obtained by nonreduction dyeing, and by more alkaline dyebath.

  • Indigo is utilized in various industries including textile dyeing, cosmetics, printing and medicinal products and its reduced form, leuco-indigo, is mainly used in these process. Chemical reducing agent (sodium dithionite, sodium sulfide, etc.) is preferred to use for the formation of leucoindigo in industry. In traditional indigo fermentation process, microorganisms can participate in the reduction of indigo and thus it has been known to reduce environmental pollution and noxious byproducts. However, in fermentation method using microorganisms it is difficult to standardize large scale production process due to low yield and reproducibility. In this study, we attempted to develop the indigo reduction process using microbial flora which was isolated from naturally fermented indigo vat or deduced by metagenomic approach. From the results of library analyses of PCR-amplified 16S rRNA genes from the traditional indigo fermentation vat sample (metagenome), it was confirmed that Alkalibacteriums (71%) was distinctly dominant in population. Some strains were identified after confirming that they become pure culture in nutrient media modified slightly. Four strains were separated in this process and each strain showed obvious reducing ability toward indigo in dyeing test. It is expected that the analyzed results will provide important data for standardizing the natural fermentation of indigo and investigating the mechanism of indigo reduction.

  • In Korea, to deal with the housing needs of the elderly, the government operates free nursing homes and residential homes. And to deal with the needs for daily necessities, imported products or products for the disabled have been supplied for the elderly. The government has recognized the importance of silver industry in this rapidly aging society and has established strategies to vitalize industries related to seniors to deal with the seniors' need for food, clothing, and shelter. This study examined the necessity of nursing home gowns, and the development of nursing home gowns that are environmentally friendly, beautiful and functional for the new silver generation. For this study, literature review and Internet search were conducted regarding the status of silver fashion, necessity of nursing home gowns, emergence of new silver generation and change in lifestyle, and value and utilization of natural dye and traditional patterns. To produce nursing home gowns, fabric was dyed using fermented indigo dying, and 3 styles of nursing home gowns for each gender were designed in consideration of seasons. Lastly, these gowns were evaluated by fashion experts, medical personnel, hospital gown and silver wear experts, and natural dye experts. The results showed that the traditional Korean designs developed in this study were excellent and the fermented indigo dye was appropriate for the nursing home gowns. The development of nursing home gowns in this study will be utilized as basic material for the development of silver wear and nursing home gowns to improve the quality of life for the seniors.

아라비아 검을 이용한 천연 쪽 염료의 면직물에 대한 직접 날염 연구 (Study on the Direct Printing of Natural Indigo Dye on Cotton Fabric Using Arabic Gum)

  • This research investigated the screen printing method for natural indigo dye on cotton fabric. We examined four types of thickening agents (arabic gum, guar gum, indalca, and CMC) based on their ability to retard the oxidation of natural indigo print paste while the paste remained on the screen frame. The results indicated that the retardation of arabic gum towards oxidation was the greatest among the four types of thickening agents. The highest K/S value of the printed cotton was observed with a dye concentration of 50g/L fermented indigo powder. The best printing results were obtained when the duration of dye efficiency was tested for the 10 minutes of the dye paste remaining on the screen with a thickening agent concentration of 26.56% that represented 530 cps viscosity. The test of colorfastness to washing and rubbing of the printed cotton resulted in grade 5, and the colorfastness to sunlight resulted in grade 4. Chinese traditional Naminwhapo printing was reproduced on cotton fabric using the natural indigo printing method derived from this study.

천연염료 발효(天然染料 醱酵) 쪽으로 염색(染色)한 모발(毛髮)의 특성 연구(特性 硏究) (A Study on the Property of Hair Dyed with Fermented Indigo)

  • Recently, it has been reported that chemical hair coloring can cause allergic reactions, the toxicity issue of chemical hair color was issued. Therefore, there is an increasing interest in the hair coloration technique using non-toxic and bio-compatible hair color gotten from natural resource. In this study, the possibility using fermented Indigo as natural hair coloring agent was investigated. Bleached hair samples were dyed using Indigo, and the effect of dyeing frequency, the physical change after dyeing, color, surface and cross-section characteristics, and tensile property were also studied. In addition, the protection property of cationic detergent finishing on damaged hair samples were also studied. The following conclusions are made; 1. While bleached hair samples with low value showed the negligible effect of repeated dyeing, bleached hair samples with high value showed the good effect of repeated dyeing. 2. Hair sample dyed with Indigo showed a high light-fastness, however, tensile strength and elongation showed very low values (high damage on hair sample). 3. Cationic detergent finishing did not affect on the colorfastness because of decoloration after dyeing, however, it increased the smoothness of hair sample, and therefore, can use as protectant of damaged hair. 4. Hair scale was damaged after dyeing. It seemed that the alkali, potassium carbonate, increased pH of dyeing bath to 11, as a result, the hair was swollen, weaken, and dissolved on the prolonged dyeing time. Dyed hair sample became stiff and fine. 5. The color difference was 4.62 (a high fastness value) in the test of sunlight exposure, shampoo, cationic detergent finishing, and acid perspiration fastness.

시판 쪽 분말염료의 색소 함량에 따른 면직물의 색상 및 항균성 비교연구 (Analysis of the Pigment Contents of Commercial Indigo Powders and Their Effect on the Color and the Antimicrobial Function of Dyed Cotton Fabrics)

  • Market available fermented indigo powders of Indian origin (FI1, FI2), Chinese origin (FC1, FC2), and raw indigo powders of Indian origin (R1, R2) were examined using TLC and HPLC analyses to investigate their pigment contents. TLC analysis gave $R_f$ values of 0.81 and 0.72 for blue and red pigments, respectively. All the powder products and the synthetic and natural indigo standards eluted at 6.9 min and 8.3 min in the HPLC chromatograms, and the peaks showed the ${\lambda}_{max}$ at 610nm and 542nm, representing indigotin and indirubin, respectively. The pigment content calculated based on the area of indigotin and indirubin peaks in the HPLC chromatograms showed that the indigotin content was higher in FC1 and FC2, while FI1 and FI2 had a higher indirubin content. The relative percentage of indirubin was the highest in R2, but the HPLC peak intensity was quite low. Despite the higher indigotin content in FC1 and FC2, cotton dyed with FI1 and FI2 (versus cotton dyed with FC1 and FC2) showed a higher blue (B) hue, the highest K/S values, and the highest antimicrobial effect.

국립중앙박물관 소장 사리기비단보자기의 복제 (Reproduction of the Silk Wrapper of Sarira Reliquary (Sarigong) in the Collection of National Museum of Korea)

  • 국립중앙박물관 보존과학팀에서는 역사관 한글실의 전시유물 교체에 활용하기 위해 봉인사(奉印寺) 부도탑신(浮屠塔身)에서 발견된 사리공(舍利孔)을 싸고 있었던 보자기인 사리기비단보자기(신수9431)에 대한 복제를 진행하였다. 바탕직물은 유물과 동일한 제직의 명주를 사용하였다. 푸른색을 내기 위해 쪽 앙금을 발효시켜 만든 쪽물, 누렇게 변색된 부분을 위해 오리나무열매로 식물염색하였고 묵서된 한글궁체는 먹으로 모사하였다.
