Studies on the Production of Acid Digestive Enzyme -Isolation and Characterization of a Fungal Strain Which Produces Acid Enzymes- (내산성(耐酸性) 소화효소제(消化酵素劑)의 생산(生産)에 관(關)한 연구(硏究) -내산성(耐酸性) 효소생산균(酵素生産菌)의 분리(分離)와 효소(酵素) 생산조건(生産條件)에 관(關)하여-)

  • A fungal strain which produced high levels of acid protease and amylase was isolated from the atmosphere for application to the manufacture of digestive enzme preparation. This study was carried out to elucidate its microbiological characteristics, environmental conditions for production of the enzymes, and relationships between the enzyme activity and acidity. 1. The isolate was identified as a fungal strain which belonged to Aspergillus niger by the manual of Rafer and Fennel, and was found to be a strain producing high levels of acid protease and amylase. 2. The optimal pH of tile enzymes produced by the strain were: protease, 2.0;, ${\alpha}-amylase$, 4 to 5; and glucoamylase, 3 to 5. 3. The optimal culture conditions for production of the enzymes were: protease (at pH 2.5), 2 to 3 days incubation on wheat bran at $30^{\circ}C$; ${\alpha}-amylase$ and glucoamylase(at pH 3.0), 3 days incubation at $30^{\circ}C$. 4. The production of acid protease and glucoamylase was increased approximately by 20 percent when 2 percent of corn starch was added to the wheat bran medium. 5. The addition of 0.3 percent ammonium sulfate to the wheat bran medium resulted in enhancing the enzyme production, especially of acid prctease.

Insecticidal Activity of Essential Oils against Whitegrub (식물정유의 굼벵이에 대한 살충활성)

  • White grub damages various crops, trees and they can also be one of the most serious pests of turf grass in golf courses. This study was conducted to determine the biocontrol of white grubs with 18 different kinds (anise, camphor, castor, cinnamon, clove oil, citronella, fennel, geranium, lavender, lemongrass, linseed, neem, peppermint, pine, rosemary, tea-tree, thyme, and turpentine) of plant essential oils in laboratory, green house and field. Anise oil (88.9%), linseed oil (100%), and tea-tree oil (88.9%) were highly effective among tested essential oils against 3rd instar of Bifurcanmala aulax in cap vial experiment. However, anise and linseed oils showed low mortality against $3^{rd}$ instar of Popillia japonica in pot greenhouse experiment. Efficacy of anise, linseed oil, and tea-tree was also different depending on target white grub in field trials. Correlated mortality showed 32.6% only in tea-tree oil treatment against pupae of Adoretus tenuimaculatus however, correlated mortality of anise, linseed and tea-tree oil were 54.8, 51.6 and 56.5% respectively against $3^{rd}$ instar of Exomala orientalis in the field trial in Adelscott Country Club in Hapcheon, Gyeongnam province.

Herbicidal Activity of Essential Oil from Palmarosa (Cymbopogon martini) (팔마로사 정유의 살초활성)

  • The objective of this study was to find herbicidal compounds in the essential oil of palmarosa (Cymbopogon martini). Of essential oils from basil (Ocimum basilicum), blackpepper (Piper nigrum), clary sage (Salvia sclarea), ginger (Zingiber pfficinale), hyssop (Hyssopus officinalis), nutmag (Myristica fragrance), palmarosa (Cymbopogon martini), fennel (Foeniculum vulgare), sage (Salvia leucantha), and spearmint (Mentha spicta), the herbicidal activity of palmarosa essential oil, which was determined by a seed bioassay using rapeseed (Brassica napus L.), was highest ($GR_{50}$ value, $201{\mu}g\;mL^{-1}$). In palmarosa essential oil, 11 volatile organic chemicals were identified by gas chromatography-mass spectometry with solid-phase micro-extraction apparatus and the major constituents were geraniol (40.23%), geraniol acetate (15.57%), cis-ocimene (10.79%), and beta-caryophyllene (8.72%). The $GR_{50}$ values of geraniol, citral, nerol, and geranyl acetate were 151, 224, 452, and $1,214{\mu}g\;mL^{-1}$, respectively. In greenhouse and field experiments, foliar application of palmarosa essential oil at the level of $80kg\;ha^{-1}$ controlled weeds effectively. Overall results of this study showed that the herbicidal activity of palmarosa essential oil could be due to geraniol and citral which had lower $GR_{50}$ values.

A Comparative Study on the Romanization of Korean and Japanese with English as the Standard of Pronunciation. (한.일 로마자 표기의 비교연구 - 영어발음기준 -)

  • The two existing romanization systems in Korea, namely the recently promulgated Ministry of Education version sponsored by the National Academy of Sciences incorporating for the most part the McCune-Reischauer system and the Hangeul Haghoe or the Korean Language Society's version, must be judged as failures: He fennel for its disregard of Korean orthography and pronunciation, use of Latin-oriented pronunciation despite its assertion that English is the standard used, and the greatest weakness of all, its use of phonetic symbols neither found in regular typewriters, teleprinters, word processors and telex machines nor understood by laymen, sometimes not even by specialists. And the latter suffers from its undue emphasis on Korean orthography, Latin-oriented pronunciation being only partly capable of representing Korean pronunciation, among other shortcomings. Since the two existing romanization systems of Korean in South Korea, and romanization system of Japanese with Latin-oriented pronunciation are woefully inadequate, in today's world where English is used as if it were the international language almost all over the world, the present writer has invented a completely new system of romanizing Korean and Japanese by the so-called pseudo-phonetic method. This method employs the roman letters only and uses thorn in place of phonetic symbols as long as the letters thus applied are believed to have constant sound value. The English pronunciation is the standard used for this system.

Antimicrobial Activity of Exracts from Spices on Lactic Acid Bacteria Related to Kimchi Fermentation (김치발효 관련 젖산균에 대한 향신료의 항균력)

  • Antimicrobial activities of the extracts from spices microorganisms related to kimchi fermentation were investigated. 32 kinds of spices such as allspice, mount, basil, bay leaves, caraway, cardamon, celery, chilli powder, chives, cinnamon, clover, coriander, cumin, dill, fennel, garlic, horseradish, marjorarm, mugwort, mustard, nutmeg, onion, oregano, paprika, parsely, pepper, peppermint, rosemary, sage, tarragon, thyme and tumeric were extracted with water, ethyl ether, ethyl acetate and ethanol. Fractions were tested for antimicrobial activities against Lactobacillus plantarum and Leuconostoc mesenteroides, The extracts of 28 kinds of spices except caraway, cardamon, paprika and tarragon by water extraction, those of 29 kinds except arrowroot, paprika and tarragon by ethyl ether extraction, those of 30 kinds except paprika and tarragon by ethyl acetate extraction, and those of 29 kinds except clover, paprika and tarragon by ethanol extraction showed strong antimicrobial activities against Lactobacillus plantarum. On the other hand, the extracts of 28 kinds of spices except caraway, cardamon, paprika and thyme by water extraction, those of 27 kinds except cardamon, cumin, paprika, tarragon and thyme by ethyl ether extraction, thou of 30 kinds except paprika and thyme by ethyl acetate extraction, and those of 28 kinds except cardamon, cumin, paprika and thyme by ethanol extraction showed strong antimicrobial activities against Leuconostoc mesenteroides.

Characteristics of Morphological and Production from Different Origin of Foeniculum vulgare Mill.

  • This study was carried out to get basic information about the morphological and production characteristics of fennel populations different origin under different ecological conditions in Hungary. The Korean population can be registered as a medium high genotype and showed a more accelerated growth characters than Hungarian taxon and the two populations have a medium homogeneity $(CV\;=\;4{\sim}22%)$. In the second year plant height in Hungarian taxon was significantly higher than that of the Korean population. The shooting dynamic of individual plants might be heterogeneous in both taxa and the Korean population was not only shorter in the height but had also less shoots by 16% than the Hungarian one. The Hungarian taxon exceeded the umbel size and numbers to the Korean one (by 3.62 cm in the average) and the variability in the populations was high $(CV\;=\;29{\sim}49%)$. Seed size was proved to be also a discriminative feature between the examined taxa. With small deviations (CV < 2) the Hungarian population produced seeds longer by 38%. Homogeneity in the populations was dependent more on the vegetation years and on the characteristics measured but less on the origins, respectively.

Identification of the Major Volatile Components from Different Plant Organs of Foeniculum vulgare Mill.

  • The various plant organs of fennel (Foeniculum vulgare Mill.) were investigated to identify their volatile components using Dynamic Headspace (purge & trap). They showed slight differences concerning the volatile components both qualitatively and quantitatively. Results revealed that trans-anethole (12.65%) was the major compound in the leaf. The highest compound was ${\alpha}-pinene$ (28.78%), and trans-anethole (7.90%) was highly detected in the stem. The maximum values were 5.64, 4.59, 1.58, 1.51, and 1.04% for ${\alpha}-pinene,\;{\gamma}-terpinene,\;{\beta}-pinene$, 1,8-cineol and fenchone, respectively in the flower. However, very little trans-anethole was detected (0.27%) in the flower. From these results, it was suggested that the major components were different depending on the plant organs. However it was demonstrated that the related plant organs like flower-fruit and leaf-stem contained the similar components.

  • Algeria is located at the Mediterranean coast of north Africa, 90% of its population is concentrated in the coastal area which is mainly devoted to agriculture. Highland steppe and vast desert climate have determined its food culture. Long arab domination has influenced food of Algeria which has also undergone certain impact of Spanish, Turkish and French occupation. A variety of agricultural products, vegetables, fruits, spices and herbs have determined cooking method and food combination of Algeria. It use neither pork nor alcohol. Its main food consists of bread made from wheat flour and couscous cooked with semoule, Mechuwi, roast lamb and chorba, mixed soup are also typical foods of this region. For climatic reason lamb and chicken are prefered. Energy efficient method is applied to cooking through using oil for saute and water for boiling. Under european influence, Algerian salad used dressing for leaf vegetables, root and other kind vegetables were boiled. Serving with cake and cookies as dessert may possibly be the influence from the French occupation. The cake and cookie are made of wheat flour or other grain flour and take a specific form to be fried sweet with honey. Herbs and spices are widely used in cooking which are easily cultivated in household: mint, basil, rosemary, bayleaf, thyme, sage, fennel, marjoram, coriander, celery. Garlic, onion, piment, red pepper, cinammon are also widely used in an ordinary cooking. Reasonable food combination and economic cooking method could be subject of Algerian food study.

Trans-anethole Suppresses C2C12 Myoblast Differentiation

  • Skeletal muscle, essential for metabolism, thermoregulation, and immunity, undergoes myogenic differentiation that results in myotube formation. Trans-anethole (TA), the major constituent in essential oil produced by anise, star anise, and fennel, whose function in skeletal muscle has not yet been elucidated. Therefore, we investigated whether TA influenced muscle differentiation in mouse C2C12 myoblasts. Cells were induced to differentiate using a differentiation medium with or without TA (50 or 200 mg/mL) daily for 5 days. We measured myotube length and diameter after differentiation days 1, 3, and 5 and analyzed the expression of myogenic markers (myoblast determination protein 1, myogenin, myocyte enhancer factor 2, muscle creatine kinase, and myosin heavy chain) and atrophy-related genes (atrogin-1 and muscle ring finger-1 [MuRF-1]) using quantitative real-time PCR. Additionally, we observed the expression of total protein kinase B (Akt) and phosphorylated Akt (p-Akt) using western blotting. Our data showed that TA significantly induced the formation of smaller and thinner myotubes and reduced the myogenic factor expression. Furthermore, the atrogin-1 and MuRF-1 expression markedly increased by TA. Consistent with these findings, TA significantly decreased the expression of total Akt and p-Akt. Taken together, these results indicate that TA inhibits myogenic differentiation of C2C12 cells via reduction of both total Akt and p-Akt. Our findings may provide valuable insights into the impact of PAA on individuals at risk of muscle atrophy.

Studies on the Effect of Spices and Flavoring on Ascorbic Acid content (조미료(調味料) 및 향신료(香辛料)가 Ascorbic acid에 미치는 조리화학적(調理化學的) 연구(硏究))

  • The L-ascorbic acid, when various kinds of flavoring and spices were added to it's solution individually or in combination, was determined by the 2,4-Dinitrophenylhydrazine method. The remaining rate of it in the solution, to which such spices and flavoring as vinegar, sugar, salt, glutamic acid-Na, welshonion, garlic, sesame, rice jelly, soy, pimento, hotbean paste, pepper, ginger and cinnamon were added individually, was higher than in the solution of itself alone. In the case of pinenuts, sesame oil and, bean paste, however, the remained total ascorbic acid was of little quantity, The rate of it's autooxidation in the solution added with fennel, cinnamon, ginger, pepper, bean paste, pinenuts, curry in individual were found high, but relatively low in garlic, sesame oil, glutamicacid-Na, sugar, honey, salt, pickled shrimps, hot bean paste, and soy. When combined spices and flavoring were added, the content of ascorbic acid showed high values in all cases, indicating that particularly high in soy-bean sprout namul, chinese radish namul, and muchim, seasoned with the most frequently used flavoring, No. 7,8,9,10 while low values in the combination of mustard muchim.
