Effects of Sown Season and Maturity Stage on In vitro Fermentation and In sacco Degradation Characteristics of New Variety Maize Stover

  • The effects of seedtime and maturity stage on nutritive value of five maize stover varieties, including conventional maize (Kexiangyu 11, CM), fodder maize (Huqing 1, FM), high oil maize (Gaoyou 115, HOM), sweet maize (Kexiangtianyu 1, SM) and waxy maize (Kexiangluoyu 1, WM), were examined based on chemical composition, in vitro gas production and in situ incubation techniques. Maize stover was sampled at d 17 and d 30 after tasseling, and designated as maturity stage 1 and stage 2, respectively. The average dry matter (DM) organic matter (OM), crude protein (CP) and fiber contents were the greatest for HOM, SM and FM, respectively. CM had the highest in vitro organic matter disappearance (IVOMD) and volatile fatty acid (VFA) concentration. The highest ammonia nitrogen ($NH_3$-N) concentration in the incubation solution, and effective degradability of DM ($ED_{DM}$) and neutral detergent fiber ($ED_{NDF}$) were observed in SM. Advanced maturity stage increased (p<0.05) DM content, $ED_{DM}$ and $ED_{NDF}$, but decreased (p<0.05) OM and CP contents, and decreased (p<0.05) b and a+b values, IVOMD and molar proportion of valerate in the incubation solution for maize stover. Maize sown in summer had greater (p<0.05) OM content, but lower DM, CP, neutral detergent fiber (NDF) and acid detergent fiber (ADF) content compared with maize sown in spring. Maize sown in summer had greater (p<0.001) IVOMD, $NH_3$-N concentration in the incubation solution and $ED_{NDF}$, but lower (p<0.01) ratio of acetate to propionate compared to maize sown in spring. The interaction effect of variety${\times}$seedtime was observed running through almost all chemical composition, in vitro gas production parameters and in situ DM and NDF degradability. The overall results suggested that SM had the highest nutrient quality, and also indicated the possibility of selecting maize variety and seedtime for the utilization of maize stover in ruminants.

The Influence of Feed Energy Density and a Formulated Additive on Rumen and Rectal Temperature in Hanwoo Steers

  • The present study investigated the optimum blending condition of protected fat, choline and yeast culture for lowering of rumen temperature. The Box Benken experimental design, a fractional factorial arrangement, and response surface methodology were employed. The optimum blending condition was determined using the rumen simulated in vitro fermentation. An additive formulated on the optimum condition contained 50% of protected fat, 25% of yeast culture, 5% of choline, 7% of organic zinc, 6.5% of cinnamon, and 6.5% of stevioside. The feed additive was supplemented at a rate of 0.1% of diet (orchard grass:concentrate, 3:7) and compared with a control which had no additive. The treatment resulted in lower volatile fatty acid (VFA) concentration and biogas than the control. To investigate the effect of the optimized additive and feed energy levels on rumen and rectal temperatures, four rumen cannulated Hanwoo (Korean native beef breed) steers were in a $4{\times}4$ Latin square design. Energy levels were varied to low and high by altering the ratio of forage to concentrate in diet: low energy (6:4) and high energy (4:6). The additive was added at a rate of 0.1% of the diet. The following parameters were measured; feed intake, rumen and rectal temperatures, ruminal pH and VFA concentration. This study was conducted in an environmentally controlled house with temperature set at $30^{\circ}C$ and relative humidity levels of 70%. Steers were housed individually in raised crates to facilitate collection of urine and feces. The adaptation period was for 14 days, 2 days for sampling and 7 days for resting the animals. The additive significantly reduced both rumen (p<0.01) and rectal temperatures (p<0.001) without depressed feed intake. There were interactions (p<0.01) between energy level and additive on ruminal temperature. Neither additive nor energy level had an effect on total VFA concentration. The additive however, significantly increased (p<0.01) propionate and subsequently had lower acetate:propionate (A/P) ratios than non-additive supplementation. High concentrate diets had significantly lower pH. Interactions between energy and additive were observed (p<0.01) in ammonia nitrogen production. Supplementation of diets with the additive resulted in lower rumen and rectal temperatures, hence the additive showed promise in alleviating undesirable effects of heat stress in cattle.

동결건조 분말된장의 흡습 거동에 대한 속도론적 연구 (Kinetic Study for Hygroscopic Behavior of Freeze Dried Soy Paste Powder)

  • 서로 다른 외관색택을 갖는 (적갈색 및 담백색) 2종의 된장을 동결건조하고 분말화 하였다. 이들의 흡습거동을 여러 수분찰성도$(0.11{\sim}0.88)$ 및 서로 다른 세온도(25, 40 및 $50^{\circ}C$)에서 조사하였으며, 품질 특성 및 저장안정성의 관점에서 동력학적 및 열역학적 해석을 하였다. 시료 분말된장의 등온수분흡착은 BET분류에 따른 typell에 속하였고, Henderson의 경험식으로 잘 설명할 수 있었다. Caurie식으로부터 계산한 저장 안전수분량은 온도에 의존하였으며 $5.5{\sim}3.98%$(건량기준)범위 이었다. 수분흡습과정은 평형수분함량과 임의의 수분함량과의 차이에 대한 1차 속도식에 따랐으며, 초기에 높은 흡습성을 나타내는 특징을 보였다. 분산성 및 색차의 변화는 저장수분활성도에 민감한 의존성을 보였으며, 최대의 저장안정성은 저장안전 수분함량수준에서 나타났다. 또한, 수분함량에 대한 열역학적 변수의 변화도 저장안전 수분함량수준에서 유의성이 있었으며, 분말된장이 저장안정성과 높은 상관성을 갖는 것으로 나타났다.

The application of Fourier transform near infrared (FT-NIR) spectroscopy in the wine industry of South Africa

  • Fourier transform near infrared (FT-NIR) spectroscopy was used as a rapid method to measure the $^{o}Brix$ content and to discriminate between different must samples in terms of their fee amino nitrogen (FAN) values. FT-NIR spectroscopy was also used as a rapid method to discriminate between Chardonnay wine samples in terms of the status of the male-lactic fermentation (MLF). This was done by monitoring the conversion of malic to lactic acid and thereby determining whether MLF has started, is underway or has been completed followed by classification of the samples. Furthermore, FT-NIR spectroscopy was applied as a rapid method to discriminate between table wine samples in terms of the ethyl carbamate (EC) content. EC in wine can pose a health threat and need to be monitored by determining the EC content in relation to the regulatory limits set by the authorities. For each of the above mentioned parameters, $QUANT+^{TM}$ methods were built and calibrations derived and it was found that a very strong correlation existed in the sample set for the FT-NIR spectroscopic predictions of $^{o}Brix$ (r = 0.99, SECV = 0.306), but the correlations for the FAN (r = 0.61, SECV = 272.1), malic acid (r = 0.58, SECV = 1.06), lactic acid (r = 0.51, SECV = 1.14) and EC predictions (r = 0.47, SECV = 3.67) were not as good. Soft Independent Modeling by Class Analogy (SIMCA) diagnostics and validation was applied as a sophisticated discrimination method. The must samples could be classified in terms of their FAN values when SIMCA was applied, obtaining results with recognition rates exceeding 80%. When SIMCA diagnostics and validation were applied to determine the progress of conversion of malic to lactic acid and the EC content, again results with recognition rates exceeding 80% were obtained. The evaluation of the applicability of FT-NIR spectroscopy measurement of FAN, $^{o}Brix$ values, malic acid, lactic acid and EC content in must and wine shows considerable promise. FT-NIR spectroscopy has the potential to reduce the analytical times considerably in a range of measurements commonly used during the wine making process. Where conventional FT-NIR calibrations are not effective, SIMCA methods can be used as a discriminative method for rapid classification of samples. SIMCA can replace expensive, time-consuming, quantitative analytical methods, if not completely, at least to some extent, because in many processes it is only needed to know whether a specific cut off point has been reach or not or whether a sample belongs to a certain class or not.

Pilot Scale 일체형 2상 혐기성소화에서의 음폐수 처리효율 평가 (Treatment Efficiency Evaluation of Integrated Two-Phase Pilot-Scale Anaerobic Digestion Using Food Waste Leachate)

  • 일체형 2상 혐기성소화 방식은 기존 분리형 2상 혐기성소화의 단점을 보완할 수 있는 기술로 산발효조와 메탄발효조가 병합된 형태의 일체형으로 구성되어 유기물 부하변동 대처 용이, 설치부지면적 감소 등의 이점이 있다. 본 연구는 음폐수를 기질로 일체형 2상 혐기성소화의 유기물 분해효율 및 바이오가스 생산량 등에 대한 실험을 실시하여 기존 분리형 2상 혐기성소화와의 효율 비교를 통해 대규모 플랜트 설치의 타당성 여부를 검토하였다. 5ton/일 규모의 Pilot Plant를 구성하여 약 130일 간 소화조 내 유기물 변화, 바이오가스 생산량 및 메탄함량 등의 실험을 실시하였다. 실험 결과, 평균 음폐수 투입량은 $4.1m^3$/일이었으며, 이때 VS 제거효율은 약 77%로 나타났다. 바이오가스는 평균적으로 투입 음폐수 ton당 약 $63.0m^3$($0.724m^3/kg-VS_{added}$)가 발생되었으며, 메탄함량은 약 61.3%로 분석되었다. 일체형 2상 혐기성소화는 기존 산발효조와 메탄발효조가 분리된 소화방식과 유기물 제거 측면에서 다소 높게 나타났다. 결과적으로 일체형 2상 혐기성소화 방식은 충분히 고농도 유기성 폐수인 음폐수 처리에 있어서 상용화가 가능하다는 결론을 내릴 수 있었다.

유기성 폐기물의 산발효 최적화를 위한 통계학적 분석 방안의 적용 (Application of Statistical Analysis for Optimization of Organic Wastes Acidogenesis)

  • 본 연구는 실험계획법과 통계학적 분석을 이용하여 세가지 유기성 폐기물(축산폐수, 하수슬러지, 음식물쓰레기)의 혼합 산발효를 위한 최적 운전 조건을 구하였다. 먼저, 세가지 기질의 최적 혼합비 도출을 위해 15회의 회분식 실험을 진행하였고, 획득한 실험 값의 통계학적 분석 결과 도출된 최적 혼합비는 chemical oxygen demand(COD) 기준으로 축산폐수, 하수슬러지, 음식물쓰레기 순으로 0.4 : 1.0 : 1.1로 나타났다. 도출된 최적 혼합비율로 준비된 기질을 대상으로 산발효 연속 운전 최적화를 수행하였다. 중요 운전 인자로서 hydraulic retention time(HRT)과 기질 농도를 설정하고, 실험계획법의 일종인 반응표면법(Response surface methodology, RSM)을 적용하였다. 3단계의 실험 계획에 의거한 연속 운전 결과, total volatile fatty acids(TVFA) 생성을 극대화 할 수 있는 최적 운전 조건은 HRT 2일, 기질 농도 29,237 mg COD/L로 밝혀졌고, 제시된 2차 경험적 모델식을 통해 최적 운전 조건에서 73%의 TVFA 증가율을 예상할 수 있었다. 도출된 모델식의 정확도 검증을 위한 실험 결과, 약 4%의 상대오차를 나타내 반응표면법을 비롯한 실험계획법과 통계학적 분석 방안이 성상 변화가 큰 실 폐기물의 최적화에도 효과적으로 적용될 수 있는 것으로 나타났다.

Nutrient Digestibility, Ruminal Fermentation Activities, Serum Parameters and Milk Production and Composition of Lactating Goats Fed Diets Containing Rice Straw Treated with Pleurotus ostreatus

  • The study evaluated replacement of Egyptian berseem clover (BC, Trifolium alexandrinum) with spent rice straw (SRS) of Pleurotus ostreatus basidiomycete in diets of lactating Baladi goats. Nine lactating homo-parity Baladi goats (average BW $23.8{\pm}0.4$ kg) at 7 d postpartum were used in a triplicate $3{\times}3$ Latin square design with 30 d experimental periods. Goats were fed a basal diet containing 0 (Control), 0.25 (SRS25) and 0.45 (SRS45) (w/w, DM basis) of SRS. The Control diet was berseem clover and concentrate mixture (1:1 DM basis). The SRS45 had lowered total feed intake and forages intake compared to Control. The SRS25 and SRS45 rations had the highest digestibilities of DM (p = 0.0241) and hemicellulose (p = 0.0021) compared to Control which had higher (p<0.01) digestibilities of OM (p = 0.0002) and CP (p = 0.0005) than SRS25 and SRS45. Ruminal pH and microbial protein synthesis were higher (p<0.0001) for SRS25 and SRS45 than Control, which also had the highest (p<0.0001) concentration of TVFA, total proteins, non-protein N, and ammonia-N. All values of serum constituents were within normal ranges. The Control ration had higher serum globulin (p = 0.0148), creatinine (p = 0.0150), glucose (p = 0.0002) and cholesterol (p = 0.0016). Both Control and SRS25 groups had the highest (p<0.05) milk (p = 0.0330) and energy corrected milk (p = 0.0290) yields. Fat content was higher (p = 0.0373) with SRS45 and SRS25 groups compared with Control. Replacement of BC with SRS in goat rations increased milk levels of conjugated linoleic acid and unsaturated fatty acids compared with Control. It was concluded that replacing 50% of Egyptian berseem clover with SRS in goat rations improved their productive performance without marked effects on metabolic indicators health.

Performance and Metabolism of Calves Fed Starter Feed Containing Sugarcane Molasses or Glucose Syrup as a Replacement for Corn

  • The aim of this study was to evaluate the effect of replacing corn grain for sugar cane molasses (MO) or glucose syrup (GS) in the starter concentrate on performance and metabolism of dairy calves. Thirty-six individually housed Holstein male calves were blocked according to weight and date of birth and assigned to one of the starter feed treatments, during an 8 week study: i) starter containing 65% corn with no MO or GS (0MO); ii) starter containing 60% corn and 5% MO (5MO); iii) starter containing 55% corn and 10% MO (10MO); and iv) starter containing 60% corn and 5% GS (5GS). Animals received 4 L of milk replacer daily (20 crude protein, 16 ether extract, 12.5% solids), divided in two meals (0700 and 1700 h). Starter and water were provided ad libitum. Starter intake and fecal score were monitored daily until animals were eight weeks old. Body weight and measurements (withers height, hip width and heart girth) were measured weekly before the morning feeding. From the second week of age, blood samples were collected weekly, 2 h after the morning feeding, for glucose, ${\beta}$-hydroxybutyrate and lactate determination. Ruminal fluid was collected at 4, 6, and 8 weeks of age using an oro-ruminal probe and a suction pump for determination of pH and short-chain fatty acids (SCFA). At the end of the eighth week, animals were harvested to evaluate development of the proximal digestive tract. The composition of the starter did not affect (p>0.05) concentrate intake, weight gain, fecal score, blood parameters, and rumen development. However, treatment 5MO showed higher (p<0.05) total concentration of SCFAs, acetate and propionate than 0MO, and these treatments did not differ from 10MO and 5GS (p>0.05). Thus, it can be concluded that the replacement of corn by 5% or 10% sugar cane molasses or 5% GS on starter concentrate did not impact performance, however it has some positive effects on rumen fermentation which may be beneficial for calves with a developing rumen.

Orotic acid 유발 흰쥐 지방간 개선에 미치는 발효당귀의 효과 (The Improvement Effect of Fermented Angelica gigas Nakai Powder Against Orotic Acid-induced Fatty Liver in Rats)

  • Monascus purpureus 균주에 의해 발효된 당귀분말을 Orotic acid-유발 지방간의 개선 효과를 알아보기 위해 식이에 5% 수준으로 첨가하여 10일간 급여한 후 혈중 및 조직 내 산화 스트레스에 미치는 영향에 대하여 검토하였다. 체중 증가량과, 식이 섭취량은 각 OA실험군 모두 감소 하였으나, 음료섭취량은 증가하였다. 간 기능 지표로 활용되는 혈중 AST, ALT, LDH, ALP 및 Cholinesterase 활성 변화 역시 정상 수치와 비슷한 수준으로 감소되었다. 각 조직과 혈청의 과산화지질(TBARS)을 측정한 결과 OA대조군에 비해 각 실험군에서 유의적으로 감소하였으나, 그 중 발효당귀분말을 투여한 FAG군에서 가장 낮은 함량을 보였다. 또한 미네랄 측정 결과 비헴철과 아연을 정상수치로 되돌려주는 기능이 있었으며, Orotic acid 지방간 유발로 인해 저하된 글루타치온 함량을 높여주는 것을 확인할 수 있었다. 이상의 실험결과 발효당귀는 간 조직 내 항산화 물질 증가에 의한 산화스트레스를 경감시킴으로써 향후 간 독성 개선 효능을 가지는 건강식품 개발 가능성이 높은 소재로 판단되어진다.

동결건조 분말고추장의 재수화시 리올로지 특성 (Rheological Properties of Rehydrated Suspensions of Freeze Dried Kochujang Powders)

  • 동결속도를 달리하여 제조한 2종의 동결건조 분말고추장 재수화 현탁액의 리올리지 특성을 원료고추장을 대조구로 온도 $25{\sim}60^{\circ}C$, 고형물 함량 $47{\sim}56^{\circ}C$, 전단속도 $0.1965{\sim}1.9650\;sec^{-1}$의 범위에서 Brookfield 단 원통 회전점도계로 측정하였다. 급속동결 분말고추장 및 완만동결 분말고추장의 재수화 현탁액과 원료고추장은 모두 항복치를 지닌 의가소성을 나타내고. 전단초기 20분까지 Tiu의 모델에 따라 2차 반응속도식으로 붕괴되는 thixotropic거동을 보여주였으며. 분말고추장 현탁액의 구조붕괴 속도가 원료고추장 보다 빨랐다. 평형구조변수는 전단속도에 크게 영향받지 않았으며 그 값은 세가지 시료 모두 비슷하였다. 급속동결분말고추장 및 완만동결 분말고추장의 재수화 현탁액과 원료고추장의 곁보기점도의 온도 의존성은 Arrhenius식과 일치하였으며. 활성화 에너지의 값은 가각 2.21, 2.18, $2.32\;Kcal/g{\cdot}mole $이었다. 세가지 시료의 점조도지수는 온도의 증가에 따라 감소하었으며, 고형물 함량의 증가에 따라 증가하였으나, 유동거동지수는 온두 및 고형물 함량에 거의 영향을 받지 않았다. 급속동결 분말고추장과 완만동결 분말고추장의 러올로지 성질은 큰 차이가 없었다.
