시설 수박 흰가루병 방제를 위한 살균제 교차방제 체계 (Alternative Fungicide Spraying for the Control of Powdery Mildew Caused by Sphaerotheca fusca on Greenhouse Watermelon (Citrullus lanatus))

  • 우리나라 수박 재배지에서 발생하고 있는 흰가루병 방제를 위한 아인산칼륨-살균제 교차방제체계를 개발하였다. 2010년 시험에서는, 우리나라에서 수박 흰가루병 방제를 위하여 등록되어 합법적으로 사용 가능하며, 농가에서 사용중인 6종류의 살균제를 이용하여 9가지 처리조합을 구성하여 시험하였다. 그 결과 클로로탈로닐(Chl)-마이클로뷰타닐(My)-페나리몰(Fen)-헥사코나졸(Hex)의 처리가 가장 효과가 우수한 것으로 나타났으나, My, Fen, Hex의 작용 기작이 동일하다는 문제점이 있었다. 따라서 2년차 시험에서는 처리조합을 개선하고 농약사용을 줄이기 위하여 이들 살균제 외에 작용기작이 다르면서도 친환경적인 약제인 아인산칼륨을 이용하여 살균제-아인산칼륨 교차 살포 조합을 선발하기 위한 포장시험을 수행하였다. 10간격처리에서, Chl-My-Fen-P는 31.8%의 방제효과를 나타낸 반면, Chl-My-Fen-P는 75.6%의 비교적 높은 방제효과를 나타냈다. 방제효과를 개선하기 위하여 3년차 시험에서는 살균제와 아인산칼륨을 7일 간격으로 살포하였다. Chl-My-Fen-Hex가 가장 높은 89.6%의 방제효과를 나타냈으나, 65.9%의 방제효과를 나타낸Chl-My-Fen-P와 달리 P-My-Fen-Hex 또한 83.5%의 비교적 높은 방제효과를 나타냄으로써, 수박 흰가루병 방제를 위하여 Chl-My-Fen-Hex의 방제체계는 P-My-Fen-Hex로 대체할 수 있음을 시사하고 있다.

Cohesion Establishment Factors Stimulate Endonuclease Activity of hFen1 Independently and Cooperatively

  • Human Fen1 protein (hFen1) plays an important role in Okazaki fragment processing by cleaving the flap structure at the junction between single-stranded (ss) DNA and doublestranded (ds) DNA, an intermediate formed during Okazaki fragment processing, resulting in ligatable nicked dsDNA. It was reported that hChlR1, a member of the cohesion establishment factor family, stimulates hFen1 nuclease activity regardless of its ATPase activity. In this study, we found that cohesion establishment factors cooperatively stimulate endonuclease activity of hFen1 in in vivo mimic condition, including replication protein-A-coated DNA and high salt. Our findings are helpful to explain how a DNA replication machinery larger than the cohesion complex goes through the cohesin ring structure on DNA during S phase in the cell cycle.

Development and Characterization of a New Cell Line from Olive Flounder Paralichthys olivaceus

  • A new embryonic cell line (OFEC-17FEN) derived from olive flounder Paralichthys olivaceus was developed. OFEC-17FEN cells were subcultured for <30 passages over ~200 days. OFEC-17FEN cells had a doubling time of 114.34 h and modal diploid chromosome number was 48. The pluripotency genes POU5f1 and NANOG were expressed in OFEC-17FEN cells. However, the lack of several pluripotency-related genes expression indicates that OFEC-17FEN cells are not stem cells. OFEC-17FEN cells transfected with plasmid pEGFP-c1 exhibited a strong green fluorescent signal at 48 h after transfection. Accordingly, OFEC-17FEN cells may be useful for both basic research and biotechnological application.

Human ChlR1 Stimulates Endonuclease Activity of hFen1 Independently of ATPase Activity

  • Human ChlR1 protein (hChlR1), a member of the cohesion establishment factor family, plays an important role in the segregation of sister chromatids for maintenance of genome integrity. We previously reported that hChlR1 interacts with hFen1 and stimulates its nuclease activity on the flap-structured DNA substrate covered with RPA. To elucidate the relationship between hChlR1 and Okazaki fragment processing, the effect of hChlR1 on in vitro nuclease activities of hFen1 and hDna2 was examined. Independent of ATPase activity, hChlR1 stimulated endonuclease activity of hFen1 but not that of hDna2. Our findings suggest that the acceleration of Okazaki fragment processing near cohesions may aid in reducing the size of the replication machinery, thereby facilitating its entry through the cohesin ring.

In Vitro Antifungal Activity of (1)-N-2-Methoxybenzyl-1,10-phenanthrolinium Bromide against Candida albicans and Its Effects on Membrane Integrity

  • Metal-based drugs, such as 1,10-phenanthroline, have demonstrated anticancer, antifungal and antiplasmodium activities. One of the 1,10-phenanthroline derivatives compounds (1)-N-2-methoxybenzyl-1,10-phenanthrolinium bromide (FEN), which has been demonstrated an inhibitory effect on the growth of Candida spp. This study aimed to explore the in vitro antifungal activity of FEN and its effect on the membrane integrity of Candida albicans. The minimum inhibitory concentration (MIC) and the minimum fungicidal concentration (MFC) of FEN against planktonic C. albicans cells were determined using the broth microdilution method according to the Clinical and Laboratory Standards Institute guidelines. Cell membrane integrity was determined with the propidium iodide assay using a flow cytometer and were visualized using scanning electron microscopy (SEM). Planktonic cells growth of C. albicans were inhibited by FEN, with an MIC of $0.39-1.56{\mu}g/mL$ and a MFC that ranged from 3.125 to $100{\mu}g/mL$. When C. albicans was exposed to FEN, the uptake of propidium iodide was increased, which indicated that membrane disruption is the probable mode of action of this compound. There was cells surface changes of C. albicans when observed under SEM.

제주도 물영아리늪 퇴적물의 화분분석에 의한 식생변천 (Vegetational History of the Mulyeongari Fen by Pollen Analysis in Jeju Island, Korea)

  • For the reconstruction of the past vegetational changes in Jeju Island, Korea, pollen analysis and radiocarbon dating on the sediments obtained from the Mulyeongari fen were carried out. By the results, the vegetational changes around the Mulyeongari fen from ca. 3,300 cal. yr BP to the present can be interpreted and reconstructed. The pollen record from the Mulyeongari fen was divided into two local pollen assemblage zones and three sub-pollen assemblage zones. Zone I (Quercus-Carpinus-Herbs Zone) was characterized by the predominance of Quercus(30~63%), Carpinus(9~35%) and herbs(40~424%). Zone II was characterized by three sub-pollen assemblage zones and the high occurrence ratio of the tree layer in comparison with Zone I. In Zone IIa (Quercus-Carpinus Zone), herbs(3~161%) were drastically decreased in predominance of Quercus(28~56%) and Carpinus(14~31%). In Zone IIb (Carpinus-Quercus Zone), Carpinus(36~48%) was preferentially increased but Quercus(29~39%) was slightly decreased. In Zone IIc (Quercus-Carpinus Zone), Carpinus(26~38%) was decreased inversely but Quercus(36~50%) was increased. In addition, Cyperaceae was also increased to 52%. Consequently, it is suggested that cool temperate southern/sub-montane vegetation composed of Quercus and Carpinus which was physiognomy of deciduous broad leaved forest was distributed around the Mulyeongari fen from ca. 3,300 cal. yr BP. In addition, Cyclobalanopsis(4~23%), Castanopsis(1~12%) and Myrica(under 1%) which are warm-temperate evergreen deciduous forest components were constantly appeared from this period. Accordingly, it can be inferred that the present vegetation type around the Mulyeongari fen was formed from ca. 180 cal. yr BP.

이탄습지 공극수내 용존유기탄소와 페놀계열 물질의 변화도 (Variations of DOC and Phenolics in Pore-water of Peatlands)

  • 습지내의 용존유기탄소의 함량과 구성은 이차생산, 다양한 생지화학적 반응, 그리고 수생태계의 기능에 중요한 영향을 미친다. 본 연구에서는 북구이탄습지 (bog, fen, swamp)의 공극수내의 용존유기탄소와 페놀계열 물질의 농도를 1997년도에 1년에 걸쳐 조사하였다. 일반적인 미생물의 활성 (토양 호흡도)와 페놀산화효소의 활성도 측정하여, 용존유기탄소와 페놀계열 물질의 변화에 대한 기작을 밝히고자 했다 용존유기탄소 농도는 25.5-45.4 (bog), 29.2-71.4 (fen), 13.5-87.6 (swamp) mg/L를 보였고, 페놀계열 물질의 경우에는 13.3-45.4 (bog),7.6-29.5 (fen),4.9-30.8 (swamp) mg/L의 변화정도를 보였다. Swamp에서의 계절적인 변화양상을 살펴보면, 낙엽생산이 용존유기탄소의 변화에 많은 영향을 미침을 알 수 있었다 Bog에서의 미생물활성도와 페놀산화효소의 활성이 가장 낮게 나타났는데 이것이 bog내의 높은 페놀계열물질의 농도를 야기시킨 것으로 사료된다. 본 연구의결과는 습지내 용존유기탄소의 양 뿐만 아니라 그 화학적인 구성이 습지 생지화학에서 중요함을 보여주었다.

북구 이탄습지의 생지화학적 반응에 있어서 체외효소의 중요성-미량기체 발생량 및 용존유기탄소 동태와의 연관성에 대하여 (Importance of Extracellular Enzyme Activities in Northern Peatland Biogeochemistry-Possible Coupling with Trace Gas Emission and DOC Dynamics)

  • 세 종류의 이탄습지 (bog, fen, swamp)에서 탄수유기물 분해에 관여하는 일군의 체외 효소 활성도. 미량 기체 ($CO_2$$N_2O$), DOC 변화 등을 12개월에 걸쳐 측정하였다 ${\beta}-glucosidase$, cellobiohydrolase, ${\beta}-xylosidase$, N-acetylglucosaminidase 등이 각각 $0.008-0.066\;{\mu}mole\;g^{-1}\;min^{-1}$, $0.003-0.021\;{\mu}mole\;g^{-1}\;min^{-1}$, $0.003-0.016\;{\mu}mole\;g^{-1}\;min^{-1}$, $0.004-0.047\;{\mu}mole\;g^{-1}\;min^{-1}$ 범위의 활성도를 보였다. 전반적으로 swamp에서의 활성도가 가장 높았고, fen과 bog의 순서로 나타났다. 세 종류 습지의 자료를 모두 합쳤을 때, 효소의 활성도는 미량 기체의 발생량이나 가용한 탄소의 양과 유의한 양의 상관관계를 보였다. 또한, 4가지 효소 활성도의 평균값은 미량기체와 가용한 탄소량의 변화에 있어서 20-40% 정도의 변이를 설명할 수 있었다. 이 결과로 비추어, 유기탄소 무기화에 관여하는 효소는 북구 이탄습지에서 미량기체 발생과 DOC 동태에 중요한 역할을 하는 것으로 사료된다.

  • A certain subclass $T_{\Omega}(n,\;p,\;\lambda,\;\alpha)$ of starlike functions in the unit disk is introduced. The object of the present paper is to derive several interesting properties of functions belonging to the class $T_{\Omega}(n,\;p,\;\lambda,\;\alpha)$. Coefficient inequalities, distortion theorems and closure theorems of functions belonging to the class $T_{\Omega}(n,\;p,\;\lambda,\;\alpha)$ are determined. Also we obtain radii of convexity for the class $T_{\Omega}(n,\;p,\;\lambda,\;\alpha)$. Furthermore, integral operators and modified Hadamard products of several functions belonging to the class $T_{\Omega}(n,\;p,\;\lambda,\;\alpha)$ are studied here.
