기수역 요각류 위내용물 유전자 분석: 소화기관 내외부 유전자의 선택적 처리방법 (Application of DNA Analysis for Identification of Prey Items on Zooplankton: Selective Treatment Method)

  • 동물플랑크톤이 식물플랑크톤을 선택적으로 섭식하는 특성에 대한 이해는 수생태계 먹이사슬 내의 물질 이동에 중요하다. 하지만 해부를 통한 위내용물 추출 방법은 소형 요각류를 대상으로 적용하기에는 적절하지 않고, 유전자가 유실되거나 위내용물이 아닌 개체 외부의 유전자로 인해 오염될 가능성이 존재한다. 본 연구에서 호 내 식물 플랑크톤 조성 및 기타 환경이 상이한 두 지점을 선정하여 모든 지점에서 지속적으로 출현하는 기수성 요각류인 Sinocalanus tenellus를 대상으로 위내용물의 유전자 분석을 수행하였다. 요각류 개체 외부의 DNA를 제거하는 데 2.5%로 희석한 시판용 표백제(차아염소산나트륨 5.4%)에 2분간 처리하여 증류수로 2회 세척한 뒤 유전자를 추출하였다. 추출된 유전자는 23S rRNA을 증폭하여 서열분석을 실시하였다. Capillary sequencing 분석 결과, 원수와 처리수 및 요각류 위내용물에서 다양한 분류군(규조강, 녹조강, 남조강, 와편모조강, 은편모조강, 황갈조강)에 속하는 식물플랑크톤이 검출되었으며, 새만금호 내 시공간 차이에 따라 상이한 경향을 보였다. 현미경을 이용하여 동정한 식물플랑크톤 군집 조성의 경우 규조강이 우점한 반면, 동일한 원수의 유전자 분석(capillary sequencing) 결과에서는 주로 녹조강, 남조강 및 와편모조강이 우점하여 다소 상반된 경향을 나타냈다. 본 연구에서 적용한 위내용물 분석에 특화된 외부 유전자 제거 전처리 방법은 농도와 처리시간 조절 등의 응용방법에 따라 다양한 동물플랑크톤 분류군에 적용이 가능할 것으로 사료된다.

발포온도와 카본블랙 함량이 천연고무 발포체의 가황거동 및 발포특성에 미치는 영향 (Effects of Foaming Temperature and Carbon black Content on the Cure Behaviors and Foaming Characteristics of the Natural Rubber Foams)

  • 본 연구에서는 발포체의 가황거동과 발포특성에 발포온도 및 카본블랙 함량이 미치는 영향을 조사하기 위해 카본블랙을 변량 배합하고 $145^{\circ}C$에서 $165^{\circ}C$까지 $5^{\circ}C$ 간격으로 발포하여 천연고무 발포체를 제조하였다. 발포온도 및 카본블랙 함량이 증가함에 따라 $t_{s2}$ 그리고 $t_{90}$의 감소 경향이 관찰되었다. 본 연구에서 천연고무의 발포 가황특성의 최적 온도는 $165^{\circ}C$ 였으며 이때 발포된 천연고무 밀도가 가장 낮은 것으로 확인되었다. $145^{\circ}C$에서 발포된 천연고무발포체의 가교속도지수가 가장 작은 값을 보였다. 발포 천연고무의 발포 및 모폴로지 특성으로부터 발포체의 밀도 특성을 확인할 수 있었다. 고무내부에 생성된 각 발포 격벽의 두께는 발포온도가 증가함에 따라 감소하는 반면 카본블랙 함량 증가와 더불어 증가함을 보였다.

Correlation of animal-based parameters with environment-based parameters in an on-farm welfare assessment of growing pigs

  • Nine pig farms were evaluated for the welfare quality in Korea using animal- and environment-based parameters (particularly air quality parameters) during the winter of 2013. The Welfare Quality® (WQ®) protocol consists of 12 criteria within four principles. The WQ® protocol classifies farms into four categories ranging from 'excellent' to 'not classified'. Each of these criteria has specific measures for calculating scores. Calculations for the welfare scores were conducted online using the calculation model in the WQ® protocol. Environment-based parameters like microclimate (i.e., temperature, relative humidity, air speed, and particulate matter), bacteria (total airborne bacteria, airborne total coliform, and airborne total Escherichia coli), concentration of gases (carbon dioxide, ammonia, and hydrogen sulfide) were measured to investigate the relationship between animal- and environment-based parameters. Correlations between the results of animal- and environment-based parameters were estimated using spearman correlation coefficient. The overall assessments found that five out of nine farms were 'acceptable', and four farms were 'enhanced'; no farm was 'not classified'. The average score for the four principles across the nine farms, in decreasing order, were 'good feeding' (63.13 points) > 'good housing' (59.26 points) > 'good health' (33.47 points) > 'appropriate behaviors' (25.48 points). In the result of the environment aspect, the relative humidity of farms 2 (93.4%), 3 (100%), and 9 (98%) was much higher than the recommended maximum relative humidity of 80%, and four out of the nine farms had ammonia concentrations greater than 40 ppm. Ammonia had negative correlations with 'positive social behaviors' and positive emotional states: content, enjoying, sociable, playful, lively, happy and it had positive correlations with negative emotional states: aimless, distressed. The concentration of carbon dioxide had negative correlations with positive emotional states; calm, sociable, playful, happy and it had a positive correlation with negative emotional state; aimless. Our results indicate that the control of the environment for growing pigs can help improve their welfare, particularly via good air quality (carbon dioxide, ammonia, hydrogen sulfide).

Behavioral changes of sows with changes in flattening rate

  • In this study, considering the difficulties for all farms to convert farm styles to animal welfare-based housing, an experiment was performed to observe the changes in the behavior and welfare of sows when the slat floor was changed to a collective breeding ground. Twenty-eight sows used in this study were between the second and fifth parities to minimize the influence of parity. Using a flats floor cover, the flattening rates were treated as 0%, 20%, 30%, 40%, and 50%. Data collection was the behavior of sows visually observed using a camera (e.g., standing, lying, fighting and excessive biting behaviors, and abnormal behaviors) and the animal welfare level measured through field visits. Lying behavior was found to be higher (p < 0.01) as the flattening rate increased, and sows lying on the slatted cover also increased as the flattening rate increased (p < 0.01). Fighting behavior wasincreased when the flattening rate was increased to 20%, and chewing behavior was increased (p < 0.05) as the flattening rate increased. The animal welfare level of sows, 'good feeding', it was found that all treatment groups for body condition score and water were good at 100 (p < 0.05). 'Good housing' was the maximum value (100) in each treatment group. As the percentage of floor increased, the minimum good housing was increased from 78 in 0% flattening rate to 96 in 50% flattening rate. The maximum (100) 'good health' was achieved in the 0% and 20% flattening rates, and it was 98, 98, and 99 in the 30%, 50%, and 40% flattening rate, respectively. 'Appropriate behavior' score was significantly lower than that of other paremeters, but when the flattening ratio was 0% and 20%, the maximum and minimum values were 10. At 40% and 50%, the maximum values were 39 and 49, respectively, and the minimum values were analyzed as 19 for both 40% and 50%. These results will be used as basic data about sow welfare for farmers to successfully transition to group housing and flat floors.

Perinatal and Postnatal Study of LBD-001, a Recombinant Human Interferon $\gamma$, in Rats

  • LBD-001, a recombinant human interferon $\gamma$ produced by genetically engineered yeast as a host system, was intravenously administered to pregnant female rats (Sprague-Dawley) from day 17 of gestation to day 21 of lactation at dose levels.of $0.35 \times 10^6$, $0.69 \times 10^6$, and $1.38 \times 10^6$ I.U./kg/day. In vasopressin-treated group, vasopressin (5 I.U./kg/day) was intravenously injected only for 5 days of perinatal period. (1) No signicant changes by the treatment of LBD-001 were observed in the body weights, food and water consumption, feeding and nurshing behaviors, and the weights of main organs of mother rats. In vasopressin-treated group, no significant changes were observed except the decrease in the food consumption on day 18 of gestation and one case of abnormal offspring with bleeding spots on the skin. (2) No significant changes in the body weights, survival rates, locomotor activity, emotional development. and the motor coordination of offsprings (F1) by the treatment of LBD-001 were observed except the fact that increase of ambulation in the female offsprings of LBD-001 ($0.69 \times 10^6$ or $1.38 \times 10^6$ I.U./kg/day)-treated groups and the increase of rearing in the males of LBD-($1.38 \times 10^6$ I.U./kg/day)-treated group, and the increase of the weight of liver and ovaries in the female offsprings in the LBD-001 ($1.38 \times 10^6$ I.U./kg/day)-treated group were observed. Altogether, the results show that LBD-001 at the dose of $1.38 \times 10^6$ I.U./kg/day or less does not significantly affect the mother rats and their offsprings (F1) except the minor influences when treated during the perinatal and postnatal period.

도시형 자기부상열차 분기기 구간의 제3궤조 전차선 시스템 개발 (The Development of Third-Rail System Applied to Turn-out Section for Urban Maglev)

  • The third-rail system is an important device supplying power directly to the Maglev train through physical contact with the collecting shoe. It is directly related to safety and reliability for the running of Maglev. However, most the third-rail system used in Korea depend on foreign product or technologies, Korea Urban Maglev in the development of appropriate power feeding is urgent. In particular, the turnout section is the weakness point in the system because bending force by turnout section movement and fatigue caused by repetitive motion as well as the expansion by temperature, the forces by Maglev collecting shoe is added th the third-rail. Therefore, this paper proposes the third-rail system appropriate for Korean Urban Maglev of turnout section. To verify the structural stability of POSCO ICT third-rail system, the finite element analysis and physical testing was performed. The third-rail is fixed on each side of the turn-out section steel structure by epoxy insulation supporter and the integral behaviors are occurred. Therefore, the maximum horizontal displacements of each support are investigated and then, it is applied to finite element model of the third-rail to investigate the moments and stress. Also, the bending test about one million times and Expansion Joint for the third-rail was performed. The third-rail system safety and reliability was identified by test line on Korea Institute of Machinery & Materials in Deajeon for under the actual usage environment such as the Maglev and turn-out operation.

Effects of Furnished Cage Type on Behavior and Welfare of Laying Hens

  • This study was conducted to compare the effects of layout of furniture (a perch, nest, and sandbox) in cages on behavior and welfare of hens. Two hundred and sixteen Hyline Brown laying hens were divided into five groups (treatments) with four replicates per group: small furnished cages (SFC), medium furnished cages type I (MFC-I), medium furnished cages type II (MFC-II), and medium furnished cages type III (MFC-III) and conventional cages (CC). The experiment started at 18 week of age and finished at 52 week of age. Hens' behaviors were filmed during the following periods: 8:00 to 10:00; 13:00 to 14:00; 16:00 to 17:00 on three separate days and two hens from each cage were measured for welfare parameters at 50 wk of age. The results showed that feeding and laying of all hens showed no effect by cage type (p>0.05), and the hens in the furnished cages had significantly lower standing and higher walking than CC hens (p<0.05). The birds in MFC-III had significant higher preening, scratching and feather-pecking behavior than in the other cages (p<0.05). No difference in nesting behavior was found in the hens between the furnished cages (p>0.05). The hens in MFC-I, -II, and -III showed a significant higher socializing behavior than SFC and CC (p<0.05). The lowest perching was for the hens in SFC and the highest perching found for the hens in MFC-III. Overall, the hens in CC showed poorer welfare conditions than the furnished cages, in which the feather condition score, gait score and tonic immobility duration of the hens in CC was significantly higher than SFC, MFC-I, MFC-II, and MFC-III (p<0.05). In conclusion, the furnished cage design affected both behavior and welfare states of hens. Overall, MFC-III cage design was better than SFC, MFC-I, and MFC-II cage designs.

Comparison of Growth Performance of Antibiotic-free Yorkshire Crossbreds Sired by Berkshire, Large Black, and Tamworth Breeds Raised in Hoop Structures

  • The objective of this study was to compare body weight, ADG, and feed:gain ratio of antibiotic-free pigs from Yorkshire dams and sired by Yorkshire (YY), Berkshire (BY), Large Black (LBY) or Tamworth (TY) boars. All the crossbred pigs in each of three trials were raised as one group from weaning to finishing in the same deep-bedded hoop, providing a comfortable environment for the animals which allowed rooting and other natural behaviors. Birth, weaning and litter weights were measured and recorded. From approximately 50 kg to market weight (125 kg), feed intake and body weights were recorded manually (body weight) or using a FIRE (Feed Intake Recording Equipment, Osborne Industries Inc. Osborne, Kansas) system with eight individual feeding stations. Feed intake data for 106 finishing pigs between 140 and 210 d of age and the resulting weights and feed conversion ratios were analyzed by breed type. Least square means for body weights (birth, weaning and to 240 d) were estimated with Proc Mixed in SAS 9.2 for fixed effects such as crossbreed and days of age within the sire breed. The differences within fixed effects were compared using least significant differences with DIFF option. Individual birth weights and weaning weights were influenced by sire breed (p<0.05). For birth weight, BY pigs were the lightest, TY and YY pigs were the heaviest but similar to each other and LBY pigs were intermediate. For weaning weights, BY and LBY pigs were heavier than TY and YY pigs. However, litter birth and weaning weights were not influenced by sire breed, and average daily gain was also not significantly different among breed types. Tamworth sired pigs had lower overall body weight gain, and feed conversion was lower in TY and YY groups than BY and LBY groups (p<0.05), however, number of observations was somewhat limited for feed conversion and for Tamworth pigs. Overall, no convincing differences among breed types were noted for this study, but growth performance in the outdoor environment was satisfactory.

Affordable method of video recording for ecologists and citizen-science participants

  • Observations and video documentation of interactions between animals living in dens, cavities, and other enclosed spaces are difficult, but they play an important role in field biology, ecology, and conservation. For example, bird parents visiting nests and feeding their nestlings may provide crucial information for testing of ecological hypotheses and may easily attract attention of participants of citizen-science ecological and conservation projects. Because of the nest concealment of cavity-nesting birds, their behaviors in the nest can only be studied by using video surveillance. Professional wildlife surveillance systems are extremely expensive. Here, we describe an inexpensive video setup that can be constructed with relatively little effort and is more affordable than any previously described system. We anticipate that the relatively low cost of about 250 USD for a battery-operated system is an important feature for citizen-science type of projects and for applications in heavily populated areas where the potential for theft and vandalism may be high. Based on our experiences, we provide methodological advice on practical aspects of using this system in the field for ecological research on birds. We highlight the low cost, easiness of construction, and potential availability to a large number of observers taking part in wildlife monitoring projects, and we offer technical help to participants of such research projects.

공압구동용 솔레노이드밸브의 동특성 해석 (Analysis of Dynamic Characteristics of Pneumatic Driving Solenoid Valve)

  • 우주발사체 추진기관 공급계에서 공압구동용 솔레노이드밸브는 제어시시템의 명령이 주어지면 구동가스 배관의 통로를 개폐해서 공압제어장치를 작동시킨다. 공압구동용 솔레노이드밸브의 제작에 앞서 설계검증 및 기본적인 작동특성을 분석하기 위해 AMESim 상용코드를 이용하여 해석모델을 수립하였다. 입구압력에 따른 작동시간을 시험결과와 비교하여 모델을 검증하였고 내부유동 해석결과 (FLUENT)를 이용하여 3차원 형상을 고려하여 모델의 정확도를 높였다. 밸브모델을 이용하여 다양한 설계변수에 따른 밸브의 개폐압력, 작동시간을 계산하여 설계인자 검증 및 작동성능을 분석하였다. 설계변수인 컨트롤밸브의 시트 형상, 주 밸브와 배출밸브의 시트 형상, 실링 직경비, 컨트롤 캐비티부피에 대해 밸브의 동특성 해석을 수행하였다. 해석을 통해 밸브 개폐작동시간, 작동성능, 개방압력을 예상하였다. 본 연구 결과는 한국형발사체 공급계 공압구동용 솔레노이드밸브의 설계/해석능력을 확보하고 밸브의 개발과정에서 효율성을 높일 수 있으며 파생형 밸브의 설계 및 선행연구에 적용할 수 있을 것으로 판단된다.
