하수오니에 왕겨 및 톱밥을 혼합한 호기성 퇴비화 (Aerobic Composting of sewage sludge Mixed Rice Hulls and Sawdusts)

  • This study was carried out to investigate the effects of moisture content, temperature, C/N ratio and pH of the sewage sludge mixed with hulle and sawdusts for making compost under aerobic condition and to improve the defect of the structure of experimental equipment heat lose and handling method. and obtained results were as follows 1.The temperature was reached 73$^{\circ}$ C around 50 hours fermentation in the condition of 0.8 L/min. of air and 60.4% of moisture content. and favorable moisture content of initial condition ranged from 50 to 65% 2.The temperature near bottom of the batch composter was decreased due to evaporate water vapor and lose the heat produced during aeration. and it is required to be improved. 3.The temperature in the batch composter from the center to the inside wall surface was gradually decreased. the temperatures of the points located in r=9cm and the wall surface were 4$^{\circ}$ C and 6$^{\circ}$ C respectively. and therefore it is required to be insulated. 4. The maximum C02 production was obtained as 7.3% per volume in the temperature of 63$^{\circ}$C at the moisture content of 60% 5.The temperature range of active microbes growth was found to be as 20$^{\circ}$C to 40$^{\circ}$C in the case of mesophiles and 50$^{\circ}$C to 65$^{\circ}$C in the case of thermophiles due to increase and decrease C02 production. 6.C/N ratio after decomposition was 1.3 to 2.6 smaller than that of initial one due to increase the amount of nitrogen. The more C/N ratio increased. the less the reaction velocity decresed. The optimum of it as found to be 30. 7.pH values after decomposition were slightly increased than that of initial ones. The reaction velocity was decreased at acid and alkall condition. Therefore it is neseseary to neutralize the medium to improve the reaction.

고속 압출 전처리 공정을 이용한 Chlorella sp. 당화 및 바이오에탄올 생산 (Saccharification and Ethanol Production from Chlorella sp. Through High Speed Extrusion Pretreatment)

  • Among various pretreatment processes for bioethanol production, extrusion pretreatment, one of cheap and simple process was investigated to efficiently produce fermentable sugars from micro alga, Chlorella sp. The biomass was pretreated in a single screw extruder at five different barrel temperatures of 45, 50, 55, 60 and $65^{\circ}C$, respectively with five screw rotation speed of 10, 50, 100, 150 and 200 rpm. The pretreated biomass was reacted with two different hydrolyzing enzymes of cellulase and amyloglucosidase since the biomass contained different types of carbohydrates, compared to cellulose of agricultural by-products such wheat and corn stovers, etc. In general, higher glucose conversion yield was obtained as 13.24 (%, w/w) at $55^{\circ}C$ of barrel temperature and 100 rpm of screw speed conditions. In treating 5 FPU/glucan of cellulase and 150 Unit/mL of amyloglucosidase, ca. 64% of cellulose and 40% of polysaccharides in the micro alga were converted into glucose, which was higher yields than those from other reported data without applying an extrusion process. 84% of the fermentable sugars obtained from the hyrolyzing processes were fermented into ethanol in considering 50% of theoretical maximum fermentation yield of the yeast. These results implied that high speed extrusion could be suitable as a pretreatment process for the production of bioethanol from Chlorella sp.

억새 바이오매스 전처리에서 압출 처리가 액상 암모니아 침지 처리에 미치는 영향 (The Effect of Extrusion Treatment on Aqueous Ammonia Soaking Method in Miscanthus Biomass Pretreatment)

  • Pretreatment of cellulosic biomass is necessary before enzymatic saccharification and fermentation. Extrusion is a well established process in food industries and it can be used as a physicochemical treatment method for cellulosic biomass. Aqueous ammonia soaking treatment at mild temperatures ranging from 60 to $80^{\circ}C$ for longer reaction times has been used to preserve most of the cellulose and hemicellulose in the biomass. The objective of this study was to evaluate the effect of extrusion treatment on aqueous ammonia soaking method. Extrusion was performed with miscanthus sample conditioned to 2mm of particle size and 20% of moisture content at $200^{\circ}C$ of barrel temperature and 175rpm of screw speed. And then aqueous ammonia soaking was performed with 15%(w/w) ammonia solution at $60^{\circ}C$ for 1, 2, 4, 8, 12 hours on the extruded and raw miscanthus samples respectively. In the combined extrusion-soaking treatment, most compositions removal occurred within 1~2 hours and on a basis of 1 hour soaking treatment values, cellulose was recovered about 85% and other compositions, including hemicellulose, are removed about 50% from extruded miscanthus sample. The combined extrusion-soaking treated and soaking only treated samples were subjected to enzymatic hydrolysis using cellulase and ${\beta}$-glucosidase. The enzymatic digestibility value of combined extrusion-2 hours soaking treated sample was comparable to 12 hours soaking only treated sample. It means that extrusion treatment can shorten the conventional long reaction time of aqueous ammonia soaking. The findings suggest that the combination of extrusion and soaking is a promising pretreatment method to solve both problems for no lignin removal of extrusion and long reaction time of aqueous ammonia soaking.

분리대두단백질이 첨가된 증편의 저장성에 관한 연구 (The Effects of SPI(Soybean Protein Isolate) on Retrogradation in Jeung-pyun)

  • This study examined Jeung-pyun(JP) Retrogradation in samples containing 3% whole protein, 7S protein, or 11S protein(w/w) that were stored at $4^{\circ}C$ for 6, 12, 24 and 72 hr. Rheometery and differential scanning calorimetry(DSC) were used in the analysis. The pH of the dough decreased during the fermentation process, but it increased after steaming. The JP prepared with soybean protein isolate(SPI) had higher pH than the control group. During storage the textural characteristics of the JP showed effects according to the additions of SPI. After 6 hr of storage, the JP samples containing soybean flour, whole protein, 7S protein, and 11S protein had lower hardness valuse. From 4 hr to 12 hr, higher springiness values were found in the samples containing whole protein, 7S protein and 11S protein. At 0 hr, the control group had the highest cohesiveness value, but after 24 hr it presented the lowest value. For gumminess, after 6 hr of storage, the control group offered the lowest value. Whereas after 12 hr of storage the whole protein group showed the highest value, and at 24 hr, the whole protein, 7S protein, and 11S protein groups had higher values. According to the DSC results, the 11S protein group had lower enthalpy values(${\bigtriangleup}H$) suggesting that adding 11S protein to JP might improve starch retrogradation. After 72 hr of storage, the control group had the highest onset temperature($T_{o}$) and peak temperature($T_{p}$) whereas the 7S and 11S protein JP samples had higher conclusion temperatures($T_{c}$). Therefore, based on the different analysis result between the control and treatment groups, the addition of SPI to Jp had effects on retrogradation.

반응표면분석법에 의한 비동결 파운드 케이크의 최적 배합비 설정 (Use of Response Surface Methods to Optimize the Formula of Non-frozen Pound Cake)

  • 비동결 파운드 케이크의 배합비를 반응표면분석법에 의해 여러 가지 동결방지제의 함량을 조절하여 결정하였다. 수분함량만이 동결방지제 함량과 선형관계를 나타내었으나, 다른 반응변수들은 quadratic모형으로 나타났다. 비동결 파운드 케이크에 제조시 함유되는 당류의 최적 배합비는 7736 : 0.0 : 20.4, 60.0 : 32.8 : 16.4, 70.8 : 11.2 : 19.6와 같이 3가지로 제시되었다. 최적배합비로 제조한 파운드 케이크의 냉동 시간은 기준 배합비로 만든 케이크보다 $45{\sim}27%$가 단축되었으며, 동결점은 $-20^{\circ}C$이하에서 나타났다. 동결방지제의 적절한 배합율을 적용하면 빙점강하에 의해 가공식품 냉동에 필요한 냉동 에너지 감소 효과를 얻으며 기존제품과 유사한 품질 특성을 구할 수 있음을 알았다.

멸치 어간장으로부터 분리한 Bacillus amyloliquefaciens HTP-8 이 생산하는 단백질 분해효소의 특성 (Characterization of Protease Produced by Bacillus amyloliquefaciens HTP-8 Isolated from Korean Fermented Anchovy Sauce.)

  • 저염상태에서 속성 발효를 통한 멸치 어간장의 대량 생산을 위하여 시중 어간장에서 단백질 분해효소 생산이 우수한 균주를 분리 동정하여, B. amyloliquefaciens HTP-8로 명명하였다. 효소생산을 위한 배양 최적온도, 초기 pH, 그리고 배양시간은 각각 $30^{\circ}C$, pH 7.0과 3일이었다. Jar fermenter 배양시 pH를 7.0으로. 조절한 경우가 조절하지 않은 경우에 비해 효소활성이 최대에 이르는 시간이 단축되었다. 효소의 부분정제는 조효소액을 80% ammonium sulfate에 의한 염석과 CM-Sephadex C-50을 이용하여 정제한 결과, 수율이 0.4%, 정제배수가 43.0배였다. 정제효소의 최적 pH와 온도는 pH 10.0과 $50^{\circ}C$였으며, pH 7.0~l2.0의 pH 범위와 $40^{\circ}$ 이하에서 안정하였다. 금속이온 중 $Ag^{+}$ /, $Ba^{2+}$ /에 의하여 효소활성이 저해되었다. 한편, 본 효소는 PMSF에 의하여 선택적으로 활성이 억제됨으로써 활성부위에 serine을 가진 serine protease에 속하는 것으로 판단되었다.

  • Poly-${\beta}$-hydroxybutyrates(PEB)는 토양에서 완전히 이산화탄소와 물로 분해되는 세포 내에 생성되는 고분자 물질로 잘 알려져 있다. Alcaligenes eutrophus의 세포 성장과 PHB의 생산성을 증가시키기 위해서 가압배양법이 사용되었다. 실험 데이타를 다양한 가압과 온도하에서 탄소 공급원과 기체성분의 영향에 관해서 분석한 결과 0.0075vvm의 기체수소를 공급한 배양이 수소를 공급하지 않은 배양에 비해 더 나은 PEB 생산성을 나타내었고, 6atm, $30^{\circ}C$에서 산소의 성분비를 2%에서 8%로 변화시켰을 때 더 높은 수율과 PEB의 생산성을 얻을 수 있었다.

Characterization of Low Temperature-adapted Leuconostoc citreum HJ-P4 and Its Dextransucrase for the Use of Kimchi Starter

  • Leuconostoc citreum HJ-P4 is a strain isolated for kimchi fermentation with its low temperature-adapted growth feature and its high dextransucrase activity. The detailed characteristics of cell growth and dextran sucrase activities were investigated at various environmental conditions such as temperatures, pHs, salts, and raw ingredients. This strain showed almost 2-fold higher maximal cell concentration ($X_{max}$) than that of the type culture Leuconostoc mesenteroides B-512F at $10^{\circ}C$. The $X_{max}$ of the strain was maximum at pH 7 and the cell growth was inhibited by salts in a dose-dependent mode up to 7%. Addition of pepper (<6%), garlic (<10%), and ginger (<2%) in kimchi gave no inhibition effect on the growth of HJ-P4. Dextransucrase synthesized by this strain retained over 80% of its maximum activity at $10^{\circ}C$ showing a comparable cold-adapted feature to its host microbe. This culture can be used as a starter culture in the industrial kimchi production giving desirable functions and predominance at low temperature.

Effect of inoculants and storage temperature on the microbial, chemical and mycotoxin composition of corn silage

  • Objective: To evaluate the effect of lactic acid bacteria and storage temperature on the microbial, chemical and mycotoxin composition of corn silage. Methods: Corn was harvested at 32.8% dry matter, and chopped to 1 to 2 cm. The chopped material was subjected to three treatments: i) control (distilled water); ii) $1{\times}10^6$ colony forming units (cfu)/g of Lactobacillus plantarum; iii) $1{\times}10^6cfu/g$ of Pediococcus pentosaceus. Treatments in triplicate were ensiled for 55 d at $20^{\circ}C$, $28^{\circ}C$, and $37^{\circ}C$ in 1-L polythene jars following packing to a density of approximately $800kg/m^3$ of fresh matter, respectively. At silo opening, microbial populations, fermentation characteristics, nutritive value and mycotoxins of corn silage were determined. Results: L. plantarum significantly increased yeast number, water soluble carbohydrates, nitrate and deoxynivalenol content, and significantly decreased the ammonia N value in corn silage compared with the control (p<0.05). P. pentosaceus significantly increased lactic acid bacteria and yeast number and content of deoxynivalenol, nivalenol, T-2 toxin and zearalenone, while decreasing mold population and content of nitrate and 3-acetyl-deoxynivalneol in corn silage when stored at $20^{\circ}C$ compared to the control (p<0.05). Storage temperature had a significant effect on deoxynivalenol, nivalenol, ochratoxin A, and zearalenone level in corn silage (p<0.05). Conclusion: Lactobacillus plantarum and Pediococcus pentosaceus did not decrease the contents of mycotoxins or nitrate in corn silage stored at three temperatures.

양돈농가에서 퇴비화시설별 수분변화량 분석에 관한 연구 (Investigation on the Amount of Water Evaporation from Composting Facilities Operated in Swine Farms)

  • 양돈농가에서 설치되어 운영되고 있는 퇴비화시설에 대한 수분 증발량에 대한 조사결과를 요약하면 다음과 같다. 1. 돼지사육 두수는 단순 퇴비사 1,433두/호, 기계교반 퇴비화 시설 설치농가는 3,500두/호 이었다. 2. 퇴비화시설의 용적은 단순 퇴비사 $0.33m^3$/두, 기계교반 퇴비화시설 설치농가는 $0.25m^3$/두로 퇴비화시설 용적이 약 24.2% 정도 적은 것으로 조사되었다. 3. 조사농가의 수거돈분의 평균 수분 함량은 단순퇴비사 86.8%, 기계교반 85.7%로 기계교반 퇴비화시설 설치농가의 돈분의 수분 함량이 약 1.3% 정도 낮은 것으로 조사되었다. 4. 1일 투입되는 수분량은 단순 퇴비사 $4.1kg/m^3$/일, 기계교반 퇴비화시설 설치농가는 $6.5kg/m^3$/일로 기계교반 설치농가에서 약 36.9% 정도 단위면적당 수분량이 높게 투입되는 것으로 조사되었다. 5. 퇴비화기간동안의 발효온도는 단순퇴비화 시설에서 최고온도가 $45^{\circ}C$ 정도 상승하였으나, 기계교반 퇴비화시설 설치농가에서의 최고 발효온도는 $70^{\circ}C$까지 상승하는 것으로 조사되었다. 6. 수분 감소량은 퇴비화 시설 $1m^3$당 단순퇴비사 3.7 kg, 기계교반 5.2 kg으로 기계교반 퇴비화 시설이 약 28.8% 높은 것으로 조사되었는데 이는 퇴비더미내 발효온도 차이에 의한 원인인 것으로 판단되어졌다. 7. 퇴비화후 비료성분 함량은 질소 함량은 각각 0.84% 및 0.86%로 비슷한 경향으로 조사되었으며, ${P_2}{O_5}$(%) 성분도 0.78% 및 0.74%로 단순퇴비화시설에서 약간 높은 경향을 보였다. 그러나 OM 및 OM/N은 처리간에 큰 차이가 없는 것으로 조사되었다. 8. 따라서 양돈농가에서의 기계교반 및 단순퇴비화 시설을 이용하여 돈분뇨를 퇴비화하는 경우 돈분의 수분함량과 발효온도 및 수분 증발가능량 등을 고려하여 퇴비화 효율을 향상시키는 노력이 필요할 것으로 생각된다.
